Identification of Amino Acid Residues Involved in Feedback Inhibition of the Anthranilate Synthase in Escherichia coli

  • The first step of the branch pathway in tryptophan biosynthesis is catalyzed by anthranilate synthase, which is subjected to feedback inhibition by the end product of the pathway. The $trpE^{FBR}$ gene from a mutant Escherichia coli strain coding for anthranilate synthase that was insensitive to feedback inhibition by tryptophan has been cloned. To identify the amino acid changes involved in the feedback regulation of anthranilate synthase, the nucleotide sequence of the mutant $trpE^{FBR}$ gene was determined. Sequence analysis of the $trpE^{FBR}$ gene revealed that four bases were changed in the structural gene while alteration was not found in the 5' control region. Among these base changes, only two base substitutions caused the alterations in amino acid sequences. From the results of restriction fragment exchange mapping, the 61st nucleotide, C to A substitution, that changed $Pro^{21}{\rightarrow}Ser$ was identified as the cause of the desensitization to feedback inhibition by tryptophan. Additional feedback-resistant enzymes of the E. coli anthranilate synthases were constructed by site-directed mutagenesis to examine the effect of the $Ser^{40}\;{\rightarrow}\;Arg^{40}$ change found in the $trpE^{FBR}$ gene of Brevibacterium lactofermentum. From the feedback inhibition analysis, the $Pro^{21}{\rightarrow}Ser$ and $Ser^{40}{\rightarrow}Arg$ mutants maintained about 50% and 90% of their maximal activities, respectively, even at the extreme concentration of 10 mM tryptophan. From these results, we suggest that the $Pro^{21}$ and $Ser^{40}$ residues are involved in the tryptophan binding in the E. coli enzyme.

  • E. coli growth is inhibited by organic acids produced in the broth. In order to reduce the inhibition, an electrodialysis unit was used. Model solutions (acetic acid plus distilled water or M-9 medium) were tested in the unit for investigating the optimum condition of current and voltage. Electrodialysis cultures were performed with the optimum condition where the highest current efficiency could be attained. The distilled water plus acetic acid gave us a higher current efficiency than the M-9 plus acetic acid. Electrodialysis efficiently removed acetic acid and so enhanced the specific growth rate of E. coli compared with the control experiment without clectrodialysis.

Effect of Inhibition on Glucosultransferase and Antimicrobial Activity of Polyphenol Fraction of Gallnut and Red Grape Husk (오배자와 적포도과피 폴리페놀 분획물의 항균성 및 Glucosyltransferase 저해효과)

  • The antimicrobial and glucosyltransferase(GTase) inhibition activity were searched for 30 species of various folk drugs and by products of food industry. Among them, two species, gallnut and red grape husk water extracts, were selected for the powerful antimicrobial and GTase inhibition activity. The polyphenol fractions of gallnut and red grape husk were showed very greater antimicrobial activity on both Gram(+) and (-), B. subtilis and E. coli. The minimum antimicrobial activity of gallnut polyphenol fraction were 1.0mg for B. subtilis and 3,0mg for E. coli. Red grape husk was 2.0mg for B. subtilis and 3.0mg for E coli. The polyphenol fractions of gall nut and red grape husk were showed powerful GTase inhibition activity. The concentrations of these fractions for 80% inhibition of GTase activity were 1.08$\times$10$\^$-3/mg/㎖ and 1.08$\times$10$\^$-2/mg/㎖, respectively. The most abundant compound in these fraction seems to be polyphenol derivatives. From these results, we think that the gallnut and the red grape husk polyphenol fraction had more antimicrobial and anti-plaque activities than artificial synthetic preservatives as an economic point of view.

Effectiveness of cephalosporins in hydrolysis and inhibition of Staphylococcus aureus and Escherichia coli biofilms

  • Importance: Staphylococcus aureus and Escherichia coli contribute to global health challenges by forming biofilms, a key virulence element implicated in the pathogenesis of several infections. Objective: The study examined the efficacy of various generations of cephalosporins against biofilms developed by pathogenic S. aureus and E. coli. Methods: The development of biofilms by both bacteria was assessed using petri-plate and microplate methods. Biofilm hydrolysis and inhibition were tested using first to fourth generations of cephalosporins, and the effects were analyzed by crystal violet staining and phase contrast microscopy. Results: Both bacterial strains exhibited well-developed biofilms in petri-plate and microplate assays. Cefradine (first generation) showed 76.78% hydrolysis of S. aureus biofilm, while significant hydrolysis (59.86%) of E. coli biofilm was observed by cefipime (fourth generation). Similarly, cefuroxime, cefadroxil, cefepime, and cefradine caused 78.8%, 71.63%, 70.63%, and 70.51% inhibition of the S. aureus biofilms, respectively. In the case of E. coli, maximum biofilm inhibition (66.47%) was again shown by cefepime. All generations of cephalosporins were more effective against S. aureus than E. coli, which was confirmed by phase contrast microscopy. Conclusions and Relevance: Cephalosporins exhibit dual capabilities of hydrolyzing and inhibiting S. aureus and E. coli biofilms. First-generation cephalosporins exhibited the highest inhibitory activity against S. aureus, while the third and fourth generations significantly inhibited E. coli biofilms. This study highlights the importance of tailored antibiotic strategies based on the biofilm characteristics of specific bacterial strains.

