Anti-inflammatory Activity of Peel Extracts in Color-fleshed Potatoes (컬러감자외피 추출물의 항염활성)

  • Potatoes were first introduced outside the Andes region four centuries ago, and have become an integral part of much of the world's food. Potatoes were first introduced into Europe in the 16th century and Korea in the early 19th century. In the nutritional aspects, potatoes contain abundant vitamins and minerals, as well as an assortment of phytochemicals such as carotenoids and natural phenols. Chlorogenic acid constitutes up to 90% of potato natural phenols. Due to the high content of potato functional compounds, it has known that potatoes are effective in the prevention of various human diseases. Recently, color-fleshed potatoes 'Hongyoung' and 'Jayoung' were developed by RDA, and it has reported that they have high content of anthocyanin. Additionally they show higher radical scavenging activity compared to white or yellow fleshed potatoes. So it will be expected that the consumption of color-fleshed potatoes grandually increase by pre-peeled potatoes and color potato chips. This study was conducted to enhance the peel of color-fleshed potatoes utilization and to determine the biological activity of peel of color-fleshed potatoes extract. The anti-inflammatory effects on ethanol extract and its solvent fraction were also evaluated. The anti-inflammatory activities of $CHCl_3$ fraction was evaluated for inhibitory activities against lipopolysacchride(LPS) induced nitric oxide(NO) and prostaglandin $E_2(PGE_2)$ production as well as inducible nitric oxide synthase(iNOS) and cyclo oxygenase-2(COX-2) protein expressions in RAW264.7 cell lines. The fraction inhibitory activity for both tests with $IC_{50}$ values showed in the ranges of $25{\sim}50{\mu}g/ml$. This result revealed that $CHCl_3$ fraction of Jayoung's peel is expected to be good candidate for development into source of anti-inflammatory agent.

A Study on the Shear Characteristics of the Decomposed Granite Soils Using Direct Shear Test (직접전단시험(直接剪斷試驗)에 의한 화강토(花崗土)의 전단특성(剪斷特性)에 관(關)한 연구(硏究))

  • This paper describes the observed behavior in the direct shear test on decomposed granite soil having the complicate engineering properties at various different levels of factors. The objectives of this study were to investigate the characteristics of the decomposed granite soil under controlled various moisture content, dry density, strain rate and soaking which give influence to the shear strength. The results were summarized as follows; 1. The shear strength was decreased remarkably with the increasing of moisture contents of A and B soil were 5-10% and 15-20% respectively. 2. Cohesion and angle of internal friction were decreased with the increasing of moisture content and increased with the increasing of dry density. 3. The shear strength was increased with the increasing of normal stress and volume change was decreased on the whole. The shear strength was generally increased with the increasing of the strain rate. 4. As dry density increases, A-soil shows the progressive failure and the decrease of volume change while B-soil shows the initial failure and the increase of volume change. 5. The relationships between the soaked and unsoaked specimens were as follows ; ${\tau}_f=0.1009+1.026{{\tau}_f}^*$ (A-soil), ${\tau}_f=0.1586+0.8005{{\tau}_f}^*$ (B-soil) 6. Angle of internal friction of the direct shear test shows larger value than that of the triaxial compression test. All effective stress path was nearly similar.

Synthesis, Sytructure, and Magnetic Properties of One-Dimensional Thiophoshates, $Al_2NiP_2S_6$ (A=Rb, Cs) (1차원 구조를 갖는 Thiophoshates, $Al_2NiP_2S_6$ (A=Rb, Cs)의 합성, 구조 및 자기적 성질)

