생산지 수집 신선 유기농 농산물 미생물 분포도 분석 (Microbial Prevalence and Quality of Organic Farm Produce from Various Production Sites)

  • 신선편이식품 소재인 유기농산물의 미생물 분포와 품질을 평가하기 위해서 풋고추, 상추, 토마토, 사과, 배, 쌀 등의 농산물을 47개 지역에서 관행농 농산물과 동시에 현장에서 시료를 수집하였다. 일반 세균수로는 유기농 고추가 평균 4.07 log CFU/g, 관행농 고추는 3.71 log CFU/g 검출되었고 상추는 유기농, 관행농에서 6.76-6.90 log CFU/g로 분석되었다. 토마토와 사과는 2종류 시료에서 각각 2.08-2.92 log CFU/g, 0.70-0.82 log CFU/g로 검출되었다. 쌀도 유기농과 관행농 시료에서 2.92-2.98 log CFU/g 범위의 세균분포를 보여주었으나 유기농 배에서는 4.48 log CFU/g, 관행농 배는 2.84 log CFU/g의 분포도를 보여 주었다. 분석시료에 따라 미생물의 분포에 많은 차이를 보여 주었으며 유기농과 관행농산물의 미생물분포는 거의 차이가 없었다. 병원성 세균인 Cl. perfringens, E. coli O157:H7, L. monocytogenes, Salmonella, S. aureus은 전혀 검출되지 않았다. 그러나 유기농산물 2개(4%)와 관행농산물 3개(6%)에서 E. coil가 1.7 log CFU/g으로 검출되었고 B. cereus는 유기농 6개(13%)가 1.97 log CFU/g수준으로, 관행농 11개(23%)에서 1.04 log CFU/g 수준으로 검출되었다. 그러므로 유기농산물과 관행농산물에서의 일반세균과 병원성 세균의 오염정도는 차이가 거의 없고 이들 병원성 미생물의 오염수준도 비교적 낮아 안전한 것으로 판단된다.

  • 최근 연구가 활발히 진행되고 있는 RF 대역에서 사용가능한 인덕터를 제조하여 특성을 평가하였다. 인덕턴스와 Q값을 높이기 위하여 5000 두께의 F $e_{78.81}$T $a_{8.47}$ $N_{12.71}$ 연자성박막을 삽입하였으며, 자성박막의 FMR 공진주파수를 인위적으로 확장시키기 위하여 pattern을 형성시켜 shape anisotropy를 증가시켰다. 또한 코일부분은 lift-off process를 이용하여 제조하였다. 제조된 인덕터의 디자인은 4턴의 rectangular spiral형태였으며, 측정된 특성은 Ti/Ag air-core의 경우 5 GHz까가지 공진이 없었으며 2GHz에서 Q값이 9, 인덕턴스 8.4nH였다. 자성박막을 이용한 경우 9 nH에 공진주파수는 약 700 MHz부근이었다.다.

  • The objects of the present study were to establish the method of purification, subunit dissociation of verotoxin-2 (VT2) produced by Escherichia coli O157:H7, and to investigate the characteristics of purified verotoxin-2 such as molecular weight and composition of amino acid. The results were summerized as follows; Verotoxin-2 was extracted by addition of polymyxin B sulfate into bacterial cell lysate prepared from Escherichia coli O157:H7(KSC109). As an initial step, the bacterial cell lysate was precipitated with 30% saturated ammonium sulfate. The precipitated crude toxin was then subjected to anion-exchange, chromatofocusing and cation-exchange chromatography. Using this scheme, we obtained highly purified toxin with a specific activity of $1.1{\times}10^9$ $CD_{50}/mg$. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis(SDS-PAGE) for purified VT2 showed two protein bands. The upper band, approximately 32 Kd, was supposed as A subunit and the lower band, approximately 7.7 Kd, was supposed as B subunit. When the toxin was separated in the subunit-dissociating solution, two peaks emerged with retention times of 15 and 28 min by HPLC. These peaks represented A subunit and B subunit, respectively. The amino acid composition of purified VT2 were made up in order of glutamic acid, histamine, asparaginic acid, histidine, lysine, alanine and leucine etc. The largest amount among the amino acid composing VT2 was methionine.

