  • The effect of nonlamellar-prone lipids, diacylglycerol (DG) and phosphatidylethanolamine (PE), on the ATPase activity of SecA was examined. When Escherichia coli (E. coli) PE of the standard vesicles composed of 60 mol% of this lipid and 40 mol% of dioleoylphosphatidylglycerol (DOPG) is gradually replaced with either dioleoylgylcerol (DOG) or dioeloyl PE(DOPE), the ATPase activity of SecA present together increased appreciably.(omitted)

  • 공업적으로 중요성이 날로 더해가는 phospholipase C (PLC) 를 Bacillus cereus를 이용하여 생산하였다. 또한, plc::gfp fusion protein 을 생산하는 재조합 E. coli 를 제조하고 배양하였으며 특히 형광센서를 이용하여 PLC 의 생산 특성을 모니터링하였다.

  • Phospoinositide-specific phospholipase C (PLC)는 세포막의 phosphoinositide를 분해하여 inositol phosphates와 diacylglycerol을 전달하는데 핵심적인 효소이다. PLC는 분자량과 1차구조의 비교에 의하여 type (PLC-$\beta$, ${\gamma}$, $\delta$)로 구분되며, 각 type마다 2-4종의 subtype이 존재하고 PLC isozyme들에 대한 현재가지의 각종 신호 전달 및 조절에 대한 연구를 종합하면: (1) PLC-$\beta$ type은 G-protein과 연결되어 신호를 전달받고, (2) PLC-${\gamma}$ type은growth factor receptor tyrosine kinase에 의하여 인산화 되어 활성화됨으로, 세포의 성장 신호를 전달하며. (3) PLC-$\delta$ type에 대한 신호 전달이나 조절은 밝혀지지 않고 있다.

Inositol이 돼지 난포란의 성숙에 미치는 영향

  • Inositol은 세포의증식 및 정보전달과정에 관여하는 phosphatidylinositol (PI)의 구성성분으로서 중요한 세포내 기능을 수행한다. P1는 세포내에서 특이적 인산화효소에 의하여 Pl4-phosphate(PIP), Pl4,5-phosphate($PIP_2$)로 변환되며 PIP$_2$는 phospholipase C(PLC)에 의하여 세포내 second messengers인 1,2-diacylglycerol (DAG)와 inositol 1,4,5-triphosphate ($IP_3$)로 변환된다. 이렇게 생산된 DAG와 IP3는 각각 protein kinase C의 활성과 $Ca^{2++}$의 동원에 관여하여 다양한 세포내 신호전달에 관여하는 것으로 보고되고 있다. 또한 mouse에서 $IP_3$의 작용에 의한 $Ca^{2++}$의 상승은 난모세포의 성숙분열이 촉진되고 돼지 난포세포에 있어서도 PI대사가 일어나고 있는 것으로 보고되고 있다. 본 연구에서는 돼지 미성숙 난모세포의 성숙과 단위발생에 미치는 inositol의 영향을 확인하기 위하여 실시하였다. inositol의 농도가 난포란의 성숙에 미치는 영향을 검토하기 위하여 난포란을 각각 $150 \mu mole$, $250 \mu mole$, $350 \mu mole$, inositol을 포함하는 Whitten's 배양액에서 44시간 성숙시킨 결과 $93.57 \pm 4.21, 93.91 \pm 2.71, 92.96 \pm 3.58%$가 성숙되어 대조구의 $87.10 \pm 4.21$보다 유의적(P<0.05)으로 차이를 나타내었다. 난포란의 등급에 따른 inositol의 영향을 확인하기 위하여 형태적으로 난구세포가 치밀한 난포란과 난구세포가 치밀하지 않은 난포란을 $250 \mu mole$ inositol을 포함하는 Whitten's 배양액에서 성숙을 유도한 결과 난구세포가 치밀한 난포란과 난구세포가 치밀하지 않은 난포란에서 inositol을 첨가하였을 때 성숙율은 각각 $95.35 \pm 2.22와 63.55 \pm 8.12$로 inositol을 첨가하지 않은 대조구보다($89.21 \pm 3.69와 48.56 \pm 8.99$) 유의적인 차이를 보였다. 난구세포가 inositol에 의한 성숙에 미치는 영향을 확인하기 위하여 난구세포를 제거한 난포란을 inositol을 포함하는 배지에서 성숙을 유도한 결과 inositol을 첨가하지 않은 처리구보다 양호한 성숙율을 보였다.

오로토산 유발 지방산의 간장 트리아실글리세롤 축적간 지방산 조성의 변화 (Change of Fatty Acid Compositions during Hepatic Triacylglycerol Accumulation in Dietary Orotic Acid-induced Fatty Liver)

  • Dietary orotic acid is known to induce the fatty liver. Fatty acid profiles in the lipid fraction of the liver and the serum in rats fed with or with orotic acid diet were analyzed. In all the hepatic lipid fraction of rats fed on the supplemented orotic acid diet, there was a significant increased in linoleic acid. In addition, linoleic acid was also increased in the triacylglycerol fraction of hepatic endoplasmic reticulum and the triacylglycerol and diacylglycerol fractions of hepatic Golgi apparatus of the orotic acid-feeding rats. In the time course study of the fatty acid profile in the hepatic triacylglycerol and diacylgycerol fractions, an increase of linoleic acid was observed similarly in the initial stages of orotic acid intake in the both fractions. However, linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats increased from day 1, but it began to decrease the increment from day 2, resulting in the lower level of linoleic acid in the serum triacylglycerol fraction of orotic acid-feeding rats than that of rat fed a orotic acid-free diet after 10 days. Oleic acid (18:1) was increased in the only cholesteryl ester fraction of helpatic. However, oleic acid level in other fractions was not changed. The compositions of 14:0, 16:0 and 18:0 was reduced in the hepatic triacylogylcerol, diacylglycerol and cholesteryl ester fractions by orotic acid-feeding. However, these saturated fatty acids were significantly increased in the serum triacylglycerol fractions. The orotic acid indcued changes in linoleic acid level in hepatic triacylglycerol may be explained by the impaired fatty acid metabolism and limited excretion of this fatty acid from liver to serum.

