Effects of glutathione s-transferase (GST) M1 and T1 polymorphisms on antioxidant vitamins and oxidative stress-related parameters in Korean subclinical hypertensive subjects after kale juice (Brassica oleracea acephala) supplementation

  • BACKGROUND/OBJECTIVES: Glutathione s-transferase (GST) is involved in the formation of a multigene family comprising phase II detoxification enzymes, involved in the detoxification of reactive oxygen species. This study evaluated whether daily supplementation with kale juice could modulate levels of plasma antioxidant vitamins and oxidative stress-related parameters. We further examined whether this modulation was affected by combined GSTM1 and T1 polymorphisms. SUBJECTS/METHODS: Totally, 84 subclinical hypertensive patients having systolic blood pressure (BP) over 130 mmHg or diastolic BP over 85 mmHg, received 300 mL of kale juice daily for 6 weeks. Blood samples were drawn before start of study and after completion of 6 weeks. RESULTS: After supplementation, we observed significant decrease in DNA damage and increase in erythrocyte catalase activity in all genotypes. Plasma level of vitamin C was significantly increased in the wild/null and double null genotypes. The plasma levels of ${\beta}-carotene$, erythrocyte glutathione peroxidase activity, and nitric oxide were increased only in the wild/null genotype after kale juice supplementation. CONCLUSIONS: The effect of kale juice was significantly greater in the GSTM1 null genotype and wild/null genotype groups, suggesting possibility of personalized nutritional prescriptions based on personal genetics.

  • A series of organosulfur compounds were synthesized with the aim of developing chemopreventive compounds active against hepatotoxicity and chemical carcinogesis. 2-(Allylthio) prazine (2-AP) was effective in inhibiting cytochrome P450 2E1-mediated catalytic activities and protein expression, and in inducing microsomal epoxide hydrolase and major glutathione S-transferases. 2-AP reduced the hepatotoxicity caused by toxicant sand elevated cellular GSH content. Development of skin tumors, pulmonary adenoma and aberrant crypt foci in colon by various chemical carcinogens was inhibited by 2-AP pretreatment. Anticarcinogenic effects of 2-AP at the stage of initiation of tumors were also observed in the aflatoxin B1 ($AFB_1$)-induced three-step medium-term hepatocarcinogenesis model. Reduction of $AFB_1$-DNA adduct by 2-AP appeared to result from the decreased formation of $AFB_1$-8,9-epoxide via suppression of cytochrome P450, while induction of GST 2-AP increases the excretion of glutathione-conjugated $AFB_1$ . 2-AP was a radioprotective agent effective against the lethal dose of total body irradiation and reduced radiation-induced injury in association with the elevation of detoxifying gene expression. 2-AP produces reactive oxygen species in vivo, which is not mediated with the thiol-dependent production of oxidants and that NF-KB activation is not involved in the induction of the detoxifying enzymes. the mechanism of chemoprotection by 2-AP may involve inhibition of the P450-mediated metabolic activation of chemical carcinogens and enhancement of electrophilic detoxification through induction of phase II detoxification enzymes which would facilitate the clearance of activated metabolites through conjugation reaction.

Effects of Scalp Management using SUKI on Heavy Metal Discharge (SUKI(silver tools)를 활용한 두피 관리가 중금속 배출에 미치는 영향)

  • The purpose of this study was to access the effects of detoxification of hair by SUKI(silver tools). The subjects of this study participated 26 persons(EG 13 and CG 13), Experimental group was performed SUKI programs combined with scalp and hair for two times per week, during 8 weeks. The results was of comparison of variations in each groups showed that there was a significant difference(6items) in the experimental group(p<.05). But 2items(Pb, Cs) was not showed it. Among the hazardous heavy metals in research, Al, Ba and Hg showed the greatest reduction rate of difference. Thus, the using of the SUKI tool has been shown to affect some heavy metal emissions (p<.05). In conclusion, it is thought that it can be used as basic data for heavy metal discharge of scalp through continuous research in the future.

