Comparison of Different Vehicles on Human Embryonic Stem Cells using Vitrification

  • Vitrification has been suggested to be an effective method for the cryopreservation of human ES cells. However, the efficiency of vitrification with different vehicles remains a matter of ongoing controversy. The objective of this study was to assess the efficiency of cryopreservation in human ES cells by vitrification using different vehicles. A human ES cell line and a variety of vehicles, including micro-droplet (MD), open-pulled straw (OPS) and electron microscopic grid (EM-grid), were employed in an attempt to assess vitrification efficiency. In order to evaluate the survivability and the undifferentiated state of the post-vitrified human ES cells, we conducted alkaline phosphatase staining and characterization via both RT-PCR and immunofluorescence assays. The survival rates of the post-vitrified human ES cells using MD, OPS and EM-grid were determined to be 61.5%, 66.6% and 53.8%, respectively. There also exist significant differences between slow-freezing and vitrification (p<0.01). However, no significant differences were detected between the vehicle types. Finally, the pluripotency of human ES cells after thawing was verified by teratoma formation. Cryopreservation using vitrification is more effective than slow-freezing, and the efficiency of vehicles proved effective with regard to the preservation of human ES cells.

Natural Purification Treatment using Soil Brick with Combined Effective Microorganisms and Emergent Plants (복합유용미생물 및 수생식물을 활착시킨 흙블록을 이용한 자연정화 처리방법 연구)

  • In this study, using soil brick with combined effective microorganisms and emergent plants was identified which it can increase the effect of conservation and improvement of water. Lab-test was consist of four kind of reactors and each of reactors were A(rawwater), B(soil brick), C(emergent plant) and D(soil brick+emergent plant). Iris pseudoacorus, Phargmites australis, Typha angustifolia and Zizania latifolia were used for emergent plant. Evaluation of application on various environment were performed on agricultural waterway and pond. The pH measurement test of soil brick was performed due to evaluate whether a strong alkaline water flows out of the soil brick. Result of lab-test, removal efficiency of D was better than removal efficiency of A presenting 20.9%, 27.9% 21.5%, 33.8% and 58.4% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Removal efficiency of soil brick on agricultural waterway was revealed to be 49.5%, 45.0%, 43.7%, 37.3% and 28.6% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. And removal efficiency of soil brick on the pond was revealed to be 12.7%, 10.5%, 9.32%, 10.4% and 36.3% for $COD_{Cr}$, $BOD_5$, TN, TP and TSS respectively. Result of pH measurement test of soil brick was neutral which was about 6 to 8.

Effective CPU overclocking scheme considering energy efficiency (에너지 효율을 고려한 효과적인 CPU 오버클럭킹 방법)

  • More recently, the Green Computing have become a important issue in all fields of industry. The energy efficiency cannot be over-emphasized. Microprocessor companies such as Intel Corporation design processors with taking both energy efficiency and performance into account. Nevertheless, general computer users typically utilize the CPU overclocking to enhance the application performance. The overclocking is traditionally considered as an evil in terms of the power consumption. In this paper, we present effective CPU overclocking schemes, which raise CPU frequency while keeping current CPU supply voltage for energy reduction and performance improvement. The proposed scheme gain both energy reduction and performance improvement. Evaluation results show that our proposed schemes reduce the processor execution time as much as 17% and total computer system energy as much as 5%, respectively. In addition, our effective CPU overclocking schemes reduce the Energy Delay Product (EDP) as much as 22%, on average.

