대전지역 일개 종합병원에서의 비예기 항체 발생현황과 분포 (Frequencies and Distributions of Unexpected Antibodies at a General Hospital in the Daejeon of Korea)

  • 수혈 준비 시 비예기 항체 선별검사를 통한 항체의 유무와 그 종류를 아는 것은 매우 중요하다. DiaMed-ID시스템을 이용하여 대전지역 일개 대학병원에 2016년 1월부터 2017년 12월까지 2년 동안 의뢰된 비예기 항체 선별검사 양성자 중에서 항체동정은 55명에서만 되었고 주로 여성에서 자주 동정되었다. Rh 36예(65.5%), Lewis 7예(12.7%), Kidd 4예(7.3%), Duffy 4예(7.3%), MNSs 3예(5.5%), Rh+Kidd 복합 1예(1.8%)가 동정되었는데, Rh 계열은 Anti-E 19예(34.5%), Anti-E/-c 4예(7.3%), Anti-C/-e 4예(7.3%), $Anti-E/-c/-Jk^b$ 1예(1.8%)가, Lewis 계열은 $Anti-Le^a$ 3예(5.5%), $Anti-Le^b$ 3예(5.5%), Kidd 계열은 $Anti-Jk^a$ 1예(1.8%), $Anti-Jk^b$ 3예(5.5%)가 동정되었다. Duffy와 MNSs 계열은 각각 $Anti-Fy^a$ 1예(1.8%), $Anti-Fy^b$ 3예(5.5%), Anti-M 2예(3.6%), Anti-S 1예(1.8%)가 동정되었다. 최근 대전의 비예기 항체 빈도와 분포가 반영된 이 연구는 효율적인 수혈준비에 도움이 될 것이다.

식중독세균에 대한 솔잎 Ethanol 추출물의 항균작용 (Antibacterial Activity of Ethanol Extract of Pine Needle against Pathogenic Bacteria)

  • 5종류의 식중독세균 (Aeromonas hydrophila, Escherichia coli O157:H7, Listeria monocytogenes, Staphylococcus aureus 196E, Salmonella typhimurium)에 대하여 솔잎 ethanol 추출물의 항균작용을 조사하였다. 솔잎 ehanol 추출물을 0-2% (w/v) 함유한 tryptic soy broth(TSB)에 각 식중독세균을 약 $10^4$-$10^{6}$ CFU/ml 정도 되게 접종하여 35$^{\circ}C$에서 48시간 배양하면서 증식억제 정도를 생균수 변화로서 비교하였다. Gram 양성균인 L. monocytogenes, S. aureus와 Gram 음성균인 A. hydrophila는 솔잎 ehanol 추출물에서는 0.5% 이상의 농도에서 증식이 억제되어 솔잎 ethanol 추출물의 항균활성이 우수하였다. 그러나 Gram 음성균인 E. coli O157:H7과 S. typhimurium은 2%의 솔잎 ethanol 추출물에서 약간 증식이 억제되었으나 대체로 두 균주 모두 솔잎 ethanol 추출물에 대하여 강한 내성을 나타내었다. 솔잎 ethanol 추출물에 의한 증식억제 효과는 S. aureus > A. hydrophila> L. monocytogenes > E. coli O157:H7의 순으로 나타났으며 S. typhimurium에서 가장 강한 내성을 나타내었다.

Detection of Antibiotic Resistance and Resistance Genes in Enterococci Isolated from Sucuk, a Traditional Turkish Dry-Fermented Sausage

