Production of Immunostimulating Polysaccharide in Angelica gigas Nakai SusDension Cell Cultures (참당귀 현탁세포배양에 의한 면역증강성 다당 생산)

  • Suspension cells of Angelica gigas Nakai were cultivated to produce extracellular polysaccharide(ECP) as immunostimulating agents. Effects of environmental conditions such as sucrose and 2,4-dichlorophenoxyacetic acid(2,4-D) concentrations on the growth and production of ECP were studied using suspension cultures of A. gigas Nakai. Final dry cell weight was increased with an increase of initial sucrose concentration from 30 to 60 g/L. The maximum production of ECP(1.2 g/L) was achieved at an initial sucrose concentration of 50 g/L on day 8. High 2,4-D concentration was effective for ECP production but not for cell growth. In addition, various fungal elicitors were investigated for the enhanced production of ECP in A. gigas suspension cultures. Among the tested fungal elicitors, Verticillium dahliae was the most effective for the production of ECP in A. gigas suspension culture.

Tylosin Production by Streptomyces fradiae Using Raw Cornmeal in Airlift Bioreactor

  • Using a 50-1 airlift bioreactor, for the effective production of tylosin from Streptomyces fradiae TM-224 using raw cornmeal as the energy source, various environmental factors were studied in flask cultures. The maximum tylosin concentration was obtained at $32^{\circ}C$ and pH between 7.0 and 7.5. When seed was inoculated after 24 h of culture, the maximum tylosin concentration, 5.7 g/l, was obtained after 4 days of culture. Various concentrations of raw cornmeal were tested to investigate the optimum initial concentration for the tylosin production. An initial raw cornmeal concentration of 80 g/l gave the highest tylosin concentration, 5.8 g/l, after 5 days of culture. Of the various nitrogen sources, soybean meal and fish meal were found to be the most effective for the production of tylosin. In particular, with the optimal mixing ratio, 12 g/l of soybean meal to 14 g/l of fish meal, 7.2 g/l of tylosin was obtained after 5 days of culture. To compare raw cornmeal and glucose for the production of tylosin in the 50-1 airlift bioreactor for 10 days, fed-batch cultures were carried out under the optimum culture conditions. When raw com meal was used as the energy source, the tylosin production increased with increasing culture time. The maximum tylosin concentration after 10 days of culture was 13.5 g/l, with a product yield from raw cornmeal of 0.123 g/g of consumed carbon source, which was about 7.2 times higher than that obtained when glucose was used as the carbon source.

Instruction Effects of Teaching Relative Clauses on Comprehension and Production in Korean EFL Classes

  • This study investigates the effects of three different types of instruction, namely form-based, comprehension-based, and production-based on the development of Korean university students' (n=137) comprehension and production of English relative clauses (RCs). The extent of improvements was analyzed by administering pre-and post-tests consisting of two comprehension tests (selecting the right form of RCs and the right picture descriptions) and one production test (combining two sentences). Findings of this study suggest that all three types of instruction increased participants' comprehension and productions of RCs. However, there appeared differential effects by the instruction type. It was found production-based instruction was most effective in promoting comprehension, followed by comprehension-based instruction. Comprehension-based instruction worked best with the development of production, suggesting that the effects of comprehension training did not only work for increasing comprehension skills, but also transfer to production skills. The type or level of tasks employed for each instruction appeared to play an important role in causing such results. Form-based instruction displayed the lowest improvements in both comprehension and production of RCs. A sentence-combination task employed for form-based instruction appear to result in mere explicit rule explanations without chances to notice rules in context or use their knowledge in practice.

Production Cost Analysis of Leaf tobacco farm Households (잎담배 재배농가의 생산비 분석)

  • This study had carried out an analysis of leaf tobacco production cost by cost items, growing stages, and farm sizes per 10a to provide the basic data for determination of the purchasing price of leaf tobacco by KT&G. Considering the survey results of 12 tobacco farm households, the composition rates of production cost by items revealed as 7-10% for land service, 5-22% for depreciation, 13-25% for material costs, 50-65% for labour cost respectively. The production cost of leaf tobacco by growing stages were shown as 15.3% in nursery bed period, 32.3% in main growing period in field, 30.8% in harvesting period and 21.6% in packing period. The magnitude of wage expenditure was appeared as harvesting stage, packing stage, growing stage on main field and nursery bed stage in order. The amount of material costs were revealed as the growing stage in main field, harvesting stage, nursery bed stage and packing stage respectively. The production costs of leaf tobacco per 10a by farm sizes were shown as 1,615,879won for small farm, 1,446,896won for medium farm and 1,454,408won for large farm respectively. The production cost of leaf tobacco had shown decreasing tendency according to increasing farm sizes. To promote the international market competitiveness of leaf tobacco producing farms, labour saving production technologies and cost effective farm size to decrease tobacco production cost should be developed.