  • The quaternary thiophosphates, $A_2NiP_2S_6$ (A=Rb, Cs), have been synthesized with halide fluxes and structurally characterized by single-crystal X-ray diffraction technique. These compounds crystallize in the space group $C_{2h}^5-P2_1/n$ of the monoclinic system with two formula units in a cell of dimensions a=5.960(2), b=12.323(4), $c=7.491(3)\AA$, $\beta=97.05(3)^{\circ}$, and $V=546.0(3)\AA^3$ for Rb2NiP2S6 and a=5.957(4), b=12.696(7), $c=7.679(4)\AA$, $b=93.60(5)^{\circ}$, and $V=579.7(5)\AA^3$ for $Cs_2NiP_2S_6.$ These compounds are isostructural. The structure of $Cs_2NiP_2S_6$ is made up of one-dimensional $_\infty^1[NiP_2S_6^{2-}]$ chains along the a axis and these chains are isolated by $Cs^+$ ions. The Ni atom is octahedrally coordinated by six S atoms. These Ni$S_6$ octahedral units are linked by sharing three m-S atoms of the $[P_2S_6^{4-}]$ anions to form the infinite one-dimensional $_\infty^1[NiP_2S_6^{2-}]$ chain. For $Cs_2NiP_2S_6$, the magnetic susceptibility reveals an antiferromagnetic exchange interaction below 8K,which corresponds to the Neel temperature ($T_N$). Above $T_N$, this compound obeys Curie-Weiss law. The magnetic moment, C, and ${\theta}forCs_2NiP_2S_6$ are 2.77 B.M., 0.9593 K, and -19.02 K, respectively. The effective magnetic moment obtained from the magnetic data is agreed with the spin-only value of $Ni^{2+}d^8$(2.83 B.M.) system.

The Effect of Aquapuncture with Hepatonics on the Recovery in Artificially Induced Hepatic Damaged Dogs (간기능강화제의 수침이 간손상 유발견의 회복에 미치는 영향)

  • The present experiment was performed in order to know the treatment effect of aquapuncture with hepatonics on recovery in artificially induced hepatic damaged dogs by carbon tetrachloride. The animals were divided into a control and two experimental groups (aquapuncture with taurine into Gan-su acupoint: Aa-I group and aquapuncture with taurine into blank acupoint in the thigh: Aa-II group). The changes of serum enzyme activities (ALT, AST and ALP), serum total protein contents, protein fractions and pathohistological findings of the liver were examined after application of aquapunctiuf treauent The results obtained through this experiment were summarized as follows : The serum ALT activities tendeded to decrease in experimental group compared with those of control group. Significances were detected at 5th (p<0.05) and 7th (p<0.05) day in Aa-I group and 7th day (p<0.05) in Aa-II group, respectively. Low value was fecund in Aa-I group compared with Aa-II groups but significance was not observed between two experimental groups. The serum AST activities in experimental group showed decreasing tendency compared with those of control group. Significances were observed at 2nd (p<0.05) and 5th (p<0.05) day in Aa-I group and 2nd (p<0.05) day in Aa-II groups respectively. Aa-I group showed lower values than those of Aa-II groups however, no significance was detected between experimental groups. The serum ALP activities of experimental group showed a slight decrease compared with those of control group, however, significance was not detected among all groups. The serum total protein content in experimental group showed tendency of increase compared with control group. Significance was found at 2nd day (p<0.05) in Aa-I group, but there was no significance in Aa-II group. Further significant increase of total protein content was seen at 1st day (p<0.05) in Aa-I group compared with Aa-II group. The change of serum albumin content in experimental group showed tendency of increase compared with control. Significant increases were detected at 1st (p<0.01) and 2nd (p<0.01) day in Aa-I group, respectively. Aa-II group showed increase compared with control groups but significance was not observed. Further significant increase was at 1st day (p <0.05) in Aa-I group compared with Aa-II group. The change of ${\beta} $-globulin in Aa-I group was slightly decreased compared with control group. Aa-II group was similar to controls but significance was not observed among all groups. The change of P-globulin content in Aa-I group showed tendency of increase compared with control and Aa-II group showed the tendency of decrease compared with control. The change of ${\gamma}$-globulin content experimental group showed tendency of increase compared with control, however, significance was not detected among groups. The change of A/G ratio in Aa-I group showed tendency of increase compared with control group and Aa-II group was similar to controls but no significance was found among groups. As for pathohistological observations the grade of hepatocellular vacuolized degeneration and necrosis in Aa-I group was milder than those of control and Aa-II groups and the change of Aa-II group was similar to that of control. Considering above finding collectively, it was thought that aquapuncture of Gan-su acupoint with hepatonics was more effective than aquapuncture of blank acupoint for the recovery of hepatic damage.

