Dwarfuess and Kunitz trypsin inhibitor (KTI) protein in soybean is useful traits for basic studies. df2 and ti gene control dwarfness and the expression of Kunitz trypsin inhibitor (KTI) protein in soybean, respectively. The objective of this research was to verify genetic linkage or independent inheritance of df2 and ti loci in soybean. The $F_2$ population was made by cross combination between "Gaechuck#2" (Df2Df2titi genotype, KTI protein absence and a normal growth type) and T210 (df2df2TiTi genotype, a dwarf growth type and KTI protein present). A total of 258 $F_2$ seeds were analyzed for the segregation of KTI protein using SDS-PAGE. And so, 198 $F_2$ plants were recorded for the segregation of dwarfness. The segregation ratio of 3 : 1 for Ti locus (201 Ti_ : 57 titi) and Df2 locus (143 Df2_ : 55 df2df2) was observed. Segregation ratio of 9 : 3 : 3 : 1 (116 Ti_Df2_: 44 Ti_df2df2: 27 titiDf2_: 11 titidf2df2) between df2 gene and ti gene was observed ($x^2$=3.53, P = 0.223). These results showed that df2 gene was inherited independently with the ti gene in soybean.
This study was conducted to investigate the effect of cone protective net and the variation of cone and seed characteristics of a unique and rare dwarf stone pine (Pinus pumila Regel) population in Mt. Seorak which is in the southern peripheral of South Korea. Iron protective net (50 mesh, 25 ${\times}$ 25 cm) was effective in reducing birds and rodents damage to conelet, thereby it was possible to safely collect seed genetic resources. Early July was observed to be the most appropriate season to cover conelet with Iron protective net. P. pumila have ovoid-triangle shaped, wingless seeds and indehiscent cones, seeds dissemination is mainly by the hoarding behavior of nutcrackers and rodents. Statistical analysis showed that there were significant differences among individuals within population, and the mean characteristics of the species were 35.3 mm, 25.6 mm, 1.38, 6.6 g and 39.3 ea in the cone length, cone width, cone index, cone weight and in number of seeds per cone, respectively. Coefficients of variations in seed weight and number of seeds per cone were relatively high (21.7%, 21.5%, respectively) compared to other traits. Based on the correlation analysis between cone characteristics and seed characteristics, the length of cone and seed showed a significant positive correlation with the seed size and the seed weight, but the width and thickness of seed showed a negative correlation with the number of seeds per cone.
The germination percentages of Quercus hybrid seeds produced in 1994 using stored pollens were low. The survival rate and height growth within 2 years differed according to crossing combinations. The survival rate and growth of seedlings from hybrid seeds produced in 1991 and 1993 were investigated. The survival rates of five-year-old and three-year-old seedlings were from 71 to 100% and from 33 to 100%, respectively. The survival rates differed according to cross combinations. The mortality of seedlings increased in proportion to the increase of percentages of dwarf seedlings. The growth of five-year-old seedlings, of which mother tree was Q. aliena, was the best. The growth of seedlings produced by artificial crossing tended to be worse than that of natural crossing.
The main objective of this experiment was to investigate the selection efficiency for clear rice by the application of the wx-carrier technique in rice. Twelve semi-dwarf waxy lines were bred through backcrosses to the two recurrent semi-dwarf parents, IR667 and IR1317, utilizing six different waxy cultivars as waxy donor parent. These waxy lines were crossed to three clear non-waxy varieties, IR24, Suweon 287 and Suweon294. Their F$_3$ seeds were separated into waxy and non-waxy and the clearness of non-waxy grains were counted. The results are summarized as follows: l. Mean clearness per plant for twelve waxy lines ranged from 3.7% to 78.9% at the 35 days after heading. It was higher in the lines which utilized IR 1317 as recurrent parent and it was lower in the lines which utilized IR667 as recurrent parent. Mean clearness per plant for recurrent parent, IRl317 and IR667 were 47.6% and 5.6% respectively. The clear non-waxy parents; Suweon287, IR24 and Suweon294 showed 83.7% 80.5% and 73.5% clearness respectively. 2. Mean clearness of F$_3$ seeds of the crosses between different waxy parents and IR24, Suweon 294 and Suweon 287 ranged 16.7-73.9%, 21.9-42.9% and 10.6-26.9%, respectively. IR24 crosses showed the highest mean clearness. 3. Highly positive correlation was found between the clearness of F$_3$ seeds of Suweon 287 crosses and those of Suweon294. 4. Significant differences were observed in mean clearness of F$_3$ seeds of the crosses between the different waxy lines, which was bred through the same number of backcrosses to the same recurrent parent, and a clear non-waxy parent. Crosses of IR1317 recurrent parent showed higher mean clearness than those of IR667 recurrent parent. 5. In some crosses, clearness was higher in homo-non-waxy than in hetero-non-waxy, but, in other crosses it was higher in hetero-non-waxy. Thus no distinct pattern in the segregation of clearness was observed along the homo-non-waxy or hetero-non-waxy. 6. From the results it was concluded that, the selection efficiency for the clear rice can be improved by choosing the proper waxy parent, as well as proper recurrent parent under the wx-carrier technique.
