The strength properties of Light-Weighted Soils(LWS) mixed with Expanded Polystrene(E.P.S) are studied. Test results show that when the initial water contents of dredged soils are under 135% and the cement contents are above 1%, Light-Weight Soils are in the appropriate strength range of 2.0 to 4.0kg/㎠. However, E.P.S contents had a little affects on the strength properties of LWS. In the view of E.P.S diagram's effects on LWS, which have expand ratio of 25, 35, 45 and 60, further studies considering of physical properties of original E.P.S beads are needed.
Tannin 및 p-benzoquinone으로 활성화시킨 다공성 섬유소 입자를 사용하여 대장균 변이주(E. coli K-12 CSH36)로부터 추출한 $\beta$-galactosidase를 고정화시켜 효소학적 성질과 그 응용성에 대하여 연구하였다. $\beta$-galactosidase는 pH 11.0으로 맞춘 tannin과 p-benzoquinone을 사용하여 6시간 동안 활성화시킨 섬유소 입자담체에 고정화 시켰을 때 그 잔여 활성도가 가장 높았다. $\beta$-galactosidase의 최적작용 pH는 5.5였으나 고정화 시켰을 때 pH 6.0으로 변화하였으며 최적작용 온도는 고정화 전이나 후에도 일정하였다. $\beta$-galactosidase의 Km 값은 $4.0{\times}10^(-4)M$으로 나타났으며 고정화 효소의 경우에는 $7.5{\times}10^(-4)M$이었다. 고정화 효소의 재사용성을 검토한 결과, 담체 활성화 물질로 tannin과 p-benzoquinone 용액을 사용하였을 때 20히 사용 후에 초기 효소 활성도의 80% 이상이 유지되는 결과를 나타내었다.
두개봉합부의 조기융합으로 알려진 Craniosynostosis는 두개봉합부 주위 조직들 사이의 조화로운 상호작용이 파괴되었을 때 야기될 수 있다. 흥미롭게도 FGF receptor들, 특히 FGFR2의 point mutation은 여러 가지 형태의 craniosynostosis 증후군과 연관되어 있어, FGFR가 두개봉합부를 포함한 두개골 성장발달과정에 중요한 유전자임을 시사하고 있다. Mouse 두개봉합부의 초기형태발생시 FGFR 유전자들의 기능을 알아보기위해, in situ hybridization 방법을 이용하여 FGFR2(BEK) 및 골아세포분화의 초기표지자인 osteopontin이, 태생기(E15-18)에서 출생후(P1-P3)까지, 두개골의 시상봉합부에서의 발현양상을 조사하였다. FGFR2(BEK)은 osteogenic fronts에 강하게 발현되었으며, osteopontin은 parietal bone의 exo-, endocranial부위에서 발현되었으나, parietal bone의 성장가장자리인 osteogenic front에서는 관찰되지 않았다. 두개봉합부에서의 FGF-mediated FGFR signaling의 역할을 좀더 심도깊게 조사하기위해 E15.5 mouse의 두개골을 이용하여 in vitro 실험을 시행하였다. 흥미롭게도 osteogenic fronts 및 시상봉합부의 간엽조직 중앙에 FGF2-soaked beads를 점적하여 36시간 기관배양한 결과, bead주위 조직들의 두께 및 세포수가 증가되었으며, osteogenic fronts 상에 FGF4 beads를 올려놓은 경우, 시상두개봉합부 중앙에 점적된 FGF4 beads나 BSA control beads에 비해, 골성장이 촉진되어 시상두개봉합부의 부분적인 소멸을 관찰할 수 있었다. 이와 더불어 FGF2 beads는 osteopontin 및 Msx1 유전자의 발현을 유도하였다. 이 결과들을 종합해 볼 때, FGF-mediated FGFR signaling이 발육중인 두개골과 두개봉합부에서 세포의 증식과 분화의 균형을 조절하는데 중요한 역할을 담당하고 있음을 시사해주고 있으며, 이 과정중 FGF signaling이 osteopontin 및 Msx1 유전자의 발현을 조절하므로써 막내골성장 및 두개봉합부의 유지기전에 기여할 것으로 사료된다.
