Comparison of Three Different Methods for Campylobacter Isolation from Porcine Intestines

  • Using 200 porcine colon tissues, the efficiencies of three isolation methods of Campylobacter from porcine intestines were compared: Method 1, direct streaking of colon mucosa; Method 2, direct inoculation of intestinal contents with a swab; Method 3, inoculation of pre-enriched medium. A total of 460 Campylobacter isolates were obtained from 178 samples (89%) by direct streaking of colon mucosa, 142 samples (71%) by direct streaking of a swab, and 94 samples (47%) by pre-enrichment of intestinal contents in Preston broth. Direct streaking of colon mucosa was superior to the other two isolation methods, in terms of rapidity and higher efficiency. When isolates were identified with various biochemical tests and PCRs specific to 16s rRNA, mapA, and ceuE, C. coli was the predominant species (87%) in porcine, whereas the rest of the isolates were identified as C. lanienae.

Isolation Enhancement between Two Dual-Band Microstrip Patch Antennas Using EBG Structure without Common Ground Plane (독립된 접지면을 갖는 EBG 구조를 이용한 이중 대역 마이크로스트립 패치 안테나 사이의 격리도 향상)

  • In order to enhance the isolation level between two dual-band E-slot microstrip patch antennas, EBG structure which operates in UMTS Tx(1.92~1.98 GHz) and Rx(2.11~2.17 GHz) band is proposed. The proposed EBG structure made with a periodic array of two different size EBG unit cells which has a modified mushroom-type for isolation improvement between two antennas. They do not share a common ground plane of the microstrip patch antenna. Overall size of the fabricated antenna is $210.5mm{\times}117mm$. The two different EBG unit cell sizes are $15.6mm{\times}4mm$ and $17.4mm{\times}4mm$, respectively. It was etched on the FR-4 substrate(thickness=3.93 mm, ${\varepsilon}_r$=4.6). The experiment results show that the isolation level between antennas in Tx/Rx band were improved by about 9 dB and 12 dB, respectively, through the use of the proposed EBG structure.

Comparison of DNA isolation methods for detection of foodborne pathogens by real-time PCR from foods (식품으로부터 식중독 세균 검출을 위한 Real-time PCR에 적합한 DNA 추출 방법 비교)

  • This study was conducted to find out the most suitable DNA isolation methods for PCR detection of foodborne pathogens. Four DNA isolation methods including Universal Genomic DNA Extraction Kit (TaKaRa), PrepMan Ultra (Applied Biosystems), boiling method and alkaline lysis method (w/PEG) were tested and compared. The Universal Genomic DNA Extraction kit (TaKaRa) was considered as the more efficient isolation method for Escherichia coli O157:H7 and Staphylococcus aureus in lettuce, fish and beef. Meanwhile to detect the foodborne pathogens directly from foods without enrichment, the four different buffers such as double-distilled water, saline, glycine-saline, glycine-saline with Tween-20 and beef extract were also evaluated. As a result, saline was more suitable buffer for E. coli O157:H7. And double-distilled water was more suitable buffer than saline for S. aureus, respectively

Design of a Metamaterial Absorber for High Isolation of a WCDMA Indoor Repeater Antenna (WCDMA 댁내형 중계기 안테나의 격리도 개선을 위한 메타 구조 기반의 흡수체 설계)

  • In this paper, an absorber based on metamaterial is proposed to improve the isolation of conventional WCDMA indoor repeater antenna. The proposed absorber is composed of Double Split Ring Resonators(DSRRs) and Complementary Spiral(CS) structure. The proposed absorber based on metamaterial is $9.6mm{\times}9.6mm{\times}1.2mm$ and absorption is about 94 % at 2.2875 GHz. The proposed antenna, which proposed absorber is applied to conventional WCDMA indoor repeater antenna, has isolation over 85 dB. Isolation is improved more than 10 dB compared with the conventional antenna. The VSWR is lower than 2 at WCDMA band from 1.92 GHz to 2.17 GHz. The radiation patterns are $60^{\circ}{\pm}10^{\circ}$ E-plane and H-plane, respectively. And, the gain is more than 6 dBi. The volume of proposed antenna with absorber based on metamaterial is $90mm{\times}90mm{\times}44.8mm$.

