Effects of Barely Processings on the Reduction of Deoxynivalenol (보리 가공에 의한 Deoxynivalenol의 감소 효과)

  • This study examined the effects of pearling, simple heating, baking and extrusion in processing of barley contaminated with deoxynivalenol (DON) on the reduction or destruction of DON. The DON contamination level of barley used in the experiment was 2.08 ppm. The DON-forming strain isolated from the barley was Fusarium graminearum, and the DON formation potential of which was 6.3 ppm on the average in Czapek-Dox broth medium. The DON removal efficiency of pearling was 56.5%, simple heating 3.8-32.7%, extrusion 53-59%, and baking 10.4%. As a large quantity of DON still remains after processing, it tis necessary to develop a new method of removing DON comletely.

Influence of Dissolved Organic Nitrogen on Organic Chloramine Formation during Chlorination (염소 소독시 DON이 유기성 클로라민 생성에 미치는 영향)

  • Although formation of organic chloramines have been studied for decades, most of them have involved model organic compounds (e.g., amino acids) but not naturally occurring organic nitrogen in water. This study investigated formation of organic chloramines during chlorination of 16 natural organic matters (NOM) solutions which were isolated from surface water and contained dissolved organic nitrogen (DON). Organic chloramine yields per chlorine consumption was $0.25mg-Cl_2/mg-Cl_2$. Upon chlorination of NOM solutions, organic chloramines were rapidly formed within 10 minutes. The average organic chloramine yields upon addition of chlorine in to NOM solutions were $0.78mg-Cl_2/mg-DON$ at 10 minutes and $0.16mg-Cl_2/mg-DON$ at 24 hours. Organic chloramine yields increased as the dissolved organic carbon/dissolved organic nitrogen (DOC/DON) ratios decreased. Chlorination of molecular weight (10,000 Da) fractionated samples showed that the influence of DON molecular weights on the organic chloramine formation was minimal.

Effects of Deoxynivalenol Reduced Barley Flours on Breadmaking Properties (Deoxynivalenol을 감소시킨 보릿가루의 첨가가 제빵 특성에 미치는 영향)

  • Deoxynivalenol (DON)-contaminated barley was treated with 0.1 M $Na_2CO_3$ solution to reduce DON content. DON content of barley was reduced from 2.08 to 0.67 ppm. Bread was made with 10, 20, and 30% DON-reduced barley flour added to white wheat flour. Farinogram showed water absorption and arrival time increased, while stability and mechanical tolerance index decreased when DON-reduced barley flour was added to white wheat flour. Gelatinization temperature, temperature at maximum viscosity, and maximum viscosity increased in amylogram with increasing addition of DON-reduced barley flour. Loaf volume of bread decreased with increasing amount of DON-reduced barley flour, while loaf weight increased. Barley flour pH increased by treatment with$Na_2CO_3$, and pH reduction decreased fermentation rate of yeast. Volume and size of gluten matrix decreased and crumb hardened in bread made with DON-reduced barley flour. Acceptabilities for color and texture were low in bread made with DON-reduced barley flour. Addition of DON-reduced barley flour at 30% reduced overall acceptability, whereas no significant difference in overall acceptability was observed when DON-reduced barley flour was added at 10 and 20%.

Removal of Dissolved Organic Nitrogen from Surface Water and Reclaimed Water by Coagulation (지표수 및 재이용수내 용존 유기질소의 응집처리)

  • During chlorination processes dissolved organic nitrogen (DON) can form toxic nitrogenous disinfection byproducts and organic chloramines which have little or no bactericidal activity. DON needs to be removed before chlorination processes to reduce the formation of those products. This study investigated the removal of DON from surface water and reclaimed water by coagulation with aluminum sulfate (alum) and a cationic polymer (polyDADMAC). Removal characteristics of dissolved organic carbon (DOC) and ultraviolet absorbance at 254 nm ($UVA_{254}$) were compared with that of DON. Coagulation with alum removed DON, DOC, and $UVA_{254}$ with similar trends, but the removal of $UVA_{254}$ was highest. A dual coagulation strategy of alum and cationic polymer improved the removal of DON. Coagulation with cationic polymer alone was not effective due to its narrow range of charge neutralization. DON in reclaimed water was easier to remove than that in surface water, and higher molecular weight fraction (>10,000 Da) of DON was preferentially removed.

Screening of Deoxynivalenol Producing Fungi from Greenhouse Horticulture by Enzyme Amplification System Immunoassay (Enzyme Amplification System Immunoassay에 의한 시설원예산물의 Deoxynivalenol 생성곰팡이의 검색)

  • In order to evaluate the safety of greenhouse horticulture products in Korea, we carried out this work by screening of Fusarium species, which produce deoxynivalenol (DON) from greenhouse horticulture in Western Gyeongnam and Northern Gyeongbuk, Korea. For this study, high sensitive enzyme-linked immunosorbent assay, ALP/NADP method, was applied to detection of DON by enzyme amplification system. From 192 samples of greenhouse horticulture soil and its products, 103 isolates of Fusarium species were obtained. The isolates were cultured at 28C for 15 days and the cultured mediums were extracted by ethyl acetate. The production of DON was verified by thin layer chromatography (TLC). As the results of TLC, 8 strains were identified as DON producing strain. We screened potential producers of DON by ALP/NADP. The levels of DON production were shown from 0.007 to 1.21 g/ml of YES medium. The maximum DON producing strain No. 32-D-3 was isolated from soil in Namhae, Korea. In conclusion, the above results indicate that DON producing fungi contaminated greenhouse horticulture products in Korea. Therefore, further studies are required to accumulate more detailed data about the contamination of DON in various cereals.

