Chemoprevention of Azoxymethane Induced Colon Cancer in Rats by Feeding Grange Juice, Soy, Wheat Bran and Flaxseed

  • Epidemiologic studies consistently demonstrate an inverse relationship between risk for colon canter and consumption of fruits and vegetables. Wheat bran, flax and soy contain dietary fiber and phytochemicals, such as lignans and isoflavones, that may inhibit colon carcinogenesis. Orange juice contains hesperidin, a flavanone glucoside that protects against colon carcinogenesis. This study determined if feeding orange juice, wheat bran, soy and flaxseed (combined diet) would inhibit azoxymethane (AOM) induced colon cancer. Cancer was initiated in male Fisher 344 rats by injecting 15 mg AOM/kg of weight at 22 and 29 days of age. One week after the second AOM injection, rats (N = 30) in the combined diet group received dry diet containing wheat bran (4%), soy with ethanol soluble phytochemirals(13%) and flaxseed (8%) and orange juice replaced drinking water. The control group remained on the control diet and received distilled water to drink. The rats were killed 28 weeks later, and colon tissues and tumors were removed for histologic analysis. Feeding the combined diet significantly reduced tumor incidence (p < 0.05), however tumor multiplicity was not changed (p > 0.05, 0.9 tumors/rat fed the combined diet vs 1.2 for controls). Also, tumor burden was only marginally reduced in rats fed the combined diet vs control rats (65 vs 210 mg of tumor/rats, respectively). The reduction in tumor incidence was associated with a decreased labeling index and proliferation zone in normal appearing colon mucosa. Therefore, this study shows that phytochemicals in wheat bran, soy, flax and orange juice reduce colon carcinogenesis, presumably by decreasing cell proliferation and enhancing cell differentiation.

Effects of vitamin C and E supplementation on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet

  • We compared the preventive capacity of high intakes of vitamin C (VC) and vitamin E (VE) on oxidative stress and liver toxicity in rats fed a low-fat ethanol diet. Thirty-two Wistar rats received the low fat (10% of total calories) Lieber-DeCarli liquid diet as follows: either ethanol alone (Alc group, 36% of total calories) or ethanol in combination with VC (Alc + VC group, 40 mg VC/100 g body weight) or VE (Alc + VE group, 0.8 mg VE/100 g body weight). Control rats were pair-fed a liquid diet with the Alc group. Ethanol administration induced a modest increase in alanine aminotransferase (ALT), aspartate aminotransferase (AST), conjugated dienes (CD), and triglycerides but decreased total radical-trapping antioxidant potential (TRAP) in plasma. VE supplementation to alcohol-fed rats restored the plasma levels of AST, CD, and TRAP to control levels. However, VC supplementation did not significantly influence plasma ALT, AST, or CD. In addition, a significant increase in plasma aminothiols such as homocysteine and cysteine was observed in the Alc group, but cysteinylglycine and glutathione (GSH) did not change by ethanol feeding. Supplementing alcohol-fed rats with VC increased plasma GSH and hepatic S-adenosylmethionine, but plasma levels of aminothiols, except GSH, were not influenced by either VC or VE supplementation in ethanol-fed rats. These results indicate that a low-fat ethanol diet induces oxidative stress and consequent liver toxicity similar to a high-fat ethanol diet and that VE supplementation has a protective effect on ethanol-induced oxidative stress and liver toxicity.

상수리첨가 식이가 흰쥐의 혈청지질 및 간장에 미치는 영향 (The Effect of Acon on the Serum and Liver of Rats)