Identification of triacylglycerols in coix seed extract by preparative thin layer chromatography and liquid chromatography atmospheric pressure chemical ionization tandem mass spectrometry

  • Here we reported a methodology for identification of triacylglycerols (TAGs) and diacylglycerols (DAGs) in coix seed by preparative thin layer chromatography (prep-TLC) and non-aqueous reversed-phase liquid chromatography (NARP LC)-atmospheric pressure chemical ionization (APCI) tandem mass spectrometry (MS/MS). Lipid components were extracted from coix seed by reflux extraction using n-hexane for 3 hr. TAGs and DAGs in coix seed extract were effectively purified and isolated from matrix interferences by prep-TLC and then analyzed by LC-APCI-MS and MS/MS for identification. TAGs were effectively identified taking into consideration of their LC retention behavior, APCI-MS spectra patterns, and MS/MS spectra of $[DAG]^+$ ions. In MS/MS spectra of TAGs, diacylglycerol-like fragment $[DAG]^+$ ions were useful to identify TAGs with isobaric fragment ions. Based on an established method, 27 TAGs and 8 DAGs were identified in coix seed extract. Among them, 15 TAGs and 8 DAGs were for the first time observed in coix seed. Interestingly, some of TAGs isolated by prep-TLC were partly converted into DAGs through probably photolysis process during storing in room temperature. Thus, degradation phenomenon of TAGs should be considered in the quality evaluation and nutritional property of coix seed. LC-APCI-MS/MS combined with prep-TLC will be practical method for precise TAG and DAG analysis of other herbal plants.

Sphingomonas chungbukensis DJ77에서 Sphingosine Kinase를 암호화하는 spk 유전자의 동정과 대장균에서의 발현 (Identification of the spk Gene Encoding Sphingosine Kinase in Sphingomonas chungbukensis DJ77 and Its Expression in Escherichia coli)

  • Sphingomonas chungbukensis DJ77의 유전체 서열분식 과정에서 969개의 nucleotide로 구성된 sphingosine kinase 유전자를 동정하였다. 이 sphingosine kinase 단백질의 아미노산 서열은 Zymomonas mobilis subsp. mobilis ZM4의 sphingosine kinase 아미노산 서열과 $55\%$의 상동성을 보였다. 또한 다중서열정렬을 통해 각각 진핵세포의 sphingosine kinase의 C2, C3, C5 domain에 속하는 3개의 conserved sequence를 발견하였다. 그 중 하나는 sphingosine kinase에서 ATP-binding site일 것으로 예상되어지는nucleotide-binding motif(GGDG)였고 나머지 둘은 아직 기능이 알려지지 않은 conserved sequences 였다. 이러한 다중서열정렬을 바탕으로 계통수를 그려본 결과, S. chungbukensis DJ77의 sphingosine kinase (SPK)는 COG1597 그룹과 유사했으며, COG1597 내에서 동일종의 diacylglycerol kinase와는 서로 다른 그룹에 속하는 것으로 나타났다. 재조합 SPK는 이종(異種)세포인 Escherichia coli내에서 성공적으로 과발현 되었으나, 세포 내에서 불용성 복합체(inclusion body)를 형성하였다.

DMH로 처리한 쥐에서 식이에 첨가한 Conjugated Linoleic Acid 함량에 따라 대장세포의 Apoptosis와 대장암 Biomarker에 미치는 영향 (Effect of Different Level of Conjugated Linoleic Acid on Apoptosis of Colonic Mucosal Cell and Biomarkers in Colon Cancer of 1,2-Dimethylhydrazine-Treated Rats)

  • The study was designed to compare the anti-carcinogenic effect of different level of conjugated linoleic acid (CLA) in 1,2-dimethylhydrazine (DMH)-treated rats by determining biomarkers (apoptosis, cell proliferation, eicosanoids, 1,2-diacylglycerol) and phospholipid fatty acid profile in colonic mucosa. Eighty male Sprague Dawley rats weighing 180-220g were randomly divided into 4 groups depending on the content of CLA, i.e. 0.0% CLA, 0.5% CLA, 1.0% CLA, 1.5% CLA. The experimental diet contained protein 21.6%, carbohydrate 54.6%, and fat 14.5% including CLA mixture at different level by weight. The experimental diet was fed for 14 weeks with the initiation of intramuscular injection of DMH, which was injected twice a week for 6 weeks to give total amount of 180 mg/kg body weight. Regardless of the amount of CLA supplemented to diet, CLA significantly increased the apoptotic index but did not have significant effect on cell proliferation in colonic mucosa. CLA was undetected in colonic mucosal phospholipid of rats fed the 0% CLA diet and increased to 5.9mg/g phospholipid in rats fed the 0.5% diet. The apoptotic index was increased by 251% and the 1,2-DAG content was decreased by 57% in rats fed 0.5% CLA. No further changes in these variables were observed when CLA in the diet was raised to 1.0% or 1.5%. However, dietary CLA decreased mucosal levels of prostaglandin (PG)E$_2$, thromboxane (TX)B$_2$, and arachidonic acid in dose-dependent manner. The present data indicate that dietary CLA can inhibit DMH-induced colon carcinogenesis by mechanism probably involving increased apoptosis.
