EDTA-Enhanced Electrokinetic Removal of Cu and Zn from Contaminated Sandy Soil (동전기 기술과 세척제 EDTA를 이용한 모래 토양으로부터 구리 및 아연의 제거)

  • EDTA-enhanced electrokinetic removal of copper and zinc from contaminated sandy soil was carried out. In desorption equilibrium tests, the required mass ratio of EDTA to metal was 10:1 to obtain over 90% of desorption from soil. The removal of heavy metals with chelating agent EDTA below pH 3 was limited because of EDTA precipitation. In electrokinetic experiments, the pH control at anode chamber was essential and 38% Cu and 56% Zn were removed under 30 mA for 1.5 days. Heavy metal removal was greatly improved by controlling anode and soil pH with circulation of anolyte with NaOH solution, in which >50% heavy metal was removed for 4 days and >70% for 9 days.

  • The purpose of this study was to observe the effect of cleansing action of irrigation solutions which was 3% $H_2O_2$ and 5% NaOCl, and 15% EDTA solution on the root canal wall. After treatment with the irrigant, each sample was dehydrated, and coated with 200~250${\circ}$A of gold, and observations were made with the use of scanning electron microscope. The results were as follows: 1. In the root canal walls irrigated with 3% $H_2O_2$ and 5% NaOCl solution without instrumentation after extirpation through barbed broach, the predentin of root canal wall was found scarely affected, and the wall was shown retaining network structure and fibrous organic matters. 2. When 15% EDTA was applied as irrigants for 60, 90 and 120 seconds after instrumentation, there was no signigicant difference of the cleansing effect of the elapsed times which were 90 and 120 seconds on the root canal wall, but in the applied time which was 120 seconds, the canal wall was the cleanest. Therefore it was thought that the most suitable application time of 15% EDTA as the irigants was 120 seconds.

Effect of dentin treatment on proliferation and differentiation of human dental pulp stem cells

  • Objectives: Sodium hypochlorite (NaOCl) is an excellent bactericidal agent, but it is detrimental to stem cell survival, whereas intracanal medicaments such as calcium hydroxide ($Ca[OH]_2$) promote the survival and proliferation of stem cells. This study evaluated the effect of sequential NaOCl and $Ca(OH)_2$ application on the attachment and differentiation of dental pulp stem cells (DPSCs). Materials and Methods: DPSCs were obtained from human third molars. All dentin specimens were treated with 5.25% NaOCl for 30 min. DPSCs were seeded on the dentin specimens and processed with additional 1 mg/mL $Ca(OH)_2$, 17% ethylenediaminetetraacetic acid (EDTA) treatment, file instrumentation, or a combination of these methods. After 7 day of culture, we examined DPSC morphology using scanning electron microscopy and determined the cell survival rate with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. We measured cell adhesion gene expression levels after 4 day of culture and odontogenic differentiation gene expression levels after 4 wk using quantitative real-time polymerase chain reaction. Results: DPSCs did not attach to the dentin in the NaOCl-treated group. The gene expression levels of fibronectin-1 and secreted phosphoprotein-1 gene in both the $Ca(OH)_2$- and the EDTA-treated groups were significantly higher than those in the other groups. All $Ca(OH)_2$-treated groups showed higher expression levels of dentin matrix protein-1 than that of the control. The dentin sialophosphoprotein level was significantly higher in the groups treated with both $Ca(OH)_2$ and EDTA. Conclusions: The application of $Ca(OH)_2$ and additional treatment such as EDTA or instrumentation promoted the attachment and differentiation of DPSCs after NaOCl treatment.

  • It was found that cupric ion selective electrode, which was prepared by mixing CuS and $Ag_2S$ with the ratio four to one and PVC, was hard and durable. The response potentials were reproducible and linear in the range from 1.0 ${\times}$ $10^{-1}M$ to 1.0 ${\times}$ $10^{-5}M$ copper (II) solution and its slope was 25.0 mV per decade concentration at $298^{\circ}K$, slightly different from Nernstian slope. The copper (II) indicating electrode was applied in precipitation titration of 1.0 ${\times}$ $10^{-2} M Cu(II)$ sample solution containing proper amounts $NaNO_3$ with 0.1 M NaOH standard solution. Also, this electrode could be used in complex titration of Zn(II), Mg(II), Ca(II) with EDTA and stability constant of EDTA complex of Ca(II) and Mg(II) was calculated by using known Cu-$EDTA^{2-}$ stability constant.

