Detection Characteristics of TL, ESR and DNA Comet for Irradiated Soybeans (열발광, 전자스핀공명 및 DNA Comet 분석에 의한 대두의 방사선 조사 여부 검지 특성)

  • The detection characteristics of gamma-irradiated $(0{\sim}4\;kGy)$ soybeans produced in Korea and China were investigated by thermoluminescene (TL), electron spin resonance (ESR), and DNA comet assay. The TL glow curves were shown at around $200^{\circ}C$ for irradiated soybeans, while that at $280^{\circ}C$ for the non-irradiated one. The normalization with a re-irradiation step at 1 kGy could verify the above detection results. The Korean soybean showed higher glow curves than Chinese did. The ESR spectroscopy for husks of irradiated soybeans revealed specific signals (g = 2.02374, 1.98715) derived from cellulose radical, which intensities were proportional to irradiation does, with the higher peaks in Chinese sample than Korean one. The DNA comet for the non-irradiated sample showed no or little tails, while those for irradiated samples above 0.5 kGy were remarkably changed in their length, size, and concentration, thus resulting in distinguishing non-irradiated from irradiated samples. As a result, TL, ESR, and DNA comet determinations were found suitable for the detection of irradiated soybean at 0.5 kGy or more, and negligible differences were observed between Korean and Chinese origins in their detection characteristics.

Synthesis, Characterization and ESR Studies of New Copper(II) Complexes of Vicinal Oxime Ligands (Vicinal Oxime 리간드의 새로운 구리(II) 착물에 대한 합성, 특성 및 ESR 연구)

  • Ethoxylacetyl oxime ligands [HL, (1) and $H_2L^1$, (3)] react with copper(II) acetate monohydrate yield octahedral and square planar complexes, respectively. The complexes have been postulated due to elemental analyses, IR, UVVis. spectra, magnetic susceptibility, conductivity and ESR spectra. Molar conductance of the complexes in DMF indicates a non-ionic character. The ESR spectra of [$(L)_2Cu(H_2O)_2$], (2) complex at room temperature and 77K are characteristic of an axial symmetry ($d_{x2-y2}$) with covalent bond character and have a large line width typical of dipolar interactions. However, [$(L^1)Cu$], (4) complex in the solid state showed spectra of marked broadening and loss of hyperfine splitting confirming spinexchange interactions between the copper(II) sites. The spectrum of the doped copper(II) complex at room temperature showed super-hyperfine splitting from coordinated nitrogen atoms and it has an axial type ($d_{x2-y2}$) with covalent bond character and an essentially square-planar arrangement around the copper(II) ion. The spectrum of [$(L^1)Cu$], (4) in frozen methanol at 77K was characteristic of the triplet state of a dimer species and the distance found between the two copper(II) centers was calculated and is equal to 4.8 ${\AA}$.

  • Cu(II) binding mechanisms with water soluble organic fractions (WSOF) extracted from an agricultural soil (W), a soil treated with sludge for 6 years ($WS_6$), a sludge-soil mixture incubated for one week ($WS_1$), and sewage sludge (SS) were studied by electron spin resonance (ESR) spectroscopy and potentiometric titrations. Cu(II)-WSOF complexes produced $g_{11}$ values which were larger than $g_{\perp}$ values, indicating that the coordination of Cu(II) complex was an elongated octahedron. At liquid $N_2$ temperature (77K), the Cu(II)-W complex showed an anisotropic ESR spectrum while the Cu(II)-SS complex showed an isotropic spectrum. These spectral results suggest that the oxygen donor ligands of W may form relatively strong bonds with $Cu^{2+}$ due to extensive chelation while ligands of SS may form little or no chelate bonds with $Cu^{2+}$. The ESR spectra of Cu(II)-SS complex also suggest that each of four in-plane ligands (e.g., $COO^-$, $H_2O$, $Cl^-$, etc.) may act independently as monodentate ligands. Oxygen donor ligands such as aromatic carboxyl groups were probably the major Cu(II) binding sites in W. Sulfonate, aliphatic carboxyl group, and N-containing ligands were probably the major binding sites in SS at pH 5. The Cu(II) complexation with N-containing groups increased as sludge was added to the soil. Much higher (6x) pyridine concentrations were required to displace W from Cu(II)-W complex as compared to the Cu(II)-SS complex.

