해양생태독성평가를 위한 열대 요각류 Nitocra sp.의 이용 가능성 (The availability of tropical copepod Nitocra sp. for marine ecotoxicological evaluation)

  • 열대지역과 같은 특정지역에서 오염물질의 독성평가 시, 그 지역의 생태환경에 적합한 위해성평가를 위해서는 그 지방 고유의 생물에 대한 독성실험이 요구된다. 따라서 본 연구는 열대에서 분리한 열대 요각류 Nitocra sp.를 독성실험생물로 사용하기 위해 이들을 안정적으로 배양/유지하기 위한 최적배양환경조건과 해양생태독성평가 가능성을 조사하였다. 최적 배양환경요인으로 수온, 염분 및 먹이에 대해 조사하였으며 생태독성평가는 급성독성과 만성독성 실험으로 나누어 실시하였다. 최적배양조건 실험데이터의 통계분석을 위해 One-way ANOVA test를 실시하였다. 최적배양환경조건을 조사한 결과, Nitocra sp.는 수온 $29^{\circ}C$, 염분 24~34‰에서 먹이로 Tetraselmis suecica를 공급하였을 때, 비교적 빠른 발달기간과 높은 생존율을 보였다. 최적배양조건을 바탕으로 구리와 비소에 대한 독성평가를 실시한 결과, 구리와 비소의 각 노출농도에 따라 민감하게 잘 반응해서 반수치사농도 즉 $LC_{50}$값과 영향을 미치지 않는 농도인 NOEC값을 얻을 수 있었다. 만성독성시험 결과, 구리와 비소노출 모두, 성비와 생산력은 유의적인 차이가 없었던 반면, 발달기간과 생존율은 농도에 따라 반응을 보였기 때문에 종말점으로 사용이 가능한 것으로 나타났다. 본 연구를 종합해 보았을 때, 열대 요각류인 Nitocra sp.는 열대 해양독성물질 평가를 위한 생태독성실험생물로 사용이 가능할 것으로 판단되며 차후 다양한 독성물질의 평가에 활용이 기대된다.

합성세제의 생태독성 평가 (Ecotoxicological Evaluation of Synthetic Surfactant)

  • In order to evaluate the ecotoxicological harzardousness of synthetic surfactants on Han river, Jung-ryang and Jin-Wi stream, we used the Ecotoxicological Risk Quotient (ERQ). The chemical harzardousness is evaluated by the balance of the toxicity and concentration in the environment.. Then, ERQ is defined as follows; ERQ = - log ( Concentration in the environment / Effective concentration in the test ) ERQ of chemical is a logtrighmic value of ratio of a chemical concentration and the toxicity in the laboratory. In case of small ERQ, tie chemical harzardousness is high. If ERQ equals O, the same biological effect as in the laboratory test will be observed in the enviromment by the chemicals. ERQ values of the chemicals were calculated using the maximum concentration in water environment which were cited from the annual report by our ministry of environment, and EC$_{50}$ of Daphnia magna (water flea; acute immobilization test) LC$_{50}$ of Oryzias latipes (fish; acue toxicity test) and EC$_{50}$ of chlorella vulgaris (alga; growth inhibition test), which were taken from the annual report of "Chemical in environment" by Japan EA. Liner alkylbenzene sulfonate (determined to MBAS) showed the high average values with more than 2.0 to three species in Han river and Jin-wi stream, and these results mean to be favorable to environmental safety. The areas of Jung-ryang stream were polluted, as the average values of ERQ were less than 2.0 with equal to three species, and attention should be paid. Therefore, they must be inspected again because their concentration in the environment may have changed during that period. The chemical harzardousness can be numerated with ERQ, and it can be a help to find the chemicals that should be kept under observation and to see whether the chemical pollution is improved or worsened. The determination of the chemical concentration in the environment and toxicity are essential for the effective use of ERQ.se of ERQ.

