Adhesion and Biofilm Formation Abilities of Bacteria Isolated from Dental Unit Waterlines (치과용 유니트 수관에서 분리한 세균의 부착 및 바이오필름 형성 능력)

  • The purpose of our study is to compare the adhesion and biofilm formation abilities of isolates from water discharged from dental unit waterlines (DUWLs). Bacteria were isolated from a total of 15 DUWLs. Twelve isolates were selected for the experiment. To confirm the adhesion ability of the isolates, each isolate was attached to a glass coverslip using a 12-well plate. Plates were incubated at $26^{\circ}C$ for 7 days, and the degree of adhesion of each isolate was scored. To verify the biofilm formation ability of each isolate, biofilms were allowed to form on a 96-well polystyrene flat-bottom microtiter plate. The biofilm accumulations of all isolates formed at $26^{\circ}C$ for 7 days were identified and compared. A total of 56 strains were isolated from 15 water samples including 12 genera and 31 species. Of the 56 isolates, 12 isolates were selected according to the genus and used in the experiment. Sphingomonas echinoides, Methylobacterium aquaticum, and Cupriavidus pauculus had the highest adhesion ability scores of +3 among 12 isolates. Among these three isolates, the biofilm accumulation of C. pauculus was the highest and that of S. echinoides was the third-most abundant. The lowest biofilm accumulations were identified in Microbacterium testaceum and M. aquaticum. Most isolates with high adhesion ability also exhibited high biofilm formation ability. Analysis of adhesion and biofilm formation of the isolates from DUWLs can provide useful information to understand the mechanism of DUWL biofilm formation and development.

  • The purpose of this study is to examine the relationship between sustainable PLM(Product Lifecycle Management) activity and performance. To ensure this purpose, we designed the S-PLM Framework which is consisted of traditional PLM activity and sustainable PLM activity. We also conducted path analysis to investigate PLM success factor on manufacturing company and to understand the relationship between these success factors. First the result of analysis of the relationship between traditional PLM activity and sustainable performance. Second, there is significantly positive relationship between sustainable activity and performance. Third, traditional PLM activity and sustainable PLM activity factor have an influence on the innovation performance factor. Fourth, sustainable performance have an effect on the management and business performance. In conclude we analyzed and verified the influence sustainable PLM establishment mechanism and the sustainable PLM activity factors. Therefore this study is to create innovative performance and to improve efficiency of Convergence PLM establishment and operation.

A Study on the Characteristics and Cleanliness of Fluidic Strip Process of Environment-Friendly Aqueous Stripper (친환경 수계 박리액의 유동박리 공정 특성 및 청정성 연구)

  • In this research, we investigated the cleanliness by optimizing the water content of the aqueous stripper in fluidic strip process. The stripping properties of the photoresist with optimized aqueous stripper were compared with the commercial organic stripper. The stripping performance was evaluated by electrical and optical characteristics on the surface of the transparent electrode that compare with stripped the transparent electrode surface and the rare surface before patterning by the photoresist. As a result of the photoresist stripping process of the organic stripper and the aqueous stripper optimized for water content, the aqueous stripper exhibited better electrical and optical characteristics than the organic stripper. In the case of the fluidic strip process with organic stripper, the photoresist dissolves in the stripper solution during stripping which can cause re-adsorption by contamination. Whereas that the aqueous stripper under development seems to decrease the photoresist dissolution in the stripper solution. Because the cyclodextrin contained in the stripper captures organic photoresist into hall of cyclodextrin which stripped through swelling and tearing. The photoresist residue captured by the cyclodextrin can be filtered. After the fluidic stripping process by different chemical stripping mechanism, the cleanliness of the organic stripper and aqueous stripper was compared and analyzed.

Korean Red Ginseng Extract inhibits Tumor Necrosis Factor-alpha-induced Monocyte Adhesion in the Human Endothelial Cells

  • Vascular inflammation is an important step in the development of cardiovascular disorder. Since it has not been known whether Korean red ginseng has a role to play on the vascular inflammation, we investigated the effects of Korean red ginseng extract (KRGE) on monocyte adhesion and its underlying signaling mechanism. Monocyte adhesion assay and Western blot were conducted on the human umbilical vein endothelial cells to study monocyte adhesion and the expression of adhesion molecules. Intracellular calcium was measured with Fura-2 fluorescent staining, and superoxide production was measured with lucigenin chemiluminescence in the endothelial cells. KRGE inhibits tumor necrosis factor (TNF)-alpha-induced monocyte adhesion on the endothelial cells at the range of $0.03{\sim}1$ mg/ml. TNF-alpha-induced vascular cell adhesion molecule-1 and intercellular cell adhesion molecule-1 expression were inhibited by the pretreatment of KRGE in the endothelial cells. KRGE also inhibits TNF-alpha-induced intracellular calcium and the superoxide production in the endothelial cells. This study first demonstrated that KRGE inhibits TNF-alpha-induced monocyte adhesion by inhibiting the adhesion molecule expression, intracellular calcium and superoxide production in the endothelial cells. Therefore, the anti-inflammatory function of KRGE may be contributed to protecting the endothelial dysfunction in the vascular inflammatory disorders.

