1.5 L 교반 탱크반응조에서 이미 그 특성 이 밝혀진 TNT 분해세균인 Stenorophomonas maltophilia의 배양을 이용하여 dinitrotoluenes (DNTs) [2,4-dinitrotoluene (2,4-DNT), 2,6-dinitrotoluene (2,6-DNT)]를 호기적 조건하에서 미생물학적으로 제거하는 시험을 하였다. 2,4-DNT와 2,6-DNT는 각각 10일과 14일의 배양기간 중에 완전히 분해되었다. 이차 탄소는 DNT리 분해에 필수적이었으며 이차 탄소가 없는 상태에서는 거의 분해되지 않았다. DNTs의 분해에 이차 탄소가 미치는 효과에 대하여 조사하였다. 이차 질소원이 없는 상태에서 DNTs는 완전히 분해되었으나 이차 탄소원이 첨가된 배지에서는 부분적으로 분해되었다. HPLC와 GC-MS가 잔존 DNTs와 중간대사산물을 확인하기 위하여 사용되었다. 그 결과, HPLC와 GC-MS chromatograms은 대상 화합물인 2,4-DNT와 2,6-DNT, 그리고 각각의 중간대사산물인 2-amino-4-nitrotoluene과 2-amino-6-nitrotoluene을 성공적으로 확인할 수 있었다.
This report presents a voltammetric assay of dinitrotoluene using a DNA immobilized onto a carbon nanotube paste electrode (PE). The cyclic voltammetry (CV) and square wave (SW) stripping voltammetry parameters of the optimized conditions were obtained. An anodic peak current appeared at 0.3 V (versus Ag/AgCl) in a 0.1-M $NH_4H_2PO_4$ electrolyte solution. The detection limit was found to be $0.6ngL^{-1}$(S/N = 10), within a deposition time of 100 sec.
White-rot fungus Irpex lacteus degraded TNT significantly in proportion to the culture time. After 48 h incubation, about 95% of TNT was degraded. Two reduced metabolites were identified as 4-amino-2,6-dinitrotoluene (4-ADNT) and 2-amino-4,6-dinitrotoluene (2-ADNT) which was further degraded.
본 연구는 TNT 분해능이 우수한 세균인 S. maltophilia OK-5를 이용하여 TNT 함유 폐수인 pink water의 미생물학적 처리 가능성에 대한 연구를 하였다. Pink water에 함유된 TNT 제거를 위해 S. maltophilia OK-5를 교반탱크 반응조에서 배양한 결과 pink water 내에 존재하는 100 mg/L의 TNT를 배양 6일 만에 완전 분해하였다. Hydride-Meisenheimer complex에서 유래하는 진한 적갈색은 배양기간 내에 증가하였으며, 이를 정량적으로 확인하였다. 본 연구에서 pink water에 잔류하는 TNT 뿐만 아니라 2,4-dinitrotoluene, 2,6-dinitrotoluene, 2,4-dinitro-6-hydroxytoluene 등의 대사산물도 HPLC 분석방법으로 측정하였으며, GC-MS를 사용하여 확인하였다. 또한 pink water에서 배양된 S. maltophilia OK-5에서 발현되는 nitroreductase (pnrB)의 유전자 발현 정량을 real time PCR로 측정하였다. 그 결과 배양 5일째 pnrB copy 수가 $10^3$ 이상 증가하는 것을 확인하였다.
The biological removal of 2,4,6-trinitrotoluene (TNT) was studied in a bench-scale bioreactor using a bacterial culture of strain OK-5 originally Isolated from soil samples contaminated with TNT. The TNT was completely removed within 4 days of incubation in a 2.5 L bench-scale bioreactor containing a newly developed medium. The TNT was catabolized in the presence of different supplemented carbons. Only minimal growth was observed in the killed controls and cultures that only received TNT during the incubation period. This catabolism was affected by the concentration ratio of the substrate to the biomass. The addition of various nitrogen sources produced a delayed effect for the TNT degradation. Tween 80 enhanced the degradation of TNT under these conditions. Two metabolic intermediates were detected and identified as 2-amino-4, 6-dinitrotoluene and 4-amino-2, 6-dinitrotoluene based on HPLC and GC-MS analyses, respectively. Strain OK-5 was characterized using the BIOLOG system and fatty acid profile produced by a microbial identification system equipped with a Hewlett Packard HP 5890 II gas chromatograph. As such, the bacterium was identified as a Stenotrophomonas species and designated as Stenotrophomonas sp. OK-5.