Experimental Investigation of Clay Fly Ash Bricks for Gamma-Ray Shielding

  • This study aims to determine the effect of fly ash with a high replacing ratio of clay on the radiation shielding properties of bricks. Some interaction parameters (mass attenuation coefficients, half value layer, effective atomic number, effective electron density, and absorption efficiency) of clay fly ash bricks were measured with a NaI(Tl) detector at 661.6 keV, 1,173.2 keV, and 1,332.5 keV. For the investigation of their shielding behavior, fly ash bricks were molded using an admixture to clay. A narrow beam transmission geometry condition was used for the measurements. The measured values of these parameters were found in good agreement with the theoretical calculations. The elemental compositions of the clay fly ash bricks were analyzed by using an energy dispersive X-ray fluorescence spectrometer. At selected energies the values of the effective atomic numbers and effective electron densities showed a very modest variation with the composition of the fly ash. This seems to be due to the similarity of their elemental compositions. The obtained results were also compared with concrete, in order to study the effect of fly ash content on the radiation shielding properties of clay fly ash bricks. The clay fly ash bricks showed good shielding properties for moderate energy gamma rays. Therefore, these bricks are feasible and eco-friendly compared with traditional clay bricks used for construction.

A Study on the Composition of Atkinson Cycle and Thermodynamically Analysis for a Diesel Engine (디젤기관에 대한 앳킨슨사이클 구성과 사이클의 열역학적 해석에 관한 연구)

  • The present study composed a diesel-atkinson cycle of high expansion as a method of achieving high efficiency in diesel cycle engines. It also interpreted the cycle engine thermodynamically analysis to determine the possibility of the improvement of thermal efficiency and clarified the characteristics of several factors . According to the result of theoretical analysis, heat efficiency was highest when expansion-compression ratio Reど:1. In addition. diesel engines with high apparent compression ratio had higher expansion-compression ratio than otto engines and consequently their effect of high expansion was high. which in turn enhanced thermal efficiency. When the atkinson cycle was implemented in a real diesel engine by applying the miller cycle through the variation of the closing time of the intake valve, the effective compression ratio and the quantify of intake air decreased and as a result, the effect of high expansion was not observed. Accordingly. the atkinson cycle can be implemented when the quantity of intake air is compensated by supercharge and the effective compression ratio is maintained at its initial level through the reduction of the clearance volume. In this case. heat efficiency increased by $4.1\%$ at the same expansion-compression ratio when the apparent compression ratio was 20 and the fuel cut off ratio was 2. As explained above, when the atkinson cycle was used for diesel cycle. heat efficiency was improved. In order to realize high expansion through retarding the intake value closing time, the engine needs to be equipped with variable valve timing equipment, variable compression ratio equipment and supercharged Pressure equipment. Then a diesel-atkinson cycle engine is realized.

Calibration of cylindrical NaI(Tl) gamma-ray detector intended for truncated conical radioactive source

  • The computation of the solid angle and the detector efficiency is considering to be one of the most important factors during the measuring process for the radioactivity, especially the cylindrical γ-ray NaI(Tl) detectors nowadays have applications in several fields such as industry, hazardous for health, the gamma-ray radiation detectors grow to be the main essential instruments in radiation protection sector. In the present work, a generic numerical simulation method (NSM) for calculating the efficiency of the γ-ray spectrometry setup is established. The formulas are suitable for any type of source-to-detector shape and can be valuable to determine the full-energy peak and the total efficiencies and P/T ratio of cylindrical γ-ray NaI(Tl) detector setup concerning the truncated conical radioactive source. This methodology is based on estimate the path length of γ-ray radiation inside the detector active medium, inside the source itself, and the self-attenuation correction factors, which typically use to correct the sample attenuation of the original geometry source. The calculations can be completed in general by using extra reasonable and complicate analytical and numerical techniques than the standard models; especially the effective solid angle, and the detector efficiency have to be calculated in case of the truncated conical radioactive source studied condition. Moreover, the (NSM) can be used for the straight calculations of the γ-ray detector efficiency after the computation of improvement that need in the case of γ-γ coincidence summing (CS). The (NSM) confirmation of the development created by the efficiency transfer method has been achieved by comparing the results of the measuring truncated conical radioactive source with certified nuclide activities with the γ-ray NaI(Tl) detector, and a good agreement was obtained after corrections of (CS). The methodology can be unlimited to find the theoretical efficiencies and modifications equivalent to any geometry by essential sufficiently the physical selective considered situation.