  • The antioxidant effects of freeze-drying acorn were examined to find how much the freeze-drying acorn can reduce peroxidized corn oil poisoning, which influenced cholesterol, triglyceride, phospholipids, lipidperoxides, GOT(glutamate oxaloacetate transaminase), GPT(glutamate pyruvate transaminase) in serum, and cholesterol, triglyceride, lipidperoxides, fatty acid of phospholipids, SOD(superoxide dismutase), catalase in liver. In this experiment, male rats of Sprague-Dawley strain were used. The rats were divided into 6 groups, which were fed differently for 5 weeks : basal diet, 10% peroxidized corn oil added to basal diet, 1% acorn flour and 10% peroxidized corn oil added to basal diet, 5% acorn flour and 10% peroxidized corn oil added to basal diet, 10% acorn flour and 10% peroxidized corn oil added to basal diet, and 0.25% tannic acid and 10% peroxidized corn oil added to basal diet. The results were as follows : It was found that the peroxidized corn oil-fed 5 weeks induced the elevation of cholesterol, triglycerides, lipid peroxides, GOT, GPT in serum, and cholesterol, triglycerides, lipid peroxides in liver as compared to the basal diet-fed rats, but the acorn flour-fed rats reduced the elevation of these components. In addition, saturated fatty acid in rat liver phospholipids induced the elevation by feeding of peroxidized corn oil and, on the other hand, the acorn flour-fed rats reduced the elevation of saturated fatty acids. The acorn flour-fed rats reduced the activity of SOD in liver while they enhanced the activity of catalase in liver as compared with the peroxidized corn oil-fed rats.

Effect of Dietary Monascus Pigment on the Liver Damage Induced with $CCl_4$in Rats

  • In the biological world, there are a number of ecological fights for survival between each organism such as plants, animals and microorganism In such events, an organism can use its natural bioactive products as defence agent against other organisms. Furthermore, natural bioactive products can be utilized for medicine or functional food. Recently, we investigate the effect of Monascus pigment extracted from a fungus, Monascus anke, on the alcohol metabolism and blood lipid profile. In the present study, it is observed that Monascus pigment supplemented dietary may have a hepatoprotective effect on rat's liver damage induced with $CCl_4$ . By treatment with $CCl_4$(3 times, I.P), liver damage was reduced more in the rats fed 2% Monascus pigment extract supplemented diet than those fed standard diet, based on the serum levels of alanine aminotransferase, microsomal glucose-6-phosphatse activity and hepaic malondialdehyde content. On the other hand, oxygen free radical generating enzymes, hepatic P-450 dependent aniline hydroxylase, xanthine oxidase, and oxygen free radical scavenging enzymes, hepatic glutathione S-transferase, catalase, superoxide dismutase activities were generally higher both in $CCl_4$ treated group and control fed 2% Monascus pigment extract supplemented diet than those fed standard diet. In conclusion, the rats fed 2% Monascus pigment extract supplemented diet showed more reduced liver damage than those fed standard diet, which may be due to the acceleration of oxygen free radical metabolism.

고지방식이 유도 비만 랫드에 대한 유황오곡충 추출물의 지질감소 메카니즘 (Lipid lowering mechanism of sulfur-fed grain larvae extract in high-fat induced obesity rats)

  • This study was carried out to determine the action mechanism of sulfur-grain larvae extract (SGE) on anti-obesity and the reduction of blood lipid level in high-fat diet induced obese model animals. Animals were classified into a normal diet group (NC, normal control), HFD (high-fat diet without SGE), HFD 15 (high-fat diet + oral administration of 15 mg of SGE extract per 100 g body weight) and HFD 30 (high-fat diet + oral administration of 30 mg of SGE extract per 100 g body weight). The body weight gain declined in HFD 15 and HFD 30 groups compared with the HFD group, even though the diet intake increased significantly. The weight of liver and adipose tissue increased significantly in HFD group compared with in the HFD 15 and HFD 30 groups. Triglyceride, total cholesterol, LDL-C and AI decreased in HFD 15 and HFD 30 groups compared with in the HFD group, but the contents of HDL-C increased significantly. Expression of SREPB-$1{\alpha}$, SREPB-2 mRNA in the liver was lower in the high-fat diet group compared with the HFD group, but the expression of LPL mRNA in adipose tissue and $PPAR{\alpha}$ increased significantly. Fat accumulation in the liver tissues and liver damage were greatly reduced in HFD 15 and HFD 30 groups compared with in the HFD group. The size of adipocytes became smaller in the HFD 15 and HFD 30 groups compared with HFD group. In conclusion, this research discovered for the first time that grain maggot has anti-obesity effects, by reducing the abdominal fat of obese model animals and lowering blood lipid level through the down-regulation of PPAR-$1{\alpha}$ and SREPB-2 mRNA and the up-regulation of PPAR-${\alpha}$ mRNA.

