Evaluaton of the Government Recommended Forage Cultivars in Korea I. Forage performance and quality of oat cultivars (목초 및 사료작물 정부장려품종의 지역적응성 평가 I. 연맥품종의 사초수량 및 사료가치)

  • Nine oat(Avena sativa L.) cultivars have been recommended as the govemment recommended forage cultivars since 1984, however, their forage performance and quality have mostly been tested at two locations, such as Suweon in the Middle Northwestem Coast Region and Sunghwan in the Middle Southwestern Coast Region. Therefore, the objective of this experiment was to retest the forage performance and quality of six government recommended oat cultivars at five scattered locations, such as Icheon, Suweon, Sunghwan, Keongsan and Kwangju fom 1992 to 1994 with the intention of improving recommendations to dairy farmers. Among the oat cultivars tested, "West" was the highest yielding cultivar at all locations in 1992-94, but the lowest yielding cultivars varied by location and year. At both Icheon and Suweon, "Cayuse", "Magnum" and "Foothill", at Sunghwan, "Cayuse" , "Ensiler" and "Foothill", at Keongsan, "Magnum", "Cayuse" and "Swan", and at Kwangju, "Cayuse", "Magnum" and "Cayuse", were the lowest yielding cultivars in 1993, 1994 and 1995, respectively. At all locations in 1992-94, "West" and "Swan" were the most advanced in growth stage, but "Cayuse", "Foothill", "Magnum" and "Ensiler" were the latest developing cultivars among the oats. Differences among the oat cultivars for the three-year mean NDF, ADF and IVDMD were small and inconsistent, but forage quality of the oats was generally lowest in the early developing cultivars and highest in the late developing cultivars. The effects of growth medium and partial shade on early growth of milkweed (Calotropis procera L.) under drought stress

  • The use of growth medium is often recommended milkweed seedlings to grow and develop after emergence, and it is affected by growth medium and local habitat conditions. The effects of growth medium and partial shade on early growth of milkweed under drought stress (Calotropis procera L.) were studied in a field experiment. A split-split plot experimental design with three replications was carried out in the nursery. The main treatment plot was divided into two levels of shade; (no shading and partial shading). Sub treatment plot1 included growth medium at four levels (G1 = clay [suitable for milkweed growth], G2 = clay + sand, G3 = clay + perlite, G4 = clay + perlite + sand) and sub treatment plot2 included drought (irrigation intervals) at six levels (D1 = 2 [control], D2 = 4, D3 = 6, D4 = 8, D5 = 10, and D6 = 12 days per for three month). The results showed that drought stress significantly decreased emergence percentage, shoot length, shoot dry weight (SDW1), root dry weight (RDW), seedling dry weight (SDW2) and vigor index (VI). The use of growth medium increased all seedling characteristics. The G3 (clay + perlite) growth medium showed the highest performance, especially in terms of emergence percentage and seedling dry weight. Partial shade improved shoot length, shoot dry weight, and vigor index. Our results showed that the best treatment for high-vigor milkweed seedlings under drought stress was G3 (clay + perlite) growth medium and partial shade.

Molecular and Cellular Analyses of NCP, a Nuclear and Centrosomal Protein in Mouse Gametes and Early Embryos

  • For many animals the centrosome consists of a pair of centrioles and surrounding pericentriolar materials (PCMs). PCMs have been known to play roles during cell division. It is known that centrioles are necessary to assemble centrosomal components. However, many types of oocytes undergo meiosis without centrioles. It is known that in nonmurine mammalian species, the sperm introduces an intact proximal centriole unlike sea urchin where two centrioles are introduced. In case of mouse sperm, the presence of centrosome is not clear In this study, a monoclonal antibody was developed to investigate centrosome during mouse germ cell and early embryo development. Results of immunostaining and Western blotting in CHO cells suggest that the monoclonal antibody recognizes a nuclear and centrosomal protein, thus called NCP. The NCP monoclonal antibody was used to screen a cDNA expression library prepared from 12.5 mouse brain to isolate NCP gene. Nucleotide size of NCP gene obtained from immunoscreening was about 5.5kb. It is determined that the NCP may be closely related with pericentriolar material -1 gene (Pcm-1) from the result of sequencing analysis. The molecular weight, 66kDa, calculated by known DNA sequence in database is consistent with that of detected from Western blotting using CHO cell lysates. Therefore, it is assumed that NCP may be alternative splicing form of Pcm-1 of which molecular weight is 228kDa. In mouse oocytes, NCP was distributed in nucleus as in CHO cells. It was shown that the NCP was localized around neck region, probably the centrosome in mouse neck region. Interestingly, dramatic change in distribution of NCP was also shown in male germ cell development. Finally, we observed the cellular distribution of NCP during early embryo development. NCP was detected in nucleus as well as centrosome foci. It is suggested that the centrioles reassembly we occurring in blastocysts and then affects the distribution of NCP.

