고온가열된 어류의 돌연변이성 검색을 위한 시료 추출방법 (Method for the Detection of Mutagenicity of Fried Fish)

  • A method was developed to detect total mutagenicity of fried fish for S. typhimurium TA98, using Ames assay. Method described herein circumvented problems associated with the sample preparation for Ames assay, i.e., a multi-purification step of sample and interference with solvent residuals. Experiment A, the best method developed in the present study, consisted of two important steps: pH adjustment of the aqueous sample solution from fried fish samples to remove impurities, and simultaneous distillation extraction (SDE) for partially purified samples to remove volatile compounds from solvents. The procedure and results were described as below. Fillet of gizzard shad (Konosirus punctatus) fish sample fried for 10 min each side on the temperature-controlled fry-pan (210$\circ$C) was homogenized in an aqueous acidic solution (pH 2) with a homogenizer, followed by filtration through Celite. The tiltrate (pH 2), removed some impurities by extraction with chloroform:methanol (2:1, v/v) mixture, was adjusted pH to 10 and then centrifuged to remove precipitate. The ethylacetate extract from the tiltrate of pH 10 was rotoevaporated and purified by SDE apparatus for 2 hours. Experiment A revealed significantly higher revertants (1928 per 25 g fried sample) than other Experiment (B, C, or D) tested. Experiment A gave good results in the mutagenicity test of fried fish sample with few purification steps using only 25 g fried sample and 650 ml of solvents; and thus this method could be a useful tool for the screening the mutagenicity or antimutagenicity of other foods as well.

Rational Design of Extractive Distillation Toward Enhanced Separation of HFPO from HFP/HFPO Mixture

  • Hexafluoropropyleneoxide ($C_3F_6O$, HFPO) is highly expensive and it may be used as a raw material for the synthesis of various fluorine based compounds. Currently, extractive distillation method has gained considerable attention to collect the HFPO from a mixture of HFPO / hexafluoropropylene ($C_3F_6$, HFP). Optimized operating conditions are studied using a theoretical method for the extraction process. Among available solvents for the purification process, the use of 1,1-dichloro-1-fluoroethane exhibits a high purity of HFPO as a top product and minimize the required heat duty. Since the boiling point of the solvent increases as the pressure in the column increases, the enhanced extractive capability of the solvent led to the high purity of HFPO at the high pressure.

Exreaction of Thiosulfinates from Garlic Using Gas-in-Liquid-Dispersion

  • Extraction yields of thiosulfinates from garlic were studied by using gas-in-liquis-dispersion(GLD) system to maxmize the thiosulfinate extraction. Extracted thiosulfinates were spectrophotometically quantified using synthesized dially1 thiosulfinate. The conditions for maximum extraction were optimized by response surface methodology. The volatile components extracted by various methods, such as hexane extraction, simultaneous steam distillation and GLD system, were compared by using gas chromatography. The results indicated that the thiosulfinate yield was increased by incresaing temperature and nitrogen gas flow rate, while the effects of bubble sizes on thiosulfinate extraction were not significant at the ranges tested. Application of GLD system resulted in extraction of more volatile components than other extraction methods, Therfore, it was suggested that GLD system was one of the efficient extraction methods among the ever introduced ones, for thiosulfinate extraciton.

초임계유체 추출과 SDE에 의한 솔 추출물의 휘발성분 비교 (A Comparison of Volatile Compounds in Pine Extracts Obtained by Supercritical Fluid Extraction with Those by Simultaneous Steam Distillation and Solvent Extraction)

