Background: Metal-responsive transcription factor-1 (MTF-1) is a key transcriptional regulator playing crucial roles in metal homeostasis and cellular adaptation to diverse oxidative stresses. In order to understand cellular pathways associated with metal regulation and stress responses in Pacific abalone (Haliotis discus hannai), this study was aimed to isolate the genetic determinant of abalone MTF-1 and to examine its expression characteristics under basal and experimentally stimulated conditions. Results: The abalone MTF-1 shared conserved features in zinc-finger DNA binding domain with its orthologs; however, it represented a non-conservative shape in presumed transactivation domain region with the lack of typical motifs for nuclear export signal (NES) and Cys-cluster. Abalone MTF-1 promoter exhibited various transcription factor binding motifs that would be potentially related with metal regulation, stress responses, and development. The highest messenger RNA (mRNA) expression level of MTF-1 was observed in the testes, and MTF-1 transcripts were detected during the entire period of embryonic and early ontogenic developments. Abalone MTF-1 was found to be Cd inducible and highly modulated by heat shock treatment. Conclusion: Abalone MTF-1 possesses a non-consensus structure of activation domains and represents distinct features for its activation mechanism in response to metal overload and heat stress. The activation mechanism of abalone MTF-1 might include both indirect zinc sensing and direct de novo synthesis of transcripts. Taken together, results from this study could be a useful basis for future researches on stress physiology of this abalone species, particularly with regard to heavy metal detoxification and thermal adaptation.
Stable isotope tracers were first applied to evaluate the Microcystis cell assimilation efficiency of bivalves, since the past identification method has been limited to tracking the changes of each chl-a, clearity, and nutrient. The filter-feeders (Sinanodonta woodiana and Unio douglasiae) were assessed under the condition of cyanobacteria (Microcystis aeruginosa) blooms through an in mesocosm experiment using $^{13}C$ and $^{15}N$ dual isotope tracers. chl-a concentration in the treatment mesocosm was dramatically decreased after the beginning of the second day, ranging from 116 to $66{\mu}g\;L^{-1}$. In addition, the incorporated $^{13}C$ and $^{15}N$ atom % in the S. woodiana bivalve showed higher values than U. douglasiae bivalves. The results demonstrate that U. douglasiae has less capacity to assimilate toxic cyanobacteria derived from diet. Our results therefore also indicate that S. woodiana can eliminate the toxin more rapidly than U. douglasiae, having a larger detoxification capacity.
The present study was performed to evaluate the effect of citric acid on the change of implant surface microstructure according to application time. Implants with pure titanium machined surface, titanium plasma-sprayed surface, and sand-blasted, large grit, acid etched surface were utilized. Implant surface was rubbed with pH 1 citric acid for $\frac{1}{2}$ min., 1 min., 1 $\frac{1}{2}$ min., 2 min., and 3min, respeaively in the test group and implant surface was not treated in the control group. Then, the specimens were processed for scanning electron microscopic observation. The following results were obtained. 1. Both test and control group showed a few shallow grooves and ridges in pure titanium machined surface implants. There were not significant differences between two groups. 2. In titanium plasma-sprayed surfaces, round or amorphous particles were deposited irregularly. The irregularity of titanium plasma-sprayed surfaces conditioned with pH 1 citric acid was lessened and the cracks were increased relative to the application time of pH 1 citric acid. 3. Sand-blasted, large grit, acid etched surfaces showed the macro/micro double roughness. The application of pH 1 citric acid didn't change the characteristics of the sand-blasted, large grit, acid etched surfaces. In conclusion, the application of pH 1 citric acid to titanium plasma-sprayed surface is improper. And pure titanium machined surface implants and sand-blasted, large grit, acid etched surface implants can he treated with pH 1 citric acid for peri-implantitis treatment if the detoxification of these surfaces could be evaluated.
Liver and kidney are specific organs which play an active role in biotransformation and detoxification mechanisms. Ant adverse effect of chemicals or heavy metal can cause the delay or fade in these mechanisms. Present study was designed to find out the protective effect of Panax ginseng extract on renal functions altered by mercuric chloride (heavy metal) in albino rat. Fifty albino rats were divided into 10 groups. Five groups for acute study and five groups for sud-acute study viz. control group (Tween 20 and distilled water), mercuric chloride treated group (0.926 mg/kg body wt. for acute and 0.044 mg/kg body wt. for sub-acute group after calculated $LD_{50}$ (9.26 mg/kg body wt.) by probit analysis (Finney, 1971), Panax ginseng extract treated group (10 mg/kg body wt. for acute and sub-acute sets), mercuric chloride treated followed by Panax ginseng extract and Panax ginseng extract followed by mercuric chloride group. All doses were given orally by gavage tube. The result revealed that the serum urea and creatinine significantly increased in mercuric chloride treated group, while significantly decreased (p<0.01) in Panax ginseng extract group after acute and sub-acute treatment. The biochemical estimation is also confirmed by nephropathological aspect. However, the Panax ginseng extract treated followed by mercuric chloride group is more prominent than the mercuric chloride treated followed by Panax ginseng extract group. It can be concluded that Panax ginseng extract had a protective nature on renal functions against mercuric chloride toxicity in albino rats.