Flight Activity and Injury Characteristics of Beet Armyworm, Spodoptera exigua(Hubner), (Lepidoptera: Noctuidae) in Southern Region of Korea (남부지방에서 파밤나방의 비산활동 및 가해특성)

  • This study investigated seasonal occurrence and daily flight activity using synthetic sex pherom mone and injury characteristics of 야et armyworm, Spodoptera exigua in Chonnam area of K Korea in 1990. Male adults were attracted to phermone trap from mid-June to late November w with the peaks at mid-late August, mid-late September and early-mid November in vinyl i house and also were attracted from mid October to late November in field. Primary peak of occurrence in vinyl house was 20 days sooner than field. It seems to have 6 generations a year in C Chonnam region based on the estimation of larval biomass. Daily male flight was active from m midnight to before sunrise with the peak at 4-6 A.M .. In injury characteristics at various host plants, damage was great in early growing stage at below 10 cm of plant height in Allium fistulsum. Chrysanthemum morifolium was damaged at mostly upper part of leaves reaching to m the extent up to 70%. In case of Gypsophila paniculata, plants injured at early growing stage do n not grow and flower normally.

Effects of Maturing Stage of Corn Hybrids on Silage Yield, Feeding Value for Dairy Cows and Milk Production in a Cold Region of Japan

  • This experiment was conducted to evaluate the effects of differently maturing corn hybrids on silage production and milk production per unit area in the northern part of Japan, where grain development occurs under decreasing ambient temperature. Both hybrids were harvested at the same time. The stages of maturity for the early-maturing hybrids (EH; 80 d relative maturity) and the mid-maturing hybrids (MH; 93 d relative day) were early dent and late dough stage, respectively. The plant yields for MH were higher than those for EH. The dry matter (DM) content of MH was lower than that for EH, and the effluent loss for MH silage was greater than that for EH silage. Therefore, the DM yields of prepared silage per area were similar for both treatments. Twelve multiparous mid-lactation Holstein cows ($58{\pm}13$ days in milk) were fed diets based on EH or MH silage in a crossover design with two 3-week periods. Cows were fed 3 kg of hay crop silage (DM basis) and either EH or MH silage ad libitum, and concentrates were supplied to meet NRC requirement for dairy cows. Silage DM intake for EH was found to be higher (p<0.05) than that for MH (10.0 vs. 9.1 kg/day). Milk production and milk composition for EH were similar to those for MH. Feed efficiency per total feed intake was similar in both treatments, although the feed efficiency per concentrate intake tended to be higher for the EH than that for the MH diet. These results indicate that differences in maturation in corn hybrids affect the effluent production of silage and the silage intake of dairy cows. It may be advantageous to plant early hybrid corn with a reduction in effluent production of silage as well as a reduction in purchased feed costs for dairy cows under the climatic conditions of the northern part of Japan.

Determination of Marginal Sowing Date for Soybean in Paddy Field Cultivation in the Southern Region of Korea

  • A double-cropping system with soybean (Glycine max) following the cultivation of potato, garlic, and onion is widely adopted in the southern region of Korea. For this system, marginal dates for planting must be determined for profitable soybean yields, because the decision to plant soybean as a second crop is occasionally delayed by harvest of the first crop and weather conditions. In order to investigate the effect of planting date on soybean yield, three cultivars (early and late maturity) were planted on seven different dates from May 1 to July 30 in both paddy and upland fields across 2012 and 2013. Soybean yields were significantly different among the planting dates and the cultivars; however, the interaction between cultivar and planting date was not significant. Based on linear regression, the maximum yield of soybean was reached with a June 10 planting date, with a sharp decline in yield for crops planted after this date. The results of this study were consistent with those of a previous one that recommends early and mid-June as the optimum planting period. Regardless of soybean ecotype, a reduction in yield of greater than 20% occurred when soybean was planted after mid-July. Frost during soybean growth can reduce yields, and the late maturity cultivars planted on July 30 were damaged by frost before completing maturation and harvest; however, early maturity cultivars were safely harvested. For sufficient time to develop and reach profitable yields, the planting of soybean before mid-July is recommended.

