The Effects of ChongMyungTang(CMT) and ChongMyungTang added Hibiscus syriacus(MCMT) Extract on the Alzheimer's Disease Model Induced by CT-105 and ${\beta}A$ (총명탕(聰明湯)과 목근피총명탕(木槿皮聰明湯)이 CT105와 ${\beta}A$로 유도된 Alzheimer's Disease 병태(病態) 모델에 미치는 영향)

  • Objective : This research investigates the effect of the CMT and MCMT on Alzheimer‘s disease. Methods : The effects of the CMT and MCMT extract on (1) amyloid precursor proteins(APP), acetylcholinesterase(AChE) mRNA of PC-12 cells treated with CT-105; (2) the AChE activity and the APP production of PC-12 cell treated with CT-105; (3) the behavior; (4) expression of $IL-1{\beta}$, $TNF-{\alpha}$, MDA, GFAP, CD68 abd CD11b; (5) the infarction area of the hippocampus in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. Results : 1. The CMT and MCMT extract suppressed the expression of APP, AChE, and mRNA in PC-12 cells treated with CT-105. 2. The CMT and MCMT extract suppressed the AChE activity, and the production of APP significantly in PC-12 cells treated with CT-105. 3. For the CMT and MCMT extract group a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 4. The CMT and MCMT extract suppressed the over-expression of $IL-1{\beta}$, $TNF-{\alpha}$, MDA, GFAP, CD68 abd CD11bCD68/GFAP, in the mice with Alzheimer's disease induced by ${\beta}A$. 5. The CMT and MCMT extract reduced the infarction area of hippocampus with Alzheimer's disease induced by ${\beta}A$ 6. The MCMT showed more excellent effects than CMT in the every experiments except PC-12 cells. Conclusions : These results suggest that the CMT and MCMT extract may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of the CMT and MCMT extract for Alzheimer's disease is suggested for future research.

  • This research investigates the effect of the Hibiscus syriacus(HSS) on Alzheimer's disease. Specifically, the effects of the HSS extract on (1) $IL-1{\beta}$, IL-6, and $TNF-{\alpha}$ mRNA of PC-12 cells treated with LPS; (2) amyloid precursor proteins(APP), acetylcholinesterase(AChE), and glial fibrillary acidic protein(GFAP) mRNA of PC-12 cells treated with CT-105; (3) the AChE activity and the APP production of PC-12 cell treated with CT-105; (4) the behavior; (4) expression of $IL-1{\beta}$, $TNF-{\alpha}$, $IL-1{\beta}$ mRNA, $TNF-{\alpha}$ mRNA, and reactive oxygen species(ROS); (5) the infarction area of the hippocampus, and brain tissue injury in Alzheimer's diseased mice induced with ${\beta}A$ were investigated. The results were summarized below ; 1. The HSS extract suppressed the expression of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ mRNA in THP-l cells treated with LPS. 2. The HSS extract suppressed the expression of APP, AChE, and GFAP mRNA in PC-12 cells treated with CT-105. 3. The HSS extract suppressed the AChE activity, and the production of APP significantly in PC-12 cells treated with CT-105. 4. For the HSS extract group a significant inhibitory effect on the memory deficit was shown for the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured stop-through latency, and distance movement-through latency. 5. Investigation into the clinical use of the HSS extract for Alzheimer's disease is suggested for future research.

  • The homogeneous road section is defined as one consisted of similar traffic characteristics focused on demand and supply. The criteria, in the aspect of demand, are the diverging rate and the ratio of green time to cycle time at signalized intersection, and distance between the signalized intersections. The criteria, in that or supply, are the traffic patterns such as traffic volume and its speed. In this study, the effective method to generate valuable data, pointing out the problems of removal method of obscure data, is proposed using data collected from Gonjiam IC to Jangji IC on the national highway No.3. Travel times are collected with licence matching method and traffic volume and speed are collected from detectors. Futhermore, the method of selecting homogeneous road section is proposed considering demand and supply aspect simultaneously. This method using outlier filtering algorithm can be applied to generate the travel time forecasting model and to revise the obscured of missing data transmitting from detectors. The point and link data collected at the same time on the rational highway can be used as a basis predicting the travel time and revising the obscured data in the future.

Is Compact Urban Spatial Structure Effective for Public Transportation Mode? (컴팩트형 공간구조가 대중교통수단의 이용활성화에 보다 효과적인가?)

