Antioxidant Activity of Onion (Allium cepa L.) Peel Extracts Obtained as Onion Byproducts (산업체 적용을 위한 양파껍질 추출물의 항산화활성)

  • This study investigated the antioxidant activity of onion peels extracted from onion byproducts by hot water treatment. Hot water extraction of freeze dried onion peel powder was analyzed for total polyphenol content, 2,2'-diphenyl-1-picryllhydrazyl (DPPH), and 2,2'-azino-bis(3-ehtylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging activities, reducing power, oxygen radical absorbance capacity (ORAC). Total polyphenol content was the highest (233.90 mg/g) in onion peel extract mix with ethanol (OPME-1). The DPPH radical scavenging activity ($IC_{50}$), reducing power, and ORAC obtainbed from onion peel extract mix with ethanol precipitation (OPMPE-1) were the highest at 1.15 mg/mL, 1.69 $A_{700}$, and $318,509{\mu}M\;TE/mL$, respectively. The ABTS radical scavenging activity was the highest at 432.78 mg amino acids (AA) eq/g in the OPM. The results of this study suggest that onion peel extracts have marked antioxidant activity, which can have significant health benefits.

Comparative study on growth of leafy vegetables grown in a hybrid BFT-aquaponics using Japanese eel, Anguilla japonica and hydroponics

  • Aquaponics is a cultivation system that combines aquaculture and agricultural hydroponics. This study investigated the productivity of leafy vegetables cultivated in the hybrid biofloc technology-aquaponics (HBFT-AP) using Japanese eel fed two kinds of diets (Mash, commercial powdered feed and extruded pellet [EP]) and hydroponics (HP).The mash was fed to the fish in a type of a dough mixed with water for 6 weeks (Exp1) and switched to the EP containing 2.7% monobasic potassium phosphate (MKP) for subsequent 6 weeks (Exp2). Leafy vegetables of 8 cultivar were employed in the experiment and water quality [dissolved oxygen (DO, mg/L), pH, water temperature (℃), electrical conductivity (EC; ㎲/cm), turbidity (NTU), TAN (NH3 + NH4+) (mg /L), NO2-N (mg/L), NO3-N (mg/L) and PO4-P (mg/L)] was measured 6 times a week. Leafy vegetable productivity (HBFT-AP vs HP) was compared in respective experiment, which was similar or somewhat higher in HBFT-AP. During the 12-week feeding trial, concentrations of nitrite (NO2-N) and phosphorus (PO4-P) were kept low in variability while total ammonia nitrogen (TAN) and NO3-N levels increased with time in HBFT-AP. At the end of two feeding trials, values of weight gain (WG, %), feed efficiency (FE, %), specific growth rate (SGR, %) and protein efficiency ratio (PER) were higher in Exp2 than in Exp1. As well, higher values in hematocrit (PCV, %), plasma K (mEq/L) and inorganic phosphorus (mg/dL) were found (p < 0.05) in Exp2 where fish were fed the EP (EP: 38.60%, 2.80 mEq/L and 7.04 g/dL; Mash: 33.20%, 1.95 mEq/L and 5.50 g/dL). Leafy vegetables in HBFT-AP using Japanese eel fed the EP with MKP 2.7% had a productivity similar (4 kinds of cultivar) or somewhat higher (4 kinds of cultivar) compared to those in HP. Also, Japanese eel fed the EP showed higher values of Pi (m/dL) and K (mEq/L) in plasma compared to those fed commercial powder diet.

