서울근교 약수터에서 분리한 Pseudomonas속균의 혈청생화학적 특성 및 항균제 감수성 (Serobiological Characteristics and Antibiotic Sensitivity of Pseudomonas spp. Isolated from Spring Waters in Seoul Area)

  • 서울시내 약수에서 Pseudomonas속균의 균종분포, 생화학적 특성, 혈청학적 특성 및 항생제 감수성을 조사하기 위하여 약수 57개소에 대하여 봄, 여름, 가을에 걸쳐 조사하였다. 총 57개 약수를 조사한 결과, 33개 (57.9%)의 약수에서 Pseudomonas속균이 검출되었다. 봄, 여름, 가을에 모두 검출된 곳은 1곳(1.8%) 이었으며, 2회에 걸쳐 검출된 곳은 9곳 (15.8%)이었으며, 1회만 검출된 곳은 13곳 (22.8%)이었다. 지역별로는 청계산이 50%의 검출율을 나타내었으며, 북한산, 35.7&, 대모산 33.3%, 도봉산 29.6%, 수락산 25.9%, 우면산 22.2%, 불암산 7.4% 순이었다. 분리된 균종은 Ps. putidark 22주(50%)로 가장 많았으며, Ps. aeruginosa, Ps. fluorescens 및 Ps. mendocins 각 6주(13.6%), Ps.aureofaciens 4주(9.1%)이었다. 분리된 Ps. aeruginosa6주의 혈청형은 모두 5종으로 A형 2주 , B, C,E 및 G형 각 2주씩이었다. 분리된 Ps. aeruginosa의 항생제에 대한 내성은 Amoxicillin 90.9%, Chloramphenicol 84.1%, Carbenicillin 81.8%, Nalidixic acid 68.2%, Neomycin 38.6%, Streptomycin 31.8%, Gentamicin 4.6%, Kanamycin 4.6% 그리고 Colistin 2.3%이었다. Ps.aeruginosa는 다른 슈도모나스균에 비해 carbenicillin에 대한 감수성이 있으나 kanamycin에 대한 내성이 높았다. Ps. aureofaciens는 다른 슈도모나스균에 비해 streptomycin에 대하여 내성이 없었다. 분리된 Ps.aeruginosa의 항생제에 대한 다제내성양상은 5제내성이 31.8%로 가장 높았으며, 내성양상에서는 NA-CB-CTE-AMC가 18.2%로 가장 높게 나타났다.

전염병관리 관련법령의 변화 추이분석 및 향후 개정방향에 관한 연구 (The amendment tendency analysis of the Korean Infectious Disease Prevention Act and a recommendation for the next amendment)

  • This Study has been carried out to make a recommendation for the next amendment of the Infectious Disease Prevention Act with a specific focus on the kind of notifyable disease. Korean, Japanese, German, U.S, English and French acts on infectious diseases prevention were reviewed, compared with and analized in regards of numbers and kinds of notifyable infectious diseases and their tendency of amendments. An criteria was designed to assess the level of validity of diseases to be designated in the act. Four items, the fatality (greater than 10% or not), the possibility to make a big epidemic, the availability of efficient vaccination and the usefulness of isolation, are used in the assessment. This index is applied to the diseases in Korean and other countries' Infectious Disease Prevention Acts. Results are as follows: 1. The Korean Infectious Disease Preventon Act has a unique way of classifying the notifyable infectious disease, that is, the first, the second and the third class. But the author cannot find the basis of classification. No other countries reviewed have the similar classification. 2. The ten diseases, cholera, plague, yellow fever, diphtheria, typhoid fever, poliomyelitis, rabies, tetanus, malaria, and meningococcal meningitis are designated as the notifyable diseases not only in Korea but also in Japan, Germany, United States, England and france. 3. Thirty seven diseases including small pox, Lassa fever, anthrax, influenza, German measles, Legionellosis, infection with E. coli O157:H7, Q-fever, brucellosis, Lyme disease are designated as legal disease at least one of the above mentioned countries. 4. The Korea has been coped with the change of the infectious disease occurrence for last fifty years in amendment of the Infectious Disease Prevention Act. 5. Japan has a special infectious surveillance system composed of 3,880 clinics throughout the whole country. 6. Germany has classified infectious diseases in five categories which are based on seriousness of disease. Any confirmed death, cases and suspected cases in class I should be reported within 24 hours. But only confirmed death and cases in class II, but not suspected cases, are reportable in Germarny. 7. Plague, bacillary dysentery, pertussis, mumps, Japanese encephaltis and Korean hemorrhagic fevers are diseases with high credits validity index among Korean legal disease. 8. German measles, anthrax, E. coli O157 : H7 infection, Lassa fever, Q-fever, brucellosis are high in validity index among those which are not designated in Korea but designated in other countries. In conclusion, the Korean Infectious Disease Prevention Act has well been coped with the changes of infectious disease occurrence for last fifty years, but the classification basis and the validity of diseases to be designated as legal diseases is worth reevaluating.