This study was carried out to investigate the possibility of establishing a reproductive system for the seed of Pedicularis hallaisanensis, which is in the endangered wild species II class in Korea. The seed of P. hallaisanensis is egg-shaped, and the seed coat is dark brown. The embryo was identified as a dwarf type by the seed section. The seed length was $0.47{\pm}0.07mm$, width $0.16{\pm}0.006mm$, and thickness $0.12{\pm}0.01mm$. The weight of one seed was $0.0003{\pm}0.0001mg$, and 1000 seeds weighed $4.59{\pm}0.02mg$. The degree of seed viability was 75.33% by the tetrazolium (TZ) assay. The highest germination rate of P. hallaisanensis seed was 71% after 4 weeks of storage at $4^{\circ}C$. However, the germination rate tended to decrease gradually over a longer storage period. The germination rates after 6 or 8 weeks of storage at $4^{\circ}C$ were 64% and 60%, respectively. We used two host plants, Artemisia princeps and Dendranthema zawadskii, to determine the effect of host plants on P. hallaisanensis seed germination. The germination of P. hallaisanensis mixed with A. princeps or D. zawadskii started at 53.5 and 62.5 days after sowing, respectively. We did not find any germination 164 days postsowing with both host plants. When A. princeps and D. zawadskii were used as host plants for P. hallaisanensis seed germination, P. hallaisanensis seed germination rates were 45.5% and 19.5%, respectively. The average time to germination was 70.2 days for A. princeps, and 46.8 days for D. zawadskii.

Gibberellin(GA) 3-$\beta$ hydroxylation is very important for the shoot elogation in the higher plants, since only 3$\beta$-hydryoxylated GAs promote shoot elogation in several plants. Fluctuation of 3$\beta$-hydryoxylase activity was examined during seed maturation using two cultivars of , P. vulgaris, Kentucky Wonder (normal) and Masterpiece (dwarf). Very immature seeds of both cultivars contain high level of 3$\beta$-hydroxylase activity (per mg protein). Both cultivars showed maximum of enzyme activity (per seed) in the middle of their maturation process. Gibberellin 3$\beta$-hydroxylase catalyzing the hydroxylation of GA20 to GA1 was purified 313-fold from very early immature seeds of P. vulgaris. Crude soluble enzyme extracts were purified by 15% methanol precipitation, hydrophobic interaction chromatogrphy, DEAE ion exchange column chromatography and gel filtration HPLC. The 3$\beta$-hydroxylase activity was unstable and lost much of its activity duting the purification. The molecular weight of purified enzyme was extimated to be 42, 000 by gel filtration HPLC and SDS-PAGE. The enzyme exhibited maximum activity at pH 7.7. The Km values for [2.3-3H] GA20 and [2.3-3H]GA9 were 0.29 $\mu$M and 0.33 $\mu$M, respectively. The enzyme requires 2-oxoglutarate as a cosubstrate; the Km value for 2-oxoglutarate was 250 $\mu$M using 3H GA20 as a substrate. Fe2+ and ascorbate significantly activated the enzyme at all purification steps, while catalase and BSA activated the purified enzyme only. The enzyme was inhibited by divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Effects of several GAs and GA anaogues on the putrified 3$\beta$-hydroxylase were examined using [3H]GA9 and GA20 as a substrates. Among them, GA5, GA9, GA15, GA20 and GA44 inhibited the enzyme activity. [13C, 3H] GA20 was converted by the partially purified enzyme preparation to [13C, 3H]GA1, GA5 and GA6, which were identified by GC-MS, GA9 was converted only GA4, GA15 and GA44 were converted to GA37 and GA38, respectively. GA5 was epoxidized to GA6 by the preparation. This suggests that 3$\beta$-hydroxylation of GA20 and epoxidation of GA5 are catalyzed by the same enzyme in P, vulgaris.

