Evaluation of Various Slow-release Nitrogen Sources for Growth and Establishment of Poa pratensis on Sand-based Systems (모래지반에서 켄터키블루그래스의 성장과 조성에 미치는 질소의 유형별 효과)

  • Nitrogen (N) is one of the most important nutrients among 17 essential nutrients for maintaining turfgrass color and quality. The slow release fertilizers were initially developed to provide a more consistent release of nitrogen over a longer period and are often used to decrease leaching potential from sandy soils. The goal of this study is to determine if various slow release N sources affect the rate at which turfgrass establishes. Six nitrogen sources were evaluated; Nitroform (38-0-0), Nutralene (40-0-0), Organiform (30-0-0), Sulfur coated urea (SCU, 37-0-0), urea (46-0- 0), and Milorganite (6-0-0). The root zone media was seeded and sodded with 'Limousine' Kentucky bluegrass (Poa pratensis L.). Sodded pots produced 182 to 518 g more clipping dry weight than seeded pots. Among seeded pots, Milorganite produced greater amount of root dry weight than any other N sources. Because the period of turfgrass growth is different between sodded and seeded plots, there were differences on clipping yield and root growth. Overall, high N rate had turf color greater than acceptable color of 6 among seeded pots throughout the study. However, low N rate didn't produce acceptable turf color throughout the study. Based on the result of this tudy, ilorganite would be ecommended for new establishment of Kentucky bluegrass an urea with less clipping yield which can lead to reduce abor.

Production of Fungal Lipids (Part 2) Isolation of Starch Utilizing Mold and Its Optimum Compositions of Growth Media (곰팡이 유지 생산에 관한 연구 (제 2 보) 전분 이용성 곰팡이의 분리 및 배지조성에 관하여)

  • A potential fungal lipid producer from starch, which was identified as Muror plumbeus, was isolated from natural sources and its optimum cultivation condition for lipid production was investigated. The Mucor plumbeus FRI 0007 showed the highest felt weight and lipid content which were 2.09 $\pm$ 0.24g per 50$m\ell$ of medium and 37.43% on dry weight basis respectively after 20 days incubation on the medium containing 21% of starch as a carbon source. The urea was the best nitrogen source as compared with sodium nitrate, potassium nitrate, magnesium nitrate, ammonium nitrate and ammonium acetate and its optimum concentration was 2.14g/$\ell$, showing 2.39 $\pm$ 0.07 g felt/50$m\ell$ of medium and 50.73% lipid content on dry weight basis after 25 days incubation. Besides the starch as a carbon source and urea as a nitrogen source, the Mucor plumbeus FRI 0007 utilized ZnSO$_4$, MgSO$_4$, NaH$_2$PO$_4$, $K_2$SO$_4$and FeCl$_3$as mineral sources. However, it did not require ail the above 5 minerals in group in-dispensably for its growth and lipid accumulation. The lipid and economic coefficient of Mucor plumbeus FRI 0007 grown on the medium containing 0.44g $K_2$SO$_4$or 5.00g MgSO$_4$/$\ell$solely were 14.96 and 15.37 and 31.12 and 26.10 which was higher than those on the medium containing the above 5 minerals.

Effect of Root Medium Formulations on Growth and Nutrient Uptake of Hot Pepper Plug Seedlings (혼합상토의 조성이 고추 플러그 묘의 생육 및 무기원소 흡수에 미치는 영향)

  • This research was conducted to evaluate the effect of root medium formulations on growth and nutrient uptake of hot pepper 'Knockgwang' in 72-cell plug trays. To achieve this, the nine root media were formulated by adjusting blending rate of perlite (PL) to coir (CO), peatmoss (PM) or coir+peatmoss (5:5, v/v, COPM). Then, the growth characteristics and tissue nutrient contents were determined at 35 and 70 days after sowing. The elevated blending rate of PL to CO increased fresh and dry weight of hot pepper at 35 days after sowing. The treatments of 20% in blending rate of PL to PM or that of 0% to COPM produced the highest fresh and dry weight among perlite treatments of PM or COPM. The results of crop growth at 70 days after sowing also showed similar trends to those of 35 days after sowing. The elevated blending rate of PL to CO or PM decreased tissue $P_2O_5$ and K contents and increased Ca and Mg contents at 35 days after sowing, With the equal blending rate of perlite, the plant tissue grown in CO had higher K contents and lower N, Ca and Mg contents than those in PM and COPM. The elevated blending rate of perlite in three organic matter also decreased tissue $P_2O_5$ and K contents at 70 days after sowing, but Ca and Mg contents were the highest in the treatment of 20% PL in CO, 40% PL in PM and 40% PL in COPM among perlite treatments in each organic matter.

