Near-Field Hydrodynamic Analysis of the Submerged Thermal Discharge Using CFD Model (CFD 모델을 이용한 수중방류 온배수의 근역 동수역학 해석)

  • The buoyancy and initial momentum fluxes make near-field dominated by buoyant jet when thermal discharge releases underwater. In order to estimate prediction capabilities of those near-field phenomena, non-hydrostatic RANS applied CFD(Computational Fluid Dynamic) model was used. Condition of model was composed based on past laboratory experiments. Numerical simulations carried out for the horizontal buoyant jet in the stagnant flow and vertical buoyant jet into crossflow. The results of simulation are compared with the terms of trajectory and dilution rate of laboratory experiments and analytic model(CorJET) results. CFD model showed a good agreement with them. CFD model can be appropriate for assessment of submerged thermal discharge effect because CFD model can resolve the limitations of near-field analytic model and far-field quasi 3D hydrodynamic model. The accuracy and capability of the CFD model is reviewed in this study. If the computational efficiency get improved, CFD model can be widely applied for simulation of transport and diffusion of submerged thermal discharge.

  • A MILD (Moderate and Intense Low oxygen Dilution) combustion, which is effective in the reduction of NOx, is considerably affected by the recirculation flow rate of hot exhaust gas to the combustion furnace. The present study used the MILD combustor, which has coaxial cylindrical tube. The outside tube of the MILD combustor corresponds to the exhaust gas passage and the inner side tube is the furnace passage. A numerical analysis was accomplished to elucidate the characteristics of exhaust gas entrainment toward the inner furnace with the changes of coanda nozzle geometrical parameters, nozzle passage gap length, nozzle passage length, nozzle angle and expansion length. The optimal configuration of coanda nozzle for the best entrainment flow rate was gap length, 0.5 mm, expansion angle, 4o and expansion length, 146 mm. The nozzle passage length was irrelevant to the exhaust gas entrainement.

Effect of Double Noise-Barrier on Air Pollution Dispersion around Road, Using CFD

  • Noise-barriers on both sides of the roadway (hereafter referred to as double noise-barriers), are a common feature along roads in Korea, and these are expected to have important effects on the near-road air pollution dispersion of vehicle emissions. This study evaluated the double noise-barrier impact on near-road air pollution dispersion, using a FLUENT computational fluid dynamics (CFD) model. The realizable k-${\varepsilon}$ model in FLUENT CFD code was used to simulate vehicle air pollutant dispersion, in around 11 cases of double noise-barriers. The simulated concentration profiles and surface concentrations under no barrier cases were compared with the experimental results. The results of the simulated flows show the following three regimes in this study: isolated roughness (H/W=0.05), wake interface (H/W=0.1), and skimming flow (H/W>0.15). The results also show that the normalized average concentrations at surface (z=1 m) between the barriers increase with increasing double noise-barrier height; however, normalized average concentrations at the top position between the barriers decrease with increasing barrier height. It was found that the double noise-barrier decreases normalized average concentrations of leeward positions, ranging from 0.8 (H/W=0.1, wake interface) to 0.1 (H/W=0.5, skimming flow) times lower than that of the no barrier case, at 10 x/h downwind position; and ranging from 1.0 (H/W=0.1) to 0.4 (H/W=0.5) times lower than that of the no barrier case, at 60 x/h downwind position.

Development of Chicken Immunoglobulin Y for Rapid Detection of Cronobacter muytjensii in Infant Formula Powder

  • The present study was aimed to produce a chicken polyclonal antibody against Cronobacter muytjensii and to develop an immunoassay for its detection. Purification of anti-C. muytjensii IgY from egg yolk was accomplished using various methods such as water dilution and salt precipitation. As a result, sodium dodecyl sulfate-polyacrylamide gel electrophoresis produced two bands around 30 and 66 kDa, corresponding to a light and a heavy chain, respectively. Indirect competitive enzyme-linked immunosorbent assay (IC-ELISA) was performed to determine the effectiveness of the chicken IgY against C. muytjensii. The optimum conditions for detecting C. muytjensii by indirect ELISA and checkerboard titration of the antigen revealed an optimum average absorbance at the concentration of 18 ${\mu}g/mL$, having ca. $10^8$ coated cells per well. The anti-C. muytjensii IgY antibody had high specificity for C. muytjensii and low cross-reactivity with other tested pathogens. In this assay, no cross-reactivity was observed with the other genera of pathogenic bacteria including Escherichia coli O157:H7, Salmonella Typhimurium, Staphylococcus aureus, Bacillus cereus, Enterobacter aerogenes, Salmonella Enteritidis and Listeria monocytogenes. In addition, detection of C. muytjensii in infant formula powder showed a low matrix effect on the detection curve of IC-ELISA for C. muytjensii, with similar detection limit of $10^5$ CFU/mL as shown in standard curve. These findings demonstrate that the developed method is able to detect C. muytjensii in infant formula powder. Due to the stable antibody supply without sacrificing animals, this IgY can have wide applications for the rapid and accurate detection of C. muytjensii in dairy foods samples.

