마(麻)의 품종별 영양성분 및 Diosgenin 함량 (Contents of Nutritional Ingredients and Diosgenin in the Tubers of Different Dioscorea spp.)

  • The principal objective of this study was to determine the available components in different varieties of Discorea spp. The Discorea spp. samples were divided into three groups by variety. The three groups were divided into Jangma(D. batatas Decaisne), Danma(D. japonica Decaisne), and Doonggunma(D. opposita Thumb), and then assessed with regard to the content of proximate compositions, minerals, sugars, crude saponin, and diosgenin. No substantial differences were noted to exist among samples in terms of carbohydrate and crude protein contents. The crude lipid contents were detected in the following order : Danma(D. japonica Decaisne, $0.71{\pm}0.15%$) > Jangma(D. batatas Decaisne, $0.65{\pm}10.12%$) > Doonggunma (D. opposita Thumb, $0.49{\pm}0.10%$). The Danma(D. japonica Decaisne) samples were determined to harbor the highest K($2.07{\pm}0.91%$), Ca($0.21{\pm}0.12%$), Fe($110.7{\pm}0.0$ mg/kg), Mn($20.6{\pm}3.4$ mg/kg) and Zn($31.2{\pm}3.9$ mg/kg) contents among all tested samples. The predominant sugar components in all samples were mannose($70.7{\pm}1.3{\sim}80.5{\pm}1.5%$), glucose($18.2{\pm}1.2{\sim}28.3{\pm}2.5%$) and sucrose($60.3{\pm}3.7{\sim}83.6{\pm}6.5%$). The crude saponin and diosgenin contents for Jangma(D. batatas Decaisne), Danma(D. japonica Decaisne) and Doonggunma(D. opposita Thumb) varieties were : $1.7{\pm}0.4%$, $3.9{\pm}0.8%$ and $3.2{\pm}0.7%$, and $13.59{\pm}1.74$ mg/g, $14.25{\pm}1.60$ mg/g and $18.00{\pm}1.92$ mg/g, respectively.

Changes of Endogenous Gibberellins in Tubers of Chinese Yam(Dioscorea opposita) during Storage Period

  • For storage periods of tubers in Chinese yam, the levels of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$ was constant, meanwhile both $\textrm{GA}_{53}$ and $\textrm{GA}_{19}$ level were always higher than that of $\textrm{GA}_{44}$ and $\textrm{GA}_{20}$$\textrm{GA}_9$ content as precursor of $\textrm{GA}_4$ was not changed during storage. $\textrm{GA}_{24}$ content was low to below 0.2 ng for 90 days after storage, $\textrm{GA}_{36}$ content as precursor of $\textrm{GA}_4$ like $\textrm{GA}_9$ was about 6-8 fold higher than that of $\textrm{GA}_9$ during storage. GA contents of the two gibberellin biosynthetic pathways were gradually increased when storage periods were progressed. Bioactive GA$_1$ content as the GA members of an early C-13 hydroxylation was always constant, and its content was very low as below 0.1ng per dry weight, meanwhile, bioactive $\textrm{GA}_4$ content as the GA members of non C-13 hydroxylation was drastically increased, also, its content was highest at 90 days after storage, and then decreased at 120 days after storage. Consequently, we suggest that $\textrm{GA}_4$ may be involved in controlling tuber sprouting in Chinese yam.

Factors Affecting the Production of In Vitro Plants from the Nodal Pieces of Chinese Yam (Dioscorea Opposita Thunb)

  • This study was carried out to establish The regeneration of healthy seedlings from the nodal segment culture of Chinese yam (Dioscorea opposita cv. Danma), cultivated in Korea. Different explants such as leaves, petioles, roots and nodal pieces, excised from the in vitro grown seedlings of Chinese yam, were cultured on MS medium supplemented with various combinations of growth regulators. All the growth regulators used induced plantlet regeneration from the nodal segments at a high frequency, while there was no induction of shoot or callus from leaf, petiole or root tissues. The medium supplemented with 0.01mg/L NAA, 0.5mg/L BA, 0.5-1.0mg/L kinetin and without plant growth regulator was effective for shoot development of buds from the nodal segment culture. The concentration of BA and NAA was an important factor in the bud induction of buds from the nodal segments of Chinese yam. Nodal segments cultured on the medium containing 1.0mg/L NAA and 0.5-1.0mg/L BA gave the best response to bud formation. The addition of GA$_3$ to the culture medium suppressed shoot induction and growth, while it increased microtuber formation. The shoot growth and microtuber formation were also affected by medium strength and solidity. The MS basal medium containing 1 g/L gelrite was suitable for microtuber formation from the nodal segment of Chinese yam.