대장균(大腸菌)의 xylRjT 변이주(變異株)의 분리(分離) 및 그 특성(特性) (Isolation and Characterization of xylR/TMutants in Escherichia coli)

  • Xylose 오페론의 조절기구(調節機構)를 밝히고 xyl 유전자(遺傳子)를 가진 재조합유전자(再組合遺傳子)들의 수용세포(受容細胞)로 사용(使用)하기 위해 대장균(大腸菌)에 NTG를 처리(處理)하여 xylose를 이용(利用)할 수 없는 xyl 변이주(變異株)를 최종(最終) 9주(株) 선발(選拔)하였다. MC-Xyl 한천평판배지(寒天平板培地)에서 백색(白色) 콜로니로 분리(分離)된 xyl 변이주(變異株)들은 모두 LB와 DM-Glc 배지(培地)에서는 친주(親株)인 E. coli JM109와 동일(同一)하게 자랐으나, DM-Xyl 배지(培地)에서는 생육(生育)하지 못했다. 그러나 xyl 유전자(遺傳子) 전체(全體)를 가진 pBX1으로 형질변형(形質變形)시킨 결과(結果) 모두MC-Xyl 한천평판배지(寒天平板培地)에서는 적색(赤色) 콜로니를 나타내고 xylose isomerase 활성(活性)도 친주(親株)와 유사(類似)하게 되살아 났다. 이들의 부귀(復歸) 돌연변이빈도(突然變異頻度)는 $10^{-8}{\sim}10^{-11}$ 이하(以下)로 유전적(遺傳的)으로 안정(安定)되었다. 분리(分離)된 xyl 변이주(變異株)와 그들의 형질전환주(形質轉換株)에 대하여 MC-Xyl과 MC-Xylu 한천평판배지(寒天平板培地)에서 콜로니의 색(色)을 관찰(觀察)하였고 xylose isomerase와 xylulokinase의 활성(活性)을 측정(測定)하여 다시 xylT 변이주(變異株) 3주(株)(DH13, DH121, DH125) xylA 변이주(變異株) 1주(株)(DH77), xylB 변이주(變異株) 1주(株)(DH43) 그리고 xylose 존재하(存在下)에서 이들 효소(酵素)들의 생성(生成)을 조절(調節)하는 xylR 변이주(變異株) 3주(株)(DH10, DH53, DH60), 마지막으로 xylR, A 유전자부위(遺傳子部位)에 변이(變異)가 일어난 것(DH35)으로 최종선별(最終選別)하였다.