해양생태독성평가를 위한 표준시험생물로서의 식물플랑크톤에 관한 연구 (Phytoplankton as Standard Test Species for Marine Ecotoxicological Evaluation)

  • A series of experiments was conducted to identify the potential of five phytoplankton species as standard test species for marine ecotoxicological tests. The candidate phytoplankton species are Skeletonema costatum, Heterosigma akashiwo, Prorocentrum micans, Isochrysis galbana, and Tetraselmis suecica. Salinity tolerance and sensitivity on potassium dichromate as a reference material were identified. Toxicity of eleven ocean dumped sewage sludges and four red tide expellent extracts were estimated by the inhibition of population growth rates (PGR) of marine diatom S. costatum, While most species revealed relatively weak tolerance on salinity, T. suecica demonstrated the highest salinity tolerance ranged from $5\~35$ psu and the others $15\~35$ psu. H. akashiwo revealed the highest sensitivity as 72h $IC_{50}$=0.76mg/L and T. suecica the lowest as 72h $IC_{50}$=8.89mg/L on potassium dichromate. Sludge extracts from industrial waste, domestic sewage and livestock farm waste sludge showed high toxicity as 72h $IC_{50}$<$2\%$ and lowest toxicity from filtration bed sludge as 72h $IC_{50}$=$30.50\%$ NOEC (No Observed Effective Concentration) of sludge extract ranged from <$0.4\%$ to $1.6\%$ and this indicated high phytotoxicity of ocean dumped sewage sludge. The test sensitivity of phytoplankton PGR inhibition was much higher than those of marine rotifer Brachionus plicatilis mortality test and bioluminescent inhibition test by marine bacteria Vibrio fischeri, and comparable with the sea urchin (Strongylocentrotus intermedius) fertilization test. As a result the phytotoxicity test using phytoplankton PGR inhibition ($IC_{50}$) must be a useful tool for marine phyto-toxicological evaluation of ocean dumped materials.

병저항성 GM(OsCK1)벼의 물벼룩(Daphnia magna)에 대한 급성독성 평가 (Acute Toxicity Evaluation to Daphnia magna of Disease Resistant(OsCK1) Rice)

  • BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oryza sativa and Streptomyces hygroscopicus into the genome of the rice, Nakdongbyeo. With the potential problems of safeties, the evaluations on non-target organisms are essentially required for the environmental risk assessment of genetically modified (GM) crops. In the present study, we conducted the evaluation of acute toxicity on Daphnia magna that commonly used as a model organism in ecotoxicological studies for non-target organism evaluation. METHODS AND RESULTS: Effect of acute toxicity to Daphnia magna by each concentration were investigated in the disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice, Nakdongbyeo, as concentration (0, 1,000, 1,800, 3,240, 5,830, 10,500 and 20,000 mg/L). The OsCK1 rice used for the test was confirmed to express the OsCK1/PAT gene by the PCR(Polymerase chain reaction) and western blot analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of Daphnia magna fed on OsCK1 rice or non-GM rice. The 48hr-$EC_{50}$ values showed no difference between OsCK1 rice (3,147.18 mg/L) and non-GM rice (3,596.27 mg/L). CONCLUSION: This result suggested that there was no significant difference in toxicity to Daphnia magna between OsCK1 rice and non-GM counterpart.

Risk assessment and evaluation of epidermal growth factor (EGF) transgenic soybean: responses of Cyprinus carpio fed on EGF transgenic soybean

  • The epidermal growth factor (EGF) transgenic soybean was developed and biosynthesis of human epidermal growth factor (hEGF) in soybean seeds was confirmed. Also, EGF transgenic soybean were found to contain a herbicide resistance selectable marker by introduction of phosphinothricin acetyltransferase (PAT) gene from the Streptomyces hygroscopicus. For biosafety assessment, the EGF transgenic soybean expressing the EGF biosynthesis gene EGF and herbicide resistant gene PAT was tested to determine effects on survival of Cyprinus carpio, commonly used as a model organism in ecotoxicological studies. C. carpio was fed 100% ground soybean suspension, EGF soybean or non-genetically modified (GM) counterpart soybean (Gwangan). Gene expression of EGF soybean was confirmed by PCR and ELISA to have EGF/PAT. Feeding test showed that no significant differences in cumulative immobility or abnormal response between C. carpio samples fed on EGF soybean and non-GM counterpart soybean. The 48 h-EC50 values of the EGF and non-GM soybean were 1,688 mg·L-1 (95% confidence limits: 1,585 - 1,798 mg·L-1) and 1,575 mg·L-1 (95% confidence limits: 1,433 - 1,731 mg·L-1), respectively. The soybean NOEC (no observed effect concentration) value for C. carpio was suggested to be 625 mg·L-1. We concluded that there was no significant difference in toxicity for non-target organisms (C. carpio) between the EGF soybean and non-GM counterparts.