Growth, Deficiency Symptom and Tissue Nutrient Contents of Leaf Perilla (Perilla frutesens Britt) as Influenced by Potassium Concentrations in the Fertigation Solution (칼륨 시비농도가 잎들깨의 생육, 결핍증상 및 무기원소 함량에 미치는 영향)

  • This study was carried out to investigate the effect of potassium concentrations in fertigation solution on growth and development of nutrient deficiency symptoms of leaf perilla (Perilla frutesens). The nutrient concentrations in above ground plant tissue, petiole sap and soil solution of root media were also determined. Potassium deficiency symptoms developed in older leaves with marginal necrosis. The brown areas on the lower leaves enlarged rapidly and the margins became scorched. Elevation of K concentrations in the fertigation solution up to 8 mM increased the crop growth in leaf length, stem thickness, and fresh and dry matter production of above ground plant tissue. However, that decreased the chlorophyll contents. The 8.0 mM K treatment which showed the greatest growth had 5.01 g in dry weight and 2.76% in K content of above ground plant tissue, suggesting that maintaining K content higher than 1.7% is necessary for good growth of Perilla frutesens. The K concentrations in petiole sap and soil solution of 8.0 mM treatment were $12,289mg{\cdot}kg^{-1}\;and\;11.65mg{\cdot}L^{-1}$, respectively. These indicated that K fertilization to maintain higher than $8,700mg{\cdot}kg^{-1}$ in petiole sap and $4.5mg{\cdot}L^{-1}$ in soil solution are necessary to ensure good crop growth.

  • A laboratory experiment was conducted to find out the effect of inoculation with plant growth of associative $N_2$-fixing bacteria on the combination of eight different rice seedling with seven different associative $N_2$-fixing bacteria in which were isolated and identified from grasses and rice in saline and reclaimed saline paddy soil. Those were examined in N-free axenic culture medium. The result of this experiment exemplified the contribution of associative $N_2$-fixing bacteria to their host plant were varied much by the combination of rice cultivars and bacterial strains. Acetylene reduction ($N_2$-fixation) activity seems to be more variable with respect to the combination of rice cultivars and bacterial strains. The relationship between acetylene reduction activity and rice performance such as dry matter weight, plant height, root length, and number of root were statistically insignificant. However, in comparision with the control, the dry matter yield of Annapuruna was increased by inoculation of all the seven bacteria strains. Among the seven rice cultivars, the average acetylene reducing activity was the highest in Annapuruna, and Shingwang, and the lowest in IR-8 inoculated with Pseudomonas sp H8. Among the seven bacterial strains the highest acetylene reducing activity was obtained by the combination of Shingwang with Pseudomonas sp H8.

Influence of Paper Mill Sludge and Briquet Ash on the Growth of Zoysiagrass (제지(製紙)슬러지와 연탄재가 들잔디의 생육(生育)에 미치는 영향(影響))

  • To investigate the potential of paper mill sludge and briquet ash as cultural media in turfgrass, zoysiagrass was grown in the mixtures prepared with paper mill sludge and briquet ash. The mixtures were consisted of sludge and briquet ash in the ratio by volume 3:1, 2:1, 1:1, 1:2, and 1:3 in the order. To compare the growth responses, some plants were grown in the medium containing sand(3), field soil(1) and peatmoss (1) by volume anti regarded as control. 1. Activated sludge mixed with sand increased plant height, fresh weight, and dry weight more than 1.5-2.0 times compared to those of the control. The highest plant growth was shown in mixtures containing 67% activated sludge. 2. Plant density per $100cm^2$ and chlorophyll content were higher in all mixtures containing activated sludge than control. 3. The growth of zoysiagrass was reduced along with the increase of non-activated sludge ratio, but no difference was found in chlorophyll content. 4. Plant height, fresh weight and dry weight were greater in activated sludge combined with sand compared to the briquet ash mixtures, but the difference of shoot density between two mixtures was not shown. 5. Even though non-activated sludge appeared not to be appropriate to the growth of seedlings, the number of tillers of plant propagated with rhizome was more increased compared to control when briquet ash content was less than 75%.

  • The effect of metallothionein expression on the metal resistance and removal by recombinant Saccharomyces cerevisiae containing the plasmid pJW9 was investigated. The recombinant strain S. cerevisiae BZ-pJ was constructed by transforming the host strain S. cerevisiae BZ3l-1-7Ba with the gene coding for a metal-binding protein, metallothionein. Introduction of the MT gene yielded an increase in the minimum inhibitory concentration (MIC) of copper more than three times compared with the host strain. The minimum inhibitory concentrations of $Cr^{2+}, Znr^{2+} and Pb^{2+}, $ were not different for the two strains. The recombinant yeast grown in a medium containing 8mM CuSO4 was able to remove copper with a capacity of 18.9mg $Cu^{2+}$/g dry cell. In a mixture of copper and zinc, the presence of copper relieved the toxic effects caused by zinc, resulting in an enhancement of the final cell density and the specific growth rate of the recombinant yeast. The capability to remove copper by the recombinant yeast was linearly proportional to the copper concentrations in the medium. The efficiency of copper removal was rather constant regardless of the initial copper concentrations. The specific removal of zinc was dependent on the zinc concentrations in media, though, and such dependence was not so pronounced as the concentration of copper.