Phase of Overhead Flooding Damage during Maturation of Rice (벼 성숙기 침관수 피해양상)

  • The typhoon and flooding injury in rice were investigated both the region of downpour at the middle of August and the region of typhoon 'Yanni' at the late of September, 1998 in Kyongbuk provincial of Korea. More than 10% of rice stem were buried by soil inundation, the heading was delayed and the high node tiller was brought out. Yield components were deteriorated and yield was reduced. As the clay in the muddy water was attached on grains of rice, the yield was greatly reduced by reason of low grain filling ratio and polished rice ratio. The injury of muddy water was less at yellow ripening stage than at early ripening stage. On the other hand, the muddy water injury was reduced by the fungicide 'Benomyl' and 'Daconil' treatments. The degree of lodging according to varieties and ripening stages was not made a great difference, while the lodging was mainly occurred in the early transplanting time. In a week after lodging, viviparous spikes were occurred in almost all varieties, the degree of viviparity was shown highly in order of early maturing, middle maturing and late maturing varieties in the field as well as in laboratory test. The viviperous spikes were greatly occurred in the lodging toward hill space within row than row space because the more grains were touched on the ground.

Growth Characteristics and Yield of New Variety Italian Ryegrass (Lolium multiflorum Lam.), "Oasis"

  • The experiment aim to breed an early-maturing variety of Italian ryegrass (Lolium multiflorum Lam.). It was conducted at the Grassland and Forage Crops Division, National Institute of Animal Science, RDA, Cheonan, Korea, from 2022 to 2023. The new variety named "Oasis" is a diploid with green leaves and has a semi-erect growth habit in late fall, and an erect growth habit in early spring. With a heading date of May 10, Oasis is categorized as an early-maturing variety. Compared with the "Florida80" as a control variety, Oasis's flag leaf was 1 mm wider and 2 cm shorter, while the plant length was 7.8 cm longer. Additionally, the ear of Oasis was 1.2 cm longer than that of Florida80, and it showed lodging resistance. The dry matter yield of Oasis (8,805 kg/ha) was higher than that of Florida80. The total digestible nutrient and crude protein contents of Oasis were 64.5%, and 9.3%, respectively, which were 1.1% and 0.6% higher than those of Florida80, respectively. The neutral and acid detergent fiber contents in Oasis were 54.2% and 30.8%, respectively, which was 1.9% and 1.4% lower than those of Florida80, respectively. These results indicate that Oasis has good dry matter yield in the most of region and especially, middle and northern region of South Korea.

  • DNA methylation is involved in tissue-specific gene control and essential for normal embryo development Octamer-binding transcription factor 4 (Oct-4) is one of the most important transcription factors for early differentiation. This study was performed whether the bovine Oct-4 is tissue specific or developmental dependent epigenetic mark, we investigated transcripts and the methylation status of CpGs of 5'-promoter region of Oct-4 in bovine preimplantation embryos. Oct-4 transcripts were highly detected in morula and blastocyst, while they were present low levels in sperm and 2- to 8-cell stage embryos. These results suggest that de novo expression of Oct-4 initiates at morula stage of embryogenesis. Here we determined that there is a tissue-dependent differentially methylated region (T-DMR) in the 5'-promoter region of Oct-4. The methylation status of the Oct-4 T-DMR was distinctively different in the oocyte from that in the sperm and adult somatic tissues and changed from zygote to blastocyst stage, suggesting that active methylation and demethylation occur during preimplantation development. Based on these results, the 5'-promoter region of Oct-4 gene is target for DNA methylation and the methylation status changes variously during embryonic development in bovine.

  • High production of milk and its components are necessary to allow maximal growth of developing piglets. In this study, transgenic pigs were produced containing the $\alpha$-lactalbumin gene, whose product is a potential limiting component in the production of milk. Two lines of transgenic pigs were produced to analyze the effects that overproduction of the milk protein $\alpha$-lactalbumin may have on milk production and piglet growth. Transgenic pigs were produced through microinjection of the bovine $\alpha$-lactalbumin gene. The gene construct contained 2.0 kb of 5 flanking region, the 2.0 kb coding region and 329 bp of 3 flanking region. Sows hemizygous for the transgene produced as much as 0.9 g of bovine $\alpha$-lactalbumin per liter of pig milk. The production of the bovine protein caused approximately a 50 % increase in the total $\alpha$-lactalbumin concentration in pig milk throughout lactation. The concentration of bovine $\alpha$-lactalbumin was highest on day 0 and 5 of lactation and decreased as lactation progressed. The ratio of bovine to porcine $\alpha$-lactalbumin changed during the sow's lactation. This ratio was 4.3 to 1 on day 0 of lactation, but by day 20 of lactation the ratio was 0.43 to 1. This suggested that the bovine transgene and the endogenous porcine gene were under slightly different control mechanisms. The higher level of total $\alpha$-lactalbumin present on day 0 of lactation was correlated with higher lactose percentage on day 0 in transgenic sows (3.8 %) as compared to controls (2.6 %) (P < 0.01). Although there was also a trend for higher lactose percentage in transgenic sows on day 5 and 10 of lactation, no significant differences were observed. These data suggest that $\alpha$-lactalbumin is limiting early in lactation of swine. Furthermore, higher concentrations of $\alpha$-lactalbumin early in lactation may boost milk output.