화장품용 방부제인 CPN-Gal은 사람 피부세포에 대하여 보다 안전하다고 알려져있다. 장기간에 걸쳐서 CPN-Gal을 생산하는 공정을 확립하기 위하여, β-gal을 생산하는 재조합 대장균을 alginate bead에 고정화 시켜서, 반복 회분식 반응으로 CPN으로부터 CPN-Gal로의 전환반응(transgalactosylation)을 조사하여 보았다. 이 전환반응은 300 ml flask에서 수행하였으며, 반응액에는 고정화된 대장균, 33.8 mM CPN, 400 g/l lactose가 포함되어있으며, pH와 온도는 7.0과 40℃ 였다. 이 때, galactose 한 분자가 lactose로부터 CPN으로 전달된다. 반복 회분식 전환반응은 4회 연속적으로 192시간 동안 성공적으로 수행이 가능하였고, 이 때, 64%의 전환수율을 보였으며, 이 결과는 선행연구 결과에 비하여 보다 우수한 결과이다. 그런데, 반복 회분식 반응이 진행되면서, 192시간 이후부터는 점진적으로 전환수율의 감소가 관찰되었다. western blotting으로 β-gal의 양을 관찰하여본 결과, β-gal의 양의 감소가 관찰되었으며, alginate beads에서 깨어진 금이 또한 발견되었다. 이러한 bead의 깨어짐 현상과 아울러서 β-gal의 불활성화, 그리고 galactose에 의한 product inhibition 등이 192시간 이후의 전환수율 감소의 원인으로 생각된다. 본 연구의 전환공정은 β-gal의 정제가 필요 없이 진행할 수 있기 때문에, CPN-Gal 합성이 보다 실용적이고, 가격 경쟁력 있게 수행될 수 있을 것으로 생각된다. 그래서, 이러한 전환공정이 CPN-Gal을 상업화하기 위한 공정으로 발전하길 기대하고 있다.

Laccase enzymes from Cerrena unicolor and Trametes versicolor were immobilized on the activated glass beads (CPG), silica gel (SG) and soil (SL). The heterogeneous matrices were activated by ${\gamma}$-aminopropyltriethoxysilane (APTES) and glutaraldehyde (GA), and their surfaces were coated by keratin (KER) on activated or non-activated CPG, SG and SL. The laccase activities were tested in the aqueous solution for the native and immobilized preparations using different pH and temperature conditions. By keratin coating on supports, in the cases of CPG-KER and SL-KER, the immobilization yield was increased from about 80% to 90%. Moreover, much less protein was immobilized in keratin coated matrices than in inorganic ones alone (e.g. on CPG-KER 57.6%, whereas on CPG alone 80.6%). Laccase immobilization on keratin coated inorganic matrices was generally more effective than that of non-coated matrices. Concerned to pH dependency, the optima pH for immobilized laccases generally shifted towards to higher values, 5.5-5.8 and even 5.9 in the case of keratin for C. unicolor and from 5.3 to 5.7 for T. versicolor, respectively, and decreased less gradually both in acidic and alkaline regions. The immobilized laccase was more stable against thermal denaturation. This seems particularly true at $75^{\circ}C$ in the case of C. unicolor, where the activity of immobilized enzyme is > 50% higher than that of the free enzyme. For T. versicolor the respective values were $65^{\circ}C$, and 50%.
Objectives : To determine the DNA sequence of the 18S rRNA gene of the Rehmannia glutinosa and analyze it phylogenetically Methods : Dried root of the Rehmannia glutinosa was ground with a mortar and pestle. Glass beads(0.5 mm in diameter), TE buffer and SDS solution were added to that. The mixture was vortexed vigorously and extracted with the mixture of phenol, chloroform and isoamyl alcohol and with the mixture of the chloroform and isoamyl alcohol. The nucleic acids were precipitated with ethanol and resuspended in TE buffer. Contaminating RNA was digested with RNAse A and the DNA was purified further with the Geneclean Turbo Kit. This DNA was used as a template for amplification of the 18S rRNA gene by PCR. The PCR product was cloned in the pBluescript SK II plasmid by blunt-end ligation and the DNA sequence of the insert was determined. This DNA sequence was analyzed phylogenetically by the BLAST program. Results and Conclusion : Vortexing the ground powder of the dried plant root with glass beads during cell lysis improved recovery of DNA. The DNA sequence of the Rehmannia glutinosa 18S rRNA gene was determined and deposited at the GenBank as the accession number DQ469606. Phylogenetic analysis of that sequence showed the relationship between the members of the family of Scrophulariaceae and also the close relationship of the Buddleja davidii to the members of the Scrophulariaceae family.
