Microleakage of restorative materials has been investigated mostly by dye penetration method. Dye penetration method was not quantitative and not measured repeatedly. Fluid filtration method was used to research purpose to understand the effects of various restorative treatments on dentin permeability. The purpose of this study was to evaluate of dentin permeability according to the process of compomer restoration treatment. In this study, C1 V cavities were prepared on buccal surface of thirty extracted human molars. The prepared cavities were etched by 37% phosphoric acid.(omitted)
The purpose of this study was to evaluate the effect of apical seal produced by ultrasonic instrumentation. 120 extracted, permanent single rooted teeth were randomly divided into 6 groups of 20 teeth and root canals were enlarged & obturated according to Table I. After canal obturation, the obturated teeth were immersed in 2.5 % Methylene blue for 72 hrs. at $37^{\circ}C$ incubator and longitudinally sectioned. The apical sealing ability was evaluated by measuring the degree of dye penetration into the canal. The results were as follows: 1. All groups showed varying depth of dye penetration. 2. There were no significant differences between Group I and N, Group II and V, Group III and VI. (P>0.05) 3. Regardless of obturation method, there were no significant differences between hand instrumented group and ultrasonic instrumented group.(P>0.05).
This experiment was performed to observe the adhesion pattern and microleakage in the gingival margin according to variation in the resin cement thickness which results from thickness of Die spacer. which is considered to effect the adaptability of the composite resin inlays. Clearfil CR inlays were fabricated on stone models with CR Sep applicated once and Nice fit twice, 4 times, and 6 times each. After 2nd curing within the CRC-100 oven, CR inlays were cemented with CR inlay cement. Dye(2% methylene blue) penetration and adhesion pattern were evaluated after sectioning of gingival margin into :3 pieces. The results were as follows ; 1. The thickness of resin cement showed unevenchanging pattern with that of die spacer, namely, it was increased until 4 times' application of Nice-Fit but was decreased with 6 times' application of that. 2. The degree of dye penetration wasn't affected by cement thickness within a limited value. 3. Most of dye penetration was shown through the interface between cement and enamel rather than the interface between cement and CR inlay. This shows that the affinity of resin cement for CR inlay was superior to the adhesive strength with tooth structure. 4. No gap was found at the interface between enamel and cement but some showed separation between dentin and cement. It is concidered that the contraction force of cement was less than the bond strength with the enamel. 5. Lots of voids were found in the CR inlay and resin cement. There was a pooling tendency of bonding agent and cement in the axiogingival line angle portion. 6. In some specimens, cracks were shown in enamel margin. From this it could be considered that cavity preparation and surface treatment weakened the tooth structure.
The purpose of this study was to evaluate the effect of the dentin pretreatment on the marginal leakage of a glassionomer cement. 1n this study, 60 molars with sound and healthy crown portion were used. The dentin surface of these teeth were exposed and polished with 600 grit silicon carbide paper. Square - shaped cavities were prepared on the flattened dentin surfaces and these were divided into 4 groups according to the dentin pretreatment procedures. Group I : Dentin pretreatment with distilled water as a control group. Group II : Dentin pretreatment with 5% sodium hypochlorite solution. Group III : Dentin pretreatment with Ketac conditioner. Group IV : Dentin pretreatment with 40% polyacrylic acid. The degrees of dye penetration in the cavity walls were assessed using a stereoscope at ${\times}40$ magnification according to the maximum dye penetration. The results were analyzed by using Mann - Whitney U test. The results were as follows : 1. All groups showed varying depth of dye penetration. 2. Distilled water group showed the most severe marginal leakage when compared with the other groups(P<0.05). 3. 40% polyacrylic acid group showed the least amount of marginal leakage compared with the other groups (P<0.05). 4. There were significant differences between Goup I(distilled water) and Group IV (40% polyacrylic acid)(P<0.05), but there were no significant differences among Group I(distilled water), Group II(sodium hypochlorite), Group III(Ketac conditioner) (P>0.05).

Polypropylene fibers are difficult to dye by conventional dyeing techniques due either to lack of dye receptor sites in their molecular structure to with the dye molecules may become attached, or to difficult of penetration of certain types of dye molecules into the structure$^1$. Although the diffusion coefficient of disperse dyes into polypropylene is quite high, the saturation values are very low.(omitted)
The multicellular layers(MCL) of human cancer cells is a three dimensional(3D) in vitro model for human solid tumors which has been used primarily for the assessment of avascular penetration of anti-cancer drugs. For anti-cancer drugs with penetration problem, MCL represents a good experimental model that can provide clinically relevant data. Calcein-AM is a fluorescent dye that demonstrates the cellular vitality in a graded manner in cancer cell culture system. In the present study, we evaluated the use of calcein-AM for determination of anti-proliferative activity of anti-cancer agents in MCL model of DLD-1 human colorectal cancer cells. Optical sectioning of confocal imaging was compromised with photonic attenuation and penetration barrier in the deep layers of MCL. By contrast, fluorescent measurement on the cryo-sections provided a feasible alternative. Cold pre-incubation did not enhance the calcein-AM distribution to a significant degree in MCL of DLD-1 cells. However, the simultaneous determination of drug penetration and cellular vitality appeared to be possible in drug treated MCL. In conclusion, these data suggest that calcein-AM can be used for the simultaneous determination of drug-induced anti-proliferative effect and drug penetration in MCL model.