A Study on the Treatment of Dyeing Wastewater Using TiO2/UV (TiO2/UV 산화기술을 이용한 염색폐수처리에 관한 연구)

  • This research uses the $TiO_2$/UV process to verify the most suitable condition and possibility to dispose dyeing wastewater that contains pigment and a large amount of pollutants. For this, this research has enforced experiments that compare photo adsorption, photolysis, and photo catalyst oxidation reaction, and also evaluated and analyzed the change of pH and $TiO_2$ dosage, irradiation rates of ultraviolet rays and the dosage change and injection method of $H_2O_2$. According to the results of the dyeing wastewater experiment of storehouse catalyst that uses the new form of $TiO_2$, the photo catalyst oxidation reaction proved to be more effective than photo adsorption and photolysis; 35%, 21% in the case of $TCOD_{cr}$ and 39%, 28% in the case of chromaticity. Taking into consideration the reaction time, amount of photo catalyst reaction and irradiation amount of ultraviolet rays, the decomposition efficiency of pH change proved to be most effective at pH 4. On the whole, the acidity area proved to be effective in dyeing water exclusion than neutral and alkalinity areas. Having evaluated the influence of $TiO_2$ dosage, not only does the decomposition efficiency continuously improve as the $TiO_2$ dosage increases but the shielding effect does not occur also when the $TiO_2$ is at a fixed state. The influence of ultraviolet irradiation amount concluded in the result that as the ultraviolet irradiation amount increases the decomposition efficiency continually increased, but in the case of chromaticity when the irradiation amount was higher than 37.8mW/cm2 the removal efficiency is slowed remarkably. The influence of $H_2O_2$ dosage evaluation reached the results that although the decomposition efficiency increases with the increase of $H_2O_2$ dosage, when above 150mg (total dosage: 1200mg) $H_2O_2$ consumes OH radical itself and reduces the decomposition efficiency. Also in the case of the $H_2O_2$ injection method rather than injecting in the whole amount of $H_2O_2$ (1200mg) needed at the beginning all at once, injecting divided quantities of $H_2O_2$ whenever the electric current density falls below 10mgfl reduces the wases of OH radical due to an excess of $H_2O_2$ and in tum heightens the decomposition efficiency.

Mixing effect on organic removal efficiency in treating low-strength wastewater using a modified anaerobic filter reactor (변형 혐기성 여상 반응조에서 교반강도가 유기물 제거효율에 미치는 영향)

  • Laboratory investigation was conducted to evaluat the mixing effects on organic removal efficiency to treat low-strength synthetic wastewater using modified anaerobic - filter reactor combining anaerobic filter and upflow anaerobic sludge blanket. Using the modified process the low-strength wastewater like municipal sewage could be treated with 85% T-COD removal efficiency at hydraulic retention time of 6 hours. At the constant organic loading of 0.5 kg COD/m 3-day, the organic removal efficiency and effluent COD concentration are increased as influent COD concentration increased from 125 mg/l to 500 mg/l. Mixing effects on organic removal efficiency are evident and optimum mixing speed is found as 50RPM. Placing the granular sludge and media on which slime layer was pre-formed into the reactor seemed to be very effective In achieving short start-up period. Therefore, the steady state was achived after 4 weeks and 1 week based on T-COD and S-COD, respectively.

  • Data Envelopment Analysis (DEA) is a methodology of computing the relative efficiency of each decision making unit (DMU) by comparing it with other DMUs having similar input and output structure. In this paper, we compare the efficiency of Korean rail transit corporations using DEA. To do this, we design a DEA-AR/AHP model, and evaluate efficiency by comparing the subway operating agencies of six big cities. The analysis reveals that Seoul Metro and Seoul city railroad construction turn out to be the most efficient groups. The result of this research can provide helpful information for effective management in a domestic subway operating agency.