  • The purpose of this thesis is to dedicate to get the objectivity of herbal medicine treatments by choosing herbs likely to work as antibacterial agents, Moutan Cortex Radicis, Caesalpiniae Lignum, Houttuyniae Herba, Forsythiae Fructus, Prunellae Herba, Scrophulariae Radix against Gardnerella vaginalis which is associated with Bacterial vaginosis, making experiments on them and getting the significant results. Each herb's efficacy on control the number of Bacterial vaginosis is noticed by using Disk Susceptibility test with six herbs medicine and Broth dilution assay of the culture. Disk Suseptibility Test : The efficiency strength is as follows in a row : Caesaipiniae Lignum, Moutan Cortex Radicis, Prunellae Herba, Forsythiae Fructus, Houttuyniae Herba, Scrophulariae Radix. MIC Prunellae Herba is 1.5 mg/ml, Caesalpiniae Lignum is 5 mg/ml, Forsythiae Fructus and Moutan Corex Radicis is 10mg/ml for G. vaginalis. As a results, herbmed which tested in this study have an antibacterial effects against G. vaginalis and that may be used for treament of Bacterial vaginosis. Assistant use of external medicine to p.o medicine is expected to bring the good effect of treatment which is used for the origin of this disease.

Production of the Monoclonal Antibodies to the Escherichia coli Heat-Stable Enterotoxin (대장균의 내열성장독소 측정법개발을 위한 단세포군항체의 생산)

  • Monoclonal antibody to the Escherichia coli heat-stable enterotoxin(ST) was produced to develop a rapid and convenient diagnostic method to the ST. The toxin was purified from culture supernatant of enterotoxigenic E. coli O148H28($ST^+/LT^+$) and conjugated to bovine serum albumin(BSA). The ST-BSA conjugate was used to immunize BALB/c mice and the immune spleen cells from these mice were fused with $P3{\times}63$ Ag8.V653 plasmacytoma cells. Hybridomas were screened by ELISA and positive hybridomas were cloned by limiting dilution. Finally, one stable clone (AS36) specific to ST was selected for further growth and characterization. Antibody titers of culture supernatant and ascitic fluid from BALB/c mice were 1:1,024 and 1:20,480 respectively in ELISA. The isotype and subclass of monoclonal antibody was IgG1 in sandwich ELISA. To test the neutralizing effect of monoclonal antibody on toxin activity of ST, mixture of ascitic fluid and ST was assayed by infant mouse assay and this monoclonel antibody was proved to be a neutralizing antibody. The titer of ascitic fluid which completely neutralized biological activity of 4 units of ST was 1:4. Purified ST was quantitatively measured by competitive ELISA and minimum amount of ST detectable by this assay was 250pg, which was an amount six-fold smaller than that detectable by infant mouse assay. Four reference strains of enterotoxigenic E. coli from WHO were detected by competitive ELISA and highly specific, sensitive and reproducible result was obtained.

