RAV1 Negatively Regulates Seed Development by Directly Repressing MINI3 and IKU2 in Arabidopsis

  • A plant-specific B3 domain and AP2 domain-containing transcription factor, RAV1 acts as a negative regulator of growth in many plant species and its transcription was down-regulated by BR and ABA. In this study, we found that RAV1-overexpressing transgenic plants showed abnormally developed ovules, resulting in reduced seed size, weight, and number in a silique. Interestingly, the endogenous expression of RAV1 fluctuated during seed development; it remained low during the early stage of seed development and sharply increased in the seed maturation stage. In plants, seed development is a complex process that requires coordinated growth of the embryo, endosperm, and maternal integuments. Among many genes that are associated with endosperm proliferation and embryo development, three genes consisting of SHB1, MINI3, and IKU2 form a small unit positively regulating this process, and their expression was regulated by BR and ABA. Using the floral stage-specific RNAs, we found that the expression of MINI3 and IKU2, the two downstream genes of the SHB1-MINI3-IKU2 cascade in the seed development pathway, were particularly reduced in the RAV1-overexpressing transgenic plants. We further determined that RAV1 directly binds to the promoter of MINI3 and IKU2, resulting in their repression. Direct treatment with brassinolide (BL) improved seed development of RAV1-overexpressing plants, but treatment with ABA severely worsened it. Overall, these results suggest that RAV1 is an additional negative player in the early stages of seed development, during which ABA and BR signaling are coordinated.

  • In this paper, we analyzed the communication distances in the flat and spherical earth model using the main specifications of the wireless communication device under development to be installed in the unmanned surface vehicle(USV). We installed the wireless communication device on the test ship and the actual communication test was performed in the South Sea of Korea, it compared the experimental result with the simulated results. If the installation altitude of the transmitting and receiving antennas is low, there is not the big difference between the two models. However, when the altitude of one of the two antennas is high, the spherical earth model has similar results to the experimental data. As the altitude of the antenna installation increases, fading occurs in a certain section. We expect that this fading can be overcome through antenna technologies such as space diversity.

Collaboration in Agrotourism Development: The Role of Local Government in Yeongdong County, Chungcheongbuk Province, Korea

  • Background and objective: There are many theories related to the development of agrotourism, and each stakeholder has its own role and function. If it is based on organizational functions, stakeholders need to be coordinated, and the state plays a major administrative role. But in the successful case of Yeongdong County, Chungcheongbuk province, the Republic of Korea, the government played an important role in the development of tourism, which grants us another perspective on the role of the government in promotion and development agrotourism activities. Methods: This article aims to examine the development of agrotourism through a cooperative approach, by clarifying the role of the local authorities in the Yeongdong region in the management and operation of the local agrotourism activities. The qualitative research method is based on a field survey conducted in Yeongdong County in 2019. Results: The study's findings show that in situations where conflicts of interest among stakeholders are prone to occur, the development of local tourism has a limited impact on the cooperative culture, and local governments play an important role. Accordingly, the local governments must actively formulate strategies to maintain and develop traditional agricultural industries and at the same time develop agrotourism activities. The government's ability to coordinate, cooperate, and participate reasonably is a key factor for the success of the sustainable development of the tourism industry. Conclusion: Based on coordination between stakeholders and a reasonable strategic direction, Yeongdong's agrotourism is being effectively deployed. The development of Yeongdong's agrotourism industry has achieved impressive results through an effective strategy, highlighting the important role of localgovernment in the consensus of local communities and collaboration with stakeholders. The role of outstanding local governments has been clearly demonstrated, especially in promoting, establishing, and contacting different organizations to effectively deploy agrotourism in the region.

Factor Analysis of Soil and Water Quality Indicators in Different Agricultural Areas of the Han River Basins (한강수계 농업지대에서 토양과 수질 지표에 대한 요인 분석)

  • Factor analysis technique was employed to screen the principal indicators influencing soil and water qualities in the intensively cultivated areas of the Han River Basin. Soil chemical parameters were analyzed for the soil samples collected at intensive farming area in Pyungchang-Gun, and water quality monitoring data were obtained from the agricultural small catchments of Han River Basin during 1996 and 1997. Among the $11{\times}11$ cross correlation matrix, 29 correlations were significant out of 55 soil quality indicator pairs. The overall Kaiser's measure of sampling adequacy(KMS) value was acceptable with 0.60. Most indicators except iron were acceptable. Among soil indicators, the first factors showing high factor loadings were pH, Ca and Mg. The factor loading was the highest for Ca. The second factor could be characterized as phosphate and micronutrient. The third factor was organic matter and EC, and the fourth factor was potassium and Fe. Out of 190 water quality indicators, 86 correlations were significant. Overall KMS value was 0.74, but the KMS values for pH, TSS, Cd, Cu and Fe were lower than 50. The first factor of EC accounts 27.1 percents of the total variance, and showed high factor loadings with Na, Ca, $SO_4$, Mg, K, Cl, $NO_3$, and T-N. The second factor showed high loadings with Zn, Fe, Mn and Cd. The third to seventh factors could be characterized as $PO_4$, TSS, inorganic nitrogen, pH and T-P, and Cu factors, respectively. The factor score for EC was the highest in Kuri, followed by Chunchon, Dunnae and Daegwanryng. The factor score for heavy metals were the highest in the Daegwanryng. The results demonstrated that the factor analysis could be useful to select the most principal factor influencing soil and water qualities in the agricultural watershed.