Effects of ginseol k-g3, an Rg3-enriched fraction, on scopolamine-induced memory impairment and learning deficit in mice

  • Background: Although ginsenosides such as Rg1, Rb1 and Rg3 have shown promise as potential nutraceuticals for cognitive impairment, their use has been limited due to high production cost and low potency. In particular, the process of extracting pure Rg3 from ginseng is laborious and expensive. Methods: We described the methods in preparing ginseol k-g3, an Rg3-enriched fraction, and evaluated its effects on scopolamine-induced memory impairment in mice. Results: Ginseol k-g3 (25-200 mg/kg) significantly reversed scopolamine-induced cognitive impairment in the passive avoidance, but not in Y-maze testing. Ginseol k-g3 (50 and 200 mg/kg) improved escape latency in training trials and increased swimming times within the target zone of the Morris water maze. The effect of ginseol k-g3 on the water maze task was more potent than that of Rg3 or Red ginseng. Acute or subchronic (6 d) treatment of ginseol k-g3 did not alter normal locomotor activity of mice in an open field. Ginseol k-g3 did not inhibit acetylcholinesterase activity, unlike donezepil, an acetylcholinesterase inhibitor. Rg3 enrichment through the ginseol k-g3 fraction enhanced the efficacy of Rg3 in scopolamine-induced memory impairment in mice as demonstrated in the Morris water maze task. Conclusion: The effects of ginseol k-g3 in ameliorating scopolamine-induced memory impairment in the passive avoidance and Morris water maze tests indicate its specific influence on reference or long-term memory. The mechanism underlying the reversal of scopolamine-induced amnesia by ginseol k-g3 is not yet known, but is not related to anticholinesterase-like activity.

  • All-trans retinoic acid (AtRA) induces growth inhibition and apoptosis in a variety of tumer cells, including MCF-7 cells. Insulin-like growth factors (IGFs) system has been reported to be associated with the development of cancer. Although MCF-7 cell with AtRA is to be the major stimulus for the cell growth and apoptosis, the mechanism of insulin-like growth factor-I (IGF-I)/insulin-like growth factor binding protein-3 (IGFBP-3) system remains to be elucidated. Thus, this study was conducted to the effect of AtRA on the gene expression and level of IGF-I and IGFBP-3. In addition, we investigated the involvement of PKC-${\delta}$ on the IGF-I and IGFBP-3 secretion in MCF-7 cell. AtRA(${\geq}10^{-7}M$) decreased the IGF-1 secretion and mRNA expressions, but increased IGFBP-3 secretion and mRNA expressions in MCF-7 cells. Especially, the treatment of AtRA at 72 hours caused a significant reduction in the IGF-I secretion and mRNA expressions but increment in IGFBP-3 secretion and mRNA expressions (p < 0.05). $10^{-7}M$ AtRA activated PKC-${\delta}$ that is one among PKC-$\iota$, ${\alpha}$, ${\lambda}$ and ${\delta}$ in MCF-7 cell. Rotllerin, a PKC-${\delta}$ inhibitor, blocked AtRA-induced inhibition of the IGF-I and mRNA expressions, and increase of lGFBP-3 and mRNA expressions in MCF-7 cell. Together, AtRA inhibited the IGF-I secretion and mRNA expressions, but increased IGFBP-3 secretion and mRNA expressions in MCF-7 cell. Furthermore, AtRA-induced alteration of IGF-I, IGFBP-3 secretion, and the gene expressions were mediated via PKC-${\delta}$ activity.

Siamese Crocodile White Blood Cell Extract Inhibits Cell Proliferation and Promotes Autophagy in Multiple Cancer Cell Lines