  • The aim of this study was to isolate enterococci in Sucuk, a traditional Turkish dry-fermented sausage and to analyze isolates for their biodiversity, antibiotic resistance patterns and the presence of some antibiotic resistance genes. A total of 60 enterococci strains were isolated from 20 sucuk samples manufactured without using a starter culture and they were identified as E. faecium (73.3%), E. faecalis (11.7%), E. hirae (8.3%), E. durans (3.3%), E. mundtii (1.7%) and E. thailandicus (1.7%). Most of the strains were found resistant to rifampin (51.67%) followed by ciprofloxacin (38.33%), nitrofurantoin (33.33%) and erythromycin (21.67%). All strains were found susceptible to ampicillin. Only E. faecium FYE4 and FYE60 strains displayed susceptibility to all antibiotics. Other strains showed different resistance patterns to antibiotics. E. faecalis was found more resistant to antibiotics than other species. Most of the strains (61.7%) displayed resistance from between two and eight antibiotics. The ermB, ermC, gyrA, tetM, tetL and vanA genes were detected in some strains. A lack of correlation between genotypic and phenotypic analysis for some strains was detected. The results of this study indicated that Sucuk manufactured without using a starter culture is a reservoir of multiple antibiotic resistant enterococci. Consequently, Sucuk is a potential reservoir for the transmission of antibiotic resistance genes from animals to humans.

  • Human papillomavirus type 16 (HPV16) has been known as a major causative factor for the development of uterine cervical carcinomas. To investigate the in vivo activity of HPV16 expressed in squamous epithelia, transgenic mice harboring HPV16 E6/E7 with human keratin 14 (hK14) promoter were generated. Grossly, hK14 driven HPV16 E6/E7 transgenic mice exhibited multiple phenotypes, including wrinkled skin that was apparent prior to the appearance of hair in neonates, thickened ears, and loss of hair in adults. 인체폐암세포의 Bcl-2 family 및 cyclooxygenases의 발현에 미치는 해면동물 Sarcotragus sp. 유래 furanoterpenoids의 영향 (Regulation of Bcl-2 Family and Cyclooxygenases by Furanoterpenoids Isolated from a Marine Sponge Swcotragus nt. in Human Lung Cancer A549 Cells.)

  • 본 연구에서 국내 연근해에 서식하는 해면 Sarcotragus sp. (Dictyoceratida속)에서 분리 추출된 7종의 furanoterpenoid계 화합물〔sarcotin A, epi-sarcotin A, ircinin-1, epi-sarcotrine B, sarcotin I, (8E,13Z,20Z) -strobilinin/ (7E,13Z,20Z) -felixinin and (7E,12E,18R,20Z)-variabilin〕의 항암 활성을 비교하기 위하여 A549 인체폐암세포를 대상으로 그들의 세포독성을 조사하였고, 이와 연관된 세포증식 억제 및 apoptosis 유발에 관여할 것으로 예상되는 중요한 유전자 몇 가지의 발현에 미치는 영향을 조사하였다. 조사된 7종의 화합물 모두 처리 농도 의존적으로 A549 폐암세포의 증식을 억제하였는데, 그중 sarcotin A 및 (7E,12E,18R,20Z)-variabilin이 비교적 높은 세포독성을 나타내었다. 이러한 세포증식의 억제는 종양억제 유전자 p53 의존적 또는 비의존적으로 Bcl-2 유전자에 대한 Bax의 발현 증가와 연관된 apoptosis 유발과 관련이 있었으며, epi-sarcotin A, ircinin-1 및 epi-sarcotrine B 처리군에서 이러한 현상은 두드러지게 관찰되었다. 또한 epi-sarcotin A와 ircinin-1은 COX-1의 발현에는 아무런 영향을 미치지 않았으나, COX-2의 발현은 선택적으로 저해하였다. 이러한 결과는 해양 해면동물에서 유래된 furanoterpenoid계 화합물이 선택 적으로 강력한 항암효과를 가질 수 있다는 것을 의미한다.