Regulation of nsdD Expression in Aspergillus nidulans

  • The nsdD gene has been predicted to encode a GATA type transcription factor with the type IVb zinc finger DNA binding domain functions in activating sexual development of A. nidulans. In several allelic mutants of nsdD producing truncated NsdD polypeptides lacking the C-terminal zinc finger, the transcription level of nsdD gene was greatly increased. Also in an over-expressed mutant, the transcription under its own promoter was reduced. These results suggest that the expression of nsdD is negatively autoregulated. When the nsdD gene was over-expressed, cleistothecia were formed in excess amounts even in the presence of 0.6 M KC1 that inhibited sexual development of the wild type. Northern blot analysis revealed that the expression of nsdD was repressed by 0.6 M KC1. These results strongly suggest that the inhibition of sexual development by salts was carried out via the nsdD involved regulatory network.

  • Studies on innovation point towards the need for both in-house research and development (R&D) capabilities along with collaborations with external organisations or linkages with other firms, universities and research institutes. The argument is that innovation is not just a firm specific factor but requires support from several other factors which in turn involves developing linkages with various actors/institutions as sources of resources and/or knowledge for innovation. This paper presents findings from an exploratory study about the Malaysian biofuel sector as an innovation system. Instead of using the neo-classical approach and proposing policy interventions based on traditional market failure rationale (Bergek et al., 2008a), the functional analysis of the technological innovation systems (TIS) approach is used to analyse the biofuel sector in Malaysia to help reveal the strengths and weaknesses in terms of its fulfillment of seven critical functions which are necessary for the generation and commercialization of a product. In addition an attempt is made to reveal the inducement and blocking mechanisms affecting the Malaysian biofuel TIS and the seeking of improved poor functionality. These functional aspects of the Malaysian biofuel sector are analysed in different phases of its development to gain an understanding of the different institutions and their functions at different phases of the development of the innovation system.

Characterization of Korean Cattle Keratin IV Gene

  • Keratins, the constituents of epithelial intermediate filaments, are precisely regulated in a tissue and development specific manner. There are two types of keratin in bovine. The type I is acidic keratin and the type II is neutral/basic keratin. 1.5 kb of 5' flanking sequence of Korean cattle Keratin IV gene, type II keratin (59 kDa), was cloned and sequenced. A symmetrical motif AApuCCAAA are located in a defined region upstream of the TATA box. Proximal SP1, AP1, E-box and CACC elements as the major determinants of transcription are identified. When it was compared to the bovine sequence from -600 bp to ATG upstream, the homology was 97% in nucleotide sequence. Several A and T sequences, located in the promoter region, are deleted in the Korean cattle. An expression vector consisted of Korean cattle Keratin IV gene promoter/SV40 large T antigen was transfected to HaCaT cell (Epithelial keratinocyte). The transformed HaCaT cells showed active proliferation when treated with PDGF (Platelet-derived growth factor) in 0.3% soft agar compared to control cells. These results indicate that Korean cattle Keratin IVgene promoter can be used as a promoter for transfection into epithelial cell.

Sustainability and International Environmental Agreements

  • This paper examines the perceptions of sustainability, which is conceptualised as cross-generational social preferences, on the formation of international environmental agreements (IEAs) in a two-stage game in two periods. There are two scenarios are considered: myopic and sustainable development scenarios. The myopic scenario assumes the decision makers only concern the present welfare. Whilst the scenario of sustainable development has two characters: cross-generational fairness and altruism. When both are taken into account, a coalition will be expanded. The numerical example indicates that the marginal cost of the total emissions is the crucial factor for the formation of IEAs. Only when the marginal cost is low, a sustainable system can be succeeded. While, the technological advancement may lead to a more efficient production per unit of emissions, it also encourages countries to emit more in total and have a lower level of welfare. The results confirm the importance of sustainability to IEAs. The lesson learnt from this study is: when decision makers are myopic, the system is unsustainable even if an IEA is formed. Only when the perception of sustainability is considered, the system could be sustainable. Regardless of the existence of IEAs, international environmental conventions shall not neglect the fundamental goal to pursue sustainable development.

Effect of Olive Flounder, Paralichthys olivaceus, Immune Genes Stimulation by Molecular Adjuvant in Vitro Culture Condition (In vitro 조건에 따른 molecular adjuvant의 넙치, Paralichthys olivaceus 면역유전자 자극 효과)

  • Adjuvant is an immune enhancer commonly used during vaccination to enhance the host immune response. In the present study, we produced the several recombinant protein from immune related gene of olive flounder (Paralichthys olivaceus). Especially, to produce the soluble type of recombinant protein, we constructed the MBP (Maltose binding protein) fusion G-CSF (Granulocyte colony stimulating factor) recombinant protein among the flounder immune related genes. To verify the immune stimulatory effect and safety of this recombinant protein (rPoGCSF), expression changes of several immune genes were tested using quantitative real-time PCR method with gene specific primer from flounder head kidney leukocytes. As a result, we confirmed that the rPoGCSF has an ability of immune stimulatory effect, also it has broad range of pH and temperature.