A Recombinant Human GM-CSF Protein Expressed as an Inclusion form in Escherichia coil Stimulates Colony Formation and Cell Proliferation in vitro

  • Granulocyte-macrophage colony-stimulating factor (GM-CSF) is a pleiotropic hematspoietic growth factor involved in the development of myeloid cells from bone marrow, and an activator of mature myeloid cells functioning in a variety of antimicrobial and inflammatory responses. Recently, recombinant GM-CSF is increasingly under clinical study for treatment of various diseases including cancer, infectious diseases and hematopoietic diseases as well as for an immune response modulator, In this study, we constructed a recombinant human GM-CSF (rhGM-CSF) expression plasmid with a pelB leader sequence and His. Tag under T7 promoter control. The expression construct was shown to produce a recombinant protein of 20 kDa in the 8M urea preparation, indicating the rhGM-CSF may be expressed as an insoluble inclusion form. The 20 kDa recombinant protein in 8M urea was transformed into the water-so1ub1e form by dialysis against PBS buffer (phosphate buffered saline). The soluble rhGM-CSF protein was shown to stimulate colony formation and cell proliferation in vitro, indicating that the rhGM-CSF could be refolded into its native form to show colony stimulating activity.

2,4,6-Trinitrotoluene (TNT)의 광분해 특성 (Photo-decomposition Characteristics of 2,4,6-Trinitrotoluene in a UV/$H_2O_2$ Process)

  • The decomposition of 2,4,6-trinitrotoluene (TNT) and the mass balance of nitrogen (N) species as products were investigated in a UV/H2O2system by varying pH, concentrations of $H_2O_2$, and $O_2$. All experiments were conducted in a semi-batch system employing a 50 mL reaction vessel and a coil-type quartz-tube reactor. In contrast with previous studies employing batch mode, TNT decomposition in the semi-batch mode was proportionally enhanced by increasing $H_2O_2$ concentration to 10 mM (0.034%), indicatingthat an inhibitory effect of excess $H_2O_2$on hydroxyl radical (${\cdot}OH$) can be negligible. N compounds are released as $NO_2^-$ in the early stages of the reaction, but $NO_2^-$ is rapidly oxidized to $NO_3^-$ by means of ${\cdot}OH$. $NH_4^+$ was also detected in this study and showed gradually the increase with increasing reaction time. In this study, $NH_4^+$ production can involve the reduction of nitro group of TNT concurrent with the production of $NO_3^-$. Of the N species originating from TNT decomposition, 12 ~ 72% were inorganic forms (i.e. [$NO_3^-$] + [$NO_2^-$] + [$NH_4^+$]). This result suggests that the large remaining N portions indicate that unidentified N compounds can exist.

VCM의 제어 및 제진 장치 응용에 관한 연구 (Study on the control of VCM and its application to the vibration isolator)

  • 엔진 및 회전기기의 진동은 선체의 피로도를 증가시켜 내구성을 저하시키는 원인이 된다. 이를 방지하기 위하여 엔진 및 회전기기와 선체사이에는 댐퍼를 부착하여 진동을 최소화 하고 있다. 일반적으로 선박에 부착하는 댐퍼는 수동형 댐퍼로 중 고주파의 특정 진동영역에 대하여 제진 특성을 갖도록 설계되어있지만 저주파영역에서는 그 효과를 기대하기 힘들다. 이에 본 연구에서는 보다 광대역의 진동신호에 대해 높은 제진특성을 얻기 위하여 VCM을 이용한 능동형 제진 장치를 개발하였다. 제진장치를 제어하기 위해 라우스휴리츠 안정조건과 한계감도법을 이용하여 PID제어기를 구성하였고, 제어기 출력은 2상한과 4상한 초퍼회로를 이용하여 VCM을 구동할 수 있도록 하였다. 시뮬레이션과 실제 실험을 통하여 제어알고리즘과 제어장치의 유용성을 확인하였다.

와전류시험에 의한 조사핵연료봉의 결함 검사 (The Defect Inspection on the Irradiated Fuel Rod by Eddy Current Test)

  • 핵연료봉의 결함 탐상을 위하여 차동환상형 와전류시험 탐촉자를 설계 및 제작하여, 핵연료봉 시험을 위한 최적 시험 조건을 구하였다. 관통결함, 내부결함, 외부결함을 낸 표준시편에 대하여 와전류시험을 수행함으로써 시험 주파수와 출력 신호의 위상 및 크기에 대한 상관 관계를 구하였다. 이를 토대로 하여 고리원자력 1호기에서 연소한 핵연료봉 G33-N2에 대하여 와전류시험을 수행하여 관통결함 신호를 얻었다. 와전류시험에서 관통결함으로 예측된 위치에서 금속조직시험을 수행하여 얻은 결과는 와전류시험 결과와 잘 일치하였다. 따라서 와전류시험법은 조사핵연료봉의 피복관에 대한 건전성을 평가하는 도구로써 그 신뢰성이 양호함을 실증하였다.

