Production of Angiotensin-I Converting Enzyme Inhibitory Hydrolysates from Egg Albumen

  • ACE (Angiotensin-I converting enzyme) inhibitory peptides derived from foods are thought to suppress high blood pressure by inhibiting ACE. We tried to make efficient production of the ACE inhibitory hydrolysate from egg albumen. A hydrolysate digested by neutrase presented the highest ACE inhibitory activity ($IC_50\;value=256.35{\mu}g/ml$) and the proper proteolysis was occurred by 1.0% enzyme addition and 4 h incubation at $47^{\circ}C$. Antihypertensive effect of neutrase hydrolysate was investigated in spontaneously hypertensive rats (SHR, n=5). Systolic blood pressure (SBP) was decrease by 6.88% (-14.14 mmHg, p<0.05) at 3 h after oral administration of 300 mg/kg body weight, and by 13.33% (-27.72 mmHg, p<0.05) by emulsified hydrolysate. These results showed that it is very effective to utilize egg albumen as a protein source for the production of ACE inhibitory peptides. However, further studies are required to investigate the methods to increase recovery yield and the isolation of active peptide is necessary for determining its sequence responsible for ACE inhibitory activity.

Effects of Dietary Addition of Bentonite on Manure Gas Emission, Health, Production, and Meat Characteristics of Hanwoo (Bos taurus coreanae) Steers

  • A study was conducted to determine the dietary effects of a clay mineral (sodium bentonite, NaB) on manure gas emission, health, production, and meat characteristics of Hanwoo steers. Two diets fed to steers included a control diet (concentrate mix and rice straw) and a treatment diet (control diet+1.0% clay mineral/concentrate mix). Dietary NaB addition considerably reduced concentrations of gases ($H_2S$, $SO_2$ and $NH_3$) in the manure of Hanwoo steers. Growing steers fed NaB had similar blood profiles with the exception of lower (p<0.05) concentrations of blood alkaline phosphatase and lactate dehydrogenase. Dietary NaB addition tended to increase (p = 0.10) live weight by 30 kg at slaughtering and did not affect carcass yield and quality traits. Dietary NaB addition increased concentrations of P (p<0.01), Mg (p<0.01), Na (p<0.01), Zn (p<0.005), K (p = 0.08), Fe (p = 0.08) and Cu (p = 0.07) in the longissimus muscle compared to the control but did not affect (p>0.05) fatty acid composition. The study demonstrated that the dietary addition of a clay mineral could be effective in improving mineral bioavailability to Hanwoo steers, which could be one of the reasons for their improved performance.

Standardization of a Mass-Production Technique for Pycnidiospores of Dydymella bryoniae, Gummy Stem Blight Fungus of Cucurbits (박과작물 덩굴마름병 Didymella bryoniae의 병포자 대량 생산 방법의 표준화)

  • Didymella bryoniae, gummy stem blight fungus of cucurbits, has been known not to produce its pycnidium in vitro without irradiation. Various methods for producing pycnidiospores of the fungus as an inoculum have been used. However, those methods have not been verified in terms of efficiency of the productivity, activity and synchronous maturation of the inoculum. Therefore, a pycnidiospore production method in vitro that is highly reliable and reproducible has to be developed to obtain a large amount of inoculum for screening disease resistant varieties or effective fungicides. Here we standardized a mass-production technique for pycnidiospores of D. bryoniae in vitro by comprehensively finding the optimal conditions such as kinds and thickness of cultural medium, growing temperature, and quality and duration of irradiation as well as examining the activity and pathogenicity of the pycnidiospores reproduced. In brief, mycelial colony on the PDA plate was cultured at 26$^{\circ}C$ for 2 days under the darkness, and then the plate was irradiated under the UV light (12 hr/a day) for 2~3 days at the same temperature(26$^{\circ}C$). Two days after UV irradiation, a great number of pycnidia was simultaneously formed. This plate was subjected to darkness again for 4~5 days to mature pycnidiospores. We could obtain a large amount of inoculum that is synchronously matured in a short period of time through the above procedures.

Biocontrol Activity of Volatile-Producing Bacillus megaterium and Pseudomonas protegens against Aspergillus flavus and Aflatoxin Production on Stored Rice Grains

  • In our previous study, three bacterial strains, Bacillus megaterium KU143, Microbacterium testaceum KU313, and Pseudomonas protegens AS15, were selected as effective biocontrol agents against Aspergillus flavus on stored rice grains. In this study, we evaluated the inhibitory effects of the volatiles produced by the strains on A. flavus growth and aflatoxin production on stored rice grains. The three strains significantly reduced mycelial growth of A. flavus in dual-culture assays compared with the negative control strain, Sphingomonas aquatilis KU408, and an untreated control. Of these tested strains, volatiles produced by B. megaterium KU143 and P. protegens AS15 markedly inhibited mycelial growth, sporulation, and conidial germination of A. flavus on agar medium and suppressed the fungal populations in rice grains. Moreover, volatiles produced by these two strains significantly reduced aflatoxin production in the rice grains by A. flavus. To our knowledge, this is the first report of the suppression of A. flavus aflatoxin production in rice grains using B. megaterium and P. protegens volatiles.

