경골어류 등목어과 Three spot gourami의 난자형성과정 (The Oogenesis of Three Spot Gourami, Belontiidae, Teleostei)

  • 경골어류 등목어과(Belontiidae)에 속하는 three spot gourami (Trichogaster trichopterus Pallas, 1770)의 난자형성과정을 광학 현미경으로 관찰한 결과는 다음과 같다. 난소는 한 쌍으로 흐린 살색의 아가미 쪽은 뾰족하고 생식공 쪽은 둥근 장타원형(장축 2 cm, 단축 1 cm)이었으며 부레와 창자 사이에 위치하고 있었다. 난원세포의 세포질은 호염기성이었고 핵 내에 다수의 인들이 분포하고 있었다. 난원세포의 세포질은 hematoxylin으로 매우 강하게 파란색으로 염색되었고 핵막을 따라서 여러 개의 인들이 분포하고 있었다. 발달함에 따라서 난원 세포의 세포질은 핵 쪽에서 세포막 쪽부터 hematoxylin의 염색 성이 낮아졌고, 제1난모세포는 난세포 세포질의 가장자리인 세포막 쪽에서 국한적으로 유구들이 형성되기 시작하였다. 제1난 모세포가 발달함에 따라서 유구들이 세포의 바깥 가장자리에서 증식되어 증가하기 시작하였고 작은 유구들이 핵막 근처에 집중 되어 형성되기 시작하였다. 제2난모세포는 발생이 진행됨에 따 라서 난막은 두꺼워져 뚜렷하게 관찰되기 시작하였고, 특히 유구 가 세포질 전체로 퍼져나갔다. 세포막 쪽의 유구 사이에 난황포 가 형성되기 시작했으며 난황포는 호산성으로 에오신에 의해서 적색으로 염색되어 관찰되었다. 성숙한 난자는 난자의 크기가 상당히 증가하였으며 난막이 뚜렷하고 핵은 염기성을 띠었다. 또한 난황포는 더욱 발달하여 적색으로 염색된 구형의 난황괴를 이루 고 있었다. 수정란을 광학현미경으로 관찰한 결과 중앙하단에 한 개의 큰 유구가 발견되었다. 이상과 같이 three spot gourami의 난자형성과정은 난세포의 크기 증가, 유구와 난황의 축적, 염기성 물질의 감소 및 큰 유구의 형성으로 요약될 수 있다.

Attribution of Cortical Granules to Formation of Fertilization Envelopes and Polyspermy Block in Urechis unicinctus

  • Cortical reaction and polyspermy block are well defined in most marine invertebrates. In Urechis species, the function of cortical granules (CGs) is not yet known, and there is controversy on whether the cortical reaction occurs, or the fertilization envelope (FE) is attributed to CG releases or functions to prevent polyspermy. This study was carried out to determine the cortical reactions and functions of the FE in Urechis unicinctus. Artificial insemination of the eggs revealed that CG release occurred to give rise to perivitelline space (PS) into the final FE. Both PS and final FE effectively blocked polyspermy. The final FE was accomplished within 10 min after sperm-egg initial binding. No massive release of CGs occurred within the early phase of 5 min after the initial binding, initially and the PS seemed to playa role to prevent polyspermy. The CG massively released its content into the PS in late phase of FE formation, and differentiated PS into five intermediate layers. The layers opened into each other by anastomosis, so that the final FE consisted of two layers, the inner layer ($15{\mu}m$ in thickness) and the outer layer ($1{\mu}m$ in thickness). The outer layer derived from vitelline layer and the inner layer consisted of PS and CG secretions. Immunofluorescence and confocal laser microscopy revealed that the spermatozoon took up residence in the egg cortex during FE formation and successive meioses of the fertilized egg. These results suggest that both PS and final FE of U. unicinctus were equivalent to the early and late block, respectively, of other marine animals.

토고숲 모기 (Aedes togoi)의 난황립 및 난황막 형성에 따른 미세구조적 변화 (Ultrastructural Changes in the Formation of Yolk Granules and Vitelline Envelope in Aedes togoi)

  • Drastic changes were observed in egg chamber during oogenesis of Aedes togoi immediately after blood meal. The egg chamber, in which the oocyte remains at previtellogenic stage before blood meal, shows very little change in size even for 3 days after emergence but increased 25 folds in volume within 60 hours after blood meal, presumably due to rapid yolk formation. Upto 6 hours after blood meal structures considered to be the precursor of the yolk granules were not observed in the space between follicle cells and oocyte. Vitelline bodies, which are similar to dense bodies secreted from follicle cells, appeared in the space at 10 hours after blood meal. Although vitelline bodies were fused to form vitelline layer, these bodies seem to contribute to the formation of yolk granules. Nurse cells are connected to oocytes by cytoplasmic bridge before blood meal, but the cells are absorbed into oocyte at 6 hours after blood meal.
