Nitrogen Management with Split Application of Urea for Direct-Seeding Rice in Wet Paddy

  • Direct-seeding has major advantages such as labor and cost saving by eliminating preparation of seed bed and transplanting. But, it required increased input of fertilizers and pesticides because of the extended paddy period. Direct seeding in wet paddy (DSWP) gives faster growth and more uniform seedling emergence than direct-seeding in dry paddy. This research had an objective to develop an efficient N management practices for DSWP with split application of N fertilizer. A paddy field experiment was conducted to evaluate effects of starter N and N-topdressing which was delayed N application until 5-leaf stage, with comparison to transplanting (TP). Total amount of N application were two levels; 110kg and 77kg/ha. The N applications were split four times during rice growth stages; starter, topdressing at 5-leaf stage, top dressing at tillering stage, and topdressing at panicle initiation stage. DSWP had more tillers/$m^2$ than TP, but with the delayed heading. The DSWP plots which received N-topdressing at 5-leaf stage without starter N had higher leaf area index (LAI) and leaf greenness than the TP plot. Also, these DSWP plots had high leaf-N concentration at the heading stage, as calculated from leaf chlorophyll meter readings. Rice yield in DSWP with N-topdressing at 5-leaf stage was significantly higher than that in TP and in DSWP with starter N. Energy and N use efficiency were improved in DSWP with N-topdressing at 5-leaf stage. But, there were no significant differences in grain yield between the two levels of total amounts of N applications, 77kg and 110kg/ha. We concluded that starter N could not be used effectively by rice seedlings, but topdressing N at 5-leaf stage was an efficient N management for rice growth and yield in DSWP system.

Effect of Nitrogen Split Application Methods under Different Soil Textures on Growth and Yield of Rice in Direct Seeding on Dry Paddy (벼 건답직파재배에서 토성별 질소분시방법이 생육에 수량에 미치는 영향)

  • This experiment was carried out at National Crop Experiment Station in 1994 to obtain basic information of growth characters and yield of rice under various nitrogen split application methods on different soil textures in direct seeding on dry paddy. Hwaseongbyeo sown on April 27 by flat drill seeding, and irrigation was done at 3 leaf stage after seedling establishment. Number of seedling stand was 159~177 seedlings per $m^2$ regardless of soil texture and nitrogen application method. Number of panicle per unit area in loam was higher than in sandy loam, and it also was higher in top dressing plots, which were 3 times application at rate of 40-30-30% (3rd leaf stage -7th leaf stage -panicle initiation stage) and 4 times application 10-30-30-30%(basal-3rd leaf stage -7th leaf stage -panicle initiation stage), than in conventional method. Leaf colour, leaf area index and dry matter production at heading stage were highest at top dressing plots among the nitrogen application methods in both sandy loam and loam. Lodging index in top dressing plots increased by low breaking weight with long culm. There were field lodging of degree 3 in top dressing plots. Rice yield in sandy loam, loam increased by 7~9%, 6~9% in top dressing of nitrogen, respectively.

  • The mechanical behavior of the brittle material samples containing the internal and edge cracks are studied under direct shear tests. It is tried to investigate the effects of stress interactions and stress intensity factors at the tips of the pre-existing cracks on the failure mechanism of the bridge areas within these cracks. The direct shear tests are carried out on more than 30 various modeled samples each containing the internal cracks (S models) and edge cracks (E models). The visual inspection and a low power microscope are used to monitor the failure mechanisms of the tested samples. The cracks initiation, propagation and coalescences are being visualized in each test and the detected failure surfaces are used to study and measure the characteristics of each surface. These investigations show that as the ratio of the crack area to the total shear surface increases the shear failure mode changes to that of the tensile. When the bridge areas are fixed, the bridge areas in between the edge cracks have less strength than those of internal cracks. However, the results of this study show that for the case of internal cracks as the bridge area is increased, the strength of the material within the bridge area is decreased. It has been shown that the failure mechanism and fracture pattern of the samples depend on the bridge areas because as the bridge area decreases the interactions between the crack tip stress fields increases.

Direct somatic embryogenesis, plant regeneration and genetic transformation of Panax ginseng

  • Somatic embryogendesis is one of good examples of the basic research for plant embryo development as well as an important technique for plant biotechnology. This paper describes the direct somatic embryogenesis from zygotic embryos of Panax ginseng is reversely related to normal axis growth of zygotic embryos by the experiment of various chemical treatments. Under the normal growth condition, the apical tips of embryo axis produced an agar-diffusible substance, which suppressed somatic embryo development from cotyledons. Although the cells of zygotic embryos were released from the restraint of embryo axis, various factors were still involved for somatic embryo development. Electron microscopic observation revealed that the ultrastructure of cells of cotyledon epidermis markedly changed before initiation of embryonic cell division, probably indicating reprogramming events into the cells embryogenically determined state. Polar accumulation of endogenous auxin or cell-cell isolation by plasmolysis pre-treatment is the strong inducer for the somatic embryo development. The cells for the process of somatic embryogenesis might be determined by the physiological conditions fo explants and medium compositions. Direct somatic embryos from cotyledons fo ginseng were originated eithrer from single or multiple cells. The different cellular origin of somatic embryos was originated either from single or multiple cell. The different cellular origin of somatic embryos was depended on various developmental stages of cotyledons. Immature meristematic cotyledons produced multiple cell-derived somatic embryos, which developed into multiple embryos. While fully mature cotyledons produced single cell-derived single embryos with independent state. Plasmolysis pretreatment of cotyledons strongly enhanced single cell-derived somatic embryogenesis. Single embryos were converted into normal plantlets with shoot and roots, while multiple embryos were converted into only multiple shoots. GA3 or a chilling treatment was prerequisite for germination and plant conversion. Low concentration of ammonium ion in medium was necessary for balanced growth of root and shoot of plantlets. Therefore, using above procedures, successful plant regeneration of ginseng was accomplished through direct single embryogenesis, which makes it possible to produce genetically transformed ginseng efficently.