Studies on the Growth of Excised Root between Indica- Japonica Hybrids and Japonica Type in Paddy Rice (통일형수도와 일본형수도에 있어서 종근.분리근의 생육에 관한 연구)

  • The experiments were carried out to compare the growth characteristics of excised seminal roots in Japonica type, Indica- Japonica hybrids and its parents. Based on the modified culture medium by Kawata et al., several levels of pH. sucrose and casamino acid were varied in the culture media. The excised root length at 27$^{\circ}C$ incubator was little different between Japonica type and Indica-Japonica hybrids, but at 30$^{\circ}C$ incubator the excised root elongation of Indica-Japonica hybrids was much better than that of Japonica type. The dry matter weight of excised root in Indica- Japonica hybrids was significantly heavier than in Japonica type at both 27 and 30$^{\circ}C$ incubators. The difference of dry root weight between Japonica type and Indica- Japonica hybrids was caused from the number of lateral roots, especially the root numbers of 25 cm length. The main and lateral seminal root growth of Japonica type grew best at the lot of 0.2 percent casamino acid. However, the lots of 0 and 0.2 percent casamino acid showed good results in the main and lateral excised root growth. respectively of Indica-Japonica hydrids. The growth of excised roots was better in Indica-Japonica hybrids than in Japonica type at even various levels of pH, sucrose and casamino acid. Between the parents of Indica - Japonica hybrids, the excised root growth of Indica type was more predominant than that of Japonica type, and the effect of casamino acid on the excised root growth in the formar was similar to that of Indica type. This result indicated that the growth characteristics of excised seminal roots in Indica Japonica hybrids were derived from the parent, Indica type.

Effect of Irrigation Frequency on Growth of Tomato Plug Seedlings in Media Containing Cellular Glass Foam and Peatmoss (Cellular Glass Foam과 Peatmoss를 함유한 혼합배지에서 관수빈도가 토마토 플러그묘의 생육에 미치는 영향)

  • Cellular glass foam (CGF), the reprocessed glass, has a possibility to be used as a medium component in plug culture of horticultural crops due to the its excellent air and water permeability as comparable to perlite. An experiment was conducted to investigate growth of plug seedlings of Lycopersicum esculentum 'Segye' as influenced by irrigation frequency in various medium combinations of CGF (2.0-4.0 mm particle size) and peatmoss. Seeds were sown in 200-cell plug trays, filled with mixtures of CGF and peatmoss either at 33:67 or 25:75 (%. v/v) and were germinated on a fogged propagation bed. The irrigation frequencies used were one, two or three times per every two days. A commercial plug medium (Tosilee medium) was used as the control, and the irrigation frequency in the control was one time per day. Growth of seedlings, and medium pH and EC were measured at 33 days after sowing. The medium composition had little influence on overall growth of seedlings. Irrigation frequency very significant affected number of leaves, leaf area, chlorophyll concentration, fresh and dry weights of shoots and roots, and dry matter. Growth of seedlings was the greatest with the highest irrigation frequency in the 25% CGF+75% peatmoss mixture.

Use of Phenolic Foam as a Medium for Production of Plug Seedlings of Paprika (Phenolic Foam 배지를 이용한 파프리카의 플러그묘 생산)

  • The study was conducted in a glasshouse to examine the possibility of producing paprika plug seedlings in a newly developed inert phenolic foam growing medium. Plug seedlings of 'Fascinato' paprika were grown in five media, Grodan rockwool (Grodan Co. Ltd., Denmark), UR rockwool (UR Co. Ltd., Korea), phenolic foam LC (Smithers Oasis Korea Co. Ltd., Korea), phenolic foam RC, and phenolic foam 3813-4 all in a pellet type. Seeds were germinated in a growth chamber ($25^{\circ}C$, 90% RH, dark) for 4-5 days and then seedlings were grown in a glasshouse with nutrient solution supplied by an overhead irrigation system. Seedling growths were measured 20 days after sowing. The medium pH was the highest in the Grodan rockwool, and medium EC was the highest in phenolic foam 3813-4, although no nutritional excess disorders were observed. Germination rates of paprika were higher than 90% in all the media. Plant height, stem diameter, T/R ratio, leaf area, and chlorophyll showed a similar to those in the rockwool medium. Number of leaves, length of the longest root and dry weights of shoot were not significantly different among treatments. Overall, phenolic foam LC and RC produced seedlings with a similar growth as the rockwool medium. The results obtained suggest that rockwool can be replaced with a new material such as phenolic foam in the commercial scale production of plug seedlings of 'Fascinato' paprika.