  • New series of Mn(II), Ni(II), Co(II), Cu(II) and Zn(II) of the 2-(benzothiazol-2-yl)-N'-(2,5-dihydroxybenzylidene) acetohydrazide have been synthesized and characterized by elemental analysis, IR, UV-vis, $^1H$-NMR, mass and ESR spectra, magnetic susceptibility and molar conductivity measurements. The spectral data and magnetic measurements of the complexes indicate that, the geometries are either square planar or octahedral. The biological activity of the ligand and its complexes against fungi (Aspergillus nigar and Fusarium oxysporium) were investigated. The metal complexes exhibited higher activity than both the parent ligand and the corresponding metal ion.

Identification Characteristics of Irradiated Dried Red Pepper during Storage by the Analysis of Electron Spin Resonance and Hydrocarbons (전자스핀공명 및 Hydrocarbon 분석에 의한 방사선처리 건고추의 저장 중 검지특성)

  • Electron spin resonance (ESR) and hydrocarbon characteristics were analyzed to establish identification conditions for irradiated dried red pepper. The ESR spectroscopy for 4 different parts (powder, pericarp, seed, stem) of the samples showed that irradiated samples signaled (g=2.024, 2.006, 1.987) a pair of peaks from a cellulose radical at intervals of 6 mT, which were not found on the non-irradiated samples. The ESR signals increased in directly proportion to the irradiation doses, which were still detectable after 12 weeks of storage at room temperature. The GC-MS analysis of hydrocarbons after fat extraction and separation by florisil column chromatography revealed that hydrocarbons, such as 1-tetradecene (14:1), 1,7,10-hexadecatriene (16:3), 1,7-hexadecadiene (16:2), 1-hexadecene (16:1), 6,9-heptadecadiene (17:2), and 8-heptadecene (17:1), were detected only from the irradiated samples immediately after irradiation and 8 months of storage. They linearly increased with the dose of irradiation, suggesting them as radiation-induced markers for irradiated dried red pepper.

Multistep Identification of γ-Irradiated Boiled-Dried Anchovies by Analysis of Thermoluminescence, Electron Spin Resonance, Hydrocarbon and 2-Alkylcyclobutanone (건멸치의 방사선 조사 확인을 위한 열발광, 전자스핀공명, Hydrocarbon 및 2-Alkylcyclobutanone의 다중분석)

  • Multistep detection was peformed to identify irradiated dried anchovy. In thermoluminescence (TL) analysis, non-irradiated samples represented a lower peak at about 28$0^{\circ}C$, but irradiated samples showed a higher peak at around 20$0^{\circ}C$. The normalization with a re-irradiation step at 1 kGy could verify the identification results. Electron spin resonance (ESR) spectroscopy for bones separated from irradiated anchovy revealed specific signals (g=2.002,1.998) derived from a hydroxyapatite radical, which intensities were in proportion to the irradiation doses and still detectable even after 6 months of storage at -2$0^{\circ}C$. Six kinds of hydrocarbons (HC) were observed in dried anchovy samples and 1,7-hexadecadiene and 1-hexadecene were only detected in irradiated anchovy at 1 kGy or more. Also among 3 kinds of radiation-induced 2-alkylcyclobutanones (2-ACB) observed, 2-dodecylcy-clobutanone and 2-tetradecylcyclobutanone were unique in irradiated anchovy. As a result, the concentration of radiation-induced HCs and 2-ACBs were dependent on the irradiation dose and detectable after 6 months of storage. However, TL and ESR analyses were found simpler than the other methods for identification of irradiated boiled-dried anchovy.