Variation and Profile of Paralytic Shellfish Poisoning Toxins in Jinhae Bay, Korea

  • To understand critical aspects of paralytic shellfish poisoning (PSP) in a chief area of bivalve production in Korea, seasonal variation in PSP toxins in bivalves collected from Jinhae Bay, Korea in 2009 was surveyed by the pre-column high-performance liquid chromatography oxidation method. We also confirmed the profiles of major bivalves such as oysters Crassostrea gigas and mussels Mytilus galloprovincialis in Jinhae Bay. PSP toxins in the bivalves showed remarkable seasonal variation. PSP toxin levels were detected from April to May in 2009, and the highest total toxin levels at all stations were recorded in May. The major toxins in bivalves were gonyautoxin [GTX] 1&4 and C 1&2; in oysters GTX 2&3 were also detected as major components. GTX 1&4, which showed the highest PSP toxin levels at each station, accounted for the highest proportions of toxin components in mussels and oysters (64.5-71.3% and 41.4-42.4%, respectively). It was also confirmed that the highest toxicity (in ${\mu}g$ saxitoxin [STX] eq/g) was derived from GTX 1&4. The highest total toxicity (in ${\mu}g$ STX eq/g) was approximately 2-8-fold higher in mussels than in oysters collected from the same station. PSP toxin levels in bivalves differed significantly according to the sample collection station. However, the profiles of toxins in the bivalves did not show significant differences during the survey period according to sample collection station. This study shows that PSP toxin levels in some samples from Jinhae Bay were above the regulatory limit in Korea during a specific period in spring.

A Study on the PCBs-containing Waste Treatment by High Temperature Incineration (고온소각에 의한 PCBs 함유 폐기물처리에 관한 연구)

  • This study is for understanding the domestic possibilities of the high temperature incineration of waste containing Polychlorinated Biphenyls (PCBs) with the analysis of normal operation case and waste gas, fly ash, dioxin about bottom ash, Total-PCBs, Co-PCBs, and the for analysis the heavy metal leaching feature included by bottom ash and fly ash, heavy metal leaching experiment was implemented. The result shows the dioxin density of the waste gas from waste containing PCBs was $0.00699{\sim}0.00763ng-TEQ/Nm^3$, which is lower than $0.0192ng-TEQ/Nm^3$ from the normal operation case. And each Co-PCBs and total PCBs shows $0.00043{\sim}0.00112ng-TEQ/Nm^3$ and $3.06{\sim}3.87ng/m^3$ respectively. The bottom ash test result shows Dioxin 0.00225~0.00630ng-TEQ/g, Co-PCBs 0.00027~0.00082ng-TEQ/g, Total PCBs 0.9~2.6ng/g, and the fly ash shows Dioxin 0.00164~0.00344ng-TEQ/g, Co-PCBs 0.00053~0.00054ng-EQ/g, Total PCBs 0.64~0.84ng/g. The bottom ash and fly ash experiments for heavy metal leaching did not show any leaching but when it comes to the ingredients of the fly ash, Pb elements shows 31.01~237.7ppm, higher than leaching criterion. The analysis of the density of all air pollution material from the waste gas shows the lower value than permissible criterion.

Physicochemical Characteristics and Antioxidant Activities of Mulberries by Greenhouse and Open Field Cultivation in Maturation Degrees (시설 및 노지 재배한 오디의 숙기별 이화학적 품질 및 항산화 활성)

  • This study was conducted in order to compare changes in the quality characteristics and antioxidant activity of mulberries depending on the degree of ripeness between greenhouse (GH) and open field (OF) cultivation. The mulberries were divided into five degrees of ripeness. Quality characteristics such as pH, acidity, antioxidant activity, and contents of free sugar, organic acid, polyphenols, flavonoids, and anthocyanins were investigated. pH level increased slightly while acidity decreased depending on the degree of ripeness. Fructose and glucose were detected in mulberries as the major free sugar. Fructose and glucose increased rapidly during ripening. Citric acid was major acid and decreased considerably during ripening. Contents of total polyphenols, flavonoids, and anthocyanins increased considerably in the last stage of ripening. Contents of polyphenols, and flavonoids in mulberry from OF were higher than from GH. DPPH and ABTS radical scavenging activities increased rapidly in mulberries of the last degree. Contents of functional components were not greatly different between GH and OF, whereas content of free sugar in GH was higher than that in OF.