산사의 장내 미생물 조절을 통한 항비만 효과 (Anti-obesity Effect of Crataegus pinnatifida through Gut Microbiota Modulation in High-fat-diet Induced Obese Mice)

  • Objectives This study was performed to evaluate anti-obesity effects of Crataegus pinnatifida (CP) on high-fat-diet induced obese mice. Methods The experimental animals were divided into four groups: normal diet (NOR) group, high fat diet (HFD) group, HFD+Xenical (XEN) group, and HFD+CP (CP) group. NOR group was fed a normal diet and the other three groups were fed high fat diet during the experiment. After the first two weeks of diet, XEN group and CP group were administered with XEN or CP for seven weeks, respectively. After that, we measured body weight, liver weight, fat weight, food intake, and serum concentrations of lipids and liver enzymes. Also the liver, intestine, fat tissue was removed to estimate the obesity-related mRNA expressions and the stool sample was collected to analyze the gut microbiota. Results We found that body weight, fat weight, and triglyceride level were decreased significantly in CP group compared to HFD group. Also CP significantly suppressed gene expressions associated with lipogenesis and inflammation, and increased gene expressions of browning of white adipose tissue and mitochondrial biogenesis. Moreover, it shifted the microbial diversity closer to that of NOR group and increased Firmicutes/Bacteriodetes ratio. Conclusions These results suggest that CP decrease body weight, fat weight and serum triglyceride. Also it inhibit inflammation and adipogenesis, altering gut microbial diversity and abundance. In conclusion, CP could be used as a therapeutic drug for obesity via gut microbiota modulation.

비알콜성 지방간 동물모델에서 갈화추출물의 간보호 효과 (Hepatoprotecive Effects of Puerariae Flos Extract on Experimental Non-alcoholic Fatty Liver Disease Models)

  • Objectives : The aim of this study is to investigate the preventive effect of Puerariae Flos ethanol extract (PE) on methionin and choline deficient (MCD)-diet-induced Nonalcoholic fatty liver disease (NAFLD) in C57BL/6J mice. Methods : In the in vivo experiments, C57BL/6J mice were divided into 4 groups; Normal group, Control group, MCD+PE 100 group, and MCD+PE 300 group. After 4 weeks, body weight, liver weight, biochemical parameters for liver function test, histological changes, reverse transcription polymerase chain reaction (RT-PCR), and Western blot were assessed. Results : Mice lost body weight with the MCD-diet and the MCD+PE 100 group and MCD+PE 300 groups lost less than the control group, though showed no statistical significance. Liver weights were decreased by the MCD diet, but MCD+PE 300 groups were increased significantly. In the liver function test, all the values were decreased with the MCD-diet, MCD+PE 100 group and MCD+PE 300 groups were increased significance. In histological findings of the livers, MCD-diet induced severe fatty accumulation in the livers, but this fatty change was reduced in the MCD+PE 100 group and MCD+PE 300 groups was inhibited respectively. In lipid accumulation factors (such as SREBP-1c, $C/EBP{\alpha}$, PPAR-${\gamma}$), MCD+PE 100 group and MCD+PE 300 groups showed inhibitory effect on liver lipogenesis by reducing associated gene expressions caused by MCD diet. Conclusions : We were able to know that Puerariae Flos ethanol extract (PE) shown hepatoprotective effects via a decrease on the hepatic lipogenesis factors in the experimental NAFLD Models.