한국인의 자가평가 건강수준 측정도구(KHP 1.0) 개발 (A Trial for Development of Health Profile (KHP 1.0) to Measure the Self-Perceived Health Status of Korean)

  • Objectives : The 1990s has seen advances in the conceptualization of self-perceived health status which has important roles for individual health and the quality of life. Many types of standardized questionnaires have been developed with the current wide use of SF-36, NHP, andEuroQol. However, the outcomes of these tools may be different with regard to regional, cultural and emotional backgrounds. The purpose of this study was to trial the development of a Korean Health Profile (KHP 1.0) to measure the self-perceived health stati of Koreans. Methods : The KHP 1.0 was designed on the basis of the Medical Outcome Study Form 36 (SF-36), the Nottingham Health Profile (NHP), and the EuroQOL. It was composed of 9 scales; physical functioning, role limitation-physical, pain, general health, energy, social isolation, sleep, role limitation-emotional, and e-motional health. Self-reported chronic disease conditions, and the Zung's Self-Rating Depression Scale (SDS), were also checked for the evaluation of clinical validity. This study was conducted, from December 2000 to January 2001, on 800 middle-aged parents, with four high school students, with 100 retest sets being conducted two weeks later. From the 800 subjects there were 588 complete responses (effective response 73.5%). The reliability of the test-retest results, and the factor analysis on the validity of the KHP 1.0 components, were evaluated using the SPSS (ver 10.0) software. Results : The reliability of the KHP 1.0 was good with Cronbach's alpha (>0.6), test-retest correlation coefficients (>0.5), but with no significant differences from the paired t-test. From the psychometric validity tests, the 9 scales of the KHP 1.0 were divided into two components; physical and mental, and trimmed to the established model with 55% of the total variance, with the exception of role limitation-emotional. The clinical validity on the basis of the comparison for the four characteristic groups; healthy, physical conditions only, mental conditions only, and physical and mental conditions were also good. Conclusions : The KHP 1.0 appears to be a valid measurement tool of self-perceived health stati of Koreans, although there are limitations, i.e. sample size was too small, a limited number of middle-aged subjects, and it was based on unconfirmed diagnoses, etc. Therefore, further study is required to standardize the assessment.

Negatively Charged Membrane을 이용한 해수 중 어류질병바이러스의 검출 (Detection of Fish Pathogenic Viruses in Seawater Using Negatively Charged Membranes)

  • After an outbreak of viral disease in an aquafarm, release of virus (es) from infected fish into environmental seawater has been suspected. In the present study, we utilized a negatively charged membrane (HA type) as an efficient method for concentration and detection of fish pathogenic viruses, specifically, megalocytivirus and viral hemorrhagic septicemia virus (VHSV) present in field-collected seawater samples or inoculated into seawater artificially. Positively charged viruses adsorbed onto the negatively charged membrane and were eluted with 1 mM NaOH (pH 10.5) following rinsing with 0.5 mM $H_2SO_4$ (pH 3.0). Megalocytivirus and VHSV particles isolated using anegatively charged HA membrane from seawater inoculated with each virus at a concentration of 10 viral particles/mL were of sufficient quantity to show positive results in atwo-step PCR (or RT two-step PCR); however, despite it being negatively charged, a cellulose acetate (CA) membraneshowed negative results. In quantitative PCR, the detection limits of the HA membrane for megalocytivirus and VHSV in seawater were 1.20E+00 viral particles/mL and 1.22E+01 viralparticles/mL, respectively. The calculated mean recovery yields from 1 L seawater spiked with known concentrations of megalocytivirus and VHSV particles were 28.11% and 23.00%, respectively. The concentrate of a 1-L sample of culturing seawater from the aquatank of flounder suffering from VHSV showed clear positive results in PCR when isolated with an HA, but not a CA, membrane. Thus, viral isolation using an HA membrane is a practical and reliable method for detection of fish pathogenic viruses in seawater.

  • A lactic acid bacteria with ${\beta}$-gal activity was isolated from Kimchi, a traditional fermented vegetable food in Korea. The isolate was identified as a Lactococcus lactis strain and named L. lactis A2. The gene encoding ${\beta}$-gal of L. lactis A2 was cloned as a 5.8 kb PstI fragment. DNA sequencing identified the complete lacA (galactoside acetyltransferase)-lacZ (${\beta}$-galactosidase) genes together with the 3' part of upstream galT (galactose-1-phosphate uridyltransferase), and the 5'region of downstream galE (UDP-galactose-4-epimerase) genes. L. lactis A2 had the same gal/lac operon structure as in L. lactis subsp. lactis 7962. Other genes of the Leloir pathway are most likely to be located in the 5'upstream of the 5.8 kb fragment on the A2 chromosome. Sequences downstream of galE were different from those of L. lactis subsp. lactis 7962.