  • The unloading compliance method is the most commonly used method to evaluate the fracture resistance characteristics of a material. In dynamic loading condition, the direct current potential drop(DCPD) method has been used because the unloading compliance method can not be applied due to the discontinuity of loading. However, even in the dynamic test using DCPD method, there is a problem that the voltage drops sharply on the initiation of crack. For the reason metioned above, the normalization method was suggested on ASTM E 1820 which is revised recently, as a new method to evaluate the dynamic fracture resistance characteristic. The nomalization method can be used to obtain a fracture resistance curve directly from a load-load line displacement. In this study, we obtained two fracture resistance curves from static test of welding part of nuclear piping both by unloading compliance and nomalization method. The two curves were almost same each other, so the adaptability of the nomalization method has been proved. We conducted a dynamic fracture resistance test for the same material. The fracture resistance curve from the dynamic test was obtained by normalization method and compared to that of the static test result.

Critical thrust force and feed rate determination in drilling of GFRP laminate with backup plate

  • Using backup plate is one of the most commonly used methods to decrease drilling-induced delamination of composite laminates. It has been shown that, the size of the delamination zone is related to the vertical element of cutting force named as thrust force. Also, direct control of thrust force is not a routine task, because, it depends on both drilling parameters and mechanical properties of the composite laminate. In this research, critical feed rate and thrust force are predicted analytically for delamination initiation in drilling of composite laminates with backup plate. Three common theories, linear elastic fracture mechanics, classical laminated plate and mechanics of oblique cutting, are used to model the problem. Based on the proposed analytical model, the effect of drill radius, chisel edge size, and backup plate size on the critical thrust force and feed rate are investigated. Experimental tests were carried out to prove analytical model.

The Correlation between Fatigue Fracture Crack Surface Friction and Crack Closure Effect in Crack Growth under Mixed-mode loading (혼합모드 하중 하에서의 균열성장 중 피로파단면 마찰과 균열닫힘효과의 상호관계)

  • Crack tip behavior of single mode loading condition(mode I)depend on tensile loading component but one of mixed mode loading condition(mode I+II) have influenced on shear loading component like the practical structure. Because crack closure is caused by shear loading component under mixed-mode loading a research on the behavior in the stage of crack initiation and propagation require to be evaluate about crack closure effect by fatigue crack surface friction. For that reasion we examined the behavior at the crack tip by direct measuring method. Measured behavior at the crack tip was analyzed through vector crack tip displacement. As a result, crack propafation equation was corrected by considering with crack closure effect. In addition we compared fatigue fracture crack surface and crack closure level.

Development of the Phage Displayed Peptide as an Inhibitor of MCP-1 (Monocyte Chemoattractant Protein-1)-mediated Angiogenesis

  • The CC chemokine, monocyte chemoattractant protein-1 (MCP-1), plays a crucial role in the initiation of atherosclerosis and has direct effects that promote angiogenesis. To develop a specific inhibitor for MCP-1-induced angiogenesis, we performed in vitro selection employing phage display random peptide libraries. Most of the selected peptides were found to be homologous to the second extracellular loops of CCR2 and CCR3. We synthesized the peptide encoding the homologous sequences of the receptors and tested its effect on the MCP-1 induced angiogenesis. Surface Plasmon Resonance measurements demonstrated specific binding of the peptide to MCP-1 but not to the other homologous protein, MCP-3. Flow cytometry revealed that the peptide inhibited the MCP-1 binding to THP-1 monocytes. Moreover, CAM and rat aortic ring assays showed that the peptide inhibited MCP-1 induced angiogenesis. Our observations indicate that the MCP-1-binding peptide exerts its anti-angiogenic effect by interfering with the interaction between MCP-1 and its receptor.

  • Fracture toughness defined near the initiation of stable crack growth is investigated by R-curve and Direct Current electric Potential Determination(DCPD) under mode I plane strain conditions for CT specimen with 25.4mm thickness of SS400 steel. Fracture toughness. $J_{IC}$lit near crack tip of CT specimen by R-curve is 17.14 $kg_{f}/mm$ and however. its value by DCPD is 22.82 $Rg_{f} mm$ The value of fracture toughness by DCPD is larger than that by R-curve. Therefore, it is suggested that the evaluation of fracture toughness by R-curve is optimum than by DCPD, when considering amount of crack growth about each of fracture toughness.

A Study on Corrosion Fatigue Crack Growth Behavior in Al-Alloy 7075-T651 (I) (Al-Alloy 7075-T651의 부식피로균열 성장거동에 관한 연구(I))

  • Fatigue crack growth rates(i.e. crack initiation and crack growth of short and long crack) are investigated using commercial plates of high strength Al alloy 7075-T651 for the transverse-longitudinal(T-L) direction in air, water and sea water. Also, the evaluation direct current potential drop(D.C.P.D) method and the fractographical analysis by SEM are carried out. Near threshold region, short crack growth rates were much faster than those of comparable long cracks, and these short crack growth rates actually decrease with increasing crack growth and eventually merge with long crack data. Fatigue crack propagation rates in aggressive media(i.e. sea water) increase noticeably over three times those in air. One of the most significant characters in this phenomenon as a corrosion-fatigue causes an acceleration in crack growth rates. Sea water environment, particularly Cl$^{[-10]}$ solution brings the most detrimental effects to aluminum alloy. The result of fractographical morphology in air, water and sea water by SEM shows obvious dimpled rupture and typical striation in air, but transgranular fracture surface in water and sea water.
