Antibacterial Activity of Korean Fig (Ficus carica L.) against Food Poisoning Bacteria (국내산 무화과의 식중독균에 대한 항균활성)

  • The antibacterial activities of methanol extract and systematic solvent fractions( -hexane, chloroform, ethyl acetate, and butanol) from Korean common type figs at different ripening stages were tested by the broth dilution method against 8 representative food-poisoning bacteria- : L. monocytogenes, S. aureus, S. enteritidis, E. coli O157:H7, E. coli, Y. enterocolitica, V. parahaemolyticus, and S. typhimurium. The methanol extracts of unripened I and II showed stronger activity than that of the ripened figs especially against L. monocytogenes, S. enteritidis, E. coli O157:H7, Y. parahaemolyticus and S. typhimurium in 10 mg/mL. The systematic solvent fractions showed stronger antibacterial activities than the methanol extract, even al the lower concentrations. The hexane fraction of ripened figs showed higher growth inhibition than those of unripened I and II against L. monocytogenes, E. coli O157:H7, Y. enterocolitica and V. parahaemolyticus. The chloroform fraction showed strong antibacterial activity in all ripening stages against E. coli O157:H7 and V. parahaemolyticus. The butanol fraction showed better inhibition activity in unripened I and II than in the ripened figs. The hexane and chloroform fractions showed inhibition activity of more than $75\%$ against E. coli O157:H7, V. parahaemolyticus in 0.5 mg/mL. Each fraction showed a little different antibacterial activity according to the ripening stages of the fruits and the tested strains. Especially, figs in the unripened II stage revealed superior activity relatively and the hexane and chloroform fractions revealed the strongest activity, followed by the butanol fraction, while the ethylacetate and water fractions hardly showed any activity.

A Study on the Source Profile Development for Diesel and Gasoline-Powered Vehicles (디젤 및 가솔린자동차 배출원의 구성물질 성분비 개발에 관한 연구)

  • The purpose of this study was to develop the $PM_{2.5}$ source profiles for diesel and gasoline-powered vehicles, which contained mass abundances in terms of mass fraction of $PM_{2.5}$ of chemical species. Seven diesel-powered vehicles and nine gasoline-powered vehicles were sampled from a chassis dynamometer exhaust dilution system. The species measured were water-soluble ions, elements, elemental carbon (EC), and organic carbon (OC). From this study, the large abundances of EC (54.5%), OC (26.0%), ${SO_4}^{2-}$ (1.5%), ${NO_3}^-$ (0.8%), and S (0.6%) were observed from the diesel-powered vehicle exhaust showing that carbons were dominant species. The gasoline-powered vehicle exhaust emitted large abundances of OC (38.3%), EC (4.2%), ${SO_4}^{2-}$ (3.6%), ${NH_4}^+$ (3.5%), and ${NO_3}^-$ (3.0%). The abundances of ${SO_4}^{2-}$, ${NH_4}^+$, and ${NO_3}^-$ from gasoline vehicle were greater than those of diesel vehicle. The emissions of P, S, Ca, Fe, and Zn among trace elements for the gasoline vehicle were greater than 1% of the $PM_{2.5}$ mass unlike those for the diesel vehicle. Particularly, the fraction of Zn was five times higher from the gasoline vehicle than that from the diesel vehicle. The source profiles developed in this work were intensively examined by applying chemical mass balance model.

The Effect of Eluent Concentration on the Separation of Nd with Ln-resin Method (란탄 레진법에서 용리액의 농도가 Nd 분리에 미치는 영향)

  • The rare earth element(REE)s play an important role in understanding of rock formation and evolution because of their similar geochemical behaviors. Sm and Nd are more useful than other REEs because Sm-Nd isotopic system has important applications for geochemical interpretation like age dating and crustal evolution. These studies require a chromatographic technique for Sm and Nd separation from the geological samples. Ln-resin method using 0.25 M HCl as the eluent is widely used for Nd separation. However, this technique has a disadvantage of the poor elemental selectivity that the Nd fraction contains Ce as a tailing of the previous fraction. This technical report is a comparison study on the effect of eluent concentration between 0.25 M HCl and 0.15 M HCl on the separation of Nd with Ln-resin method for improving the technique of Nd separation. The results showed that the separation of Ce and Nd using 0.15M HCl as the eluent was not effective compared to the separation using 0.25 M HCl. In this experiment, we could confirm that the dilution of eluent might not be effective on the high purity separation of Nd with Ln-resin method.