  • Changes in certain fish behaviors are known to be very sensitive indicators of sublethal exposure to environmental contaminants. Therefore, behavioral toxicity tests, if properly designed, can be very useful to assess the influence of hazardous chemicals on fish. However, quantitative analysis of xenobiotic-mediated changes in locomotor behavior in fishes are rare, due mainly to the methodological difficulties. In general, fish movement has been known to be hypertrophic or hypotrophic according to the chemicals. As a study of fish behavioral toxicities, we qualified the swimming movement of Oryzias latipes using computer-automated video tracking system. Oryzias latipes was exposed to cadmium of 128 mg/L for 1 hour in a limited aquaria, then the total swimming distance, the average swimming velocity, the histogram of turning angles, and the turning frequency were analyzed. Fish treated with cadmium showed decreased swimming activities, decreased velocity, and decreased turning frequency, which means hypotrophic activity. From these results, the computer-automated video tracking system of this study seems to be a good tool for the evaluation of the potential ecotoxicological studies.

조류를 이용한 시화호 및 시화호 유입수 내 중금속 물질의 독성 평가 연구 (Toxicity Assessment of Heavy Metals in Shihwa Lake and Its Tributaries using the Algae)

  • This research investigates how using algae as an ecotoxicological test species is easier than using daphnia for identifying toxic causative substances. From the results of the ecotoxicity measurements on the Shihwa lake and its tributaries, heavy metals were considered as one of major factors in causing toxicity. The algae ecotoxicity value was 9.6 while the daphnia ecotoxicity value was 0.8 in the Jeongwang stream. By using algae as the test species, we could identify the toxicity that causes heavy metals which might otherwise have been missed with only daphnia. The results from the EDTA addition test showed that zinc and copper were the main toxic causative substances in the Jeongwang stream and Gunja stream.

Ecotoxicological Evaluation of Sewage Sludge Using Bioluminescent Marine Bacteria and Rotifer

  • Bioassay using the marine bacteria, Vibrio fischeri and rotifer, Brachionus plicatilis, and chemical analyses were conducted to assess the toxicity of the various sewage sludges, one of the major ocean dumped materials in the Yellow Sea of Korea. Sludge elutriates extracted by filtered seawater were used to estimate the ecotoxicity of the sludge. Chemical characterization included the analyses of organic contents, heavy metals, and persistent organic pollutants in sludge. Bacterial bioluminescent inhibition (15 min), rotifer mortality (24 hr) and rotifer population growth inhibition (48 hr) assay were conducted to estimate the sludge toxicity. EC50 15 min (inhibition concentration of bioluminescence after 15 minutes exposed) values by Microtox(R) bioassay clearly revealed different toxicity levels depending on the sludge sources. Highest toxicity for the bacteria was found with the sludge extract from dyeing waste and followed by industrial waste, livestock waste, and leather processing waste. Clear toxic effects on the bacteria were not found in the sludge extract from filtration bed sludge and rural sewage sludge. Consistent with Microtox(R) results, rotifer neonate mortality and population growth inhibition test also showed highest toxicity in dyeing waste and low in filtration bed and rural sewage sludge. High concentrations of persistent organic pollutants (POPs) and heavy metals were measured in the samples from the industrial wastes, leather processing plant waste sludge, and urban sewage sludge. However, there was no significant correlation between pollutant concentration levels and the toxicity values of the sludge. This suggests that the ecotoxicity in addition to the chemical analyses of various sludge samples must be estimated before release of potential harmful waste in the natural environment as part of an ecological risk assessment.