The adverse health effects of a number of environment pollutions are related to the formation of free radicals. Induction of antioxidant defensive system in the response to an oxidative attack is an essential element of the cell to survive. CYP2E1 is easily induced by organic solvents and induces continuous formation of reactive oxygen species (ROS). ${\gamma}$-Glutamyltranspeptidase (${\gamma}$GT) plays an important role in glutathione metabolism and xenobiotic detoxification. To evaluate the characteristic of oxidative stress which induces GGT expression and to understand human antioxidant defensive response against oxidative stress induced by CYP2E1, we studied regulation of ${\gamma}$GT enzyme expression in response to various oxidative stresses in human HepG2 cells. The ${\gamma}$GT activity was not modified after exposure of acute oxidative stress inducing agents (ferric nitrilotriacetate, cumene hydroperoxide, ADP-Fe, O-tetradecanoylphorbol-13-acetate, tumor necrosis factor-alpha). To induce continuous exposure of cells to ROS, HepG2 cells were transfected by human CYP2E1 gene transiently. The CYP2E1 activity was verified with chlorzoxazone hydroxylation. Transfection of CYP2E1 showed continuous 60% increase in intracellular ROS and 240 % increase in microsomal ROS. CYP2E1 overexpressing cells showed increased ${\gamma}$GT activity (2.5-fold). The observed enhancement of ${\gamma}$GT activity correlated with a significant increase of ${\gamma}$GT mRNA (2.1-fold). Treatment with antioxidant strongly prevented the increase in ${\gamma}$GT activity. The CYP2E1 overexpression did not modify toxicity index and increased glutathione levels. These results show that continuous exposure of cells to ROS produced by CYP2E1 up-regulates ${\gamma}$GT; This may be one of the adaptive antioxidant responses of cells to oxidative insult. Present study also suggests that the induction of ${\gamma}$GT could be used as a marker of oxidative stress induced by exposure to organic solvents.
A study on drug abuse of young people who failed in college entrance examinations and preparing the next examination and those enrolled at colleges but trying to transfer to other colleges was conducted from February 3 to May 31, 1999 in order to determine current drug abuse status of them and factors affecting their behavior related to drug abuse. A total of 1,771 people were selected from college entrance preparation institutes in Seoul, Pusan, Taegu, Taejon and Kwangju and interviewed by using questionnaires. The results are summarized as follows; 1. 49.78% of them experienced drinking, 22.8% smoking, and 6.7% drug previously. More males smoked cigarettes than females. The young in the age group of 25 to 29 years old, and the young living in small towns, and subjects who finished junior colleges and those without religions drank more than those in other categories. The subjects in the age group of 20 to 24 years old and those with one parent, and those living in small towns used more drugs than those in other categories. 2. 60.1% of those with drug using experience used the drug in their or friends' homes. 53.6% of them used drugs with their friends, junior or senior classmates. 36.6% of them purchased drugs from drug stores. 76.0% of them spent their pocket money for buying drugs. 3. The drugs (excluding alcohol and tobacco) they used were bond and butane gas (29.6%), stimulants (20.6%), marijuana 02.0%) LSD (4.8%), cocaine, heroine and opium (4.2%), and others (28.8%). 4. 21.5% of those with drug using experience had detoxification treatment previously. The ajmor sufferings they experienced were distraction (42.6%), fear 01.7%), and others. 5. 51.0% of the subjects experienced drug related health education in schools, and 14.6% had that kind of education through social program. 6. 64.3% of the subjects got information about drug abuse through mass media, 16.4% through printed materials, and 11.4% through public health professionals. 7. The knowledge level of drug abuse was 16.48:1:2.76 points out of 24 (68.6 points out of 100).
To improve the erythritol productivity of Penicillium sp. KJ81, mutants were obtained using UV irradiation and NTG treatment Among these mutants, Penicillium sp. KJ-UV29 revealed no morphological changes, yet was superior to the wild strain in the following three points: (1) Penicillium sp. KJ-UV29 produced more erythritol than the wild strain under the same conditions, (2) no foam was produced during cultivation, unlike the wild strain, and (3) the mutant produced a Significantly lower amount of glycerol. Penirillium sp. KJ-UV29 produced as much as 15.1 g/L of erythritol, whereas the wild-type Penirillium sp. KJ81 only produced 11.7 g/L. Penicillium sp. KJ-UV29 only generated 6.1 g/L of glycerol, compared to 19.4 g/L produced by the wild strain. When investigating the optimal culture conditions for erythritol production by the mutant strain Penicillium sp. KJ-UV89, sucrose was identified as the most effective carbon source, and the mutant was even able to produce erythritol in a 70% sucrose-containing medium, although a 30% sucrose medium exhibited the highest productivity. The production of erythritol by Penirillium sp. KJ-UV29 was also significantly increased by the addition of ammonium carbonate, potassium nitrate, and sodium nitrate. Accordingly, under optimal conditions, Penicillium sp. KJ-UV29 produced 45.2 g/L of erythritol in a medium containing 30% sucrose, 0.5% yeast extract, 0.5% (NH$_4$)$_2$C$_2$O$_4$, 0.1% KNO$_3$, 0.1% NaNO$_3$, and 0.01% FeSO$_4$ with 1 vvm aeration and 200 rpm agitation at 37$^{\circ}C$ for 7 days in a 5-L jar fermentor.