Biomolecules specific to explosives can be exploited as chemical sensors. Peptides specific to immobilized dinitrotoluene (DNT) were identified using a phage display library. A derivative of DNT that contained an extended amine group, 4-(2,4-dinitrophenyl)butan-1-amine, was synthesized and immobilized using a self-assembled monolayer surface on gold. Filamentous M13 phages displaying random sequences of 12-mer peptides specific to the immobilized DNT-derivate were isolated from the M13 phage library by biopanning. A common peptide sequence was identified from the isolated phages and the synthesized peptides showed selective binding to DNT. When the peptide was immobilized on a quartz crystal microbalance (QCM) chip, it showed a binding signal to DNT, while toluene barely showed significant binding to the QCM chip. These results demonstrate that peptides screened by biopanning against immobilized DNT can be useful for quick and accurate detection of DNT.

Single-chain variable fragments of antibodies (scFv) specific to 2,4-dinitrotoluene (DNT) were isolated from a phage library displaying synthetic human scFv fragments with 6 diversified complementary determining regions (CDRs). A DNT derivative that contained an extended amine group was synthesized and conjugated to the NHS-group that was linked to magnetic beads. Phages specific to the immobilized DNT derivatives were isolated from the library after 4 rounds of sequential binding and elution processes. The displayed scFv fragments from the isolated phages showed consensus CDR sequences. One DNT-specific scFv was expressed in E. coli and purified using Ni-affinity chromatography. The purified DNT-specific scFv binds specifically to the immobilized DNT-derivative with $K_D$ value of $6.0{\times}10^{-7}$ M. The scFv and DNT interaction was not disrupted by the addition of 4-nitrotoluene or benzoic acid. These data demonstrate that the screened scFv from the phage displayed library could be used for selective and sensitive detection of explosives such as TNT.
본 연구에서는 marsh와 pond의 조합으로 구성된 회분식과 연속식 습지 반응조를 이용하여 화약물질인 TNT(2,4,6-trinitrotoluene)를 처리하는 실험을 수행하였다. 회분식 실험결과 10 mg/L의 TNT가 20일 이내에 marsh와 pond 반응조 내에서 모두 제거되었다. TNT의 분해에 따른 주요 분해 부산물들은 2,4-diamino-6-nitrotoluene(2,4-DANT), 2,6-diamino-4-nitrotoluene(2,6-DANT), 4-diamino-2,6-nitrotoluene(4-ADNT), 그리고 2-diamino-4,6-nitrotoluene(2-ADNT)이었으며 시간이 경과하면서 이들 부산물도 점차 감소하는 경향을 보였다. 다음으로 Marsh와 pond의 조합으로 이루어진 연속식 반응조를 이용하여 TNT를 처리한 결과 회분식 반응조와 비슷한 경향을 보였으며, 2일 이내에 수중에서 거의 모든 TNT가 제거되었다. 주요 부산물은 marsh와 pond 단일조합 반응조에서는 ADNT(mono amino-dinitrotoluene)계열의 부산물이었던 반면, marsh-pond, pond-marsh 연속식 반응조에서는 분해가 된 DANT(diamino-mononitrotoluene)가 주된 부산물로 관찰되어 혐기성과 호기성 조건을 동시에 가지는 연속식 조합 습지 시스템이 TNT의 분해에 더 효과적임을 알 수 있었다. 또한 습지의 유입수와 처리수의 독성을 평가하기 위해 Microtox Assay를 통하여 분석한 결과, marsh와 pond의 연속식 복합 반응조의 처리수는 $EC_{50}$를 계산할 수 없을 정도로 독성이 감소됨을 알 수 있었다. 이들 실험결과를 통하여 marsh와 pond의 조합으로 혐기성과 호기성을 동시에 마련한 인공습지 시스템을 이용하여 수중에 존재하는 TNT를 안정적으로 처리할 수 있었다.
