A Study on Flame Extinction in Oxymethane Combustion (메탄 산소 연소에 있어서 화염 소화에 대한 연구)

  • Oxy-methane nonpremixed flames diluted with $CO_2$ were investigated to clarify impact of radiation heat loss and chemical effects of additional $CO_2$ to oxidizer stream on flame extinction. Flame stability maps were presented with functional dependencies of critical diluents mole fraction upon global strain rate at several oxidizer stream temperatures in $CH_4-O_2/N_2$, $CH_4-O_2/CO_2$, and $CH_4-O_2/CO_2/N_2$ counterflow flames. The effects of radiation heat loss on the critical diluent mole fractions for flame extinction are not significant even at low strain rate in nonpremixed $CH_4-O_2/N_2$ diffusion flame, whereas those are significant at low strain rate and are negligible at high strain rate (> $200s^{-1}$) in $CH_4-O_2/CO_2$ and $CH_4-O_2/CO_2/N_2$ counterflow flames. Chemical effects of additional $CO_2$ to oxidizer stream on the flame extinction curves were appreciable in both $CH_4-O_2/CO_2$ and $CH_4-O_2/CO_2/N_2$ flames. A scaling analysis based on asymptotic solution of stretched flame extinction was applied. A specific radical index, which could reflect the OH population in main reaction zone via controlling the mixture composition in the oxidizer stream, was identified to quantify the chemical kinetic contribution to flame extinction. A good correlation of predicted extinction limits to those calculated numerically were obtained via the ratio between radical indices and oxidizer Lewis numbers for the target and baseline flames. This offered an effective approach to estimate extinction strain rate of nonpremixed oxy-methane flames permitting air infiltration when the baseline flame was taken to nonpremixed $CH_4-O_2/N_2$ flame.

Fungicidal Efficacy of a Fumigation Disinfectant with Ortho-phenylphenol as an Active Ingredient against Trichophyton mentagrophytes, Candida albicans and Aspergillus niger (Ortho-phenylphenol을 주성분을 하는 훈증소독제의 Trichophyton mentagrophytes, Candida albicans 그리고 Aspergillus niger에 대한 살진균 효과)

  • Objectives: This study evaluated the fungicidal efficacy of a fumigant containing 20% ortho-phenylphenol against Trichophytone mentagrophytes (T. mentagrophytes), Candida albicans (C. albicans) and Aspergillus niger (A. niger). Methods: Five replicates of each carrier were contaminated by depositing 0.05 mL of each fungal suspension. After drying, two carriers without exposure to the fumigant and three carriers with exposure to the fumigant were left in a sealed room ($25m^3$) at $21{\pm}0.5^{\circ}C$ and $60{\pm}10%$ relative humidity for 15 hours. Immediately after removal from the test room, each carrier was transferred into recovery diluent and suspended, diluted and inoculated. After incubation, the numbers of each colony were counted, and the parameter values (N, T, d) were calculated. Results: The working culture suspension number (N value) of T. mentagrophytes, C. albicans and A. niger were $1.0{\times}10^8$, $1.2{\times}10^8$ and $5.7{\times}10^7CFU/mL$, respectively. All the colony numbers on the carriers exposed to the fumigant (n1, n2, n3) were higher than 0.5N1 (the number of fungal test suspensions by pour plate method), 0.5N2 (the number of fungal test suspensions by filter membrane method) and 0.5N1, respectively. In addition, all mean numbers of test strains recovered on the control-carriers (T value) were over $10^6CFU/mL$. For the fungicidal effect of the fumigant, all numbers of fungal reductions after exposure of the fumigant (d value) were 4 logCFU/mL. Conclusions: The present study showed that fumigant containing 20% ortho-phenylphenol has effective fungicidal activity against T. mentagrophytes, C. albicans and A. niger.

