Responses of Alkoxyresorufin Dealkylases and Glutathione S-transferase Activities of Surf Clam, Pseudocardium sachalinensis, Injected with Sea-Nine 211 Antifoulant (Tin-free 방오제인 Sea-Nine 211을 주사한 북방대합에서 alkoxyresorufin 탈알킬화효소와 글루타치온 포합효소 활성의 변화)

  • To evaluate the extent of Sea-Nine 211 (4,5-dichloro-2-n-octyl-3(2H) isothiazolone), one of the alternating organic booster compound used in antifouling paint to replace TBT, on marine benthic bivalve, we injected Sea-Nine 211 to clam, Pseudocardium sachalinensis, and then determined some xenobiotics metabolizing enzyme activities, especially EROD (ethoxyresorufin deethylase) and MROD (methoxyresorufin demethylase), in digestive gland during 4 day-exposure period. Moreover, the results were compared with those of TBT exposed clam. CYP1A1 dependant EROD activity in both the Sea-Nine 211 and the TBTC exposure groups showed no significant differences compared to those of the solvent control group. CYP1A2 dependant MROD activity in Sea-Nine 211 exposure group was significantly induced, but no significant difference was obtained in the TBTC exposure group. These results indicate that Sea-Nine 211 demonstrated a tendency to induce MROD activity, while TBTC inhibits the activities of this enzyme.

Substitution of soybean meal with detoxified Jatropha curcas kernel meal: Effects on performance, nutrient utilization, and meat edibility of growing pigs

  • Objective: The study was conducted to investigate the effects of replacing soybean meal (SBM) with different levels of detoxified Jatropha curcas kernel meal (DJM) in growing pig diets on growth performance, nutrients digestibility and meat edibility. Methods: A total of 144 pigs with initial body weight of $20.47{\pm}1.44kg$, were randomly allocated to 6 dietary treatments with 6 replications per treatment and 4 pigs per replication for a period of 79 days. Six diets (DJM0, DJM15, DJM30, DJM45, DJM60, and DJM75) were formulated using DJM to replace 0%, 15%, 30%, 45%, 60%, and 75% of SBM. From d 37 to 42, feces and urine were total collected from six barrows in each treatment. At day 79, thirty-six pigs were slaughtered for sampling. The feed intake and weight gain were recorded, while the intestinal morphology, digestive enzyme activities, nutrient digestibility and the content of residual phorbol esters in muscles were determined. Results: The results showed that increasing the replacement of SBM with DJM decreased the parameters including body weight, average daily gain, average daily feed intake, gain-to-feed ratio, weight and villus heights of duodenum, villus height and villus height/crypt depth of jejunum, digestive enzymes (protease, amylase, lipase, and trypsin) activities, and nutrients digestibility (nitrogen deposition, digestibility of nitrogen, energy digestibility, and total nitrogen utilization) (linear, p<0.05; quadratic, p<0.05) and there was no significant difference among DJM0, DJM15, and DJM30 in all measured indices. The highest diarrhea morbidity was observed in DJM75 (p<0.05). Phorbol esters were not detected in pig muscle tissues. Conclusion: The DJM was a good protein source for pigs, and could be used to replace SBM up to 30% (diet phorbol esters concentration at 5.5 mg/kg) in growing pig diets with no detrimental impacts on growth performance, nutrient utilization, and meat edibility.

Changes of Antigen Binding Activities of Hen′s Egg Yolk Immunoglobulins after Proteolysis (효소 분해에 의한 난황 항체의 항원 결합력 변화)

  • Yolk immunoglobulins (yIgG) from hen's egg were purified. To investigate the stability of yIgG to digestive enzymes, the changes of antigen binding activities (ABC) after in vitro proteolysis were examined by competitive ELISA. After 30 min exposure to pepsin, the ABC of yIgG was lost. However, comparing with native yIgG, the ABC of pepsin digested yIgG was decreased, but considerable amount of ABC was remained after 30 min exposure to pepsin in 50% saccharose solution. Therefore, the stability of yIgG to pepsin digestion was improved by the addition of saccharose to yIgG solution. The ABC of yIgG was considerably remained after exposure to trypsin and chymotrypsin for 8 hr. YIgG showed especially good stability to chymotrypsin proteolysis.