  • In order to develop a lactic acid bacterial powder which can be used as a probiotic for human and animal, a lactic acid bacteria which has high resistance against low pH and ox-gall, and shows a good growth inhibition against E. coli, was isolated from an animal intestine and characterized. The isolated strain was identified as Enterococcus faecium. It had more than 90% of survival at low pH for 2 hours and almost 100% of survival in the presence of 0.3% ox-gall. When co-cultured with E. coli in MRS broth, all of the E. coli cells were killed within 24 hours. The final powdered product of the isolated strain was manufactured after a freeze drying process using an industrial media, and then checked its stability. Its storage stability was 80% for 11 months at 18$\circ$C.

  • A modified E. coli trp operon, $trpL({\Delta}att)\;trpE^{FBR}$, was conjugally transfered into Klebsiella pneumoniae $KC_{100}\;(Phe^-,\;Tyr^-,\;Trp^-,\;Rif^r,\;Kam^r)$ by in vivo cloning using the hybrid plasmid $R_{6}K::$ Mucts 61 with a transfer frequency of $5.2{\times}10^{-7}$. Two K. pneumoniae transconjugants, $KUA_{701}\;and\;KUA_{702}$, were isolated. The characters of attenuation control-free and resistance to feedback-inhibition which are characteristics of donor C. coli trp operon were normally expressed in the $KUA_{701}.\;However,\;KUA_{702}$ retained only the feedback-inhibition resistant character. $Trp^+$ phenotype and ampicillin resistant character were completely stable in the transconjugants, but streptomycin resistant character was lost in the transconjugants.

  • Aim of the present study was the development of a bioassay which enables the detection of toxic effects of heavy metal ions to a bacterium, Escherichia coli. Inhibition effects of the metals on growth rates of the bacterium were studied in the absence or presence of fulvic acid. This method does not clearly differentiate among metals, but does detect overall AGB inhibition rate (toxicity) for 5 different heavy metals. The toxicity of the metals in the absence of fulvic acid in the same testing conditions was significantly increased in following order: Hg < Pb, Zn < Cd < Cu, whereas the inhibition rate (toxicity) in the presence of FA was shown to be increased In following order: Cd < Pb, Hg < Cu < Zn. The results of the present study indicate that this simple and fast biomonitoring assay with direct exposure of E coli. might be a valuable supplement to analytical methods of contaminated media.

Studies on the Anti-rotaviral and Anti-bacterial Effects of Phellinus linteus Mushroom Rice (상황미를 이용한 항로타바이러스 효과와 항균성 측정에 관한 연구)

  • This study was conducted to evaluate the inhibitory effects of Phellinus linteus mushroom rice on food-borne pathogens (Staphylococcus aureus 305, Listeria monocytogenes ATCC 19114, Escherichia coli 0157:H7 ATCC 42894, Escherichia coli O55) and human rotavirus (KU, S2, YO, K-21). The results obtained are summarized as follows: The inhibitory effects of Phellinus linteus mushroom rice against food-borne pathogens; inhibition zone diameters for S. aureus 305, L. monocytogenes ATCC 19114 and E. coli 0157:H7 ATCC 42894 were 18mm, 20mm and 13mm, respectively. E. coli O55 did not form an inhibition zone. The inhibitory effects of l/3% Phellinus linteus mushroom rice on MA-104 cells using the MTT assay were, KU $90.52{\pm}18.42%,\;S2\;94.74{\pm}8.68%,\;YO\;59.77{\pm}8.68%$ and $K-21\;97.56{\pm}12.50%$. Phellinus linteus mushroom rice has inhibitory effects on the food-borne pathogens S. aureus 305, L. monocytogenes ATCC 19114 and E. coli 0157:H7 ATCC 42894, and human rotavirus (K-21, S2, YO, KU).

  • The effect of patulin, a mycotoxin, on the growth of Escherichia coli cell was investigated. E. coli cell elongation usually shown in SOS-response for DNA repair was induced by 20 mg of patulin per ml. After staining the E. coli chromosome with fluorescence dye(DAPI, 4', 6-diamino-2-phenyl-indole), chromosomal DNA partitioning was not affected by patulin. The observation indicateds that patulin acts as a DNA damaging agent which is effective for E. coli cell elongation introduced by the inhibition of septum formation.

  • The effect of low concentrations of allspice (Pimenta dioica L.) in culture broth as an antibacterial agent against Escherichia coli 0157:H7 and Staphylococcus aureus 196E was tested at 35,5 and -20$^{\circ}C$. Tryptic soy broth (TSB) containing 0∼2% (w/v) of allspice was inoculated with 10$\^$5/∼10$\^$6/ cells/$m\ell$ of E. coli and S. aureus and incubated at each temperature. The growth of E. coli was not inhibited at 0.1∼1.0% allspice and growth occured at 2% allspice but only after a prolonged lag period. Growth of S. aureus was inhibited with increasing concentration of allspice at 35$^{\circ}C$. Growth of S. aureus occured at the presence of 0.1∼0.3% allspice but the viability of S. aureus at 0.5∼2.0% allspice was decreased during storage at 35$^{\circ}C$. During refrigerated storage at 5$^{\circ}C$, inhibition of E. coli and S. aureus was increased with the progress of time and increasing spice concentration. During frozen storage at -20$^{\circ}C$, antibacterial activity of allspice against E. coli was increased with increasing storage time and spice concentration while that activity against S. aureus was effective during early period of storage. There was no major changes in population of S. aureus in TSB with different concentration of spice frozen at -20$^{\circ}C$. Viable counts of E. coli and S. aureus at 0.l% of allspice was less than that of control during frozen storage.