옥수수중 Deoxynivalenol의 검출을 위한 효소면역측정법의 개발

  • In order to develop an enzyme-linked immunosorbent assay (ELISA) for deoxynivalenol (DON) in com, we produced a specific monocl- onal antibody and established ELISA conditions. After the spleen cells from mice immunized with DON-bovine serum albumin conjugate were fused with S$_{p}$2/0 myeloma cells, a hybridoma cell 3G7 producing anti-DON antibody was screened by ELISA. From the standard curve of competitive direct ELISA (cdELISA) using 3G7 monoclonal antibody and DON-HRP conjugate, the detection range of DON showed 3-3,000 ng/ml (ppb). The monoclonal antibody showed some cross-reactivities against DON analogues such as 15 acetyl-DON (110%), nivalenol (5.0%), 3 acetyl-DON (1.7%), fusarenon-x (0.72%), and T-2 (0.59%). When the cdELISA was applied to the spiked coms after extracting with 60% methanol and diluting 5- fold with washing buffer, the assay recoveries of DON were 313, 163, 106, and 88.9% (av., 168%) in the levels of 200, 600, 2,000, and 6,000ng/g, respectively. For the quantitation of DON in coms, 30 samples kept under two different storage conditions of cold and room temperature were assayed by cdELISA. The mean detection concentrations were 595 (detection range, 0-2,750) and 2,448 (detection range, 0-4,500) ppb, respectively.

Screening of Deoxynivalenol Producing Strains from Agricultural Products by Immunoanalytical Method (면역분석기법에 의한 농산물에서의 Deoxynivalenol생성균 검색)

  • In order to evaluate the safety of agricultural products in Korea, we carried out work by screening of Fusarium species. which can produce deoxinivalenol(DON) from agricultural products in Western Gyeongnam, Korea. From 215 samples of soil agricultural products, 129 strains of Fusarium species were obtained. The production of DON was verified by thin layer chromatography(TLC) As the results of TLC, 25 strains were identified as DON producing strain. But only 10 strains were identified as DON producing strains by enzyme-linked immunosorbent assay (ELISA). The maximum DON producing strain No.41 was isolated from corn. In conclusion. the above results indicate that DON producing fungi contaminated agricultural products in Korea. Therefore further studies are required to accumulate more detailed data about the contamination of DON in various agricultural products.

Paying Back to Good Deeds: A Text Mining Approach to Explore Don-jjul as Pro-consumption Behavior

  • More consumers are choosing pro-consumption for social change, but scholars know little about why and how consumers engage in pro-consumption behaviors. A newly emerged pro-consumption behavior called "Don-jjul," which appeared during the COVID-19 pandemic in South Korea, refers to compensating businesses that have engaged in altruistic actions by boosting their sales. This study used Latent Dirichlet Allocation (LDA) of topic modeling, sentiment analysis, and in-depth interviews to investigate the perceptions, motivations, and emotions regarding Don-jjul. As a result, the study revealed pro-consumers' perceptions of Don-jjul as "collective pro-consumption for contributing to social well-being." Don-jjul has two main motives: "supporting underdogs with difficulties" and "compensating good businesses economically." We also found two ambivalent emotions evoked by Don-jjul: "respect for good business owners" and "concerns regarding the misuse of Don-jjul." The results contribute to pro-consumption research for social well-being, providing business opportunities for retailers and CSR managers with a deep understanding of pro-consumers.

Inhibitory effect of the extract of Catalpa ovata G. Don. on endothelial adhesion molecule expression (개오동나무 추출물의 내피세포 부착분자 발현 억제 효과)

  • Objectives : Catalpa ovata G. Don (Bignoniaceae) has been shown to possess a variety of pharmacological activities. However, the effect of Catalpa ovata G. Don on endothelial adhesion molecule expression has not been reported. Methods : To examine the effect of Catalpa ovata G. Don on the expression of adhesion molecules in human umbilical vein endothelial cells (HUVECs) stimulated with tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), we used various methods such as Western blot analysis, reverse tranascription-polymerase chain reaction (RT-PCR), and luciferase activity assay. Results : 1. The extract of Catalpa ovata G. Don inhibited the expression of intracellular adhesion molecule-1 (ICAM-1) and vascular cell adhesion molecule-1 (VCAM-1) in HUVECs stimulated with TNF-${\alpha}$. 2. The extract of Catalpa ovata G. Don reduced TNF-${\alpha}$-induced adhesion of leukocytes to HUVECs. 3. In addition, The extract of Catalpa ovata G. Don inhibited the promoter activities of ICAM-1 and VCAM-1. Conclusions : These results that Catalpa ovata G. Don may be beneficial in the treatment of inflammatory such as atherosclerosis.

Formation of Don-am district and Adaptation of Hanok Area in Seoul (돈암지구 가구(街區)의 형성과 도시한옥의 적응)

  • The Don-am district is a residential area that was supplied in 1936 as a land readjustment project(Tojiguhoekjeongri). The Don-am district was newly supplied with residential areas and urban hanoks were built in large numbers. The Don-am district was influenced by urban planning and legislation at that time. These affected in the layout and plan of urban hanok. Residential block in the Don-am district were developed sequentially from the late 1930s to the 1960s. Residential block were divided by modern construction company and sold by individual lots. The blocks supplied to the Don-am district made uniformly the corner out-off(Ga gak) for creating a vehicle-centered road. So urban hanoks located in the corner plot was transformed in response to the road. Residential blocks in Don-am district was divided into three to four rows. Therefore, alleys were created inside the block. Newly made alleys consist of a privately owned road(Sa-do), a public road(Gong-Do), and open space in the site. And the alleys were used as an entry space for sharing with neighboring. Urban hanoks of Don-am district have had changed and adapted to the formation of these alleys.