Improved Detection of Viable Escherichia coli O157:H7 in Milk by Using Reverse Transcriptase-PCR

  • A sensitive reverse transcriptase-PCR (RT-PCR) method to detect viable Escherichia coli O157:H7 in milk was established. The primer sets were designed based on the nucleotide sequences of the rfbE (per) and wbdN genes in the O157 antigen gene cluster of E. coli O157:H7. RT-PCR using five different primer sets yielded DNA with sizes of 655, 518, 450, and 149-bp, respectively. All five of the E. coli O157:H7 strains were detected by RT-PCR, but 11 other bacterial species were not. The sensitivity of RT-PCR was improved by adding yeast tRNA as a carrier to the crude RNA extract. The RT-PCR amplifying the 149-bp DNA fragment was the most sensitive for detecting E. coli O157:H7 and the most refractory to the bactericidal treatments. Heat treatment at $65^{\circ}C$ for 30 min was the least inhibitory of all bactericidal treatments. Treatment with RNase A strongly inhibited the RT-PCR of heated milk but not unheated milk. This study described RT-PCR methods that are specific and sensitive with a detection limit of 10 E. coli O157:H7 cells, and showed that pre-treating milk samples with RNase A improved the specificity to detect viable bacteria by RT-PCR.

소아에서 발생한 대장의 염증성 질환에서 E-cadherin의 발현 (E-cadherin Expression in Colonic Epithelium of Various Colitis in Children)

  • 목 적: 소아에서 다양한 원인에 의해 발생한 대장염에서 점막의 형태학적 변화와 세포 접합에 다양한 변화가 있으리라 예상되어 세포 간의 결합을 유지하는 E-cadherin의 변화를 살펴보았다. 방 법: 1998년 1월부터 2003년 8월까지 부산대학교병원 소아청소년과에서 하부 위장관 내시경술과 대장점막 조직 검사를 통해 대장염으로 진단된 39명을 대상으로 하였다. 파라핀 블록에서 면역조직화학염색법을 이용하여 E-cadherin의 세포 내 발현을 조사하였다. 주변의 정상 조직과 비교하여 E-cadherin의 발현이 동일한 강도와 양상을 가진 세포가 50% 이상인 경우를 정상으로 판정하였고, 발현이 정상인 세포가 50% 미만이거나 염색 분포의 이상이 있거나 전혀 염색되지 않은 경우를 이상으로 판정하였다. 결 과: 1) 본 연구에서 비특이성 대장염 15예(38.5%), 크론병 7예(17.9%), 감염성 대장염 5예(12.8%), 음식 단백 과민성 직결장염 5예(12.8%), 궤양성 대장염 3예(7.7%), Henoch-Schonlein purpura 대장염 2예(5.1%), 그외 베체트병, 허혈성 대장염 1예가 포함되었다. 2) 모든 종류의 대장염에서 상피세포 E-cadherin 발현 감소가 관찰되었으며, 77%의 대상 표본에서 E-cadherin 발현감소가 있었다. 3) 활동성 염증이 심한 부위에서 Ecadherin 발현 감소가 현저하였으며 병변부에서 떨어진 상피세포에서는 정상 발현을, 궤양 주위나 재생 상피가 있는 부위는 심한 발현 감소를 보였다. 결 론: 모든 종류의 염증성 대장 질환에서 E-cadherin 발현 감소가 있었다. 이러한 변화는 염증과 궤양이 있는 부위에서 상피세포 접합을 느슨하게 함으로써 상피세포의 재생을 위한 세포의 이동을 용이하게 하기 위한 작용이라고 판단된다.

유기산이 Escherichia coli O157:H7, Salmonella 및 Listeria monocytogenes의 증식에 미치는 영향 (Inactivation of Escherichia coli O157:H7, Salmonella and Listeria monocytogenes by Organic Acid)

  • The inhibitory effect of the food processing agent on growth of Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes was performed with organic acid, and combination of citric acid, acetic acid, propionic acid and vanillic acid. The minimun inhibitory concentration(MIC) of propionic acid was 5,000 ppm in E. coli O157:H7, 2,500 ppm in Salmonella Enteritidis and Listeria monocytogenes. MIC of citric acid was 10,000 ppm in E. coli O157:H7 and Salmonella Enteritidis, 2,500 ppm in Listeria monocytogenes. MIC of acetic acid was 2,500ppm, while in vanillic acid was 5,000 ppm in Escherichia coli O157:H7, Salmonella Enteritidis, and Listeria monocytogenes. MIC of combined organc acid in E. coli O157:H7 were 2,500ppm in PC, 1,250 ppm in PA, PV, CA, CV and AV. MIC of combined organc acid in Salmonella Enteritidis were 2,500 ppm in PC, PA, PV, CA, and CV, 1,250 ppm in AV. MIC of combined organc acid in Listeria monocytogenes were 1,250 ppm in all treatment group. MIC of combined treatment of three organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes were 1,250 ppm in PCA, PCV, PAV and CAV. The inhibitory effect of organc acid in E. coli O157:H7, S. Enteritidis and L. monocytogenes could be confirmed from the result of this experiment. Therefore, it was expected that the food process would increase or maintain by using organic acid.