Design of a Rectangular Waveguide Antenna for Automotive Side and Rear Radar Sensor Applications (차량 측후방 레이더 센서용 직사각형 도파관 안테나 설계)

  • In this paper, a waveguide antenna is designed and fabricated for side and rear radar sensor of vehicles in UWB (Ultra Wide Band) high band (center frequency : 9.5 GHz, -10 dB bandwidth : 600 MHz (6.4 %)). For the radar antenna, a probe fed rectangular waveguide antenna having simple structure and wide bandwidth is used. An important performance factor in this antenna is the isolation between transmitting (TX) and receiving (RX) waveguide antennas because this radar system uses TX and RX antennas separately. Thus the isolation between two antennas was simulated for E-plane and H-plane array. As a result, it was verified that the isolation of the H-plane array of the antennas is better than E-plane array, due to the TE10 mode. Therefore, H-plane arrayed waveguide antennas were mounted on a T-shaped radar module and performance of antennas was investigated. The -10 dB bandwidth of the TX and RX antenna mounted on T-shaped radar module was measured as 1000 MHz (10.52 %) and 1090 MHz (11.47 %) respectively and the isolation is less than -50 dB in the operation band. The peak gain is 7.65 dBi for the TX antenna and 7.26 dBi for the RX antenna and the beamwidth of H-plane of TX and RX antenna was measured as $64^{\circ}$ and $65^{\circ}$ respectively. Consequently, we verified that the proposed waveguide antenna is appropriate for a vehicle radar applications.

The Effects of Multidimensional Social Isolation on Physical and Mental Health: Analysis of Interaction Effects of Age Groups (다차원적 사회적 고립이 신체적·정신적 건강에 미치는 효과: 연령집단의 상호작용효과 분석)

  • Along with the well-established evidence on the negative effect of social isolation on physical and mental health, increasing attention has been paid to multi-dimensional nature of social isolation. One line of study on social isolation has discussed different pathways between objective and subjective social isolation and health. Another stream of the research focused on the possibly non-linear association between social isolation and health by age cohort groups. Drawing from the two lines of research, this study aimed at empirically examine to what extent objective and subjective social isolation are associated with physical and mental health independently and how the associations vary by three age cohorts(i.e. the middle-aged, the young old, the old-old). Data came from the first wave of Korean Social Life, Health and Aging Project (KSHAP) (N= 814). Findings showed 1) objective subjective isolation were significantly related with worse physical and mental health, interestingly, subjective social isolation was associated with mental health only, 2) pattern of association between social isolation and physical health varied by age cohorts. Specifically, compared to the middle-aged, the young old with higher objective social isolation exhibited lower level of physical health, while the old-old with higher subjective social isolation were likely to experience lower physical health. Based on the findings, we discussed implications and suggestions for future research and relevant policy/program development for ameliorating objective and subjective social isolation.

Soil-Structure Interaction Analysis for Base-Isolated Nuclear Power Plants Using an Iterative Approach (반복법을 이용한 면진적용 원전구조물의 지반-구조물 상호작용 해석)

  • The nuclear accident due to the recent earthquake in Japan has triggered awareness of the importance of safety with regard to nuclear power plants (NPPs). An earthquake is one of the most important parameters which governs the safety of NPPs among external events. Application of a base isolation system for NPPs can reduce the risk for earthquakes. At present, a soil-structure interaction (SSI) analysis is essential in the seismic design of NPPs in consideration of the ground structure interaction. In the seismic analysis of the base-isolated NPP, it is restrictive to consider the nonlinear properties of seismic isolation devices due to the linear analysis of the SSI analysis programs, such as SASSI. Thus, in this study, SSI analyses are performed using an iterative approach considering the material nonlinearity of the isolators. By performing the SSI analysis using an iterative approach, the nonlinear properties of isolators can be considered. The difference between the SSI analysis results without iteration and SSI with iteration using SASSI is noticeable. The results of the SSI analysis using an effective linear (non-iterative) approach underestimate the spectral acceleration because the effective linear model cannot consider the nonlinear properties of isolators. The results of the SSI analysis show that the horizontal response of the base-isolated NPP is significantly reduced.