p53 유전자의 변화는 인간의 여러 암에서 가장 흔하게 발견되며 종양세포내에서는 이러한 변형된 p53 단백질의 양의 증가가 초래된다. 세포내에 축적된 p53 단백질의 발견은 인간의 암증세를 판단할 유용한 기중이 되기도 한다. 본 연구에서는 이러한 면역조직화학 검사에 쓰일 수 있는 폴리클로날 항체를 만들기 위햐여 사람의 p53 유전자를 glutathione S-transferase 와의 융합 단백질의 형태로서 대장균내에서 발현시켰다. p53 의 아미노산 1-158번을 코딩하고 있는 NeoI fragment 와 아미노산 159-393 번을 코딩하는 NocI-BamHI fragment 를 BamHI linker 를 이용하여 in frame 으로 pGEX-2T 의 BamHI 자리에 삽입하여 재조합 플라스미드 pGTNS 와 pGTNL 을 각각 만들었다. 또 PCR 에 의한 증폭에 의햐여 아미노산 38-145번을 코딩하는 유전자 부위를 증폭하였으며 BamHI 과 PvuII 로 절단하여 pGEX-2T의 BamHI 과 SmaI 자리에 삽입함으로써 pGTBP 를 제조하였다. 이들 재조합 균주들을 IPTG 로 4시간 induction 한 후 세포 추출물로부터 glutathione Sepharose bead 를 이용하여 융합단백질을 분리하였다. Bead 에 결합된 단백질은 10% SDS-polyacrylamide gel 에서 전기영동하였으며, 각각의 분자량은 54 kDa, 53 kDa 와 40 kDa 였다. 이러한 방법으로 1리터 배양으로부터 약 1mg 의 단백질을 정제하였다.
본 연구는 환경샘플 중 병원균을 진단하기 위한 목적을 가진다. 최소 챔버 칩에서 환경 샘플 중 병원균을 농축하고 mRNA를 증폭하여 효과적이고 간단한 진단방법을 고안하였다. PDMS로 면적 $1.5\;cm{\times}\;1.5\;cm$, 높이 $100\;{\mu}L$의 칩을 제작하여 유리에 부착시켰다. RNase에 의한 진단 오류 또는 실패를 막고자 RNase away 처리를 하고, RNA와 PDMS의 결합을 막기 위해 BSA 처리를 하였다. 수질에 있는 병원균은 매우 적은 농도로 존재하므로 농축의 과정이 필요하다. 농축의 방법에는 여러 가지가 있으나 본 연구에서는 유리 비드를 칩 내에 삽입하고 저농도의 시료를 주입함으로서 고농도로 농축을 하는 방법을 사용하였다. 그러나 부피가 작은 칩 내에서 수행하기에는 내부 압력이 작용하여 문제가 발생하여 $100\;{\mu}m$의 유리 비드를 사용하고 유리비드의 칩 내부 이탈을 방지하기 위하여 댐을 만들어 농축에 가장 적합한 칩의 형태를 잡았다. 시료의 주입속도에 따라 내부 압력이 상승하여 댐의 기능이 상실하여 유리 비드가 이탈하게 되므로 그것을 방지하기 위하여 칩 내에 댐을 강화하여 만들고 내부압력 증가가 방지되는 최적의 댐을 개발하여 시료의 주입 속도 5 mL/min까지 유리 비드의 이탈을 막았다. 유리 비드에서의 RNA 농축은 pH 5에서 효과적이고 pH가 증가할수록 유리 비드와 RNA의 결합이 끊어지는 현상을 보였으므로 시료에 pH 5의 버퍼를 첨가하여 농축을 진행하고 중성의 NASBA 용액을 주입하여 유리비드에서 탈착된 농축된 고농도의 RNA를 증폭하였다. NASBA는 항온 수조에서 온도에 변화 없이 $41^{\circ}C$에서 1시간 30분 동안 진행하며 증폭된 mRNA는 직접 확인하였다. 이 방법은 LOC 기술을 적용하여 저농도의 시료를 효과적으로 측정할 수 있도록 편리한 바이오 칩을 개발함으로써 대용량의 샘플 중 극 저농도의 대장균을 효과적으로 검출할 수 있는 장점을 가지고 있다.
The adsorptive removal properties of synthetic A4 zeolite were investigated against a total of 16 offensive odors consisting of reduced sulfur compounds (RSCs), nitrogenous compounds (NCs), volatile fatty acids (VFAs), and phenols/indoles (PnI). Removal of these odors was measured using a laboratory-scale impinger-based adsorption setup containing 25 g of the zeolite bed (flow rate of $100mL\;min^{-1}$). The high est and lowest breakthrough (%) values were shown for PnIs and RSCs, respectively, and the maximum and minimum adsorption capacity (${\mu}g\;g^{-1}$) of the zeolite was observed for the RSCs (range of 0.77-3.4) and PnIs (0.06-0.104), respectively. As a result of sorptive removal by zeolite, a reduction in odor strength, measured as odor intensity (OI), was recorded from the minimum of approximately 0.7 OI units (indole [from 2.4 to 1.6]), skatole [2.2 to 1.4], and p-cresol [5.1 to 4.4]) to the maximum of approximately 4 OI units (methanethiol [11.4 to 7.5], n-valeric acid [10.4 to 6.5], i-butyric acid [7.9 to 4.4], and propionic acid [7.2 to 3.7]). Likewise, when removal was examined in terms of odor activity value (OAV), the extent of reduction was significant (i.e., 1000-fold) in the increasing order of amy acetate, i-butyric acid, phenol, propionic acid, and ammonia.