Effect of Diluents on the Cold Storage of Sperm in Scapharca broughtonii (Schrenck) (피조개, Scapharca broughtonii (Schrenck) 정자의 냉장보존에 미치는 희석액의 효과)

  • The effects of diluents composition on cold storage for Scapharca broughtonii (Schrenck) sperm were examined in the percentage of sperm activity and survival rate. Various diluents of glucose solutions (10 mM Hepes-pH 7.8), 600 mM NaCl, stein solution, Ringer's solution (230 mM NaCl, 8 mM KCl, 2 mM $CaCl_2$, 3.7 mM $MgCl_2$, 0.2 mM $NaHCO_3$, 10 mM Hepes-pH 7.8), 20%, 25% ASW (NaCl 2.7 g + KCl 0.07 g + $CaCl_2$ 0.12 g + $MgCl_2$ 0.46 g + $NaHCO_3$ 0.05 g + distilled water 100 ml) were used to store th sperm at $4^{\circ}C$. The storage effect was evaluated using sperm activity and survival rate. Ringer's solution was found to be better diluents which maintained high activity and survival rate of sperm for a storage period of 7 days. Optimal pH of diluents to store the sperm at $4^{\circ}C$ is 7.5.

  • This study was conducted to improve the recycling process of waste fluorescent lamp, and investigate the possibility of using the waste fluorescent lamp glass as a raw material for glass beads, the leaching method of rare earth from the waste phosphor powder, and the possibility of solvent extraction of rare earth from the rare earth leaching solution. The waste phosphor contained 28.9% yttrium oxide, 3.46% cerium oxide, 1.95% europium oxide, 1.76% terbium oxide, and 1.43% lanthanum oxide. As a result of the trial production of glass beads using waste fluorescent lamp glass, it was judged that the production yield and quality were excellent, so that waste fluorescent lamp glass could be used as a raw material for glass beads. The soda roasted waste phosphor was leached in water and thereby the aqueous solution was blown with CO2 to drop the pH to about 7, Then, Al, Si and residual N2CO3 were dissolved, and NaAlCO3(OH)2 and SiO2 were precipitated in the aqueous solution. In the solvent extraction of cyanex272-hydrochloric acid, cyanex272-sulfuric acid, D2EHPA-hydrochloric acid, D2EHPA-sulfuric acid, Ionquest290-hydrochloric acid, Ionquest290-sulfuric acid, p507-hydrochloric acid using xylene as a diluent, the extraction yield of Y, Eu, Ce, La, and Tb are close to 100%. However, in this conditions, the difference in extraction yield for each element, that is, selectivity is 16% or less.

Effects of Triladyl-egg Yolk Diluents on the Viability of Frozen Korean Black-goat Spermatozoa from Cauda Epididymis and Electro-ejaculated Semen (Triladyl-난황 희석제가 한국 재래 흑염소의 정소상체 및 전기자극 유래 정자의 융해 후 생존성에 미치는 영향)

  • To preserve genetic materials, cryopreservation of the semen from live animals is the main technique to establish cryo-banking system which could be used for artificial insemination and embryo transfer. However, the population of Korean black goat (KBG) becomes to dwindle in number and is now faced genetic erosion by crossbreeding with non-native breeds in small KBG farms. In this study, simple freezing method was used to preserve frozen semen from KBG using spermatozoa of cauda epididymis (CE) and electro-stimulated semen (ES). The negative effects of seminal plasma on fresh sperm was confirmed using precipitation test of Triladyl egg yolk diluent and sperm viability after thawing was compared between CE and ES spermatozoa. When seminal plasma of fresh ES semen was washed with semen washing media (SWM), the rates of live sperm shown no significant difference between CE and ES spermatozoa before freezing. However, the survival rate of frozen/thawed CE sperm was higher than ES ($74.6{\pm}10.6%$ vs $53.8{\pm}5.2%$) with significant difference (p < 0.05). The results of longevity test on frozen/thawed sperm from CE showed healthier sperm than ES. Therefore, spermatozoa from CE could be used for cryo-banking system in KBG lines. The more studies are needed to increase survival rate of ES semen.