Background: Continuous exposure to high temperatures can lead to heat stress. This stress response alters the expression of multiple genes and can contribute to the onset of various diseases. In particular, heat stress induces oxidative stress by increasing the production of reactive oxygen species. The liver is an essential organ that plays a variety of roles, such as detoxification and protein synthesis. Therefore, it is important to protect the liver from oxidative stress caused by heat stress. Korean ginseng has a variety of beneficial biological properties, and our previous studies showed that it provides an effective defense against heat stress. Methods: We investigated the ability of Korean Red Ginseng and Korean black ginseng extracts (JP5 and BG1) to protect against heat stress using a rat model. We then confirmed the active ingredients and mechanism of action using a cell-based model. Results: Heat stress significantly increased gene and protein expression of oxidative stress-related factors such as catalase and SOD2, but treatment with JP5 (Korean Red Ginseng extract) and BG1 (Korean black ginseng extract) abolished this response in both liver tissue and HepG2 cells. In addition, JP5 and BG1 inhibited the expression of inflammatory proteins such as p-NF-κB and tumor necrosis factor alpha-α. In particular, JP5 and BG1 decreased the expression of components of the NLRP3 inflammasome, a key inflammatory signaling factor. Thus, JP5 and BG1 inhibited both oxidative stress and inflammation. Conclusions: JP5 and BG1 protect against oxidative stress and inflammation induced by heat stress and help maintain liver function by preventing liver damage.

Sulfur is an essential element in plants, including amino acids, vitamin synthesis, and acting as an antioxidant. However, the interaction between endogenous sulfur and proline synthesis has not been yet fully documented. White clover (Trifolium repens L.) is known as a species highly sensitive to sulfate supply. Therefore, this study aimed to elucidate the role of sulfur in regulating proline metabolism in relation to ammonia detoxification and hydrogen peroxide (H2O2) accumulation in white clover. The detached leaves of white clover were immersed in solution containing different concentration of sulfate (0, 10, 100, and 1000 mM MgSO4). As MgSO4 concentrations were increased, the concentration of H2O2 increased up to 2.5-fold compared to control, accompanied with H2O2 detection in leaves. Amino acid concentrations significantly increased only at higher levels (100 and 1000 mM MgSO4). No significant difference was observed in protein concentration. Proline and ∆1-pyrroline-5-carboxylate (P5C) concentrations slightly decreased at 10 and 100 mM MgSO4 treatments, whereas it rapidly increased over 1.9-fold at 1000 mM MgSO4 treatment. Ammonia concentrations gradually increased up to 8.6-fold. These results indicate that exogenous sulfur levels are closely related to H2O2 and ammonia synthesis but affect proline biosynthesis only at a higher level.
Turfgrass, the most widely grown ornamental crop, is severely affected by fungal pathogens including Sclerotinia homoeocarpa, Rhizoctonia solani, and Magnaporthe poae. At present, turfgrass fungal disease management predominantly relies on synthetic fungicide treatments. However, the extensive application of fungicides to the soil increases residual detection frequency, raising concerns for the environment and human health. The bacterial volatile compound, 2,3-butanediol (BDO), was found to induce plant resistance. In this study, we evaluated the disease control efficacy of a combination of stereoisomers of 2,3-BDO and commercial fungicides against turfgrass fungal diseases in both growth room and fields. In the growth room experiment, the combination of 0.9% 2R,3R-BDO (levo) soluble liquid (SL) formulation and 9% 2R,3S-BDO (meso) SL with half concentration of fungicides significantly increased the disease control efficacy against dollar spot and summer patch disease when compared to the half concentration of fungicide alone. In field experiments, the disease control efficiency of levo 0.9% and meso 9% SL, in combination with a fungicide, was confirmed against dollar spot and large patch disease. Additionally, the induction of defense-related genes involved in the salicylic acid and jasmonic acid/ethylene signaling pathways and reactive oxygen species detoxification-related genes under Clarireedia sp. infection was confirmed with levo 0.9% and meso 9% SL treatment in creeping bentgrass. Our findings suggest that 2,3-BDO isomer formulations can be combined with chemical fungicides as a new integrated tool to control Clarireedia sp. infection in turfgrass, thereby reducing the use of chemical fungicides.