Treatment of Scalp Arteriovenous Malformation

  • Objective : The object of this study is to present the treatment experience of the 6cases of scalp arteriovenous malformations[AVMs] focus on treatment strategy. Methods : Six patients with scalp AVM were treated during past 12years. We analysis the clinical characteristics of the lesions, treatment methods and management outcomes. Results : The lesions were located on temporal in 2patients, parietal in 2patients, frontal and occipital area in each one. Four of six patients had a trauma history on scalp. The presenting symptoms were progressive enlarged pulsating mass with or without bruit. Four of the six lesions had the large fistula in the lesion. Two patients were treated with surgical resection alone, three patients with proximal feeding artery balloon[s] occlusion followed by surgical resection, and one patient with coil embolization through trans-venous route alone. We obtained good results in all patients. Conclusion : Most of scalp AVM can be completely cured by Judicious selection and a combination of treatment modalities, i.e., surgery only, or embolization only, or embolization plus surgical therapy. Although embolization became a primary therapy for this sort of scalp AVM recently, the selection of treatment modality should be chose based on the size, angioarchitecture, and clinical presentations of the lesion.

Physicochemical and Rheological Properties of a Novel Emulsifier, EPS-R, Produced by the Marine Bacterium Hahella chejuensis

  • The rheological properties of an exopolysaccharide, EPS-R, produced by the marine bacterium Hahella chejuensis strain 96CJ 10356 were investigated. The $E_{24}$ of $0.5\%$ EPS-R was $89.2\%$, which was higher than that observed in commercial polysaccharides such as xanthan gum ($67.8\%$), gellan gum ($2.01\%$) or sodium alginate ($1.02\%$). Glucose and galactose are the main Sugars in EPS-R, with a molar ratio of ${\~}1:6.8$, xylose and ribose are minor sugar components. The average molecular mass, as determined by gel filtration chromatography, was $2.2{\times}10^3$ KDa, The intrinsic viscosities of EPS-R were calculated to be 16.5 and 15.9 dL/g using the Huggins and Kraemer equations, respectively, with a 2.3 dL/g overlap. In terms of rigidity, the conformation of EPS-R was similar to that of caboxymethyl cellulose ($5.0{\times}10^{-2}$). The rheological behavior of EPS-R dispersion indicated that the formation of a structure intermediate between that of a random-coil polysaccharide and a weak gel. The aqueous dispersion of EPS-R at concentrations ranging from 0.25 to $1.0\%$ (w/w) showed a marked shear-thinning property in accordance with Power-law behavior. In aqueous dispersions of $1.0\%$ EPS-R, the consistency index (K) and flow behavior index (n) were 1,410 and 0.73, respectively. EPS-R was Stable to pH and salts.

Expression, Purification and Properties of Shikimate Dehydrogenase from Mycobacterium Tuberculosis

  • Tuberculosis, caused by Mycobacterium tuberculosis, continues to be one of the main diseases to mankind. It is urgent to discover novel drug targets for appropriate antimicrobial agents against this human pathogen. The shikimate pathway is onsidered as an attractive target for the discovery of novel antibiotics for its essentiality in bacteria and absence in mammalian cells. The Mycobacterium tuberculosis aroE-encoded shikimate dehydrogenase was cloned, expressed and purified. Sequence alignment analysis shows that shikimate dehydrogenase of Mycobacterium tuberculosis exhibit the pattern of G-X-(N/S)-V-(T/S)-X-PX-K, which is highly conserved within the shikimate dehydrogenase family. The recombinant shikimate dehydrogenase spectrum determined by CD spectroscopy showed that the percentages for $\alpha$-helix, $\beta$-sheet, $\beta$-turn, and random coil were 29.2%, 9.3%, 32.7%, and 28.8%, respectively. The enzymatic characterization demonstrates that it appears to be fully active at pH from 9.0 to 12, and temperature $63^{\circ}C$. The apparent Michaelis constant for shikimic acid and $NADP^+$ were calculated to be about $29.5\;{\mu}M$ and $63\;{\mu}M$. The recombinant shikimate dehydrogenase catalyzes the substrate in the presence of $NADP^+$ with an enzyme turnover number of $399\;s^{-1}$. Zymological studies suggest that the cloned shikimate dehydrogenase from M. tuberculosis has a pretty activity, and the work should help in the discovery of enzyme inhibitors and further of possible antimicrobial agents against Mycobacterium tuberculosis.