Synthesis of Tetrafluoroethylene from the Pyrolysis of Chlorodifluoromethane in the Presence of Steam (과열 수증기를 이용한 클로로디플루오르메탄 열분해 반응에 의한 테트라플루오르에틸렌의 합성)

  • The thermal pyrolysis of chlorodifluoromethane (R22) for producting tetrafluoroethylene (TFE) has been studied using the tubular reactor designed by the authors. The reaction temperature over $600{\sim}850^{\circ}C$, residence time over 0.005~0.6 sec, and steam/R22 ratio 3 to 30 were varied through experiments to analyze the effect of these variables on the conversion of R22 and selectivity for TFE. We have provided the guidelines for the optimal operation and design for the pyrolysis reactor. With increasing the dilution ratio, not only the conversion of R22 but also the selectivity for TFE increase. The optimum range of reaction temperature was $700{\sim}750^{\circ}C$ and the residence time 0.07~0.1 sec. In the kinetic study, first order rate equation was fitted well with the experimental data. This indicates that the main reaction step is a $CF_2$ generation from R22 pyrolysis. The range of activation energy for the rate constant was obtained 44.7~48 kcal/mol.

Rheological Study on Concentrated Kochujang (Korean Red Pepper Paste)-Water Suspensions (농후 고추장-물 현탁액의 리올로지 연구)

  • The rheological properties of concentrated Kochujang (Korean red pepper paste)-water suspensions were investigated. The rheological behavior of Kochujang-water suspension was illustrated by Herschel-Bulkley equation. All Kochujang-water suspensions with 12.5-100% of water addition showed pseudoplasticity in yield stress at the shear rate range of $0.25-179.20sec^{-1}$., but at the shear rate above $5.16 sec^{-1}$, more shear thinning behavior was observed with increasing shear rate. Also, pseudoplasticity of the suspension became weak by increasing water addition and decreasing temperature, The temperature dependence of consistency index on Kochujang-suspension was very low and the values of consistency index was lowered by increasing solid contents, but as heating, the values of that was increased. It was suggested that the main components responsible for rheological behavior of the suspensions at high temperature might be undigested starch or protein in Kochujang. Also, the changes of rheological behavior with vinegar addtion was only due to dilution effect.

  • The ESCORT model is applied at Mokpo coastal zone to analyze the effect of fresh water discharge released from the gates of the Youngsan River sea-dyke. Applicability and validity of both 2-D and 3-D hydrodynamic modules could be guaranteed by simulating hydrodynamic feature with ebb dominance characteristics. Then, effects of the released discharge on the hydrodynamics are investigated. And analysis of the effects on diffusion phenomena show that 3-D model is essential for such diffusion modelling. Moreover, vertical salinity distributions near the gates are examined, and the influence range of fresh water is also estimated, which shows that dilution of fresh water is going on slowly because of poor flushing.

Environmental Contamination and Bioavailability of Toxic Element around the Daduk Mine Area, Korea (다덕광산 주변지역에서의 독성원소들의 환경오염 및 인체흡수도)

  • In order to investigate the extent and degree of arsenic and heavy metal contamination and the bioavailability of toxic elements around the abandoned mine in Korea, an environmental geochemical survey was undertaken in the Daduk mine. After appropriate preparation, tailings, soil, stream sediment, crop plant and fingernail samples were analysed for As, Cd, Cu, Pb and Zn by ICP-AES and ICP-MS. Elevated levels of 8,782 mg/kg As, 8.3 mg/kg Cd, 489 mg/kg Cu, 3,638 mg/kg Pb and 919 mg/kg Zn were found in tailings from the Daduk mine. These significant concentrations can impact on soils and sediments around the tailing ponds. Mean concentrations of As, Cd, Pb, Cu and Zn in soils are significantly higher than those in world average soil, especially for As and Pb. Element concentrations in sediments decrease with distance from the tailing ponds due to a dilution effect by the mixing of uncontaminated sediments. Arsenic and Cd are elevated in rice grains and stalks, and Cu and Zn concentrations in chinese cabbage, sesame and bean leaves are higher than the upper limit values for normal plant. Arsenic concentration in fingernails of farmers are higher than the normal level with a maximum value of 1.5 mg/kg. The post-ingestion bioavailability of toxic heavy metals in some paddy and farmland soils has been also investigated using the SBET (simple bioavailability extract test) method. The method utilises synthetic leaching fluids closelyanalogous to those of the human stomach. The quantities of As, Cd, Cu, Pb and Zn extracted from paddy soils after 1 hour indicated 15.9, 65.4, 46.2, 39.4 and 29.4% bioavailability, respectively and for farmland soils, 12.4, 26.0, 31.2, 29.3 and 19.4% bioavailability, respectively. The results of the SBET indicate that regular ingestion of soils by the local population could pose a potential health threat due to long-term toxic element exposure.