Effects of Equilibration Time, Precooling and Straw Loading Method on Survival of Mouse Embryos Frozen by Vitrification (생쥐 수정란의 유리화 동결보존에 있어서 동결전 처리방법에 관한 연구)

  • This study was carried out to investigate the effect of equilibration time, precooling and straw loading method on the post-thaw survival rates of mouse embryos cryopreserved by vitrification method. The effect of the vitrification procedure on the damage of the embryos was also examined by the straining of nuclei using Hoechst 33342. The results obtained were summarized as follows; 1. The equilibration in Medium-1 for 10 minutes was considered to be the optimum equilibration time. Embryos equilibrated in Medium-1 for 10 minutes(81.0%) showed significantly(P<0.05) higher survival rates than those equilibrated for 5 minutes(40.0%) or 15 minutes(74.1%). 2. The survival rate of embryos cryopreserved using the double Medium-2 column(81.0%) was significantly(P<0.01) higher than that using the single Medium-2 column, whish was considered to be due to the double Medium-2 column method being more reliable for preventing contamination of diluent solution of 1M sucrose. 3. The survival rate of compacted morula stage embryos cryopreserved with the precooled and non-precooled Medium-2 was not significantly(P<0.05) different. 4. The number of blastomeres of embryos at late blastocyst stage after culture of mouse morulae for 24 to 28hours was significantly(P<0.05) decreased by freezing embryos using vitrification(53.3${\pm}$1.6 vs 41.4${\pm}$1.5), which was considered to be due to the damage of embryos during vitrification and the delay of embryo development by handling in vitro. 5. The vitrification procedure is considered to be a very simple and efficient method for cryopreservation of embryos at early developmental stage.

A Study of Carry Over Contamination in Chematology (이월오염에 대한 연구)

  • Carry over contamination has been reduced in some systems by flushing the internal and external surfaces of the sample probe with copious amount of diluent. It between specimens should be kept as small as possible. A built-in, continuous-flow wash reservoir, which allows the simultaneous washing of the interior and exterior of the syringe needles, addresses this issue. In addition, residual contamination can further be prevented through the use of efficient needle rinsing procedures. In discrete systems with disposable reaction vessels and measuring cuvets, any carry over is entirely caused by the pipetting system. In analyzers with reuseable cuvets or flow cells, carry over may arise at every point through which high samples pass sequentially. Therefore, disposable sample probe tips can eliminate both the contamination of one sample by another inside the probe and the carry over of in specimen into the specimen in the cup. The results of the applicative carry over experiment studied on 21 items for total protein (TP), albumin (ALB), total bilirubin (TB), alkaline phosphatase (ALP), aspratate aminotranferase (AST), alanine aminotranferase (ALT), gamma glutamyl transferase (GGT), creatinine kinase (CK), lactic dehydrogenase (LD), creatnine (CRE), blood urea nitrogen (BUN), uric acid (UA), total cholesterol (TC), triglyceride (TG), glucose (GLU), amylase (AMY), calcium (CA), inorganic phosphorus (IP), sodium (Na), potassium (K), chloride (CL) tests in chematology were as follows. Evaluation of process performance less than 1% in all tests was very good, but a percentage of ALB, TP, TB, ALP, CRE, UA, TC, GLU, AMY, IP, K, Na, and CL was 0%, implying no carry over. Other tests were ALT(-0.08%), GGT(-0.09%), CK(0.08%), LD(0.06%), BUN(0.12%), TG (-0.06%), and CA(0.89%).

Cryopreservation of Pacific Oyster, Crassostrea gigas Sperm (굴, Crassostrea gigas 정자의 냉동보존)

  • This study aims to find out a suitable cryoprotective agent (CPA) for cryopreservation and its optimum concentration in order to conduct planned artificial seed production of Pacific oyster, Crassostrea gigas and to preserve superior sperm. For this, we tried to understand toxicity and the effect of cryopreservation by CPA type and concentrations first and then looked into cell damage of the sperm after thawing. Toxicity analysis on the sperm of Pacific oyster according to different CPA and immersion time shows that dimethyl sulfoxide (DMSO) comes first when it comes to survival rate and mobility followed by ethylene glycol (EG), glycerol and methanol. To identify the optimum CPA and its level, filtered seawater was used as a diluent before cryopreservation for 30 days. As a result, cryopreserved sperm of Pacific oyster with 15% of DMSO showed the highest survival rate and activation. Also, we observed the cryopreserved and thawed sperm with Scanning electron micrographs by CPAs and concentrations. Consequently, DSMO showed the lowest cell damage followed by EG, methanol, glycerol and the level was 15, 20, 10, 5% respectively. In a nutshell, it is proven that the optimum CPA and its level is 15% of DMSO.