Production of Laccase by Trametes sp. CJ-105 (Trametes sp. CJ-105에 의한 Laccase 생산)

  • For Trametes sp. CJ-105, a kind of white-rot fungi which was collected from the mountain of Korea and was proven to be effective in decolorizing a wide range of structurally different synthetic dyes, the optimum conditions for mycelial growth and laccase(E.C. 1.10.3.2) production were investigated. Among various carbon sources, glucose showed the highest potential for the mycelial growth and laccase production, the optimum concentration being 2% glucose. For the nitrogen source, asparagine was good for the mycelial growth, while ammonium tartrate for laccase production(optimum concentration: 0.04%). The addition of thiamine and biotin increased both th emycelial growth and laccase production. When 2,5-xylidine was added as an inducer after the first day of culture, the production of alccase was seven-times higher than that in the absence of the inducer. The optimum pH and temperature conditions for laccase production by Trametes sp. CJ-105 were pH 5.0 and $25^{\circ}C$, respectively. In the 5L fermentation, the production of laccase reached a maximum of 340U/ml at the time when the ammonium ion was being rapidly depleted.

Production and Characterization of Fibrinolytic Enzyme: Optimal Condition for Production of the Enzyme from Bacillus sp. KP-6408 Isolated from Chungkook-jang (새로운 혈전용해 효소의 생성 및 특성: 청국장에서 분리한 Bacillus sp. KP-6408로부터 효소 생성의 최적조건)

  • A bacteium, KP-6408, capable of hydrolyzing fibrin was isolated from Chungkook-jang, which was possibly identified as a strain of Bacillus sp. The effects of culture condition and medium composition on the enzyme production were investigated. Among nitrogen sources tested, yeast extract was the most effective for the enzyme production, and the level of the concentration for the optimal enzyme production was 0.2%(w/v). For carbon sources, glucose was the best for the enzyme production with the level of 2.0%(w/v). The enzyme was maximally produced by cultivating the enzyme production with the level of 2.0%(w/v). The enzyme was maximally produced by cultivating the organism at the liquid medium of the initial pH 8.0 and temperature of 4$0^{\circ}C$. In Chungkook-jang fermentation, the enzyme was maximally produced when incubated at 35$^{\circ}C$ for 24 hrs using soybean as a solid medium. The addition of various rice starch to the soybean in Chungkook-jang fermentation lowered the enzyme production.

Cultural Conditions for the Production of Organic Acid During (Aspergillus awamori var. kawachii에 의한 쌀 Koji 제조시 유기산의 생산조건)

  • This study was carried out to investigate the influences of cultural conditions of koji on the production of organic acid during rice-koji making by Aspergillus awamori var. kawachii which is now widely used as koji-mold in brewing Takju and Yakju in Korea. The optimum temperature for the germination of the conidia of the mold was 35'8, and the time required for germination at this temperature was 8 hours. Rapid germination occurred when the water content of steamed rice was above 40%, but germination retardation occurred markedly below 35%. The optimum cultural temperature for the production of organic acid was 32$^{\circ}C$, and the production of organic acid was markedly restricted at 36$^{\circ}C$ and 4$0^{\circ}C$. It was effective for the high production of both saccharogenic amylase and organic acid to shift the cultural temperature from initial 36$^{\circ}C$ to 32$^{\circ}C$ after 20~25 hours of cultivation. Initial water content suitable to the production of organic acid was 40% in steamed rice, but its production was markedly restricted below 30% of water content. When the quantity of conidial inoculation was too small, the production of organic acid was low in initial phase, but it was retrived at later period. Acid production was markedly restricted together with the increase in koji thickness.