Phage Display 방법을 이용한 B형 간염 바이러스의 Terminal Protein 특이 scFv 항체 생산 (Terminal Protein-specific scFv Production by Phage Display)

  • Background: One of the important factors in the prognosis of chronic hepatitis B patient is the degree of replication of hepatitis B virus (HBV). It has been known that HBV DNA polymerase plays the essential role in the replication of HBV. HBV DNA polymerase is composed of four domains, TP (Terminal protein), spacer, RT (Reverse transcriptase) and RNaseH. Among these domains, tyrosine, the $65^{th}$ residue of TP is an important residue in protein-priming reaction that initiates reverse transcription. If monoclonal antibody that recognizes around tyrosine residue were selected, it could be applied to further study of HBV replication. Methods: To produce TP-specific scFv (single-chain Fv) by phage display, mice were immunized using synthetic TP-peptide contains $57{\sim}80^{th}$ amino acid residues of TP domain. After isolation of mRNA of heavy-variable region ($V_H$) and light-chain variable region ($V_L$) from the spleen of the immunized mouse, DNA of $V_H$ and $V_L$ were obtained by RT-PCR and joined by a DNA linker encoding peptide (Gly4Ser)3 as a scFv DNA fragments. ScFv DNA fragments were cloned into a phagemid vector. scFv was expressed in E.coli TG1 as a fusion protein with E tag and phage gIII. To select the scFv that has specific affinity to TP-peptide from the phage-antibody library, we used two cycles of panning and colony lift assay. Results: The TP-peptide-specific scFv was isolated by selection process using TP-peptide as an antigen. Selected scFv had 30 kDa of protein size and its nucleotide sequences were analyzed. Indirect- and competitive-ELISA revealed that the selected scFv specifically recognized both TP-peptide and the HBV DNA polymerase. Conclusion: The scFv that recognizes the TP domain of the HBV DNA polymerase was isolated by phage display.

Corynebacterium ammoniagenes에서 purF 유전자의 조절 및 이에 특이적인 조절 단백질의 분리 (Regulation of Corynebacterium ammoniagenes purF and Isolation of purF-Specific Regulatory Proteins)

  • Corynebacterium ammoniagenes의 purF 유전자의 발현을 purF의 프로모터 추정 부위에 cat 유전자를 융합시킨 transcriptional fusion 플라스미드를 제작하여 분석하였다. 유전자 purF는 adenine과 guanine에 의해 20~30%의 전사 저해효과를 나타내지만, hypoxanthine에는 저해를 받지 않는 것으로 나타났다. 또한 purF의 발현은 대수기중반에 최대에 달한 후 정체기 후반부까지 일정한 것으로 나타났다. 동시에, C. glutamicum에서 사용되는 강력한 프로모터인 $P_{180}$이 Escherichia coli의 $P_{tac}$보다 C. ammoniagenes에서 모든 성장 단계에서 40~50%의 향상된 프로모터 활성을 나타내었고 대수기 후반부에 최고 활성에 달해, C. ammoniagenes의 연구에도 활용 가능함을 확인하였다. DNA-affinity purification에 의해 C. ammoniagenes의 purF 프로모터에 결합하는 단백질로서 C. glutamicum의 Crp-family transcriptional regulator (NCgl0120)와 상동성이 높은 단백질을 검출하였다. 이 단백질은 크기가 40.1 kDa으로서 PAGE에서 관찰된 단백질 크기와 일치하였다. 이에 상응하는 C. ammoniagenes의 단백질은 400개의 아미노산으로 구성되어 있고, 42 kDa의 단백질을 만들며, pI는 4.9일 것으로 추정되었다. 이는 기존에 알려져 있는 E. coli 및 Bacillus subtilis의 PurR과 각각 14.1%, 15.8%의 아미노산 상동성을 보여, PurR과는 다른 종류의 단백질일 것으로 여겨진다.