  • Cancer represents one of the most significant threats to human health on a global scale. Hence, the development of effective cancer prevention strategies, as well as the discovery of novel therapeutic agents against cancer, is urgently required. In light of this challenge, this research aimed to evaluate the effects of several potent bioactive peptides and proteins contained in crocodile white blood cell extract (cWBC) against LU-1, LNCaP, PC-3, MCF-7, and CaCo-2 cancer cell lines. The results demonstrate that 25, 50, 100, and $200{\mu}g/ml$ cWBC exhibits a strong cytotoxic effect against all investigated cell lines ($IC_{50}$ $70.34-101.0{\mu}g/ml$), while showing no signs of cytotoxicity towards noncancerous Vero and HaCaT cells. Specifically, cWBC treatment caused a significant reduction in the cancerous cells' colony forming ability. A remarkable suppression of cancerous cell migration was observed after treatment with cWBC, indicating potent antimetastatic properties. The mechanism involved in the cancer cell cytotoxicity of cWBC may be related to apoptosis induction, as evidenced by typical apoptotic morphology features. Moreover, certain cWBC concentrations induced significant overproduction of ROS and significantly inhibited the $S-G_2/M$ transition in the cancer cell. The molecular mechanisms of cWBC in apoptosis induction were to decrease Bcl-2 and XIAP expression levels and increase the expression levels of caspase-3, caspase-8, and p53. These led to a decrease in the expression level of the cell cycle-associated gene cyclin-B1 and the arrest of cell population growth. Consequently, these findings demonstrate the prospect of the use of cWBC for cancer therapy.

Effects of Activin on Testosterone-primed Immature Rainbow Trout Gonadotropin Release in vitro (Testosterone 처리한 미성숙 무지개송어 뇌하수체의 세포배양계에서 생식소자극초르몬 분비에 대한 Activin의 효과)

  • The present studies were conducted to evaluate the effects of activin-A on gonadotropins (GTHs) release in testosterone-treated immature rainbow trout Oncorhpynchus mykiss. The administration of testosterone elevated pituitary level of GTH II but not of GTH I. In this study using primary cultures of dispersed pituitary cells in static incubation, dose-dependent increases in GTH II release was observed in the activin-treated group at day 3 of incubation (long-term incubation), but not at day 1 of incubation (short-term incubation). Dopamine, a potent inhibitor of gonadotropin-releasing hormone (GnRH)-stimulated GTH II release in rainbow trout, was only partially effective in decreasing actvin-induced GTH II release. Furthermore, salmon GnRH (sGnRH)-stimulated GTH II release was not potentiated by the pretreatment with activin. However, the control mechanisms of GTH I release by activin and other hormones were not observed in the all tested experiments. The results of these studies support the contention that in contrast with the usual stimulatory effects of activin on GTH release in mammals, activin exerts long-term stimulatory actions on GTH II release in rainbow trout. The control mechanism of GTH I release, however, is a question that remains to be elucidated.

A Pedestrian Network Assignment Model Considering Space Syntax (공간구문론(Space Syntax)을 고려한 통합보행네트워크 통행배정모형)

  • In Space Syntax, the greater the degree of integration between separate links, the greater the links' accessibility from the target network. As such, planning pedestrian walks so that links with high degrees of integration are connected, or else inducing high integration value land use are both valid options. The travel distribution model reflects how walking demand, or more specifically, the pedestrian, partakes in route choosing behavior that minimizes select criteria, notably level of discomfort, as measured using travel distance and time. The model thus demonstrates travel patterns associated with demand pertaining to minimization of discomfort experienced by the pedestrian. This research introduces a method that integrates Space Syntax and the pedestrian travel distribution model. The integrated model will determine whether regions with high degrees of integration are actually being used as pivots for pedestrian demand movement, as well as to explain whether the degree of integration is sustained at an appropriate level while considering actual movement demand. As a means to develop the integrated model, a method that combines display of the visibility of the space syntax network and road-divided links is proposed. The pedestrian travel distribution model also includes an alternative path finding mechanism between origin and destination, which allows for uniform allocation of demand.

Effect of Pinelliae Rhizoma Herbal Acupuncture on the Release of Thymus and Activation-Regulated Chemokine(TARC) in Human Bronchial Epithelial Cell (반하(半夏) 약침액(藥鍼液)이 사람 기관지 상피세포의 TARC 분비에 미치는 효과)

  • Objective : Chemokines are important for the recruitment of leukocytes, which is essential in host defense to the sites of infection. The thymus and activation-regulated chemokine (TARC) is a CC chemokine which potentially plays a role via a paracrine mechanism in the development of allergic respiratory diseases. Methods : The objective of this study is to investigate the effect of Pinelliae Rhizoma Herbal Acupuncture Solution (PHS) on the secretion of TARC of human bronchial epithelial cell. Methods : Enzyme-linked immunosorbent assay (ELISA) was performed to detect the secretion of TARC. The cytotoxicity was measured by MTT assay. Results : PHS significantly inhibited the secretion of TARC with a dose-dependant manner. The effective dosage did not have the cytotoxicity on human bronchial epithelial cell by MTT assay. Conclusion : Results of our study imply that PHS would play an important role in modulation of TARC in Human bronchial epithelial cells.