Extension of Low Temperature Combustion Regime by Turbocharging Using Diesel and Biodiesel Fuels (과급에 의한 디젤 및 바이오디젤의 저온연소 운전영역 확장에 관한 연구)

  • Due to its oxygen (O) content, biodiesel (BD) is advantageous in that it lowers PM emissions in CI engines. Therefore, BD is considered one of the best candidates for low temperature combustion (LTC) operation because its use can extend the regime for simultaneous reduction of PM and $NO_x$. Thus, in this study, LTC operation was realized using BD and diesel with a 5~7% $O_2$ fraction. Engine test results show that the use of BD increased the efficiency and reduced emissions such as PM, THC, and CO; furthermore, IMEP reduced by 10~12% owing to the lower LHV of the fuel. In particular, smoke was suppressed by up to 90% because O atoms in the BD enhanced the soot oxidation reaction. To compensate the IMEP loss, turbocharging (TC) was then tested, and the results showed that the power output increased and PM was reduced further. Moreover, TC in BD engine operation allowed a similar level of reduction in both $NO_x$ and PM at 11~12% $O_2$ fraction, suggesting that there is a potential to widen the operating range by the combination of TC and BD.

Spatial Variability and Contents of Metals in the Surficial Sediments of Youngil Bay, East Coast of Korea (한국 동해안 영일만 표층 퇴적물의 금속 함량과 공간 변화 특성)

  • Bottom sediments from Youngil Bay, East Coast of Korea, were analyzed for grain composition as well as elemental compositions and total organic carbon (TOC) content in order to investigate the spatial variability and content of metal elements. Grain size distribution of the sediments seems to be controlled by anticlockwise current pattern with bottom topography of the study area. Spatial variability of TOC and all elemental contents reflects those of grain size, but an exception was found in the harbor area (Old-Port): their contents are high in the central part of the bay with the muddy sediment and decrease toward the sand-dominated coastal zone. However, contents of Ca, Sr, K are high in the sand-dominated coastal zone and contents of some heavy metals (Cd, Cu, Zn) are high in the Old-Port area and the mouth of Hyeongsan River. The correlation matrix and R-mode factor analyses reveal that four important factors controlling the distribution of metals in the bay are sediment grain size (or quartz dilution effect), the formation of sulfide minerals associated with decomposition of organic matters under anoxic geochemical environment, calcium carbonate (mainly shell fragments) and coarse-grained feldspar mineral. According to the metal content of labile fraction an CER (concentration enrichment ratio) value, high accumulation of some heavy metals in the harbor area seems to result not formed by early diagenetic processes under anoxic environment.

  • Papenfussiella kuromo (PK) is a marine plant and an abundant ecological resource for the future; it is found in almost 80% of the terrestrial biosphere. The aim of this study was to investigate the antibacterial activity of PK against methicillin-resistant Staphylococcus aureus (MRSA), multidrug-resistant pathogen. The minimum inhibitory concentrations (MICs) of PK hexane fraction (PKH) against 7 strains of MRSA ranged from 1.0 to 2.0 mg/mL. In the checkerboard dilution method, a synergistic effect of the PKH and the antibiotics (oxacillin and norfloxacin) was seen. PKH markedly reduced the MIC of each of the 4 antibiotics against MRSA. The time-kill assay showed that the synergistic activity of PKH and an antibiotic reduced the bacterial counts below the lowest detectable limit after 24 h. These findings suggest that PKH has antibacterial activity, and may be important baseline data in future extensive studies of living marine resources as a source of compounds active against MRSA.

  • Destructive methods are used for the turnup determination of an irradiated PWR fuel. One of the methods includes U, Pu, Nd-148 and Nd-(145+146) determination by an isotope dilution mass spectrometry using triple spikes (U-233, Pu-242 and Nd-150). The method involves two sequential ion exchange resin separation procedures. Pu is eluted from the first anion exchange resin column (Dowex AG 1$\times$8) with 12 M HCl-0.1 M HI mixed solution, followed by U elution with 0.1 M HCl. Nd is isolated from other fission products on the second anion exchange resin column (Dowex AG 1$\times$4) with a nitric acid-methanol eluent. Each fraction is analysed by thermal ionization mass spectrometry. The difference between Nd-148 and Nd-(145+146) method is found with an average 2.07%. The results are compared with those by the heavy element method using U and Pu isotopes and by the destructive y-spectrometric measurement of Cs-137. The dependences of isotope composition of U and Pu on burn-up, and correlation between those isotopes are illustrated graphically.

