Ultrastructure of the fertilized egg envelopes in Ancistrus cirrhosus, Loricariidae, Teleostei

  • We examined the morphology of fertilized egg and ultrastructures of fertilized egg envelopes of Ancistrus cirrhosus belong to Loricariidae using light and electron microscopes. The fertilized eggs formed a mass on the spawning place and were yellowish, spherical, non-transparent, demersal, adhesive, and a narrow perivitelline space. But, the adhesiveness of fertilized eggs was disappeared after spawning excluding contact parts. The micropyle with funnel shape was surrounded by 15-19 furrow lines of egg envelope in a spoke-like pattern. The outer surface of egg envelope has smooth side and inner surface of egg envelope was rough with grooves. Also, the total thickness of the fertilized egg envelope was about 32.58 ± 0.85 ㎛ (n = 20), and the fertilized egg envelope consisted of three layers, an outer adhesive electron-dense layer, a middle layer with low electron density and an inner electron-dense layer with grooves in counter structure from other most teleost. Collectively, these morphological characteristics and adhesive property of fertilized egg, and ultrastructures of micropyle, outer surface, and section of fertilized egg envelope are showed species specificity.

High Density Lipoprotein from Egg Yolk (EYHDL) Improves Dyslipidemia by Mediating Fatty Acids Metabolism in High Fat Diet-induced Obese Mice

  • We investigated the effect of high density lipoprotein from egg yolk (EYHDL) on serum, hepatic and fecal lipid and fatty acids (FAs) levels and on gene expression involved in FAs metabolism. Male KM mice were fed either normal diet (ND; n=20), high fat diet (HFD; n=20), or high fat diet containing EYHDL (EYHDL; 0.6 mg/g, every day by oral gavage, n=20) for 100 days. At the end of the experiment, the effects of treatments on biochemical parameters, FAs profiles and involved gene expression were analyzed. Our results revealed that EYHDL markedly suppressed the body weight gain, accumulation of abdominal fat tissues, serum concentrations of LDL-cholesterol (LDL-C) and triglycerides, hepatic triglycerides and cholesterol accumulation, while increased serum concentration of HDL-cholesterol (HDL-C). EYHDL intake also increased total cholesterol (TC) excretions compared with HFD group. Moreover, it alleviated the severity of fatty liver and improved glucose and insulin tolerance compared with HFD. More importantly, EYHDL partially normalized FAs profiles in serum, liver and fecaces and neutralized the HFD-induced upregulation of SREBP-1c, Acaca, Fasn, GPAT and Scd1. In conclusion, our findings indicate that EYHDL may have the potential to improve metabolic disturbances that occur in HFD mice and can be considered as an appropriate dietary recommendation for the treatment of metabolic syndrome (MetS).

Direct Purification of Lysozyme from Hen Egg White Using High Density Mixed Mode Adsorbent

  • The high density mixed mode adsorbent known by the trade name of Mimo-AD was used to purify lysozyme directly from the hen egg white (HEW). The homogenized hen egg white was treated with the adsorbent in a stirred vessel for lysozyme adsorption, and then the adsorbent, easily separated from the HEW by sedimentation, was packed into a column. The remaining HEW and contaminant proteins were removed by washing with pH 11 distilled water in an expanded-bed state, and subsequently the elution was performed with pH 12 distilled water in a packed-bed state. By this simple and rapid adsorption, washing, and elution procedure, lysozyme was purified to>95% with an overall recovery yield of 66%. This process offers a great potential for industrial application by allowing the extraction of lysozyme while retaining the commercial value of HEW.

개량조개, Mactra chinensis의 난발생에 미치는 수온, 염분 및 수용밀도의 영향 (Effect of Temperature, Salinity and Density on the Egg Development of the Sunray Surf Clam, Mactra chinensis)