Alkaline Pretense Activity of the Tissue Extracts from Some Different Kinds of Fish (어류의 조직중에 분포하는 알카리성 단백질분해효소의 활성조건)

  • To check the differences of the digestive enzymes by the bait habits and the proteolytic activities of the fissile extracts from the fish, omnivorous filefish (Navodon modestus), carnivorous cat shark (Scilliorhinus tarazame) and bloodsucking hag fish (Eptatretus burgeri) were sampled for this experiment. The activity of crude alkaline protease extracted from the muscle and the internal organs of the samples was determined with casein as substrate. The activity of the proteolytic enzymes showed remarkable differences by the organs of the fish. The optimum condition of the pretenses from the muscle revealed in range of pH 7.8-8.3, at $60-65^{\circ}C$, while those of the enzymes from the internal organs were at about pH 8.2, $45-55^{\circ}C$, but those of hag fish were at about pH 6.7, $45-55^{\circ}C$. The proteolytic activity of the enzyme of alimentary canal in filefish and in hag fish was 57 and 11 times stronger than that of muscle, respectively. The crude enzyme from the alimentary canal of file fish showed the strongest proteolytic activity in samples submitted and that of cat shark was the lowest. The activity of pancreatic alkaline protease in cat shark was 50 fold higher than that of muscle alkaline protease in the fish.

Digestive Enzymatic and Nucleic Acidic Responses of Olive Flounder Paralichthys oilivaceus Larvae Fed Cyclopoid Copepod Paracyclopina nana (기수산 Cyclopoid 요각류 Paracyclopina nana를 섭취한 넙치 Paralichthys olivaceus 자어의 핵산 함량과 소화효소적 반응)

  • We investigated the changes in growth, digestive enzymes activities, nucleic acids contents and RNA/DNA ratio of flounder Paralichthys olivaceus larvae (C for Paracyclopina nana, A for Artemia, and M for Mix of C and A) for 14 to 28 DAH. Body length of flounder larvae showed the best in the C trial at 28 DAH. The change of nucleic acids contents showed faster in C and M trials than A trial. And RNA/DNA ratio showed the significantly faster changes in C trial than A trial. High metamorphosis rates were also observed in C and M trial. $\alpha$-amylase activities increased gradually up to 28 DAH in all trials. Total alkaline protease (TAP) activities of A trial showed the highest value to 9 mU/larvae at 26 DAH. But others trials showed lower to $5{\sim}6$ mU/larva than A trial. TAP:$\alpha$-amylase activity ratio did not significantly changed to $0.025{\sim}0.053$ in A trial during the experiments. But, C and M trials tended to gradually decrease from $0.078{\sim}0.083$ (initial) to $0.013{\sim}0.018$ (final). Therefore, it shown the ratio gradually decreased of TAP:$\alpha$-amylase activity, stability of TAP activity, and rapid change of nucleic acids in trials grown positively. Thus, because P. nana could continuously supply the optimal nutrients for flounder larvae, we suggested the supplement of the copepod to an efficient feed of the flounder larvae.

Cellulase Activity of Symbiotic Bacteria from Snails, Achatina fulica

  • Cellulase is the key enzyme in the use of cellulose-based biomaterials. Because of its structure, cellulose is difficult to be degraded by enzymes. In order to utilize cellulose-based biomaterials efficiently, evolutionary wisdom of how to use enzymes accurately and harmoniously in a biological system is needed, such as the cellulose digestive system in animals. In this study, the symbiotic bacteria from snails, Achatina fulica, were identified and their cellulase activity was evaluated. The 16S rRNA sequence analysis of 100 aerobic bacteria showed that they belonged to 9 genus and almost half of the bacteria were Lactococcus spp. Among 100 identified strains, only two Aeromonas sp. strains showed cellulase activity. Aeromonas sp. KMBS020 had both endo-${\beta}$-glucanase and ${\beta}$-glucosidase activities but Aeromonas sp. KMBS018 had ${\beta}$-glucosidase activity only. None of the 100 bacterial colonies had any cellobiohydrolase activity.

Post Feeding Trypsin Activity in the Digestive Organs and the Gastric Evacuation Rate of Litopenaeus vannamei (Boone) (먹이섭취 후 흰다리새우, Litopenaeus vannamei (Boone) 소화기관의 trypsin 활성 및 배설률)

  • The tryptic enzyme activities from hepatopancreas, foregut, midgut and feces were examined to optimize the feeding method in whiteleg shrimp, Litopenaeus vannamei. The highest tryptic enzyme activity was found in hepatopancreas. The enzyme activities of hepatopancreas were 4 times higher than those of foregut per mg dry weight at 30 minutes feeding. Post feeding period, the activities of hepatopancreas increased continuously up to 30 hours after feeding. Trypsin activities of foregut showed about 3 times higher than did those of midgut. Average activity of foregut reached the pick with $303{\pm}68\;(mean{\pm}SE)$ nmol/mg/min at two hours after feeding and kept the activity up to 4 hours after feeding and thereafter the activity decreased. Average tryptic enzyme activity of midgut increased to $96{\pm}26nmol/mg/min$ up to two hours after feeding and it decreased to $52{\pm}17nmol/mg/min$ at five hours after feeding eventhough the gastric evacuation rate was still 50% by then. Foregut clearance occurred in 30 minutes after feeding and midgut weight increased up to 2 hours after feeding. Also we found that the maximal food ingestion in foregut was equivalent to the average 0.3% of its body weight by 30 minutes after feeding. Up to 5 hours after feeding, the weight ratio of midgut to body weight reduced, but still the weight ratio of foregut to body weight kept the similarity until then. These indicated that the tryptic enzyme activity and the clearance rate are different among the digestive organs and between forgot and midgut during the post feeding period in whiteleg shrimp.