  • 본 연구에서 행하여진 온도$(35,\;55^{\circ}C)$ 및 압력(100, 300bar) 범위에서 초임계 추출된 솔 추출물의 수율은 300bar, $35^{\circ}C$와 300 bar, $55^{\circ}C$의 경우 각각 5.0%와 5.3%이었으며 100 bar, $35^{\circ}C$와 100 bar, $55^{\circ}C$에서 각각 3.7% 및 2.8%이었다. 300 bar에서는 wax류 등의 추출로 인하여 얻어진 추출물들은 상온에서 고체 상태이며 노란 색이었고, 100 bar에서 얻어진 추출물은 모두 액체 상태이고 무색에 가까웠다. 관능검사 결과 100 bar, $35^{\circ}C$에서의 초임계 추출물이 SDE 추출물보다 1% 유의수준에서 우수함이 판명되었다. 그러나 100 bar, $35^{\circ}C$에서의 추출물과 300 bar, $55^{\circ}C$ 추출물 간에는 5% 유의수준에서 차이가 없었다. 가장 많이 함유되어 있는 휘발성 향기 성분은 limonene으로써 $32.6{\sim}43.4%$를 차지하였고 ${\beta}-pinene(10.8{\sim}18.5%),\;{\beta}-myrcene(11.5{\sim}17.3%),\;{\alpha}-pinene(5.3{\sim}11.7%)$의 순이었다. 한편 향의 품질 특성에 결정적인 역할을 하는 것으로 알려진 함산소 terpene의 경우 100 bar, $35^{\circ}C$의 경우가 가장 많아서 3.6%를 차지하였으며 300 bar, $35^{\circ}C$의 경우에도 3.4%로서 300 bar, $55^{\circ}C$의 2.5%, 100 bar. $55^{\circ}C$의 1.8%보다 많았다. SDE 추출물의 경우는 6.7%를 차지하여 초임계 추출물보다도 훨씬 많은 양을 나타내었다.

해안가에 자생하는 순비기나무 열매의 증류추출을 통한 하이드로졸 성분분석 (Analysis of Hydrosol Components through Distillation Extraction of the Sunbigi Tree(Vitex rotundifolia L.f.) Fruit of the Wild Birch Tree Native to the Coast)

  • 우리나라 남부와 서해 해안가를 중심으로 자생하고 있는 순비기나무는 자원 활용 측면에서 많은 활용 가치를 가지고 있다. 예로부터 민간에서 민간약재로 이용하거나 한약재로 사용되었다. 잎과 줄기에서도 강한 향이 나지만 열매에서도 강한 향이 나서 순비기나무 열매를 이용해 에센스오일을 추출하여 성분을 분석한 연구는 일부 있지만 하이드로졸을 추출하여 이에 대한 향기성분의 연구는 거의 없다. 이유는 열매가 딱딱하여 일반적인 추출방법으로는 함유된 유효 성분을 확인하는 것이 어렵기 때문이다. 따라서 본 연구는 추출방법을 달리하여 딱딱한 열매를 고온으로 추출하여 얻은 하이드로졸의 향에 함유된 성분을 분석한 결과는 다음과 같다. 1. 플라보노이드 함량이 가장 많은 추출조건은 ethanol로 30.57mg/g 이었고 다음으로 열수추출물로 18.26mg/g, 물 추출물이 가장 낮은 9.69mg/g이었다. 2. 순비기나무 열매를 증류추출한 결과 하이드로졸의 향기는 3-Methyl-2-butenoic acid, cyclobutyl ester, Eucalyptol, L-alpha-Terpineol, 1H-Cycloprop[e] azulen-7-ol, decahydro-1,1,7-trimethyl-4-methylene-, [1ar-(1a.alpha.,4a.alpha.,7.beta.,7a.beta.,7b. alpha.)]가 많은 것으로 나타났다.

에탄올의 농도가 계피가 향기성분 용출에 미치는 영향 (Effect of Ethanol Concentration on Extraction of Vlolatile Components in Cinnamon)

  • In order to select the optimum ethanol concentration for extraction of volatile components in cinnamon, the dried cinnamon was extracted with water and 30∼90% ethanol. The volatile components of cinnamon extracts were isolated by the simultaneous distillation extraction method using Likens and Nickerson's extraction apparatus, and analyzed by GC-MS. In cinnamon bark powder 45 components were detected and 21 components were identified. The major component of cinnamon bark powder was cinnamic aldehyde. In water extract of cinnamon, volatile components were not extracted sufficiently. The volatile components of cinnamon were increased with the increment of ethanol concentraction upto 70%. The volatile component of 70% ethanol extract showed similar pattern and amount to cinnamon bark powder. But in 90% ethanol extracts, the number and amount of volatile component were reduced. The above data suggested that 70% ethanol was the most effective solvent for volatile components extraction of cinnamon.

