  • Pseudothrombocytopenia is usually associated with anticoagulant ethylene diaminetetraacetic acid (EDTA). The platelet clumping that occurs in EDTA-dependent pseudothrombocytopenia (EDPT) can sometimes be prevented by the use of other anticoagulants such as heparin or sodium citrate. As an alternative, we used kanamycin before or after the withdrawal of EDTA-anticoagulated blood in a 6-year-old boy with EDPT. Kanamycin used supplementarily during the differentiation of EDPT effectively prevented platelet clumping.

PDI-like Enzyme in Human Follicular Fluid Converts 72 kDa Gelatinase into GA110 (사람 난포액에 존재하는 72 kDa Geletinase로부터 GA110을 만드는 PDI-like PDI-like Enzyme)

  • Previously, we discovered a new MMP-2 isoform GA110, of which appearance in human follicular fluid(FF) and serum was increased by EDTA. The present study was conducted to investigate how GAI 10 can appear by EDTA. To examine possible involvement of protein disulfide isomerase(PDI), an enzyme responsible for the dimerization of protein via disulfide formation, effect of PDI inhibitor on the appearance of GA110 by EDTA was investigated. When PDI inhibitor added to FF before EDTA treatment, the gelatinolytic activity of GA110 was abolished in a concentration dependent manner. By contrast, the activity of 72 kDa gelatinase increased. However, the PDI inhibitor added to FF after EDTA treatment, the gelatinolytic activity of GA110 was unaffected. To find out the nature of the enzyme which converts 72 kDa gelatinase into GAI 10, chromatographic separation method of FF proteins was done. Using hydroxyapatite column, fractions rich in 72 kDa gelatinase were isolated and pooled. By using this pool as substrate for the 72 kDa converting enzyme, protein fractions containing the converting activity were obtained from chromatographic separation of FF onto glutathione sepharose fast flow column. When immunoblotting was performed on this enzymatically active protein fractions against polyclonal anti-PDI antibody, distinct immunoreactivity was observed, although appeared in smaller molecular weight region. Based on these observations, it is suggested that the appearance of GAI 10 in FF by EDTA treatment could be due to an activation of PDI-like enzyme, which dimerizes 72 kDa gelatinase into GAI 10 via the formation of disulfide bond between molecules.

  • The cooked surf clam meat was treated with BHA or EDTA, and then dehydrated by means of hot-air dehydration or sun drying. The dehydrated products were packed in glass bottle and stored for three months in the dark place. After three month storage, the pigment loss and water absorbing capacity of dehydrated products were compared with those of directly after dehydration. Both of the sun dried and hot-air dehydrated surf clam products showed a great deal of pigment loss during storage. The BHA treatment prior to dehydration of surf clam meat had good effects on the pigment retention, and the EDTA treatment had a weak effect on it during storage. The rate of rehydration of dehydrated surf clam products had markedly decreased during storage. And it could be seen that there was little difference among the rate of rehydration of the BHA or EDTA treated product and untreated one during storage.

  • We investigated the physicochemical and functinal properties of rapeseed(Brassica napus var. Youngsan) protein prepared by combining various soluvent and purification procedures, such as ultrafiltration (UF) concentration and acid-washin. The lightness value(YCIE) of each protein was greadully improved and its hydrophobicity increased by the degree of purification. The analysis of each protein by sodium dodecyl sulfate-plyacrylamide gel electrophoresis (SDS-PAGE) had nine bands revealed without difference, the considerable portion of which were of $1.96~1.59{\times}10^4$ dalton molecular weight. The content of amino acid increased a little more in the other processed proteins than in the control, and decreased considerably in the proteins extracted by the mixed solvent of 1% sodium hexametaphosphate(SHMP) and 0.25M ethlene diamine tetraacetic acid (EDTA). The better proteins were purified, the lower the kinematic viscosities were in their values. The water absorption and foaming properties were scarcely different according to the processes. The oil absorption and the emulsion activity index normally increased according to the degree of purification. The properties of heat coagulation revealed high values only in the proteins processed by EDTA while they showed considerably low values the other proteins.

  • Wool has excellent properties, such as heat retention, absorbency, and elasticity, but it has a disadvantage in washability because the fabric will felt and shrink greatly. Felting causes the interlocking of the fiber surface scales with one another. Therefore, the studies on wool finishing have been focused on shrink proofing. Precedent researches on wool shrink proofing are mostly on eco-friendly method. using enzyme. The purpose of this study is to examine the effect of L-cysteine, EDTA in papain treatment of wool fabrics. The specific contents of study are as follows. Depending on pH, temperature, treatment time, enzyme concentration and L-cysteine, EDTA concentration, weight loss, tensile strength, whiteness, SEM were examined. Each papain treatment conditions depending on L-cysteine, EDTA were optimized from these properties. Papain had very low activation without activators. The optimum conditions of papain treatment were pH 7.5, temperature $75^{\circ}C$, time 30minutes(L-cysteine), 180minutes(EDTA) and papain concentration 5%(o.w.f.). In the use of papain 5%(o.w.f.), the activators optimum concentration was L-cysteine 2%(o.w.f.), EDTA 7%(o.w.f.)