Efficient isolation, culture and regeneration of Lotus corniculatus protoplasts

  • This paper reports an improved protocol for isolation, culture and regeneration of Lotus corniculatus protoplasts. A range of parameters which influence the isolation of L. corniculatus protoplasts were investigated, i.e., enzyme combination, tissue type, incubation period and osmolarity level. Of three enzyme combinations tested, the highest yield of viable protoplasts was achieved with the combination of 2% Cellulase Onozuka RS, 1% Macerozyme R-10, 0.5% Driselase and 0.2% Pectolyase. The use of etiolated cotyledon tissue as a source for protoplast isolation proved vital in obtaining substantially higher protoplast yields than previously reported. Culture of the protoplasts on a nitrocellulose membrane with a Lolium perenne feeder-layer on the sequential series of PEL medium was highly successful in the formation of microcolonies with plating efficiencies 3-10 times greater than previous studies. Shoot regeneration and intact plants were achieved from 46% of protoplast-derived cell colonies.

Reproductive Isolation between Moroco oxycephalus and M. lagowskii (Pisces; Cyprinidae) in Korea

  • To clarify taxonomic status of the two sibling species, Moroco oxycephalus and M. lagowskii reproductive isolation mechanisms were investigated at sympatric area located in Kansung-up, Kosung-gun, Kangwon-do, Korea. Genetic analysis was performed to reveal mating system and intensity of Hybridization between the two species. The frequencies of hybrids were increased since 1989, and then the observed hybrid frequencies ($H_O$) did not significantly differ from the expected hybrid ($H_E$) in 1998 and 1999. However, based on histological analysis of two parents and their hybrid s gonads, the hybridizations between M. oxycephalus and M. lagowskii produced mostly fertile females but sterile males in accordance with Haldane s rule. Although it was suspected that pre- and postmating isolation mechanisms were affected between the two species, M. oxycephalus and M. lagowskii seemed to be strongly isolated with microhabitat at sympatry until 1997. Since 1998, hybrid frequencies were increased by habitat disturbance. However, their hybrid frequencies would be reduced by postmating isolation mechanisms. Therefore, the two species are considered to be distinct species recently diverged.

Effect of Bouc-Wen Model and Earthquake Characteristics for Responses of Seismically Isolated Nuclear Power Plant by Lead-Rubber Bearing (납-고무 받침에 의해 면진된 원전구조물의 응답에 대한 Bouc-Wen 모델 및 지진특성의 영향)

  • In order to modeling seismic isolation system such as lead-rubber bearing (LRB), bilinear model is widely used by many researchers. In general, an actual force-displacement relationship for LRB has a smooth hysteretic shape. So, Bouc-Wen model with smooth hysteretic shape represents more accurately actual hysteretic shape than bilinear model. In this study, seismic responses for seismically isolated nuclear power plant (NPP) with LRB modelled by Bouc-Wen and bilinear models are compared with those of NPP without seismic isolation system. To evaluate effect of earthquake characteristics for seismic responses of NPP isolated by LRB, 5 different site class earthquakes distinguished by Geomatrix 3rd Letter Site Classification and artificially generated earthquakes corresponding to standard design spectrum by Reg. Guide 1.60 are used as input earthquakes. From the seismic response results of seismically isolated NPP, it can be observed that maximum displacements of seismic isolation modelled by Bouc-Wen model are larger than those by bilinear model. Seismic responses of NPP with LRB is significantly reduced than those without LRB. This reduction effect for seismic responses of NPP subjected to Site A (rock) earthquakes is larger than that to Site E (soft soil) earthquakes.