In vitro Antioxidant and Anti-inflammatory Effects of Ethanol Extracts from Sprout of Evening Primrose (Oenothera laciniata) and Gooseberry (Actinidia arguta) (달맞이순과 다래순 에탄올 추출물의 in vitro 항산화효과 및 항염증효과)

  • To investigate the biological benefits of Korean traditional vegetables, anti-oxidative and anti-inflammatory effects of ethanol extracts from blanched and dried sprouts of evening primrose (Oenothera laciniata, OL) and gooseberry (Actinidia arguta, AA) were measured. Total polyphenol and flavonoid contents of OL were higher than those of AA; OL contained 60.4 mg tannic acid/g dry weight and 31.9 mg rutin/g dry weight, while AA contained 33.0 mg tannic acid/g dry weight and 20.3 mg rutin/g dry weight. The $IC_{50}$ value for DPPH radical scavenging activity was $58.2{\mu}g/mL$ for OL ethanol extract and $122.1{\mu}g/mL$ for AA ethanol extract. The reducing power upon $500{\mu}g/mL$ of ethanol extract treatment was as strong as $52.1{\mu}g$ ascorbate eq./mL for OL and $45.3{\mu}g$ ascorbate eq./mL for AA. Regarding anti-inflammatory effects, inhibition rate against 5-lipoxygenase (LOX) and cyclooxygenase (COX)-2 activities were 29.5% and 79.5% for OL, as well as 11.5% and 39.1% for AA, respectively at a concentration of $250{\mu}g/mL$. Lipopolysaccaride ($1{\mu}g/mL$)-treated RAW 264.7 macrophage cells subjected to OL ethanol extract at various concentrations ($0{\sim}25{\mu}g/mL$) showed significantly reduced synthesis of nitrite oxide (NO), prostaglandin (PG) E2, and IL-6 in a dose-dependent manner without cytotoxicity, although TNF-${\alpha}$ synthesis was not affected. In conclusion, both OL and AA sprouts showed strong antioxidative activity, whereas OL showed very strong anti-inflammatory activity via effective reduction of NO, PGE2, and IL-6 synthesis in LPS-activated macrophage cells.

Effects of High and Low Sodium Diet on Plasma Amino Acid Levels in Korean Adult Women (고나트륨과 저나트륨 식이시 성인 여성의 혈중 아미노산 농도변화에 관한 연구)

  • This study was performed to determine the effect of dietary Na levels on plasma amino acid levels. Plasma amino acid levels were compared in 20 adult healthy women subjects who were given high Na diet (290.48 mEq/day : NaCl 17g) or low Na diet (51.26 mEq/day : NaCl 3g) for subsequent 6 days. Plasma essential amino acids levels were significantly decreased (36%) while plasma non-essential amino acids levels were significantly increased (22%) when subjects were given low Na diet (p<0.001). Among essential amino acids, threonine was decreased (74%) significantly when subjects were given low Na diet (p<0.01). Among nonessential amino acids, serine (49%), proline (20%) and aspartic acid (14%) were increased (p<0.01), while arginine (48%) and glutamic acid (27%) were decreased (p<0.001). In conclusion, dietary Na contents seemed to be an important factor to affect plasma amino acid levels. It would be appropriate to decrease the dietary Na intakes level considering the various clinical effects of dietary Na on the body fluid. For the patients who need low Na diet, it would be suggested that the level of dietary proteins should be carefully considered along with dietary Na manipulation. Physicochemical Characteristics and Antioxidant Activities of Deoduck (Codonopsis lanceolata) with Different Aging Temperatures and Periods