  • 개량조개의 종묘생산을 위한 생물학적 기초자료를 얻고자 난발생에 영향을 미치는 수온, 염분 및 수용밀도에 관하여 실험한 결과, 수온에 따른 난발생의 각 단계에 이르기까지의 수온 (TW, $^{\circ}C$)에 따른 발생속도 (h, 시간)는 $18^{\circ}C$, $23^{\circ}C$, $28^{\circ}C$$33^{\circ}C$에서 D형 유생까지 각각 33시간 30분, 20시간 40분, 18시간 15분 및 15시간이 소요되어 수온이 높을수록 빨랐으며, 그 관계식은 다음과 같다. 2세포기 : 1/h = 0.1051WT - 1.4782 ($r^2=0.9926$) 8세포기 : 1/h = 0.037WT - 0.3686 ($r^2=0.9648$) 낭배기 : 1/h = 0.008WT - 0.0521 ($r^2=0.9134$) 담륜자 : 1/h = 0.0041WT - 0.0235 ($r^2=0.9072$) D형유생 : 1/h = 0.0024WT - 0.0102 ($r^2=0.9611$) 개량조개의 수온과 난발생 속도와의 관계에서 추정된 난발생의 생물학적 영도는 평균 $8.0^{\circ}C$였으며, 개량조개 수정란은 D형 유생까지의 발생률을 고려할 때 발생 가능한 수온은 $18-28^{\circ}C$, 적정 수온은 $23^{\circ}C$였다. 개량조개의 수정란이 D형 유생으로 발생 가능한 염분은 20-35 psu로 나타났고, 염분에 따른 수정란의 D형 유생까지 발생률을 고려할 때 최소 25 psu 이상이며 적정 염분은 30-35 psu였다. 수정란으로부터 D형 유생까지의 발생률을 높일 수 있는 수정란의 수용밀도는 사육수 1 ml 당 40개 이하로 나타났다.

Oceanographic indicators for the occurrence of anchovy eggs inferred from generalized additive models

  • Three generalized additive models were applied to the distribution of anchovy eggs and oceanographic factors to determine the occurrence of anchovy spawning grounds in Korean waters and to identify the indicators of their occurrence using survey data from the spring and summer of 1985, 1995, and 2002. Binomial and Gaussian types of generalized additive models (GAM) and quantile generalized additive models (QGAM) revealed that egg density was influenced mostly by ocean temperature and salinity in spring, and the vertical structure of temperature, salinity, dissolved oxygen, and zooplankton biomass during summer in the upper quantiles of egg density. The GAM and QGAM model deviance explained 18.5-63.2% of the egg distribution in summer in the East and West Sea. For the principle component analysis-based GAMs, the variance explained by the final regression model was 27.3-67.0%, higher than the regular models and QGAMs for egg density in the East and West Sea. By analyzing the distribution of anchovy eggs off the Korean coast, our results revealed the optimal temperature and salinity conditions, in addition to high production and high vertical mixing, as the key indicators of the major spawning grounds of anchovies.

  • The egg's interior quality is one of the most important criteria for commercial producers and consumers. Internal quality is complex, including aesthetic factors such as taste, freshness, nutritional and processing values, and the genetic influences upon these upon these factors ranges from none to considerable. The rate of cholesterol synthesis in the hen is very high compared to other animals and humans. Genetic selection, diet drugs and other chemicals can alter cholesterol concentration in the plasma of laying hen, but attempts to manipulate the cholesterol concentration in the egg yolk are generally unsuccessful since the cholesterol can only be changed to a small extent. Factors which may affect the degree of pigmentation of the yolk include the type of xanthophyll and its concentration in the feed, the feed composition, and the health of the hen. Several feed ingredients interact with carotenoid pigment to improve or reduce their deposition rates in yolks. Egg weight is determined by genetics, body size prior to first egg housing density, environmental temperature, lighting program, total feed consumption, calcium, phosphorus, niacin, water, methionine, total sulfur amino acids, energy, linoleic acid, fat and protein levels. Eggs need to be promote levels. Eggs need to be promoted a versatile commodity and new processed egg items need to be developed. Organic eggs are laid by hens which were raised in chemical and drug frdd environments. There are still difficulties in producing these eggs due to the availability of organic poultry feeds and cost of organic grains.

Ultrastructure of the fertilized egg envelope from Melanotaenia praecox, Melanotaeniidae, Teleostei

  • We examined the morphology of fertilized egg and ultrastructures of fertilized egg envelopes of dwarf rainbowfish (Melanotaenia praecox) belong to Melanotaeniidae using light and electron microscopes. The fertilized eggs were spherical with adhesive filament, transparent, demersal, and had a narrow perivitelline space and small oil droplets. The size of fertilized egg was 1.02 ± 0.18 mm (n = 30), and there were two kinds of adhesive filament on the fertilized eggs. The long and thick (diameter 12.22 ± 0.52 ㎛, n = 20) adhesive filaments were only at the area of animal pole, and short and thin (diameter 1.99 ± 0.23 ㎛, n = 20) adhesive filaments were around the long filaments. A micropyle was conical shaped with adhesive filament and located near the animal pole of egg. The outer surface of fertilized egg was rough side. Also, the total thickness of the fertilized egg envelope was about 7.46 ± 0.41 ㎛ (n = 20), the fertilized egg envelope consisted of two layers, an inner lamellae layer and an outer layer with high electron-density. And the inner layer was 8 layers. Collectively, these morphological characteristics and adhesive property of fertilized egg with adhesive filaments, and ultrastructures of micropyle, outer surface, and section of fertilized egg envelope are showed species specificity.

