A Simple and Efficient Subtractive Cloning Method

  • In subtractive hybridization, target sequences in the tester are enriched by hybridizing with an excess amount of driver, followed by removing the tester hybridized with the driver. All of existing subtractive cloning methods are designed to remove the tester/driver hybrid. The removal of hybrid, however, is often unsatisfactory For various reasons. In this study we developed a subtractive enrichment protocol in which the tester/driver can be completely removed by selecting only the tester/tester after hybridization. In this protocol both the tester and driver DNAs are ligated with same linker DNAs and amplified by polymerase chain reaction (PCR). The tester DNA is then digested with two different enzymes and used in subsequent hybridization with an excess driver. After hybridization, the DNA is ligated with the adaptor that is only compatible with the tester/tester. Since only the tester/tester can have the new adaptor, no tester/driver can be amplified by PCR in this protocol. Unlike other methods, a 100% subtraction efficiency can be achieved even though the enzymatic treatments used in the enrichment procedure are incomplete. Furthermore, only the hybridized tester DNA can have the new adaptor and be amplified by PCR, resulting in 100% denaturation in effect. The efficacy of this novel method was verified with the model system in which a known amount of the target sequence is included.

  • The existing embedded tester used TCP and shared file system for log processing. In addition, the existing processing method was treated as 1-N structure. This method wastes resources of the tester for exception handling. We implemented a log processing message layer that can be distributed by messaging system. And we compare the transmission method using the message layer and the transmission method using TCP and the shared file system. As a result of comparison, transmission using the message layer showed higher transmission bandwidth than TCP. In the CPU usage, the message layer showed lower efficiency than TCP, but showed no significant difference. It can be seen that the log processing using the message layer shows higher efficiency.

  • This study found a reliable evaluation for four kinds of pb-Free plating solutions and it's layers, through pure Sn, SnAg, SnBi and SnCu. These four kind of solutions are widely used to pb-Free plating. Hull-cell, Harring-blum, coverage, throwing power, current efficiency, stability, life-time, composition, hardness, roughness, abrasion, scratch, solderability, corrosion, contact angle, morphology, SIR(Surface insulation resistance) and Whisker test were experimented. Also, Using ICP, XRF, FE-SEM, EDS, temperature/humidity chamber, solderability tester, hardness tester, roughness tester, abrasion tester, salt spray tester, contact angle tester, SIR tester, and microscope. In this paper could be shown the systematic and various analysis for reliability about four kinds of pb-Free plating solutions, processes and it's deposit surface.

  • A new live-line insulator tester was developed to detect faulty insulators in 154kV power transmission lines. This paper is the first part of the two-part paper and deals with its tester development. The developed tester has the following characteristics. First, it automates parts of the insulator inspection process by using a sensor and an actuator, and secondly, it sustains its weight by itself, not by a lineman. Finally, it measures the insulation resistance of an insulator together with its distribution voltage to provide more information for its analysis and diagnosis. These leads to the improvement of its operation efficiency, measurement reliability and usability. Its effectiveness was validated by live-line field tests in actual power lines.

Design of Low Noise Axial Flow Fan Using Specific Sound Presssure Level (비소음 측정을 이용한 저소음 축류홴 설계)

  • Experimental investigation was conducted to study the effects of pitch angle maximum camber on the performance and noise of an axial-flow fan used in outdoor-unit of air -conditioner. For this study the axial-flow fan whose pitch angle can be varied was made and the Specific sound Pressure Level and other coefficients were measured using the anechoic fan tester. It is found that pitch angle affects more severly than the maximum camber on the fan performance. On the while the maximum camber affects much on the specific sound power level. Present results show that it is important to choose the optimum pitch angle and maximum camber to design the high-performance and low-noise axial-flow fan and specific noise measured in the anechoic fan tester can be sued effectively for the design of low-noise fan.

  • The purpose of this research is to help IPTV (Internet Protocol Television) commercialization using newly produced educational contents in the area of entertainment and education which currently popular in the market. It is called, Stimulus $Tester^{TM}$, endow reaction time from the feedback of learning system, using a non-direct method, for example, a remote controller. Reaction time is the learning efficiency promotion mechanism that learner ascertain the learning condition of oneself by the time with solved questions from the solving the question in given time. Reaction time also play a key role that the learner may go through course which distribute the point to PC from Server. If this system is ready, we expect that the educational industry will gradually spread out. To verify the learning efficiency of this system, we concluded that the learning improvements, by an Internet-based and a paper-based test, of the increase by 51%, from 2.47min to 1.27min, during reaction of 7 days.

  • A standard fire extinguishing tester, with which we could conduct the fire extinguishing ability rating of a water-based fire extinguishing agent such as water and loaded stream, has been developed in this work. It could help us enhancing the efficiency and reliability of the fire extinguishing test of loaded streams, and at last increasing the productivity of fire protection related firms. Furthermore, our country could take the lead in making a new standard for the fire extinguishing test of loaded streams. As a result, it is expected that loaded streams could be improved by using it. In addition the standard fire extinguishing tester could be made moderately in our industry, and supplied at home and worldwide.

  • In this paper, a fuzzy controller using thermoelectric for temperature environmental tester is developed. The new structure of fuzzy controller based temperature environmental tester is proposed and implemented to maintain a stable temperature and improve temperature change speed. In order to evaluate the efficiency, an experiment is setup to compare PID controller with our proposed controller. The experimental results, we proved that our proposed fuzzy controller has better performance than PID controller.

  • The operation condition of recently designed pistons for high power and high speed diesel engine become more severe due to the increment of combustion pressure and temperature. So, in order to overcome high temperature, the application of the mono-metal cast aluminum alloy piston featuring an enclosed cast-in open cooling gallery has increased. In this research, it is developed a PCJ (piston cooling jet) rig tester, described the test procedure and validated the performance of sample piston cooling gallery design. Then the test rig will be used for developing the design technology of piston cooling gallery. The test rig is composed with oil reservoir and pumping system, oil jet system, piston fixing and moving system, collecting oil measuring system, and data measuring and recording system. It will be measured collecting efficiencies under conditions of a few piston positions, oil jet pressures and oil viscosities for a piston cooling gallery. Furthermore, the PCJ rig tester will be used for the optimum design of the oil cooling gallery which being applied to increase the cooling efficiency of pistons in diesel engines satisfying the EURO V emission regulation and the more.

A Study on Improving Filter Performance Using Coanda Effect (코안다효과를 이용한 필터에 관한 연구)

  • Modern people spend a lot of time indoors. Oil mist has a very fatal effect on the cook's health. Range Hood's filter plays a very important role in removing oil mist from kitchen. In this study, we applied coanda effect to increase the collection efficiency of filter. Original, Bottom, Slide Three shapes were simulated, and the two shapes were measured and compared using a wind tunnel tester. Pressure loss is measured below 2.0mmAq for all three filters, making them easy to use indoors. The simulation results were 15% for the original filter, 22% for the bottom filter, and 30% for the slide filter. The wind tunnel tester obtained 26% of the original filter and 30% of the slide filter. Research is needed to increase the collection efficiency by 50%.