Optimization Mixture Ratio of Petasites japonicus, Luffa cylindrica and Houttuynia cordata to Develop a Functional Drink by Mixture Design (혼합물 실험계획법에 의한 머위 및 부원료의 혼합비율 최적화)

  • This study was performed to determine the optimal ratio of Petasites japonicus, Luffa cylindrica, and Houttuynia cordata, all of which are supposed to have anti-respiratory disease effects, such as against rhinitis. The experiment incorporated a mixture design and included 12 experimental points with center replicates for three different independent variables (Petasites japonicus 30~70%; Luffa cylindrica 10~30%; and Houttuynia cordata 10~30%). Based on this design, the mixture was extracted in hot water at 121℃ for 45 min and anti-allergy and anti-microbial activities were observed. The response surface and trace plot described for the anti-allergy activity showed Petasites japonicas was a relatively important factor. The correlation coefficient (R2) value 82.10% for the inhibition effect of degranulation was analyzed by the regression equation. The analysis of variance showed the model fit was statistically significant (p<0.05). The optimal ratio of the mixture was Petasites japonicus 0.75%, Luffa cylindrica 0.11%, and Houttuynia cordata 0.14%. The anti-microbial activity for each extraction of the mixture was valid on gram-positive, such as Staphylococcus aureus (KCCM 40881) and Staphylococcus epidermidis (KCCM 35494), while it was less effective on gram-negative, such as Escherichia coli (KCCM 11234) and Pseudomonas aeruginosa (KCCM 11328).

Potentials of Synbiotics for Pediatric Nutrition and Baby Food Applications: A Review (소아 영양 및 유아식 응용을 위한 신바이오틱스의 잠재력: 총설)

  • Probiotic, prebiotic, and synbiotic substances as well as microorganisms were added to infant formula in an attempt to influence the intestinal microflora with an aim to stimulate the growth of lactic acid bacteria, especially bifidobacteria and lactobacilli. Over the last 10 years, new synbiotic infant formulas containing probiotics and prebiotics have been proposed in order to simulate the effect of breast-feeding on the intestinal microflora. Owing to their synergistic effect, the new synbiotics are expected to be more helpful than using probiotics and prebiotics individually. Maintenance of the viability of the probiotics during food processing and the passage through the gastrointestinal tract should be the most important consideration, since a sufficient number of bacteria ($10^8cfu/g$) should reach the intended location to have a positive effect on the host. Storage conditions and the processing technology used for the manufacture of products such as infant formula adversely affect the viability of the probiotics. When an appropriate and cost-effective microencapsulation methodology using the generally recognized as safe (GRAS) status and substances with high biological value are developed, the quality of infant formulas would improve. The effect of probiotics may be called a double-effect, where one is an immunomodulatory effect, induced by live probiotics that advantageously alter the gastrointestinal microflora, and the other comprises anti-inflammatory responses elicited by dead cells. At present, a new terminology is required to define the dead microorganisms or crude microbial fractions that positively affect health. The term "paraprobiotics" (or ghost probiotics) has been proposed to define dead microbial cells (not damaged or broken) or crude cell extracts (i.e., cell extracts with complex chemical composition) that are beneficial to humans and animals when a sufficient amount is orally or topically administered. The fecal microflora of bottle-fed infants is altered when the milk-based infant formula is supplemented with probiotics or prebiotics. Thus, by increasing the proportion of beneficial bacteria such as bifidobacteria and lactobacilli, prebiotics modify the fecal microbial composition and accordingly regulate the activity of the immune system. Therefore, considerable attention has been focused on the improvement of infant formula quality such that its beneficial effects are comparable to those of human milk, using prebiotics such as inulin and oligosaccharides and potential specific probiotics such as bifidobacteria, which selectively stimulate the proliferation of beneficial bacteria in the microflora and the indigenous intestinal metabolic activity of the microflora.