Effects of Dietary Fiber from Mozuku (Cladosiphon novae-caledoniae kylin) Residue on Antioxidant Activity and Anticancer in HT-29 Human Colon Cancer Cells according to Extraction Condition (추출법에 따른 큰실말(Cladosiphon novae-caledoniae kylin) 부산물 식이섬유의 항산화 효과 및 HT-29 대장암 세포에 대한 항암 작용)

  • In this study, the dietary fiber contents of Mozuku (Cladosiphon novae-caledoniae kylin) residue and the extraction condition (HCl, $H_2SO_4$, NaOH, $Na_2CO_3$, $Na_2EDTA$) of the dietary fiber was investigated. We examined that the contents of the total polyphenols and flavonoids in the dietary fiber from Mozuku residue, and the potent anti-cancer effect was also tested through the growth inhibition in human colon cancer cells (HT-29) in vitro. It was effective to extract soluble dietary fiber with 1.5% $Na_2EDTA$ and 0.05 N HCl in Mozuku residue. The extraction time and temperature affected the yields of soluble dietary fiber. The contents of the total polyphenols and flavonoids in the dietary fiber from Mozuku residue were the highest in 1% NaOH extract (Total polyphenols $34.4{\pm}0.055$ mg gallic acid/g dry basis, total flavonoids $34.7{\pm}0.023$ mg naringin/g extract dry basis). In DPPH radical scavenging activity, 1% NaOH extract showed the most potent antioxidant activity. In the result of viability in human colon cancer cells, growth inhibition was observed in D.W., 0.05 N HCl, and 0.5% $Na_2CO_3$ extracts in a dose-dependent manner. These results demonstrated that soluble dietary fiber from Mozuku residue significant antioxidant activity and anticancer in human colon cancer.

Iron Nutritional Status by ALAD Genotype and Intervention Study for Rural Area Residents (농촌 여성들의 ALAD 유전형질별 철분영양상태와 철분제 섭취에 따른 영양상태의 변화)

  • Previous studies have suggested that delta-aminolevulinic acid dehydratase (ALAD) phenotype differently affect mineral metabolism. The objective of this study was to determine the effectiveness of 6-month iron supplementation as syrup of NaFeEDTA in improvement of iron status according to ALAD genotype. One hundred thirty adult women living in rural areas of Asan were provided NaFeEDTA syrup once a week for 6 months at the dose of 64mg Fe/week. Three hundred control subjects were observed during the study period. Fasting blood was obtained for analyzing hemoglobin (Hb) and zinc protophorphyrin (ZPP) and serum was analyzed for ferritin, iron and total iron capacity (TIBC) levels before and after iron supplementation. Ninety percent of ALAD 1-1 (ALAD1) and 10% of ALAD 1-2 (ALAD2) genotype were observed in the control group. However in the intervention group, 98% showed ALAD1 while only 2% was ALAD2, which is significantly lower proportions of ALAD2 compared to the control group (p<0.01). The iron status of Intervention group significantly improved except for ferritin and TIBC regardless or ALAD genotype, while the control group did not show any changes in iron status except for ZPP. ZPP concentration of the control group significantly increased in both ALAD1 and 2 while the intervention group showed significantly decreased ZPP after supplementation in ALAD1. Iron supplementation in the form of NaFeEDTA seems to be effective in reduction of ZPP levels although ALAD2 did not show significant changes due to the small number. However, it is difficult to make a conclusion from these results, and more specified further investigation is needed with more participants.

Effect of EDTA on the In Vitro Development of Parthenogenetic Mouse Eggs (EDTA가 생쥐 단위발생란의 체외 발달에 미치는 영향)

  • To investigate the effect of EDTA on the in vitro development of parthenogenetic eggs of ICR strain mice, those were cultured in 35mm culture dishes containing NaHCO3-BMOC-3 medium supplemented with 10, 50, 100, or 500$\mu$M of EDTA at 37$^{\circ}C$ for 96hrs. under the atmosphere of 5% CO2 and 95% air. EDTA supplementation of 10, 50, or 100$\mu$M to medium significantly(P<0.01) increase morula and blastocyst formation rate compared with controls in haploid(19.8, 25.9, 39.0% vs. 0.0%). And compared with 10, or 50$\mu$M of EDTA supplementation, significantly(P<0.01) higher morula and blastocyst formation rate resulted from EDTA supplementatin of 100$\mu$M. Both the nuclear number and diameter of blastocysts developed from parthenogenetic eggs were not affected by the morphological types when they were cultured, or the supplementary concentrations of EDTA. The nuclear number of blastocysts developed from haploid, diploid, and immediately cleavaged eggs was 44.8$\pm$1.2, 45.2$\pm$1.5, and 45.4$\pm$1.8, respectively. And the diameter of those eggs ranged 104.4$\pm$1.8, 104.3$\pm$1.2, and 103.8 1.3$\mu$m, respectively.