발효유제품내에서 Escherichia coli O157:H7과 Salmonella ser. typhimurium의 생존 (Survival of Escherichia coli O157:H7 and Salmonella ser. typhimurium in Fermented Milk Products)

  • Escherichia coli O157:H7 and Salmonella ser. typhimurium are pathogens involved in food poisoning in numerous countries. This study aimed to obtain knowleges on the survival of E. coli O157:H7 KSC 109 and S. ser. typhimurium ATCC 14028 in fermentedmilk products which were on sale in Suwon Yakult supplier. To the final concentration of 103~104 cfu/$m\ell$ of E. coli O157:H7 KSC 109 or S. wer. typhimurium ATCC 14028 in the fermented milks, Metchnikoff, Ace, Yakult, Mastoni and Super 100 were inoculated with these pathogens and then were stored at 4$^{\circ}C$ and viable cells of these pathogens were periodically counted. The results showed that the survival of two pathogens differed in the different types of fermented milks tested. Number of suriviving E. coli O157:H7 KSC 109 and S. ser. typimurium ATCC 14028 cells (initial inoculum, 103~104 cfu/$m\ell$) were decreased to 101, 102 cfu/$m\ell$ in Ace after 100 hours, and were decreased gradually to 101 cfu/$m\ell$ in Yakult after 250 hours. In the other fermented milks, viable cells of E. coli O157:H7 KSC 109 was not drastically decreased but those of S. ser. typhimurium ATCC 14028 was decreased gradually to 102 (Mastoni), and to 101 cfu/$m\ell$ (Super 100) after 250 hours. It appeared that S. ser. typhimurium ATCC 14028 was more susceptible than E. coli O157:H7 KSC 109 at low pH. Vibale cells of E. coli O157:H7 KSC 109 was not drastically decreased in most of fermented milks tested except Ace and Yakult, but in general, S. ser. typhimurium ATCC 14028 was drastically decreased in most of the fermented milks. The major ingibition factor against these pathogens in the fermented milks during storage at 4$^{\circ}C$ appeared to be the acidity and the metabolites produced by the starters bacteria used in fermented milk products.

A Study on the Effect of Online Activation Business Transaction Factors of Fresh Food Shopping Mall on e-Customer Relationship Quality and e-Customer Loyalty

  • Purpose - For the development of fresh food shopping malls, consumers should continue to experience loyalty and favorability for the company's products or brands, and this should lead directly to purchase so that active word-ofmouth and recommendation should be encouraged. Therefore, the purpose of this study is to investigate the effect of e-service quality and e-ERM on e-loyalty with customer satisfaction and commitment as mediators. Research design, data, and methodology - This study was conducted by sample survey method on 320 online customers who have experience in using major online fresh food shopping malls for more than one year. Data analysis methods were frequency analysis, confirmatory factor analysis, reliability analysis, correlation analysis, and structural equation model analysis. Result - Hypothesis 1 through Hypothesis 7 were all supported. The results of this study suggest that e-service quality and e-CRM of online fresh food shopping malls have a significant effect on satisfaction and commitment. Therefore, the conclusion has been derived that the focus of this study, that such satisfaction and commitment have a significant effect on e-customer loyalty. has been supported theoretically and empirically. Conclusion - This study suggests that studies on customer loyalty based on activation commerce factors related to fresh food in online shopping malls will be an index that can reflect on customer's needs corresponding with future trends of not only online shopping malls but also offline shopping malls.