능이버섯의 향기특성 (Aroma Characteristics of Neungee(Sarcodon aspratus))

  • 능이버섯의 향기성분을 조사하기 위하여 SDE, SFE, headspace 로 추출하고 GC, GC-MS로 동정하였으며, 능이버섯의 향기특성을 조사하기 위하여 GC-olfactometry 방법중 AEDA법을 이용하였다. 그 결과 SDE 추출물에서는 33개, SFE추출물에서는 26개, headspace 추출물에서는 17개의 화합물이 동정되었다. 확인된 화합물의 주성분은 1-octen-3-ol, 1-octen-3-one, 3-octanone, 2-octen-1-ol, 3-octanol, 1-octanol 등의 $C_8$ 화합물과 benzeneacetaldehyde등이었다. AEDA 결과 능이버섯의 주된 향기특성으로는 생능이버섯내(unknown), 생버섯내(1-octen-3-ol), 곰팡이내(1-octen-3-one), 흙내(3-octanol), 풀내(1-octanol), 강한 풀내(2-octenal), 단 꽃내(benzeneacetaldehyde), 목재내(L-linalool), 치즈내(3-hydroxy-2-butanone), 상큼한 단내(2-decanone), 약간의 불쾌한 고기내(3-methyl thiopropanol), 시큼한 우유내(propanoic acid) 등으로 나타났다.

추출방법에 따른 편백 정유의 향기 성분 비교 및 아토피 개선에의 응용 (Comparison of Volatile Compounds of Chamaecyparis obtusa Essential Oil and its Application on the Improvement of Atopic Dermatitis)

  • Volatile flavor compounds of Chamaecyparis obtuse essential oil were extracted by simultaneous steam distillation extraction (SDE) and supercritical fluid extraction (SFE) and analyzed by GC-MS. A total of 48 and 50 components were identified in essential oil by SDE and SFE, respectively. Monoterpenes, oxygenated monoterpenes, sesquiterpenes, oxygenated sesquiterpenes, and diterpenes in essential oil by SDE were 37.24, 10.9, 9.61, 0.22, and 0.22%, respectively. In the case of SFE, they were 19.1, 23.3, 22.66, 1.31, and 10.57%, respectively. Antioxidant activities were increased with the increase of essential oil up to $80{\mu}L/mL$, irrespective of extraction method. Especially, when the essential oil concentration extracted by SDE was increased from 20 to $80{\mu}L/mL$, the antioxidant activity was increased from 10.5 to 55.1%. However, over $80{\mu}L/mL$ of essential oil, an equilibrium state was maintained. In the case of essential oil extracted by SFE, it was decreased compared to that of SDE. For the improvement of atopic dermatitis, various cosmetics such as an ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE were tested. About over 90% was useful for the improvement of atopic dermatitis after 4 weeks of clinical trial targeting 40 female adults. These results demonstrate that ato-cide soap, ato-cide spray, and ato-cide lotion containing essential oil extracted by SFE could be used in functional cosmetics.

초임계 이산화탄소를 이용한 유자 과피로부터 향기성분의 추출

  • 유자과피의 향기성분 중 대부분을 차지하는 limonene은 추출물의 함량을 결정하는데 많은 영향을 준다. 일반적으로 Citrus속의 과실류의 향기 성분들 중 가장 많이 함유되어 있는 limonene의 경우 초임계 이산화탄소에 대한 용해도는 압력 140${\sim}$150 bar, 온도 $35{\sim}40^{\circ}C$일 때 최대이고 유자 또한 다량의 limonene을 함유한 과실류로서 초임계 이산화탄소 압력 2000 psi와 온도 $40^{\circ}C$에서 향기성분 중 limonene의 추출율이 향상되어 함량이 786.6088 mg/g으로 최대로 나타났다.

Solid Phase Micro Extraction을 이용한 산초의 휘발성 성분 분석 (Analysis of Volatile Components of Sancho(Zanthoxylum schinifolium)by Solid Phase Micro Extraction)

  • Test of the optimum condition of solid phase micro extraction(SPME) was performed by use of 5 vol-atile components in dilute aqueous solution. Volatile components of Sancho(Zanthoxylum schinifolium) were isolated by SPME method and were analyzed by GC/MSD and compared with volatile compone-nts isolated by simultaneous distillation extraction (SDE) method. Total 31 components were identified by comparing gas chromatography retention time and mass spectral data. The major compounds were limonene geranyl acetate $\beta$-phellandrene phellandral mycene linalool rose oxide caproic acid and caprylic acid SPME sampling procedure was found to be a good method for qualitative analysis of the volatile components.