The seeds of Malus sieboldii were collected in late fall of 1988 and stored in cold stratification condition for 15 weeks. Growth regulators such as $GA_3$, BA and ABA were treated to the seed to know germination capacity and seedling growth. The results were as follows. 1) Higher germination was achieved in $GA_3$ and BA plots, But ABA treatment showed lower germination percentage and physiological dwarf and also induced to the secondary dormancy condition in partly. 2) Average length of time for germination was delayed over 2 days in ABA plot when comparing with the other plots. 3) Average length of plumule and radicle also was inhibited by ABA treatment. This means lower seedling vigor. 4) Fresh and dry weight of the seeding were higher in BA plot while ABA plot was lower and undesirable seedling was produced. From above mentioned results, it is believed that ABA treatment to the seed induced the secondary dormancy and physiological dwarf pattern. Therefore, utilization of BA can be produced the normal seedling, the authors believed.
This study was conducted to find proper material of brushing stimulation for height suppression of tomato plug seedlings. The tomato seeds were sown in 40-cell plug tray filled with commercial seedling medium and brushing stimulation was started at 18 days after sowing. Acrylic, polypropylene film, and weaving film were used by materials of brushing stimulation and, non-treatment and diniconazole treatment were used as the control. In acrylic treatment, the plant height was the shortest and the stem diameter was the thickest. Leaf growth was the lowest in diniconazole treatment. However, the SPAD value was the greatest in diniconazole treatment. The dwarf rate was the greatest in acrylic treatment. In acrylic treatment, the T/R ratio was the lowest and compactness was the greatest. In conclusion, use of the acrylic as material for brushing stimulation has higher dwarf rate than diniconazole treatment, and has advantages height suppression and seedling quality.

This study was conducted to clarify effects on the germination, survival rate of seedlings, growth and variation induction of variants in gamma rays irradiated seed of lilac pink (Dianthus superbus var. longicalycinus) and nursery spiraea (Caryopteris incana). Seed germination and its germination rate were delayed over 90 Gy for lilac pink and 80 Gy for nursery psiraea. Fifty percent lethal time (LT50) level for lilac pink and nursery psiraea were 150 Gy and 100 Gy, respectively. Lilac pink and nursery psiraea were grown dwarf over 120 Gy treatment. We have produced 17 variants of lilac pink in $M_1$ generation, and selected the promising 4 variants in $M_2$ generation. We have also made 7 variants of nursery psiraea in $M_1$ generation, and selected the favorable 8 variants in $M_2$ generation.

This study was conducted to select new variants with different characteristics in plant height, leaf color, and petal color of plantain lily (Hosta plantaginea) and Japanese farfugium (Farfugium japonicum). After 10-30Gy ${\gamma}-ray$ irradiation to seeds of both species, seed germination, variant induction, growth of variants, and survival ratios were estimated. Days required for seed germination were lengthened and % seed germination decreased as the dosage of ${\gamma}-ray$ increased over 20Gy. Lethal dose ($LD_{50}$) for plantain lily and Japanese farfugium were 30Gy and 25Gy, respectively. Plantain lily and Japanese farfugium treated by more than 20Gy ${\gamma}-ray$ showed dwarf characteristics in growth, and variations in leaf and petal colors. Total 166 variants in plantain lily were induced by ${\gamma}-ray$ irradiation in a $M_1$ generation, among which 12 promising variants were selected in a $M_2$ generation. Total 57 variants of Japanese farfugium were induced by ${\gamma}-ray$ irradiation in a $M_1$ generation, among which nine promising variants were selected in a $M_2$ generation.