  • The production of micro components is one of the leading technologies in the fields of information and communiation, medical and biotechnology, and micro sensor and micro actuator system. Microfabrication (micromachining) techniques such as X-ray lithography, electroforming, micromolding and excimer laser ablation are used for the production of micro components out of silicon, polymer and a limited number of pure metals or binary alloys. However, since the first development of microfabrication technologies there have been demands for the cost-effective replication in large scale series as well as the extended range of available material. One such promising process is micro powder injection molding (PIM), which inherits the advantages of the conventional PIM technology, such as low production cost, shape complexity, applicability to many materials, applicability to many materials, and good tolerance. This paper reports on a fundamental investigation of the application of W-Cu powder to micro metal injection molding (MIM), especially in view of achieving a good filling and a safe removal of a micro mold conducted in the experiment. It is absolutely legitimate and meaningful, at the present state of the technique, to continue developing the micro MIM towards production processes for micro components.

Basic Design of Deep Subsea Manifold Frame Structure for Oil Production (심해저 원유 생산용 매니폴드 프레임 구조 기본 설계)

  • Amanifold is one of the essential subsea oil and gas production components to simplify the subsea production layout. It collects the production fluid from a couple of wellheads, transfers it to onshore or offshore storage platforms, and even accommodates water and gas injection flowlines. This paper presents the basic design procedure for a manifold frame structure with novel structural verification using in-house unity check codes. Loads and load cases for the design of an SIL 3 class-manifold are established from a survey of relevant industrial codes. The basic design of the manifold frame is developed based on simple load considerations such as the self weights of the manifold frame and pipeline system. In-house software with Eurocode 3 embedded, called INHA-SOLVER, makes it possible to carry out code checks on the yield and buckling unities. This paper finally proves that the new design of the manifold frame structure is effective to resist a permanent and environment load, and the in-house code is also adaptively combined with the commercial finite element code Nastran.

  • Until a recent date, Toyota Production System (called TPS) was introduced by many domestic companies to remove waste and reduce manufacturing cost. However, cases of substantial and effective improvement after the introduction are not much. Even though many companies have actively conducted TPS during that time, the outcome is not satisfactory. In this paper, we show the problems and core contents to consider in applying of TPS as follows. First, the innovative organizational culture formed by active participation of employees and leadership of the CEO is very important for a successful introduction of TPS above all. Second, it is necessary to prepare various training programs optimized for the field in order to continuously improve the competency of employees in each class, and to train skilled personnel through that programs. Third, it is necessary to improve the maturity level of TPS application through the construction of correct evaluation system on accomplishment of the production system. In addition, the problems that occur should be solved through the continuous improvement activities. These results will help to TPS introduction of the domestic small-medium companies. Therefore, this study will contribute to strengthen and improve the global competitiveness in the related industries.

A Basic Study on Leaf and Stem Production of Angelica acutiloba

  • This study was conducted to develop effective production system in greenhouse for leaves and stems of Angelica acutiloba by using connect pots in 2003 and 2004. Seed germination rate and plant biomass of Angelica acutiloba collected in 2004 were higher than those harvested in 2003. Germination rate of Angelica acutiloba seeds collected in 2003 was 10%, while germination rate of seeds collected in 2004 was above 90%. Especially, plant growth and yield of Angelica acutiloba grown in connect pots sized with 4 ${\times}$ 4 ${\times}$ 5cm(length ${\times}$ width ${\times}$ height) were the highest. These results indicate that leaf and stem production of Angelica acutiloba can be improved by using connect pots and optimizing seed collecting time in greenhouse.

내열성 Chitinase 생산균주의 분리 및 효소생산 특성

  • A strain capable of producing thermostable chitinase suitable for chitooligosaccharide production was isolated from high temperature environment and identified as Bacillus licheniformis. The chitinase from Bacillus licheniformis KFB-Cl4 was only induced by addition of colloidal thitin into the basal medium as carbon source, showing the decrease of the chitinase production by supplernental addition of other carbon sources into the medium containing 1.0% colloidal chitin. Among organic and inorganic nitrogen sources, yeast extract was the most effective for the increase of total activity and specific activity, and had high affinity for the enzyme production. The optimum temperature of cell growth and thermostable chitinase production was 55$\circ$C. The optimum culture medium was composed of 1.2% colloidal chitin, 0.15% K$_{2}$HPO$_{4}$, 0.05% KH$_{2}$PO$_{4}$, 0.01% MgSO$_{4}$-7H$_{2}$O, 0.1% yeast extract (pH 6.5). Bacillus licheniformis KFB-C14 produced the thermostable chitinase of 3.89 units per ml culture fluid and 7.4 units per mg protein under rotary shaking at 150 rpm for 40 hr.

Production of Compaction from Penicillium sp. Y-8515 (Penicillium sp. Y-8515에 의한 Compactin 생산)

  • A strain producing high levels of compaction was isolated from soil and identified as Penicillium sp. Y-8515 based on the morphological characteristics and ribosomal RNA sequence analysis. Optimization of several different carbon and nitrogen sources for the effective production of compaction was performed resulting in the medium compositions containing 5%(w/v) glucose, 1.0 % soybean meal, 0.5% yeast extract, 0.5%(NH$_4$)$_2$$SO_4$, 0.25%,$ NaH_2$$PO_4$, 0.25% $CaCO_3$. The fixed con-centration of glucose(5%, w/v) and relatively lower concentrations(less than 2.5%, w/v) of soybean meal stimu-lated the transformation of the growth morphology from filamentous to pellet form. Comparing to that by filamentous form, the production of compactin by pellet form increased up to 1.5 folds. In a fed-batch fermentation, continuous feeding of the mixture of glucose and nitrogen source at the ratio of 10:1 showed 3.5-fold more produc-tion yield of compaction comparing to the batch mode.