Physico-chemical and Microbial Properties of Sausages Affected by Plant Scale and Cooking Treatments during Refrigerated Storage (가열조건 및 공장 규모에 따른 소시지의 냉장저장 중 이화학적 및 미생물적 품질특성)

  • Purpose: The objective of this study was to examine the effect of plant scale and cooking conditions on the quality characteristics of sausages during refrigerated storage. Methods: Sausages used in this study were classified into two groups: those submitted to $1^{st}$ cooked treatments and those submitted to $2^{nd}$ cooked treatments. The pH, volatile basic nitrogen (VBN), gas production ratio, and microorganisms were measured in triplicate. Results: The change of quality in the products was assessed every 7 days by measuring pH, VBN levels, total microbes, coliform bacteria, Escherichia coli, and pathogenic bacteria in the products. Pathogenic bacteria such as Staphylococcus aureus, Listeria monocytogenes, Clostridium perfringens, and E. coli were not detected in the sausages with $1^{st}$ cooked treatments. The results showed that the pH of the sausages decline as storage time increased. The pH value of the sausages with $2^{nd}$ cooked treatments changed gradually. VBN levels were generally lower in products with $2^{nd}$ cooked treatments than in those with $1^{st}$ cooked treatments, but they varied with the type of products. On the $35^{th}$ day, the number of total microbes ranged between 6.13-7.12 log CFU/g in products with $1^{st}$ cooked treatments and 3.44-6.92 log CFU/g in products with $2^{nd}$ cooked treatments, showing fewer bacteria in the latter products. Conclusions: $1^{st}$ cooked treatments were effective in microbial control, but $2^{nd}$ cooked treatments could prolong the shelf life of the sausages, indicating a need for differential management of each product.

Antioxidant Activity of Sansa (Crataegi fructus) and Its Application to the Pork Tteokgalbi (산사의 항산화 활성과 돈육 떡갈비로의 적용)

  • This study was performed to investigate the antioxidant effect of Sansa (Crataegi fructus) extract in vitro, and to evaluate the functional effects of Sansa powder addition on the quality properties and storage characteristics of Tteokgalbi. Total polyphenol and flavonoid contents of Sansa extract were found to be 127.00 mg/g and 54.05 mg/g, respectively. The DPPH radical scavenging activity of Sansa extract was high and it was similar to the BHA and BHT. The Tteokgalbi was prepared by 0% (N), 0.1% (S1), 1% (S2), and 2% (S3) of the Sansa Powder. Addition of Sansa powder decreased the protein and lipid contents, but the ash content was significantly increased (p<0.05). Increasing the amount of Sansa powder in the pork Tteokgalbi tended to increase the water holding capacity (WHC) values and the cooking loss (p<0.05). The addition of Sansa powder increased the hardness and chewiness values, but did not affect the cohesiveness and springiness values. In the sensory evaluation, the S3 Tteokgalbi had the best score in color. Values of pH, total microbial counts, thiobarbituric acid (TBA) and volatile basic nitrogen (VBN) values decreased significantly added Sansa powder relative to the normal (p<0.05). The S3 Tteokgalbi was significantly (p<0.05) more effective for delaying lipid peroxidation than the other groups. Sansa powder addition increased the L (lightness) and a (redness) values. Therefore, the results demonstrate that adding the Sansa powder to the pork Tteokgalbi tended to improve antioxidative and antimicrobial effects during the chilled storage period.