Effect of Sample-loading on Fractionation Efficiency (FE) in a Large Scale Splitter-less Gravitational SPLITT Fractionation (GSF)

  • Gravitational SPLITT fractionation (GSF) provides separation of colloidal particles into two subpopulations in a preparative scale. Conventionally, GSF is carried out in a thin rectangular channel having two inlets and two outlets at the top and bottom of the channel, respectively. And the channel is equipped with two flow-splitters, one between the top and bottom inlets and another between the top and bottom outlets. A large scale splitter-less GSF system had been developed, which was designed to operate in the full feed depletion (FFD) mode. In the FFD mode, there is only one inlet through which the sample is fed, thus preventing the sample dilution. In this study, the effect of the sample-loading (in the unit of g/hr) on the fractionation efficiency (FE, number% of particles in a GSF fraction that have the sizes expected by theory) of the new large scale splitter-less FFD-GSF system was investigated. The system was tested in the sample-loading range of 3.0-12.0 g/hr with polyurethane latex beads (PU) and sea-sediment. It was found that there is an optimum range in the sample-loading for a FFD-GSF separation. It was also found that there is a general tendency of FE decreasing as the concentration of the sample suspension increases.

Safety Assessment of Intravenous Administration of Trastuzumab in 100ml Saline for the Treatment of HER2-Positive Breast Cancer Patients

  • Background: The infusion rate is considered to affect incidence and severity of infusion reactions (IRs) caused by protein formulations. Trastuzumab (TRS) is approved for 90-minute infusion as the initial dose followed by 30-minute infusion with 250 ml saline. In the study, we evaluated the safety of TRS intravenously administered over 30 minutes with 100 ml saline to reduce burden of patients, safety of infusion with 250 ml saline already being established. Materials and Methods: Women with HER2 positive breast cancer, ${\geq}18$ years and ${\geq}55%$ left ventricular ejection fraction (LVEF), were registered in the study. Patients received 8mg/kg of TRS 250 ml over 90 minutes followed by 6mg/kg of TRS 100ml over 30 minutes in a three-week cycle. Results: A total of 31 patients were recruited, 24 for adjuvant therapy and seven with metastases. The median age was 59 years (range 39 to 82). The total number of TRS doses ranged from 5 to 17 with the median of 15. Mild IR occurred in two patients at the first dose. However, no IR was observed after reducing to 100 ml saline. No decrease of LVEF, increase of serum brain natriuretic peptide or any other adverse events were reported. Conclusions: Intravenous infusion of TRS with 100 ml saline over 30 minutes in breast cancer patients can be considered safe based on results from the study. It can be given on an outpatient basis as with the currently recommended dilution in 250 ml saline.

A Study on the Development of Source Profiles for Fine Particles (PM2.5) (미세입자(PM2.5)의 배출원 구성물질 성분비 개발에 관한 연구)

  • The Purpose of this study was to develop the P $M_{2.5}$ source Profiles, which are mass abundances (fraction of total mass) of a chemical species in P $M_{2.5}$ source emissions. The source categories studied were soil, road dust, gasoline and diesel vehicles, industrial source, municipal incinerator, coal-fired power plant, biomass burning, and marine. The chemicals analyzed were ions. elements. and carbons. From this study, soil source had the crustal components such as Si, hi, and Fe. In the case of road dust. Si, OC, Ca, Fe had large abundances. The abundant species were S $O_4$$^{2-}$, C $l^{[-10]}$ , N $H_4$$^{+}$, and EC in the gasoline vehicle and EC, OC, C $l^{[-10]}$ , and S $O_4$$^{2-}$ in the diesel vehicle. The main components were S $O_4$$^{2-}$, S N $H_4$$^{+}$, and EC in the industrial source using bunker C oil as fuel, C $l^{[-10]}$ , N $H_4$$^{+}$, Fe, and OC in the municipal incinerator source, and Si, Al, S $O_4$$^{2-}$, and OC in the coal -fired power plant source. In the case of biomass burning, OC, EC, and C $l^{[-10]}$ were mainly emitted. The main components in marine were C $l^{[-10]}$ , N $a^{+}$, and S $O_4$$^{2-}$.EX> 2-/.