Motility of Rooster Spermatozoa under Different Thawing Conditions (닭 동결정액 융해방법이 정자 운동성에 미치는 영향)

  • In this study, to increase the survival rate of frozen/thaw rooster semen, standard protocols of semen thawing procedures were tested by computer-assisted sperm assay (CASA). We tested 4 different thawing protocols for frozen semen, $5^{\circ}C$ for 2 min, $35^{\circ}C$ for 30 s, $54^{\circ}C$ for 13 s, and $70^{\circ}C$ for 7 s. The pooled semen from 5 to 8 Ogye rooster line was diluted in the HS-1 diluent and frozen in 8% methylacetamide (MA) in liquid nitrogen vapors. To determine standard thawing method, straws were plunged into different temperatures and times. The resulting motilities were recorded by the CASA system. The results of this study showed that the best viability of the spermatozoa was shown by exposure at $5^{\circ}C$ for 2 min. Moreover, the longevity test of thawed sperm at $5^{\circ}C$ for 2 min also supported the higher viability under low temperature preservation of $17^{\circ}C$ for 1 hr. Further research is needed to increase the motility of thawed rooster semen for field application. In addition, the in vivo tests for different rooster lines are also needed for the establishment of avian genetic resource bank.

In-Vitro Fertilization and Culture of Pig Oocytes Matured In-Vitro by Liquid Boar Sperm Stored at 4$^{\circ}C$

  • This study was carried out to investigate the effects of liquid boar sperm stored at 4$^{\circ}C$ on sperm motility, normal acrosome, and in-vitro fertilization and culture of pig oocytes matured in-vitro. The sperm-rich fraction (30~60 ml) of ejaculate was collected into an insulated vacuum bottle. Semen was slowly cooled to room temperature (20~23$^{\circ}C$) by 2 h after collection. Semen was transferred into 15 ml tubes, centrifuged at room temperature for 10 min at 800$\times$g, and the supernatant solution was poured off. The concentrated sperm was resuspended with 5 ml of lactose, egg yolk and N-acetyl-D-glucosamine (LEN) diluent to provide 1.0$\times$10$^{9}$ sperm/ml at room temperature. The resuspended semen was cooled in a refrigerator to 4$^{\circ}C$ and preserved for 5 days to examine sperm motility and normal acrosome. The medium used for oocyte maturation was modified tissue culture medium (TCM) 199. After about 22 h of culture, oocytes were cultured without cysteamine and hormones for 22 h at 38.5$^{\circ}C$, 5% $CO_2$ in air. Oocytes were inseminated with liquid boar sperm stored at 4$^{\circ}C$ for 2 days after collection. Oocytes were coincubated for 6 h in 500 ${mu}ell$ mTBM fertilization media with 0.2, 1, 5 and 10$\times$10$^{6}$ /ml sperm concentration, respectively. At 6 h after IVF, oocytes were transferred into 500 ${mu}ell$ Hepes-buffered NCSU-23 culture medium for further culture of 6, 48 and 144 h. There were significant differences in sperm motility and normal acrosome among preservation days and incubation times, respectively. The rates of sperm penetration and polyspermy were higher in 5 and 10$\times$10$^{6}$ sperm/ml than in 0.2 and 1$\times$10$^{6}$ sperm/ml. Male pronuclear formation was lower in 0.2$\times$10$^{6}$ sperm/ml than in 1, 5 and 10$\times$10$^{6}$ sperm/ml. Mean numbers of sperm in penetrated oocyte were highest in 10$\times$10$^{6}$ sperm/ml compared with other sperm concentrations. The rate of blastocysts from the cleaved oocytes (2~4 cell stage) was highest in 1$\times$10$^{6}$ sperm/ml compared with other sperm concentrations. In conclusion, we found out that liquid boar sperm stored at 4$^{\circ}C$ could be used for in-vitro fertilization of pig oocytes matured in-vitro. Also, we recommend 1$\times$10$^{6}$ ml sperm concentration for in-vitro fertilization of pig oocytes.