  • Reactions of $(Et_4N)_2[MoOCl_5]$ with multidentate ligands containing nitrogen or/and oxygen donor atom (EDTA, DTPA, IDA, CyDTA, OX) produce a series of binuclear molybdate (V) complexes. The prepared Mo (V) complexes has been identified by Elemental Analysis, Infrared Spectra, Proton Magnetic Resonance Spectra, and Electronic Spectra. The electrochemical reduction mechanism has been studied by Cyclic voltammetry, Controlled Potential Coulometry, and Spectrophotometry in pH 3.571-10.375 acetate, borate, phosphate/sodium hydroxide, phosphate, ammonium/ammonia buffers. The cyclic voltammogram of the Mo-EDTA, DTPA, IDA, CyDTA complexes at pH < ca. 6.00 have shown two reduction waves. The first reduction wave shows two electron process and the second reduction wave shows two electron process. The cyclic voltammogram of the Mo-EDTA, DTPA, IDA, CyDTA complexes at pH < ca. 8.00 has shown one reduction wave. This reduction wave show four electron process. The cyclic voltammogram of the Mo-OX complex at pH < ca. 7.2 has shown one reduction wave. This reduction wave show four electron process.

COMPARISON OF ANTIBACTERIAL EFFECT OF Listerine(R) WITH VARIOUS ROOT CANAL IRRIGANTS (수종의 근관세척액과 Listerine(R)의 항균성 비교 연구)

  • The purpose of this study is to compare the antibacterial effect of $Listerine^{(R)}$ on two microorganisms (P. gingivalis and E. faecalis) with various root canal irrigants (NaOCl, CHX, EDTA) and to identify possibility of using $Listerine^{(R)}$ as a root canal irrigant. Porphyromonas gingivalis ATCC 3327 and Enterococcus faecalis ATCC 29212 were used in this experiment. For the test irrigants, 0.5%, 1%, 2.5%, 5.25% NaOCl, 0.1%, 0.2%, 1%, 2% CHX, 0.5M EDTA (18.6% EDTA) and $Listerine^{(R)}$ were prepared. Distiled water was used as control. Two methods-1) Comparison of turbidity in broth and 2) Agar diffusion test-were used to determine the extent of antibacterial effect of $Listerine^{(R)}$ and to compare it with that of NaOCl, CHX, and EDTA. All solutions tested were effective against two bacterial strains compared with control (p < 0.001). Any concentration of NaOCl, CHX, and EDTA showed similarly high effectiveness against all bacterial strains. In all experiment, $Listerine^{(R)}$ showed significantly low antibacterial effect compared with the other root canal irrigants (p < 0.05). In conclusion, the results reflect remarkably low antibacterial effect of $Listerine^{(R)}$ as compared with root canal irrigants in general so it is not suitable for the root canal irrigant.

Chemical Cleaning of Copper Corrosion Product Using EDTA.2Na Salt and Effect of Surface Treatment by NALCO-39L (EDTA.2Na를 이용한 구리 부식생성물의 화학세정 및 NALCO-39L에 의한 표면처리효과)

  • This study was carried out to investigate the effect of chemical cleaning of corrosion product on cooling system made of copper as a basic material and using cooling water as pure water. We studied chemical cleaning condition that minimizes the influence on basic material by means of EDTA solution so as to eliminate the slurry in cooling system. In addition, the proper amount of NALCO-39L (Nitrite-Borate-BZT mixture) as a inhibitor was determined in order to protect the copper in cooling system against corrosion after chemical cleaning and the effect of corrosion resistance on the copper surface treated was excelent in comparison with surface untreated. As a result, we found that the main components of sludge in cooling system produced by corrosion of copper were $Cu_2O$, CuO, Cu, and Fe. The optimum condition of chemical cleaning was 400ppm EDTA solution at $60^{\circ}C$. Inhibitor concentration needed to treat the surface of pure copper was 15~20ppm per unit area and corrosion rate of copper treated with 500ppm inhibitor solution for 72 hrs at $60^{\circ}C$ was remarkably decreased as compared with that of pure copper.