  • The purpose of this study was to find the characteristics of travel behavior and accessibility in terms of spatial structure. We analyzed travel behaviors and accessibility using a mode choice model and the Complementary Accessibility Index(CAI). The urban spatial structures that were compared were a compact city (CC) versus a sprawled city (SC), and high residential density districts (HD) versus low residential density districts (LD). First, CC and HDs residents had a shorter commuting distance than the CC and LDs residents. Second, behavior models showed that the use of Private cars for commuting in SCs was found to be greater than private car use in CCs, and that public transportation modes would be encouraged in CCs. Third, changes associated with the time and cost of commuting by private car generally affect the demand for public transportation modes in the CC. Also, analysis of cross elasticity suggests that changes of subway travel time affect the demand for buses very elastically. Fourth, the CAI of SC and LD were superior to the CC and HD even though the SC inefficient urban forms in terms of spatial structure. So, the spatial distribution of population density was also found to be an important factor affecting accessibility and energy savings.

Feasibility Study of Laparoscopic Gastrostomy Tube Placement in Beagle Dogs (비글견에서 복강경을 이용한 위관삽입술의 유용성 평가 연구)

  • Aim of this study is demonstrate the feasibility of Laparoscopic gastrostomy (LG) tube placement in dogs by comparing with percutaneous endoscopic gastrostomy (PEG) tube placement, based on operative time, complications and gastro-peritoneal adhesion evaluation. Eight intact male beagle dogs were used in this study. Tri-Funnel Replacement Gastrostomy tube (Bard Inc., USA) of 20 Fr was used for LG technique and PEG kit (Ponsky "Pull" PEG Kit$^{(R)}$, Bard Inc., USA) with soft silicone retention dome consisting of a 20 Fr gastrostomy tube was used. Feeding via gastrostomy tube was performed in two weeks, maintenance energy requirement (MER) divided into 3 separate feeding. LG and PEG were evaluated at intraoperative, postoperative and postmortem period. Mean operative time for the PEG group was significantly shorter when compared with the LG group (p < 0.05). Successful maintenance of gastrostomy tube was confirmed in all dogs. Gastric and peritoneal wall adhesions were formed successfully in each group. The mean adhesion length (AL) and width (AW) were significantly larger in LG group compared with in PEG group (p < 0.05). The mean adhesion distance (AD) was not significantly different between two groups (p = 0.182). Consequently, LG is an effective minimally invasive, safe and easy to perform technique for providing enteral nutritional support in dogs.

An Extraction Method of Number Plates for Various Vehicles Using Digital Signal Analysis Processing Techniques (디지털 신호 분석 기법을 이용한 다양한 번호판 추출 방법)

  • Detection of a number plate consists of three stages; division of a number plate, extraction of each character from the plate, recognition of the characters. Among of these three states, division stage of a number plate is the most important part and also the most time-consuming state. This paper suggests an effective region extraction method of a number plate for various images obtained from unmanned inspection systems of illegal parking violation, especially when we have to consider the diverse surrounding environments of roads. Our approaching method detects each region by investigating the characteristics in changes of brightness and intensity between the background part and character part, and the characteristics on character parts such as the sizes, heights, widths, and distance in between two characters. The method also divides a number plate into different types of the plate. This research can solve the number plate region detection failure problems caused by plate edge damages not only for Korean domestic number plates but also for new European style number plates. The method also reduces the time consumption by processing the detection in real-time, therefore, it can be used as a practical solution.

Semi-active storey isolation system employing MRE isolator with parameter identification based on NSGA-II with DCD

  • Base isolation, one of the popular seismic protection approaches proven to be effective in practical applications, has been widely applied worldwide during the past few decades. As the techniques mature, it has been recognised that, the biggest issue faced in base isolation technique is the challenge of great base displacement demand, which leads to the potential of overturning of the structure, instability and permanent damage of the isolators. Meanwhile, drain, ventilation and regular maintenance at the base isolation level are quite difficult and rather time- and fund- consuming, especially in the highly populated areas. To address these challenges, a number of efforts have been dedicated to propose new isolation systems, including segmental building, additional storey isolation (ASI) and mid-storey isolation system, etc. However, such techniques have their own flaws, among which whipping effect is the most obvious one. Moreover, due to their inherent passive nature, all these techniques, including traditional base isolation system, show incapability to cope with the unpredictable and diverse nature of earthquakes. The solution for the aforementioned challenge is to develop an innovative vibration isolation system to realise variable structural stiffness to maximise the adaptability and controllability of the system. Recently, advances on the development of an adaptive magneto-rheological elastomer (MRE) vibration isolator has enlightened the development of adaptive base isolation systems due to its ability to alter stiffness by changing applied electrical current. In this study, an innovative semi-active storey isolation system inserting such novel MRE isolators between each floor is proposed. The stiffness of each level in the proposed isolation system can thus be changed according to characteristics of the MRE isolators. Non-dominated sorting genetic algorithm type II (NSGA-II) with dynamic crowding distance (DCD) is utilised for the optimisation of the parameters at isolation level in the system. Extensive comparative simulation studies have been conducted using 5-storey benchmark model to evaluate the performance of the proposed isolation system under different earthquake excitations. Simulation results compare the seismic responses of bare building, building with passive controlled MRE base isolation system, building with passive-controlled MRE storey isolation system and building with optimised storey isolation system.