A Herbological Study on the Plants of Aspidiaceae in Korea (한국산 면마과(綿馬科) 식물에 관한 본초학적 연구)

  • Objectives : This study was aimed to developing Korean herbalogy of the plants belonging to Aspidiaceae in Korea. Methods : The herbological books and papers published at home and abroad were researched. The total list of Aspidiaceae was made and Medicinal plants in Aspidiaceae was classified. The medicinal properties, action and applications of medicinal plants were investigated. Results : 1. There are totaled to 17 genera and 130 species in Aspidiaceae in Korea and among them medicinal plants are 11 genera, 31 species, some 24% in total. 2. Athyrium genus is main kind enough that it has 32 species among 130 species in Aspidiaceae, and medicinal plants of Dryopteris is 4 species. 3. The rhizome is the main medicinal parts if medicinal plants in the Aspidiaceae, which is used in 24 species. 4. According to nature and flavor of medicinal plants in the Aspidiaceae, they were classified into cool 15 and cold 8 species ; bitter taste 19, and little bitter taste 15 in the order. 5. According to meridian propism of medicinal plants in the Aspidiaceae, they were classified into liver meridian 5 species, stomach meridian 3 species. 6. According to the properties and principal curative action, they were classified into drugs for clearing away heat 25 species, drugs for detoxification 22 species, drugs for promoting diuresis 16 species in the order. 7. The number of toxic species in the Aspidiaceae was examined to be 5 species. Conclusions : There are totaled to 17 genera and 130 species in Aspidiaceae in Korea and among them medicinal plants are 11 genera, 31 species, some 24% in total.   • Plant biomass, or lignocellulose, is one of the most abundant natural resources on earth. Lignocellulosic biomass, such as agricultural and forestry residue, serves as a renewable feedstock for microbial cell factories due to its low price and abundant availability. However, the recalcitrance of lignocellulosic biomass requires a pretreatment process prior to microbial fermentation, from which fermentable sugars including xylose and glucose are generated along with various inhibitory compounds. The presence of furan derivatives, such as 5-hydroxymethyl-2-furaldehyde and 2-furaldehyde (furfural), hampers the microbial conversion of lignocellulosic biomass into value-added commodities. In this study, furfural tolerance was improved by investigating the detoxification mechanism in non-model yeast. The genes encoding aldehyde dehydrogenases were overexpressed to enhance furfural tolerance and resulted in improving cell growth and lipid production that can be converted into biofuel. Taken together, this approach contributes to the understanding of the reducing toxicity mechanism of furfural by the aldehyde dehydrogenases and provides a promising strategy that the use of microorganism as an industrial workhorse to treat efficiently lignocellulosic biomass as sustainable plant derivatives.

Characterization of Mineralogical Changes of Chrysotile and its Thermal Decomposition by Heat Treatment (열처리에 따른 백석면의 광물학적 특성 변화와 열분해 과정 연구)

  • Chrysotile is a 1:1 sheet silicate mineral belonging to serpentine group. It has been highlighted studies because of uses, shapes and structural characteristics of the fibrous chrysotile. However, it was designated as Class 1 carcinogen, so high attentions were being placed on detoxification studies of chrysotile. The objectives of this study were to investigate changes of mineralogical characteristics of chrysotile and to suggest detoxification mechanism of chrysotile by thermal decomposition. Samples for this study were obtained from LAB Chrysotile mine in Canada. The samples were heated in air in the range of 600 to $1,300^{\circ}C$. Changes of mineralogical characteristics such as crystal structure, shape, and chemical composition of the chrysotile fibers were examined by TG-DTA, XRD, FT-IR, TEM-EDS and SEM-EDS analyses. As a result of thermal decomposition, the fibrous chrysotile having hollow tube structure was dehydroxylated at $600-650^{\circ}C$ and transformed to disordered chrysotile by removal of OH at the octahedral sheet (MgOH) (Dehydroxylation 1). Upon increasing temperature, it was transformed to forsterite ($Mg_2SiO_4$) at $820^{\circ}C$ by rearrangement of Mg, Si and O (Dehydroxylation 2). In addition, crystal structure of forsterite had begun to transform at $800^{\circ}C$, and gradually grown 3-dimensionally to enstatite ($MgSiO_3$) by recrystallization after the heating above $1,100^{\circ}C$. And then finally transformed to spherical minerals. This study showed chrysotile structure was collapsed about $600-700^{\circ}C$ by dehydroxylation. And then the fibrous chrysotile was transformed to forsterite and enstatite, as non-hazardous minerals. Therefore, this study indicates heat treatment can be used to detoxification of chrysotile.