Ellagic acid, a functional food component, ameliorates functionality of reverse cholesterol transport in murine model of atherosclerosis

  • BACKGROUND/OBJECTIVES: High levels of plasma low-density lipoprotein (LDL) cholesterol are an important determinant of atherosclerotic lesion formation. The disruption of cholesterol efflux or reverse cholesterol transport (RCT) in peripheral tissues and macrophages may promote atherogenesis. The aim of the current study was to examine whether bioactive ellagic acid, a functional food component, improved RCT functionality and high-density lipoprotein (HDL) function in diet-induced atherogenesis of apolipoproteins E (apoE) knockout (KO) mice. MATERIALS/METHODS: Wild type mice and apoE KO mice were fed a high-cholesterol Paigen diet for 10 weeks to induce hypercholesterolemia and atherosclerosis, and concomitantly received 10 mg/kg ellagic acid via gavage. RESULTS: Supplying ellagic acid enhanced induction of apoE and ATP-binding cassette (ABC) transporter G1 in oxidized LDL-exposed macrophages, facilitating cholesterol efflux associated with RCT. Oral administration of ellagic acid to apoE KO mice fed on Paigen diet improved hypercholesterolemia with reduced atherogenic index. This compound enhanced the expression of ABC transporters in peritoneal macrophages isolated from apoE KO mice fed on Paigen diet, indicating increased cholesterol efflux. Plasma levels of cholesterol ester transport protein and phospholipid transport protein involved in RCT were elevated in mice lack of apoE gene, which was substantially reduced by supplementing ellagic acid to Paigen diet-fed mice. In addition, ellagic acid attenuated hepatic lipid accumulation in apoE KO mice, evidenced by staining of hematoxylin and eosin and oil red O. Furthermore, the supplementation of 10 mg/kg ellagic acid favorably influenced the transcriptional levels of hepatic LDL receptor and scavenger receptor-B1 in Paigen diet-fed apoE KO mice. CONCLUSION: Ellagic acid may be an athero-protective dietary compound encumbering diet-induced atherogenesis though improving the RCT functionality.

육의감비탕 열수 추출물이 비만 및 고지혈증 유도 생쥐에 미치는 효과 (Effects of Water Extract of Yukeuigambi-tang on Obesity and Hyperlipidemia in Mice Induced by High Fat Diet)

  • This experimental study was designed to investigate the inhibitory effects of hot water extract of Yukeuigambi-tang (YEGT) on high-fat diet-induced obesity and hyperlipidemia in C57BL/6 mice. Animals were divided into normal, control, YEGT(sample A 3.8 g/kg, and sample B 7.6 g/kg) treated group. Obesity with hyperlipidemia was induced by high fat diet treatment for 6 weeks. YEGT was given to the animals by oral gavage. The effect of YEGT on the body weight change, total cholesterol, HDL-cholesterol, LDL-cholesterol, triglyceride, and leptin levels in serum were evaluated. In respect to the body weight change, sample B significantly reduced body weight compared with control group in the 6th weeks. In the total cholesterol, triglyceride, and leptin level, sample B significantly reduced all of them compared with control group; sample A significantly reduced triglyceride and leptin levels compared with control group. The above results suggest that YEGT is effective to prevention and treatment of obesity and hyperlipidemia.

단삼투여가 고지방식이로 유발된 비만형 당뇨병 동물모델에 미치는 영향 (The Effects of Salvia miltiorrhiza on High Fat Diet-induced Obese Diabetic Mouse Model)

  • Objectives : Obesity is an important cause of diabetes, and lipotoxicity causes insulin resistance. In this study, we investigated the effects of Salvia miltiorrhiza on high fat diet-induced obese type 2 diabetic mouse models. Methods : Diabetes was induced in ICR male mouse (23~25 g) with Surwit's high fat, high sucrose diet. Mice were divided into 4 groups (n=10) of normal, control, Salvia miltiorrhiza, and metformin. After 8 weeks, body weight, OGTT, fructosamine, lipid profile, serum level of adiponectin and leptin, epididymal fat pad, liver weight and epididymal adipocyte size were measured. Results : Salvia miltiorrhiza significantly reduced oral glucose tolerance levels, fructosamine serum level, epididymal fat weight, and epididymal adipocyte size. Salvia miltiorrhiza also increased HDL-cholesterol, adiponectin and leptin serum levels. Conclusions : These results show that Salvia miltiorrhiza improves insulin resistance. Therefore we suggest that Salvia miltiorrhiza would be an effective treatment for obese type 2 diabetic patients.