초임계 이산화탄소에 의한 난황분의 추출 (Supercritical Carbon Dioxide Extraction of Dried Egg Yolk)

  • Investigations were performed on the effects of extraction temperature, pressure, time on solubility and selectivity of egg yolk lipid and cholesterol, and color and fatty acid composition of the residue in supercritical carbon dioxide(SC-Co$_2$) extraction. Lipid and cholesterol solubility increased as the increase of Co$_2$ density and was found to strongly depend on the extraction pressure rather than the extraction temperature. The relative concentration of cholesterol in the extract increased with an increase in temperature and decreased with an increase in pressure and extraction time. extraction of dried eg yolk for 3hr at 4$0^{\circ}C$/276 bar removed 46.1% of cholesterol from the residual egg yolk with a yield of 63.2%. SC-Co$_2$ extraction produced a lighter color egg yolk with less redness and yellowness. As the extraction time increased, the resultant residual egg yolk became more saturated with fatty acids. SC-$CO_2$ extraction offers a safe, natural method for removing cholesterol from dried egg yolk.

난각의 음향반응에 영향을 주는 인자 (Factors Affecting Acoustic Responses of Egg Shell)

  • A nondestructive quality inspection technique using acoustic impulse response method was studied to investigate the feasibility of egg shell inspection. An experimental system was built to generate impact force, to measure the response signal and to analyze the frequency spectrum. This system includes an impulse generating unit, an egg holding seat, a microphone with preamplifier, and a digital oscilloscope connected to Personal Computer by RS-232C interface. The factors such as impulse generating method, egg holding method, and sensor location were evaluated by analyzing the power spectrum density of the measured signal. The results obtained are summarized as follows : 1. From the sampled eggs, the proper conditions for detecting damaged eggs were found as followings; ceramic for the impact ball material, rubber for egg seat material, 20 degrees for an impact angle of pendulum, 10mm for the distance between egg and sensor, the sharp side for impacting part, and 180 degrees for the location of sensor. 2. Examination of the Fourier transformed analysis in beth normal and damaged eggs revealed that those factors such as the resonant frequency, a number of peak frequencies and the magnitude of power spectrum were important to detect damaged eggs.

Effect of dietary sesame (Sesame indicum L) seed meal level supplemented with lysine and phytase on performance traits and antioxidant status of late-phase laying hens

  • Objective: This study was performed to investigate the effects of supplementing sesame seed meal (SSM) with phytase and lysine on performance, egg quality, blood biochemical and antioxidant status of laying hens. Methods: A total of 960, 56-wk-old laying hens were divided into 12 dietary groups with eight replicates per group (10 birds per replicate). A completely randomized design with factorial arrangement 2×3×2 consisted of two levels of lysine supplement (0% and 10% over requirement), three SSM levels (0%, 10%, and 20%) with or without phytase (0 and 300 g/ton). The feeding trial lasted 10 weeks. Results: Birds fed diets with 10% SSM had higher feed intake than groups fed 0% and 20% SSM. The addition of phytase to experimental feeds, improved feed conversion ratio, increased egg weight and mass (p<0.01). Egg quality criteria was not affected by supplementing phytase; however, supplementing 300 g/ton phytase to hens diet, led to a significant (p<0.05) increase in egg shell strength. Egg yolk cholesterol and serum low-density lipoprotein cholesterol, atherogenic index and total cholesterol were decreased (p<0.01) by diet containing 20% SSM. The high-density lipoprotein cholesterol was increased (p<0.05) in serum of hens fed 20% SSM than the other groups. It was also observed that total antioxidant capacity and total superoxide dismutase content of hens fed 20% SSM was significantly higher than control group (p<0.05). Conclusion: As from results, dietary supplementation of SSM and phytase had no negative effects on laying hens performance or egg quality while improving the egg oxidative stability.