돼지고기가 생체내(生體內) 중금속(重金屬)의 축적억제(蓄積抑制)에 미치는 영향

  • This study was performed to investigate the effect of pork on the cadmium detoxification in rats. Ninety male Sprague-Dawley rats weighing $125.3{\pm}1.4g$ were divided into five groups based on cadmium treatment(0, 25, 50, 100, 250ppm) and five levels of Cd in AIN-76 purified diet had been fed for 8 weeks. Cadmium was supplemented with a form of $CdCl_2$.. During following 8 weeks of intoxication, casein was replaced by pork and the effect of pork on cadmium- detoxification was compared with casein. After 8 weeks of Cd intake had resulted in apparent cadmium intoxication; reduced growth rate, enlarged kidney and testis, decreased hematocrit value and hemoglobin content in response to the supplemented Cd levels in the diets. Discontinuing cadmium feeding, the body weights were relieved. Pork-fed groups seemed to have higher body weight than casein-fed groups. Hemoglobin content and hematocrit value became normal range at detoxification stage. The weights of liver, kidney, and testis were decreased along with cadmium intake. However, organ weight ratio was not affected by cadmium. Cadmium accumulation in liver and kidney showed a tendency to increase in the cadmium-exposed groups. The levels of metallothionein were also significantly elevated in the tissues of liver in response to the levels of Cd supplemented(p<0.05). Cadmium concentration in kidney was two times higher than that in liver. Cadmium removal rate of liver was higher than that of kidney. Cadmium accumulation of the pork-fed group was lower than that of casein. Especially, the factors which affected the cadmium contents in kidney were $Cd^{***}$ and $Cd{\times}pork^{***}$. Metallothionein(MT) was increased with cadmium, and MT was not likely to be affected by pork. Based on the findings from gross lesion, rats fed 250ppm of Cd were externally emaciated, had exposed penis and observed atrophies of kidney and testis. Histopathological observation seemed that the liver of groups feeding Cd supplemented diets showed cellular degeneration and accumulation of eosinophilic materials in the capillaries. In kidney, rats fed Cd diets had shown tubular epithelium degeneration and lesions of basophilic materials, while testis were weakened in numbers of spermatid and sporadically enlarged of giant cells. But the rats administered cadmium-detoxified diet supplemented pork for 7 weeks were shown individually decreased lesions compared with the rats supplied with casein diet.

Expression and Characterization of Escherichia coli Prolidase with Organophosphorus Compounds

  • A relatively high homology between Escherichia coli prolidase and Alteromonas organophosphorous acid anhydrolase suggests that E. coli prolidase may have an activity to d egrade toxic organophosphorous compounds. To confirm this suggestion, we cloned and expressed a prolidase gene (pepQ) of E. coliBL2l. The recombinant E. coli prolidase that consisted of 443 amino acid residues exhibited activity and stereochemical selectivity against organopho sphorous compounds, although its activity was two to three orders of magnitude less than that of the other organophosphorous acid hydrolase isolated from Pseudomonas diminuta.

Activities of Oxidative Enzymes Related with Oxygen Tolerance in Bifidobacterium sp.

  • To study the relationship between oxygen tolerance and enzyme activity in the oxygen metabolism of bifidobacteria, the activities of catalase, superoxide dismutase (SOD), NADH oxidase and NADH peroxidase from six typical bifidobacteria and other bacteria were assayed by spectrophotometry. Catalase activity was hardly detected in any of the bifidobacteria tested. SOD activity was detected in every species including the Clostridium species. In particular SOD activity was notably high in the aerosensitive Bifidobacterium adolescentis. This fact indicates that SOD activity is not a critical factor to ensure aerotolerance. Aerosensitive B. adolescentis showed very low NADH oxidative enzyme activity whereas other aerotolerant bifidobacteria exhibited considerable activity for the enzymes. It seems that detoxification of $H_2O_2$ by NADH oxidative enzymes might be an important factor in improving for aerotolerant bifidobacteria survival rates in an oxygen environment.

S-(N,N-Diallyldithiocarbamoyl)-N-acetylcysteine: Synthesis and Biochemical Properties Associated with Chemoprevention (S-(N,N-Diallyldithiocarbamoyl)-N-acetylcysteine의 합성 및 발암억제와 관련된 생화학적 특성)

  • Dithiocarbamate and mixed disulfide containing allyl functions were designed and synthesized as putative chemopreventive agents, i.e. N,N-diallyldithiocarbamate (DATC) and S-(N,N-diallyldithiocarbamoyl)-N-acetylcysteine (AC-DATC). DATC and AC-DATC were administered and the activities of cytosolic glutathione S-transferase (GST), glutathione reductase (GR) and microsomal N-nitrosodiethylamine (NDEA) deethylase were assayed in order to test the effects of these organosulfur com-pounds on the detoxification and metabolic activation system of NDEA. The amounts of hepatic glutathione (GSH and GSSG) was also determined. The administration of DATC to rats led to an increase in the activity of GR and to an inhibition of CYP2E1-mediated NDEA deethylation. AC-DATC induced the activity of GR and GST, increased the hepatic GSH content and inhibited the rate of NDEA deethylation. The level of GSSG was decreased as a consequence of the increased activity of GR. These effects may contribute to possible antimutagenic and anticarcinogenic action of the dithiocarbamates investigated.
