Sleep-Promoting Effect of Ecklonia cava: Ethanol Extract Promotes Non-rapid Eye Movement Sleep in C57BL/6N Mice

  • We investigated the effects of Ecklonia cava ethanol extract (ECE) on sleep architecture and sleep profiles. ECE was orally administered at a dose of 100, 250, or 500 mg/kg to C57BL/6N mice and its effects were measured by recording electroencephalogram (EEG) and electromyogram. Administration of ECE (250 and 500 mg/kg) significantly induced non-rapid eye movement sleep (NREMS) without affecting rapid eye movement sleep. The increase in NREMS by ECE (500 mg/kg) was significant (P < 0.05) during the first 2 h after administration. In addition, ECE had no effect on EEG power density (an indicator of sleep quality) in NREMS. These results suggest that ECE induces NREMS in a manner similar to physiological sleep.

Preparing Bi-component Dye of Unripe Diospyros kaki THUNB. Fruit and Ecklonia cava and Investigating Its Dyeing Propeties on Fabric (풋감과 감태의 이성분 복합염료 제조와 섬유 염색성 고찰)

  • This paper proposes a bi-component dye, including the unripe fruit of Diospyros kaki THUNB and Ecklonia cava, to substitute for traditional persimmon dyeing because fabrics dyed with persimmon juice become stiffer and natural persimmon is insufficient for dyeing. This study examined the color difference and fabric stiffness depending on the ratio of Ecklonia cava for in a one-bath dye solution with Diospyros kaki THUNB and showed that 6% of Ecklonia cava in the bi-component dye was the optimum for decreasing the fabric stiffness. Based on these results, a bi-component dye constituting of 94% Diospyros kaki THUNB and 6% Ecklonia cava was prepared. The particle size was found to be smaller than both single dyes and it maintained a similar amount of Catechin to Diospyros kaki THUNB dye. Finally, cotton fabric dyed with a bi-component dye was much improved in terms of the fabric hand and the surface color was similar to that of the traditional persimmon-dyed fabric. These results could help to develop the natural persimmon dyeing industry.

Cellular protective effect of Ecklonia cava extract on ultra-fine dust (PM2.5)-induced cytoxicity (초미세먼지(PM2.5)로 유도된 in vitro 세포 독성에 대한 감태(Ecklonia cava) 추출물의 보호 효과)

  • To evaluate the protective effect of Ecklonia cava on ultra-fine dust ($PM_{2.5}$)-induced cytotoxicity, we investigated the in vitro antioxidant activity and cell viability after exposure to $PM_{2.5}$. E. cava was extracted using water and 80% ethanol, and antioxidant activity was determined using the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS)/2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and lipid peroxidation inhibition assays. The 80% ethanol extract showed relatively higher antioxidant activity than the water extract. The cell protective effects were determined by measuring the intracellular reactive oxygen species (ROS) content and viability of nasal epithelial (RPMI-2650), lung epithelial (A549), and brain neuroblastoma (MC-IXC) cells. Results showed that the 80% ethanol extract inhibited ROS production more than the water extract. In contrast, both extracts showed similar effects on cell viability in the $PM_{2.5}$-induced cell death assay. Thus, Ecklonia cava may act as an effective resource for preventing $PM_{2.5}$-induced cytotoxicity in nasal, lung, and brain cells.

Anti-hyperlipidemic Effect of Polyphenol Extract (SeapolynolTM) and Dieckol Isolated from Ecklonia cava in in vivo and in vitro Models

  • The inhibitory effect of polyphenol extracts (Seapolynol$^{TM}$, SPN) of the marine brown algae Ecklonia cava and dieckol, a major component of SPN, on hyperlipidemia was investigated in ICR mice fed a high-fat diet (HFD) for five weeks. For analysis of the anti-hyperlipidemic effects of SPN and dieckol, these two agents were given orally on a daily basis to HFD-fed mice for four weeks, starting one week after the beginning of HFD feeding. Groups administered with SPN as well as dieckol showed lower body weight gains than the HFD only group. Administration of SPN and dieckol also resulted in a significant reduction of the level of total cholesterol (TCHO), triglyceride (TG), and low-density lipoprotein (LDL) cholesterol in the serum of HFD-fed mice. In Oil Red O staining using 3T3-L1 preadipocytes, it was shown that both SPN and dieckol markedly inhibited lipid accumulation of 3T3-L1 cells. Furthermore, SPN and dieckol (50 ${\mu}g$/mL) significantly inhibited 3-hydroxyl-methyl glutaryl coenzyme A (HMGCoA) reductase activity in vitro. Taken together, these results suggest that polyphenols of Ecklonia cava (SPN) and dieckol reduce body weight gain and fat accumulation in HFD-induced obese mice, and that their hypolipidemic effect is related to the inhibition of adipogenesis of adipocytes and HMGCoA reductase activity.

Concentration of Fucoxanthin from Ecklonia cava Using Supercritical Carbon Dioxide (초임계 이산화탄소를 이용하여 감태로부터 Fucoxanthin 농축)

  • Fucoxanthin has been concentrated efficiently using supercritical carbon dioxide. First, fucoxanthin was extracted from Ecklonia cava using solvents. Three solvents, such as medium chain fatty acid (MCFA), medium chain fatty acid ethyl ester, and fatty acid from coconut oil were tested, and MCFA was selected as a suitable solvent to extract fucoxanthin from Ecklonia cava. The concentration was conducted at various pressures and temperatures. In order to concentrate fucoxanthin from extracts, MCFA was eliminated from the extract using supercritical carbon dioxide, and fucoxanthin was concentrated successfully in residue. Optimal conditions for concentrating the fucoxanthin from fucoxanthin extract were $40^{\circ}C$ at 9.7 MPa, $45^{\circ}C$ at 11.0 MPa, and $50^{\circ}C$ at 12.4 MPa, respectively. The density of carbon dioxide affected the concentration of fucoxanthin from fucoxanthin extract. In this optimal conditions, the density for concentrating the fucoxanthin was 600 g/L.

A Study on the Growth Effects of Ecklonia cava By-product (감태 가공부산물을 이용한 성장효과에 관한 연구)

  • We investigated the beneficial effects of Ecklonia cava by-product (ECB), a residual product obtained after polyphenol extraction from Ecklonia cava, on normal rats. Male Sprague-Dawley rats were divided into the following three groups: Control group (NC), received basal diet; ECB 0.1 group, received basal diet supplemented with 0.1% ECB for 21 days; ECB 0.5 group, received basal diet supplemented with 0.5% ECB for 21 days. The productivity, serum immunoglobulin level, expression levels of muscle-related genes, and cecal microflora were measured in all the treatment groups to evaluate the potential use of ECB as a feed additive. The ECB 0.1 group exhibited enhanced expression of Myod, Myog, and Igf1 genes, which increased the body weight of rats. Additionally, treatment with ECB increased the cecal Lactobacillus spp. counts and the serum immunoglobulin G levels in rats. Thus, ECB can effectively increase body weight and can be a potential feed additive for improving growth.