  • To assess a potential possibility of Deoduck as functional food resources, this study was performed to determine the changes in chemical components and antioxidant activities on Deoduck with various aging conditions; aging temperatures were 60, 70, and $80^{\circ}C$, and aging periods were 5, 10, 15, 30, and 50 days. We determined pH, total acidity, browning index, 5-hydroxymethyl-furfural, total phenolic contents, DPPH and ABTS radical scavenging activities of aged Deoduck. Total acidity of aged samples increased during aging treatment, at higher temperature and longer time. The pH value of aged Deoduck ranged from 4.97 to 3.76. Aged Deoduck at $60^{\circ}C$ decreased slowly than 70 and $80^{\circ}C$, and these results were similar in total acidity. 5-HMF and total phenolic contents increased when increased aging temperature and periods. The DPPH and ABTS radical scavenging activities of Deoduck were ranged from 0.374 to 1.560 mg TEAC eq/g and from 0.302 to 1.745 mg trolox eq/g, respectively.

Reduction of Perchlorate and Nitrate by Citrobacter Amalonaticus Strain JB101 : Kinetics and the Applicability of MBR (Citrobacter Amalonaticus Strain JB101에 의한 과염소산염과 질산염의 환원 : Kinetics 및 MBR을 이용한 처리 가능성)

  • This study was performed to evaluate the characteristics of the competition between two electron acceptors, perchlorate and nitrate, with Citrobacter Amalonaticus strain JB101. In addition, the applicability of membrane bioreactor(MBR) for perchlorate removal was evaluated. The maximum growth rate of strain JB101 on perchlorate and nitrate are 0.27 and 0.58 $hr^{-1}$, and maximum substrate utilization rates were 35.1 mg $ClO_4^-/g$ protein-day and 45.6 mg $NO_3^-/g$ protein-day, respectively. Nitrate was a competitive inhibitor for perchlorate, and strain JB101 prefer nitrate to perchlorate as electron acceptor. Complete removal of perchlorate could be achieved up to the surface leading rate of 4.6 g $ClO_4^-/m^2-day$ with the MBR fed with 20 mg $ClO_4^-/L$(HCMBR). When 5 mg/L of nitrate was added to the same influent, perchlorate removal efficiency decreased to 96.5%, while nitrate was completely removed. For the MBR fed with 0.7 mg/L of perchlorate (LCMBR), the maximum perchlorate removal efficiency was 100% up to the loading rate of 0.23 g $ClO_4^-/m^2-day$. Membrane fouling was found to be a problem at high leading rate for both MBRs. The acetate consumption ratio per perchlorate was $13.7{\sim}51.7\;e^-eq./e^-eq.$ in LCMBR, while the value was $2.5{\sim}3.6\;e^-eq./e^-eq.$ in HCMBR. This difference could be related to the acetate consumption with oxygen as electron acceptor. Therefore, the amount of acetate addition must be determined considering the concentrations of other electron acceptors in the influent.

Effects of Histamine on Hemodynamic Parameters and EKG in Dogs (히스타민이 혈류역학 및 심전도에 미치는 영향에 관한 연구)

  • The effects of histamine on cardiovascular system in 6 dogs were analyzed. Mongrel dogs, 10 to 16 kg in body weight, were anesthetized with Nembutal (30 mg/kg) and arterial blood pressure, heart rate, central venous pressure, electrocardiogram were recorded and measured plasma potassium concentration. Histamine $(100{\mu}g/ml)$ was infused slowly at the rate of 0.25 ml/min through the external jugular vein until BP was 80/60 and maintained restored BP for more than 5 min. The process repeated $4{\sim}5$ times. At each time before and after infusion every items were recorded and measured. 1. Arterial blood pressure was 142/105 (mean 117) mmHg in control and decreased to 90/60 68) after histamine infusion. 2. Heart rate changed from 175 beat/min to 150 and central venous pressure from 6.2 to 5.2 cm $H_2O$. 3. Plasma potassium concentration was 4.3 mEq/L and slightly increased to 4.7 mEq/L but it was not significant statistically. 4. Most characteristic changes revealed in EKG especially in T-waves. Height, Width, Steepness, and Slimness were increased $1.5{\sim}3.7$ times than control level and Pointedness decresed 0.5 times than before.