콩나물 부패병 방제를 위해 토양으로부터 분리한 길항균 Pseudomonas fluorescens의 이용 (Isolation and Utilization of Antagonistic Pseudomonas fluorescens from Soils for the Protection of Soybean Sprouts Rot)

  • 부패된 콩이나 콩나물에 존재하는 33종$(K-1{\sim}K-33)$의 각종 미생물로부터 콩나물 부패에 관여하는 세균(K-17)과 진균(K-28)을 각각 1종류씩 분리하였으며, 이들은 각각 Erwinia carotovora와 Fusarium sp.로 동정하였다. 콩나물 부패균에 길항하는 미생물은 각종 토양 균원시료로부터 분리한 350여종의 세균을 이용하여 콩나물 부패세균인 K-17 균주와 진균인 K-28 균주에 각각 길항력을 나타내는 J-232 세균을 최종 선발하였다. 길항균 J-232는 Pseudomonas fluorescens로 동정되었다. 콩나물 부패균 E. carotovora K-17과 Fusarium sp. K-28의 배양액으로 재배한 콩나물은 시루 아랫부분이 부패하였으나, 길항균 P. fluorescens J-232는 PDA 한천배지에서도 부패균들의 생육을 억제하였을 뿐만아니라 길항균 배양여액을 단독으로 콩나물재배에 처리할 경우에도 시루 내 콩나물은 부패되지 않았다. 콩나물 부패균 E. carotovora K-17과 Fusarium sp. K-28의 배양액에 길항균 P. fluorescens J-232의 배양액을 원액 또는 500배까지 희석하여 침지한 후 재배한 콩나물도 전혀 부패되지 않았으며 생육이 약간 촉진되는 경향이었다.

즉석조리식품에서의 Bacillus cereus오염실태조사 및 생육 pattern 분석 (Isolation and Growth Pattern of Bacillus cereus from Ready-to-Eat Foods.)

  • 국민 다소비 식품인 생선회, 패류 등의 횟감류 118건, 김밥, 햄버거 등의 도시락류 82건 및 기타 ready-to-eat(RTE) 식품 40건, 총 240건을 대상으로 Bacillus cereus의 오염실태를 조사한 결과, 횟감류 16건(13.6%), 도시락류 17건(20.7%), 기타 RTE 식품에서 3건(7.5%), 총 36건(15.0%)에서 B. cereus가 검출되었다. B. cereus의 검출은 polymyxin B를 첨가한 TSB배지를 사용한 증균배양 후, MYP agar에서 전형적인 집락을 선별하였으며, 선별된 균주들을 대상으로 그람염색, oxidase test, 혈액한천배지에서의 rhizoid growth, $\beta$-용혈성 유ㆍ무등을 통해 추정 균주들을 분리하였다. API 50CHB/20E system을 통한 생화학적 분석과 motility시험 등을 행하여 최종적으로 B. cereus로서 동정ㆍ확인하였다. 확인된 36균주 중 28균주(77.8%)가 BCET-RPLA 시험을 통해 BHI broth에서 설사형 독소를 생성하는 것으로 나타났다. 아울러 항생제 감수성 시험 결과 분리균들은 ampicillin (100%), penicillin G (100%) 및 rifampicin (69.5%)에 내성을 보였으며, gentamicin (100%), vancomycin (100%), bacitracin (86.1%), chloram-phenicol (97.2%), erythromycin (69.4%), kanamycin (86.1%) 및 streptomycin (97.2%)에 감수성을 나타내었다. B. cereus의 생육은 온도와 시간에 밀접한 관련성을 보였는데, 냉장온도인 1$0^{\circ}C$이하에서는 균의 생육이 상당히 저해된 반면, 2$0^{\circ}C$ 및 3$0^{\circ}C$에 보관시에는 급격한 증식을 보여 기온이 상승하는 하절기에 특히 많은 주의가 필요하다고 판단되었다.