Gene Transfer in Normal and Ischemic Tibialis Anterior Muscle of Rat by In Vivo Electroporation

  • The purpose of this preliminary study is to improve the efficiency of gene transfer of nonviral plasmid DNA by in vivo electroporation in ischemic hindlimb muscle, tibialis anterior. Hindlimb ischemic model was aseptically made by excision of left femoral artery. Each $50\;{\mu}g$ of pEGFP-C1 and pGL3-control in $100\;{\mu}l$ 0.9% NaCl was injected in tibialis anterior muscle. In vivo electroporation was applied on the same site with 10 mm-distance 2 needle array electrodes and ECM830. In 3 groups of normal rat with different electric field strength 0, 200 and 800 V/cm, the expression of pEGFP-C1 was comparatively evaluated. In 8 groups of normal rats, the expression of pGL3-control was evaluated in 0, 40, 50, 80, 100, 150, 200 and 300 V/cm of electric field strength. In 5 groups of ischemic models, the expression of pGL3-control was analyzed on 0, 4, 7, 10 and 14 days elapsed after making ischemic models. In 9 groups of ischemic rats, the expression of pGL3-control was analyzed in the electric field strength 0, 60, 70, 80, 100, 150, 200, 250 and 300 V/cm. GFP expressions in normal tibialis anterior were high in the extent and degree in order of electric field strength of 200, 800 and 0 V/cm. Luciferase value was highest in $50{\sim}100\;V/cm$ electric field strength. In the case of ischemic models, luciferase expression was significantly increasing in the order elapsed time after making the model. The degree of luciferase expression was higher in cases of application of in vivo electroporation than in that of non-application and was highest in $100{\sim}150\;V/cm$. In conclusion, in vivo electroporation is effective in transfer and expression of plasmid DNA in normal and ischemic tibialis anterior of rat.

  • Earthworks are the fundamental steps in a construction job, and are mainly comprised of smaller tasks performed by construction machinery. The productivity of the construction job can be improved by optimizing excavation, filling, and other such operations. Earthworks involve a lot of mechanical work performed by the collaboration between various kinds of construction equipment, which in turn leads to higher fuel consumption. Actual earthworks depend mostly on the intuition and experience of the driver of the machines, thus leading to inefficiency and environmental problems caused by unnecessary emission of carbon, Recently automated and information-oriented technologies are consistently being researched towards the improvement of efficiency of earthworks in the construction industry. The present research involves the introduction and understanding of the decision-making elements of heuristics which can be applied to the earthwork planning. A method is also suggested for creating an effective work path for construction machine to perform task packages (TP) for cutting and filling processes. A simulation test is performed to verify the effectiveness of suggested methods in terms of space interference and total moving distance of construction equipment.

  • Objective : This experiment was designed to investigate the effect of the GYZB hot water extract & ultra-fine powder on the Alzheimer's disease model induced by amyloid ${\beta}$ protein (${\beta}A$). Method : We measured the effects of the GYZB hot water extract on expression of $IL-1{\beta}$, IL-6 mRNA and production of IL-6, $TNF-{\alpha}$ in the BV2 microglial cell line treated with lipopolysaccharide (LPS). The effects of the GYZB hot water extract & ultra-fine powder on (1) the behavior, (2) expression of $IL-1{\beta}$ and $TNF-{\alpha}$, (3) glucose in serum, (4) the infarction area of the hippocampus, and brain tissue injury in mice induced with Alzheimer's diseased by ${\beta}A$ were investigated. Results : The GYZB hot water extract suppressed the expression of $IL-1{\beta}$ and IL-6 mRNA and significantly suppressed the production of IL-6 and $TNF-{\alpha}$ in the BV2 microglial cell line treated with LPS. The GYZB hot water extract & ultra-fine powder showed a significant inhibitory effect on the memory deficit of the mice with Alzheimer's disease induced by ${\beta}A$ in the Morris water maze experiment, which measured stop-through latency and distance movement-through latency. The GYZB ultra-fine powder significantly suppressed the expression of $IL-1{\beta}$ and $TNF-{\alpha}$ protein, and the GYZB hot water extract significantly suppressed the expression of $TNF-{\alpha}$ protein in the microglial cell of mice with Alzheimer's disease induced by ${\beta}A$. The GYZB hot water extract & ultra-fine powder reduced the infarction area of hippocampus in the mice with Alzheimer's disease induced by ${\beta}A$. Conclusions : These results suggest that GYZB hot water extract & ultra-fine powder may be effective for the prevention and treatment of Alzheimer's disease. Investigation into the clinical use of GYZB for Alzheimer's disease is suggested for future research.