Studies on the Production of Protease by Aspergillus oryzae KC-15 and Characteristics of the Enzymes (Aspergillus oryzae KC-15에 의한 protease의 생산 및 그 효소의 특징에 관한 연구)

  • This experiment was conducted to investigate the conditions for production and the characteristics of pretenses. Aspergillus oryzae KC-15, which is selected as a superior strain for the production of the protease, was used in this study. The results obtained were as follows: 1. The optimum culture time for the production of acid, neutral and alkaline protease on wheat bran medium were about 48, 48 and 72hr, respectively. The protease-produced by the strain were mainly alkaline and neutral one, but the production of acid protease was feeble extremely. 2. The addition of NaH$_2$PO$_4$, Na$_2$HPO$_4$, glucose, rice powder and Na-glutamate respectively to wheat bran media were effective for the production of alkaline and neutral protease, and the addition of (NH$_4$)$_2$HPO$_4$, glucose and rice powder respectively were effective for the production of acid protease. 3. Characteristics of professes(equation omitted) 4. As a heat resistance agent, NaH$_2$PO$_4$was the most effective one. The optimum amount of NaH$_2$PO$_4$was 10mg for alkaline and neutral protease, and 5mg for acid protease. 5. The heat resistance of the Protease by NaH$_2$PO$_4$was not recognized mostly above 6$0^{\circ}C$. 6. After the treatment of enzyme solution with 10mg of NaH$_2$PO$_4$for 30 minutes at 55$^{\circ}C$, the residual activities measured for alkaline, neutral and acid protease were 58, 57 and 55% respectively.

Effective Tillering Pattern and Grain Yield on Different Seeding Dates in Barley (보리 파종기에 따른 유효분벽의 양상과 수량)

  • This study was aimed to provide understanding on the eco-physiological response of barley tillers as affected by difference in seeding date. Yield and yield contribution rates of tillers were investigated with the data of field experiments in the former Wheat and Barley Research Institute of Suwon, Korea from September 1982 to July 1984. When barley was sowed 15 days earlier than or at the locally recommended sowing date (i.e. October 5), it produced more mainstem leaves than the that sowed 15 days later than the local recommendation. The effective tillers (i.e. ear-bearing tillers) were observed in concurrently occurring leaves up to 10/0 (abbreviation of the 10th mainstem leaf) from early and optimum sowed crops, while 9/0 from late sowed crops. Tillering followed the production rule of Gatayama(l952) which has the linear leaf appearance pattern. Early sowing produced more effective tillers, but the highest production of effective ears (i.e. ears heavier than 0.505g) was noticed in sowing at the recommended date. The tillers in the axil of first leaf in mainstem (abbreviated as 1) produced more effective ears than other mainstem tillers (i.e. primary tillers such as C, 2, 3 and 4). The tillers from the axil of first leaf, whether they were primary or secondary, always performed better in the production of effective tillers, grains per ear, grain weight, and grain yield per ear than those from the axil of coleoptile or prophyll. Other tillers from subsequent leaves were also inferior in production with the order of their appearance, thus making first leaf tillers as the best performer(e.g. 1 against C, 2, 3, 4 of primary tillers, 11 and 21 against 1P, 12, 13, 2P, 22 of secondary tillers). Even though the first leaf tiller from the first mainstem leaf(i.e. 11) emerged at a same time with the fourth mainstem leaf tiller(i.e. 4), it was always a better producer of ears and grains. The above observations of hierarchy among tillers were persistent irrespective of cropping conditions and treatments. Sowing at the recommended date produced more effective tillers and grains per ear than early or late sowing. In early and late sowings, more grains per effective ear were observed by early sowing, whereas more grains per ear were produced by late sowing. The order of performance in production of effective tillers and ears per plant was as follows: optimum > early > late sowings. In optimum sowing the mainstems were the highest in grain weight per ear, while the first leaf tillers were the ones in early sowing probably due to winter damage on mainstem ears. Yield contribution by the tillers was greater with the following order irrespective of sowing dates: mainstem, 1, 2, 3, C, 11, 12 and Cl. The contribution of CP, 2P, 21 and 31 varied with sowings.

Manipulating Isoflavone Levels in Plants

  • Metabolic engineering for production of isoflavones in nonlegume plants could distribute the health benefits of these phytoestrogens in more widely-consumed grains. Series of investigation to check the ability of the heterologous isoflavone synthase enzyme to interact with the endogenous phenylpropanoid pathway have been conducted. Overall, results provide possibility of production of isoflavonoids in several plant tissue systems including soybean and nonlegumes. In tissue that undergoes naturally enhanced synthesis of anthocyanins, genistein production was enhanced. In a monocot cell system, introduced expression of a transcription factor regulating genes of the anthocyanin pathway was effective in conferring the ability to produce genistein in the presence of the isoflavone synthase gene. However, in this case the intermediate accumulated to high levels indicating an inefficiency in its conversion. Introduction of a third gene, chalcone reductase, provided the ability to synthesize an additional substrate of isoflavone synthase resulting in production of the isoflavone daidzein. These research efforts provide insight into requirements for metabolic engineering for isoflavone production in nonlegume dicot and monocot tissues.