Increase of Spoken Number of Syllables Using MIT(Melody Intonation Therapy) : Case Studies on older adult with stroke and aphasia (MIT(Melodic Intonation Therapy) 중심의 음악활동을 이용한 실어증을 가진 뇌졸중 노인의 음절 수 증가에 대한 사례 연구)

  • Most of stroke patients have not only physical difficulty but speech and neurological disorder because of hemiplegia and such unexpected changes cause psychologic disadaptability and absent-mindedness. Particularly, lowering of physical ability can lead to serious emotional problem from failure or frustration in daily life. Generally, treatment of patient with stroke put emphasis on physical rehabilitation but actually this patient had considerable speech disorder such as aphasia or articulation disorder. Moreover, failing of recognition function, mental disorder as hypochondria, and even visual and auditory disorder are represented. So it is effective to integrate verbal remediation and other treatments in medical care environment. In particular, patients with language disorder very often wither psychologically therefore it is efficient to use of music therapy that gives opulent emotion to aphasia patients. And primarily to investigate the effects of 10 sessions treatments; change in spoken total number of syllables, to confirm their own value by success of given task and reassure about themselves ability. All of 10 sessions stages were scored by MIT manual and its improvement were measured, that is, accomplishment was analyzed within each level in order to prove detail change of spoken total number of syllables. The result of this program organized from 2 syllables to 4 syllables is summarized as follows. Subject A completed in preliminary stage Level I, in 2 syllables case advanced to Level III in fifth session and to Level IV in seventh session, in 3 syllables case advanced to Level III in seventh session and to Level IV in ninth session, and in 4 syllables case showed 8% low success rate in first session but after repeated practice increased considerably in sixth session and in advanced to Level III in eighth session to Level IV in tenth session. Subject B also completed in preliminary stage Level I, in 2 syllables case advanced to Level III in forth session and to Level IV in sixth session, in 3 syllables case advanced to Level III in fifth session and to Level IV in seventh session, and in 4 syllables case showed 10% low success rate in first session and increased considerably in fifth session and in advanced to Level III in seventh session but could not reach to Level IV until tenth session. As a result, it was shown that music therapy using MIT was not statistically meaningful but improved spoken total number of syllables and success rate of task had improved as a whole. Therefore, music intervention using MIT it has positive affect on verbal ability of patients with Broca's Aphasia and their language rehabilitation.

Characterization of Chitin Deacetylase Produced from Mucor rouxii (Mucor rouxii가 생산한 Chitin deacetylase의 특성)

  • In order to degrade chitin by enzymatic hydrolysis, it is required from screening highly active deacetylase. To this end, we examined three fungal strains and it turned out that Mucor rouxii produced highly active deacetylase, this enzyme exhibited the highest enzymatic activity against colloidal chitin. The conditions for growing Mucor rouxii are as follows; the effective carbon source, nitrogen source, adequate initial pH, temperature and incubation time were $2\%$ glucose, $1.33\%$ yeast extract, $0.66\%$ pepton, 4.5, $25{\pm}2^{\circ}C$ and 48hr, respectively. The optimum pH and temperature for purified enzyme activity were 5.5 and $40^{\circ}C$, respectively. The purified enzyme was stable at pH ranging from 4.5 to 5.5. However, the enzyme activity was decreased to less than $50\%$ at pH blow 45 and above 7.5. At temperatures above $50^{\circ}C$, the enzyme activity was decreased remarkably. The enzyme was inhibited by LiC1, $HgCl_2$, and $BaCl_2$, but stimulated by $CaCl_2$ and $ZnC1_2$, The activity of purified enzyme was increased by L-cysteine and 2-mercaptoethanol, while decreased by O-phenanthroline, p-CMB, EDTA, and iodoacetate. The $K_m$ and the $V_{max}$ value of purified enzyme were $1.2\%$ and 59.5 U/mg, respectively. The deacetylation activity of purified enzyme was not detected at optimal reaction condition when chitin particle suspension was used.