Detection Characteristics of TL, ESR and DNA Comet for Irradiated Peanuts by Origins (TL, ESR및 DNA Comet분석에 의한 원산지별 땅콩의 방사선 조사 검지 특성)

  • Gamma-irradiated peanuts, Korean and Chinese origins, were investigated on detection properties by thermoluminescence (TL), electron spin resonance (ESR), and DNA comet assay (single cell gel electrophoresis). TL measurement showed that the non-irradiated sample revealed a glow curve with low intensity at about 25$0^{\circ}C$, while the irradiated samples showed higher intensity around at 18$0^{\circ}C$. TL ratio (TL$_1$/TL$_2$) of area for TL$_1$ glow curve to TL$_2$ was below 0.05 for the non-irradiated sample and 0.2 or more for the irradiated ones, thus identifying each other. ESR spectroscopy for the irradiated peanuts using outer skin showed negligible signals induced by irradiation, indicating ESR is little applicable to the detection of irradiated peanuts. In DNA comet assay, the non-sample had no or very short tails, whereas the irradiated samples revealed the cells with long tails. Significance in the increase of their lengths depending on irradiation dose (r=0.761/Korean, r=0.768/Chinese) was also found. There was no remarkable difference in detection properties by origins of samples in all determinations, It is concluded that TL analysis or DNA comet assay is suitable for detection of irradiated peanuts and a combined method is recommendable for enhancing the reliability of detection results.

  • The identification characteristics of inadiated dried onion and garlic at 1, 4 and 7 kGy were investigated by analyzing their photostimulated luminescence (PSL), thermoluminescence (TL) and electron spin resonance (ESR). The PSL results showed that the photon counts of non-irradiated dried onion were less than 700 (negative) and those of irradiated samples at 1kGy or more were over 5000 (positive), making it possible to discriminate non-irradiated onions from irradiated ones. However, the intermediate counts (700-5000) were observed in irradiated garlic at 1 and 4 kGy, even though positive counts were measured in 7 kGy group. Radiation-induced TL glow curves $(TL_1)$ were measured at $150^{\circ}C$ range in both irradiated onion and garlic samples at 1 kGy or more, which could identify irradiated groups. Furthermore, determination of TL ratios $(TL_1/TL_2)$ for both samples could verify the above results. It was found, however, that ESR spectroscopy was not suitable for the detection of irradiated dried onion and garlic.

Detection of Gamma-irradiated Imported Foods (Perilla seeds, Almonds, and Soybeans) by Using PSL, TL, and ESR (PSL, TL 및 ESR 분석을 통한 수입산 들깨, 아몬드 및 대두의 방사선 조사 여부 판별 특성)

  • This study examined radiation detection properties of imported perilla seeds, almonds, and soybeans. Food samples were irradiated at doses of 0.5, 1, 2, and 4 kGy and analyzed by photo-stimulated luminescence (PSL), thermoluminescence (TL), and electron spin resonance (ESR). The results of PSL showed that photon counts of irradiated perilla seeds and soybeans were $32,959counts{\cdot}60sec^{-1}$ and $7,234counts{\cdot}60sec^{-1}$, respectively, at an irradiation dose of 0.5 kGy, and the photon count of irradiated almonds was $5,581counts{\cdot}60sec^{-1}$ at an irradiation dose of 1 kGy. The results of TL showed that this technique is useful for detecting irradiated samples; the TL ratios ($TL_1/TL_2$) measured for the food samples at an irradiation dose of 0.5 kGy were 0.2301 for perilla seeds, 0.4595 for almonds, and 0.4827 for soy beans. Lastly, results of ESR spectroscopy for only soybeans revealed specific signals derived from free radicals captured in the cellulose. In conclusion, PSL, TL, and ESR methods can be used for the detection of gamma-irradiated soybean samples, whereas gamma-irradiated perilla seeds and almonds can be confirmed by a serial detection with a TL method after PSL.

  • Three hydroquinone dimethyl ether derivatives have been synthesized and characterized by X-ray diffraction. The electron donating properties were evaluated by using UV-vis spectroscopy, cyclic voltammetry and by ESR spectroscopy. The microcrystalline cation-radical salts of the three donor molecules were also isolated by using antimony pentachloride, a single electron Lewis acid oxidant.