Effects of Fibrolytic Enzyme Addition on Ruminal Fermentation, Milk Yield and Milk Composition of Dairy Cows (Fibrolytic Enzyme 첨가가 반추위 발효 성상 및 착유우의 유량 및 유성분에 미치는 영향)

  • We evaluated the effects of adding fibrolytic enzyme into ruminant diets on ruminal fermentation (in vitro) and lactational performances of dairy cows (in vivo). Through the in vitro experiment that was carried out with different contents of NDF (34, 38, 43%) in diets, digestibilities of NDF in the rumen appeared not significantly different by the addition of enzyme but were different by NDF content in diets showing higher digestibility in NDF 43% diet. It could be attributed by the relatively higher amount of hemicellulose in the current experimental diets than in conventional diets that might have been digested easily by the addition of fibrolytic enzyme in the rumen. The addition of fibrolytic enzyme tended to increase NDF digestibilities to a little extent both in 0.05 and 0.1% enzyme levels. Ruminal pH, NH3-N concentrations and VFA production in the rumen were not affected by the addition of fibrolytic enzyme. Activities of CMCase and xylanase were higher in enzyme treated diets of both NDF 34 and 38%. In particular, the activities of xylanase that slowly decreased from 0 to 12 hr but rapidly after 24 hr indicates that the major action of the enzyme in the rumen occurs in early period of incubation. Through an in vivo experiment, fibrolytic enzyme addition into the diets of dairy cows indeed affected lactational performance of milk yield. The cows fed enzyme treated diets produced 8% (1.9kg/d) more amounts of milk than with no enzyme addition. Milk composition of milk fat and protein was not affected by enzyme addition. Overall, the results of this in vivo study indicates that fibrolytic enzyme can be used to improve milk production in lactating cows. In respect that animals in different treatments of this study had the same amounts of intake, the increased milk yield with enzyme addition may be attributed to the improved utilization of nutrients in the digestive tract.

Effects of Wolguk-whan Water Extract on Acute Oxidative Liver Injury Induced by Acetaminophen (월국환(越鞠丸) 물 추출물이 Acetaminophen으로 유도된 마우스의 급성 간손상에 미치는 효과)

  • Objectives : Wolguk-whan has been used as a prescription of natural drug for the treatment of stress digestive system disease. Recently, we reported that Wolguk-whan methnol extract (WGWM) exerted a significant protective effect against oxidative damage to the liver of ICR mice. This study was purposed to investigate the effects of Wolguk-whan water extract (WGWW) on liver injury induced by oxidative stress. Methods : In order to investigate the effects of WGWW on acute liver injury, ICR mice were pretreated with WGWW for 6days, starved for 24hrs, and administerated acetamirtophen(500mg/kg, i.p.). In the liver homogenates, lipid peroxide and glutathione(GSH) levels were measured. In addition, activities of hepatic enzyme, such as catalase, glutathione peroxidase(GSH-Px), glutathione S-transferase(GST) were measured in the hepatic mitochondrial and cytosolic fractions. Results : In vivo administeration of WGWW showed effective inhibition of acetaminophen induced lipid peroxidation, and showed elevations of GSH level, catalase, GSH-Px, GST activities. Conclusions : These results suggested that WGWW might suppress the formation of oxidative metabolites, and prevent acetaminophen induced hepatotoxicity.

Production of Bioactive Peptides from Milk (우유에서 생리활성 펩타이드의 생산)

  • Milk-derived bioactive peptides have been found to exhibit various physiological activities such as angiotensin-converting enzyme (ACE) inhibitory, antibacterial, and antioxidative effects. Bioactive peptides can be used in the formulation of functional foods, nutraceuticals, and natural drugs because of their beneficial effects. However, the degree of variability in the composition, functionality, and sensory properties of such peptides has greatly limited their use in the food industry. In this review, we discuss the main peptides obtained from milk proteins and summarize findings from previous studies on the production and biological activities of these peptides. In addition, we compare the methods used to separate and identify the structure of the bioactive peptides and highlight current investigations into engineering and implementation of technologies that would allow more efficient isolation of bioactive peptides for functional food production. To improve human health, further molecular biology studies will also be required to elucidate the complex network of interactions between food microorganisms and the digestive system.