Effect of Sugarcane Bagasse Soda-AQ Pulp Bleaching Properties by Type of Chelate Compounds and Simultaneous Process of (DQ) Stage (이산화염소 표백단계와 킬레이트 처리단계 동시 진행 시 킬레이트 종류가 사탕수수 Soda-AQ 펄프 표백에 미치는 영향)

  • Pulp made from sugarcane bagasse (SCB) was bleached in element chlorine free (ECF) sequence. The peroxide bleaching process for the final bleaching process has been introduced in order to reduce the use of chlorine dioxide. Prior to peroxide bleaching, different chelating chemicals were applied. When 4.5% of the total chlorine dioxide was used, bleached SCB pulp using additional DTPA chelate stage (DEDQP) resulted in 87.0% of the ISO brightness. However, bleached pulp using simultaneous stage of DTPA chelate and chlorine dioxide (DE(DQ)P) was reached at 83.9% of the ISO brightness. The viscosity of DEDQP bleached pulp was 25.6 cPs, and the one of DE(DQ)P bleached pulp was 21.9 cPs. Decreasing of chelate effect by chlorine dioxide led to a decrease in the final brightness and a lower viscosity. But simultaneous stage of EDTA chelate and chlorine dioxide (DE(DQ)P) led to higher final brightness (87.0% ISO) and higher viscosity (25.8 cPs) than those of the $DEDQ_{EDTA}P$ bleached pulp (86.4% ISO, 25.2 cPs).

Effective Method for Lend Extraction from Pb - Contaminated Soil with EDTA (EDTA를 사용한 납 오염 토양에서의 효율적인 납 추출)

  • The effects of major cations in soils, soil : extractant ratio, and EDTA : lead stoichiometric ratio on the extraction efficiency of lead using EDTA were studied for 4 different actual lead-contaminated soils and one artificially lead-contaminated soil. Extraction of lead from the lead-contaminated soil was not affected by a soil : extractant ratio as low as 1 : 3 but instead was dependent on the quantity of EDTA present. Results of the experiments showed that the extraction efficiency for each soil was different, but if sufficiently large amount of EDTA was applied, all the lead may be extracted except for a soil from lead mining area. The differences in extraction efficiencies nay be due to the major cations present in soils which may compete with lead for active sites on EDTA. The total molar amount of major cations extracted was as muck as 20 times more than the added molar amount of EDTA. For some of the soils tested, the extraction efficiency of lead may be affected by being occluded in the Fe and Mn oxides present in the soil. While major cations present in the soil may be one of the factors affecting lead extraction efficiency, the type of lead species present may also play a role. When these factors affect severely, the using of EDTA to extract lead from lead-contaminated soil might be non-effective method.

Alteration of Physical and chemical Characteristics of Waterlogged Archaeological Woods After Cleaning (세척 후 수침고목재의 물리.화학적 특성 변화)

  • Alteration of physical and chemical characteristics and the effect of removal of mineral substances in waterlogged archaeological woods by different cleaning processes were examined using oak wood(Quercus spp.) that was excavated from wetland near Gwangju, Korea. Cleaning methods employed in the present work were (1) tools, (2) deaeration, (3) EDTA and (4) ultrasonic cleaning, which are being currently applied in the field of preservation treatment. Cleaning process were performed independently or continuously. Composition of mineral substances in the waterlogged archaeological wood was almost same as the that of soil in which waterlogged archaeological woods were buried. In case of independent cleaning, tools cleaning efficiently removed the mineral substances on surface. Surface color become brighter after cleaning with EDTA. In contrast, deaeration and ultrasonic cleaning did not show any significant removal of mineral substances. In continuous cleaning process, tool cleaning as the first step treatment showed the same effect as shown in independent cleaning. Although deaeration as the second step cleaning did not remove the mineral substances, it could be assumed to contribute the infiltration of dimensional agents by homogenization of wood. EDTA treatment (the third step cleaning) removed the iron(Fe) and increased the whiteness of wood color. The ultrasonic treatment (the fourth step cleaning) removed the sodium(Na) remained after EDTA treatment and the fine mineral substances.

Effect of Organic and Inorganic Zinc Sources on Zinc Availability under Flooded Soil Condition (담수토양의 아연 유효도에 미치는 아연비종별 효과)

  • In order to evaluate effectiveness of zinc source on uptake of zinc by rice plant, a pot experiment carried out using ZnEDTA and $ZnSO_4$ in a glasshouse. The results obtained were summarized as follows: The effectiveness of zinc source on uptake of zinc by rice plants was different with different soil. Namely ZnEDTA was more efficient than $ZnSO_4$ in increasing yield of zinc and concentration of zinc in the plant grown on Jecheon calcareous soil. In case of Keumgok acid soil, however, no superiority of ZnEDTA treatment was found. The plant growing status of ZnEDTA treated calcareous soil was also shown that larger number of tillers per hill, bigger height of the plant and good yield of dry matter production than those of the plant grown on $ZnSO_4$ treatment. Regardless of the zinc source, smaller number of tillers per hill, shorter height of the plant, and the lowest yield of dry matter were shown in the treatment of zinc and straw mixed with both soils. It seemed that more applied zinc was to be fixed by the straw application resulting in poor zinc yield. The extent of the zinc fixation showed markedly in Jecheon calcareous soil.