PURPOSE. Porcelain fused to metal (PFM) crowns provide the best treatment option for teeth that have a large or defective restoration. More than 20% of teeth with PFM crowns or bridges require non-surgical root canal treatment (NSRCT). This may be due to the effect of restorative procedures and the possible leakage of bacteria and or their by-products, which leads to the demise of the tooth pulp. Thus, this study was planned to compare the ability of the restorative materials to seal perforated PFM specimens. MATERIALS AND METHODS. The study evaluates the ability of amalgam, composite or compomer restorative materials to close perforated PFM specimen's in-vitro. Ninety PFM specimens were constructed using Ni-Cr alloys and feldspathic porcelain, and then they were divided into 3 groups: amalgam (A), composite + Exite adhesive bond (B) and compomer + Syntac adhesive bond (C). All the PFM samples were embedded in an acrylic block to provide complete sealing of the hole from the bottom side. After the aging period, each group was further divided into 3 equal subgroups according to the thermocycling period (one week for 70 cycles, one month for 300 cycles and three months for 900 cycles). Each subgroup was put into containers containing dye (Pelikan INK), one maintained at $5^{\circ}C$ and the other at $55^{\circ}C$, each cycle for 30 sec time. The data obtained was analyzed by SPSS, 2006 using one way ANOVA test and student t-test and significant difference level at (P<.01). RESULTS. The depth of dye penetration was measured at the interfaces of PFM and filling materials using Co-ordinate Vernier Microscope. The lowest levels of the dye penetration for the three groups, as well as subgroups were during the first week. The values of dye leakage had significantly increased by time intervals in subgroups A and C. CONCLUSION. It was seen that amalgam showed higher leakage than composite while compomer showed the lowest level of leakage.

Objectives: The purpose of this study is to determine the exposure risk to tattoo components by analyzing skin absorption using the in vitro method. Tattoos are commonly used for cosmetic purposes, and the skin of not only the operator but of the people who are undergoing the cosmetic procedure is continuously exposed to hazardous chemicals. Methods: Skin permeation risk determination was conducted by the in vitro Franz diffusion cell method according to the ingredient types of tattoo dyes, such as volatile organic compounds (VOCs), non-volatile organic compounds and heavy metals, using hairless mouse full skin and human cadaver epidermis. Results: The major components with good skin penetration for each type of tattoo dye ingredient were clarified. Among the tatto dye ingredients, 1,2-Dichlorobenzene, Zn, Al, Pb and Ti showed good skin penetration. Most of the skin transmission rates were higher in hairless mouse full skin than in human cadaver epidermis. Conclusion: A possible exposure risk to hazardous substances in tattoo dyes was confirmed from this study. These results are expected to provide a positive contribution to the establishment of management regulations for tattoo dyes.
Penetration degree of several dyes into dentinal tubules in vivo was compared with in vitro, and experimental sensitivity of those dyes was investigated in 64 canines of 16 cats. Dentin 1mm below cusp tip was exposed by cross-sectioning with diamond disc. Pulp of 32 canines were extirpated through cervical cavities. In the presence of smear layer or after add-etching with 37% phosphoric acid to the exposed dentin surface, acrylic collar and resin cap was attached to keep dyes. 52 mmol/$\ell$ Evans' blue, 2% Methylene blue, 10 % Silver nitrate and 5% Fluorescene were then applicated on each 4 canines respectively for 30 minutes. After rinsing, the canines were ground-sectioned longitudinally and linear dye penetration was measured under microscope. The results obtained were as follows ; 1. Evans' blue and Methylene blue penetrated significantly(P<0.05) more in vitro than in vivo only in the cases that exposed dentin surfaces were not etched with acid. 2. Silver nitrate penetrated significantly(P<0.05) more in vitro than in vivo in both cases that exposed dentin surfaces remained intact and were etched with acid. 3. The penetration degree of Fluorescene did not show statistical significance between in vivo and in vitro or in acid-etched and in not acid-etched cases.
In the prior study, wool gabardines were treated with alkaline proteases which were some kinds of enzyme to decompose protein, and their tensile strengths were determined, and the surface of the fibers were also observed using a scanning electron microscope. Enzylon ASN 30 and Alkalase 2.5L DX did not show much effect on the weight loss of wool, however, the weight loss of wool increased considerably with treating Esperase 8.0L. Pretreatment of wool with dichloloisocyanuric acid before protease-treatment increased the weight loss of wool to a great extent. In this study, the enzyme treated wools dyeing behaviors with acid dye, Milling Cyanine 5R, were mainly investigated. The protease-treatment remarkably increased not only the rate of dyeing but also the saturation dye uptake. From these results, it seemed likely that the structural relaxation of adhesive filler of interscale or intercellular cement facilitated the dye penetration into the fibers, at the same time, the change in the inner structure of the wool fibers by the protease made the fixation of the dyes more efficient.
