  • Tetrofosmin is a ligand that forms a lipophilic, cationic complex with Tc-99m. Tc-99m tetrofosmin was developed as a myocardial perfusion imaging agent and also used to depict tumors. Mediastinal tumors is also detected by Tc-99m tetrofosmin. We report a case of extracardiac mediastinal activity detected by Tc-99m tetrofosmin scintigraphy, which revealed thymoma.

  • Simultaneous determination of three corticosteroids (clobetasol propionate, betamethasone dipropionate, fluticasone propionate) in moisturizers was performed by using liquid chromatography (LC) coupled with tandem mass spectrometry (MS/MS). Sample preparation was conducted by the liquid-liquid extraction (LLE). Moisturizers include emulsifying agent and it forms micelles. In order to improve the extraction efficiency of corticosteroids trapped in micelle, newly developed-optimized extraction conditions which can remove the matrix effect from moisturizers was applied with various pH conditions in LLE extraction stage of sample preparation. Thus, the addition of 10 μL of 1 M HCl into moisturizers sample before extraction could improve the extraction efficiency. For the quantitative analysis, SRM table that contained specific transition of all of target corticosteroids was created. The developed method was validated for linearity, accuracy, precision, limit of detection (LOD), limit of quantization (LOQ) and recovery. Over the 0.99 r2 value was obtained in calibration standard range. Effective accuracy and precision were also obtained. LODs were below 31 ng/mL and LOQs were estimated below 94 ng/mL for all corticosteroids tested.

Rapid and Easy Detection of Porcine Epidemic Diarrhea Virus (PEDV) by in situ Hybridization (In Situ Hybridization에 의한 돼지 유행성설사증 (Porcine Epidemic Diarrhea)의 진단)

  • Molecular diagnostic techniques have been used to identify porcine epidemic diarrhea virus (PEDV), a causative agent of acute enteritis in swine, but they were difficult to be petformed and time-consuming. To detect PEDV in a rapid and easy way, we developed biotinylated cDNA probe for N gene encoding the nucleoproteins of PEDV. Formalin-fixed and paraffin-embedded tissues from 24 naturally infected pigs were used for the experiment. The ISH produced a positive reaction in all cases. When intestinal tissues were hybridized with PEDV probe, strong signals were seen in the villus enterocytes of the jejunum and ileum. Hybridization signals were also found in the duodenum from one pig and in colon from dnother. In conclusion, ISH with a biotinylated cDNA probe was provided to be a useful diagnostic method for detecting PEDV effectively in routinely processed tissue sections.

  • This paper analyzed the electrical field strength of porcelain suspension insulator used various testing methods in order to present the causes and countermeasure for the puncture failure by the inner minute defect. A power frequency puncture test and a step-front-of-wave flashover voltage test were conducted. The failed specimen of these samples was analyzed material characteric observed the cross-sectional picture by the scanning electron microscope, and tested the autoclave expansion for the portland cement using the metal bond agent. The porcelain made a progress the failure by the thermal expansion coefficient of the different material. The inner detecting technique on behalf of the puncture failure protection is presented such as the steep-front-of-wave flashover voltage test.

A Green Fluorescent Protein-based Whole-Cell Bioreporter for the Detection of Phenylacetic Acid

  • Phenylacetic acid (PAA) is produced by many bacteria as an antifungal agent and also appears to be an environmentally toxic chemical. The object of this study was to detect PAA using Pseudomonas putida harboring a reporter plasmid that has a PAA-inducible promoter fused to a green fluorescent protein (GFP) gene. Pseudomonas putida KT2440 was used to construct a green fluorescent protein-based reporter fusion using the paaA promoter region to detect the presence of PAA. The reporter strain exhibited a high level of gfp expression in minimal medium containing PAA; however, the level of GFP expression diminished when glucose was added to the medium, whereas other carbon sources, such as succinate and pyruvate, showed no catabolic repression. Interestingly, overexpression of a paaF gene encoding PAA-CoA ligase minimized catabolic repression. The reporter strain could also successfully detect PAA produced by other PAA-producing bacteria. This GFP-based bioreporter provides a useful tool for detecting bacteria producing PAA.

  • Background: Curcumin has has been reported to exert anti-inflammatory, anti-oxidation and anti-angiogenic activity in various types of cancer. It has also been shown to induce apoptosis in leukemia cells. We aimed to unravel the role of the redox pathway in Curcumin mediated apoptosis with a panel of human leukemic cells. Materials and Methods: In this study in vitro cytotoxicity of Curcumin was measured by MTT assay and apoptotic effects were assessed by annexin V/PI, DAPI staining, cell cycle analysis, measurement of caspase activity and PARP cleavage. Effects of Curcumin on intracellular redox balance were assessed using fluorescent probes like $H_2DCFDA$, JC1 and an ApoGSH Glutathione Detection Kit respectively. Results: Curcumin showed differential anti-proliferative and apoptotic effects on different human leukemic cell lines in contrast to minimal effects on normal cells. Curcumin induced apoptosis was associated with the generation of intracellular ROS, loss of mitochondrial membrane potential, intracellular GSH depletion, caspase activation. Conclusions: As Curcumin induces programmed cell death specifically in leukemic cells it holds a great promise as a future therapeutic agent in the treatment of leukemia.

  • A simple prepared paste electrode (PE) of DNA immobilized on a carbon nanotube was utilized for monitoring the antibacterial agent oxytetracycline (OTC), using square-wave anodic stripping voltammetry (SWASV) and cyclic voltammetry (CV). Given these conditions, SWASV and CV working ranges were observed within 1-10 ngL-1 OTC. In the SWASV and CV for OTC concentrations of 0.1 mgL-1, the relative standard deviations (n=15) were 0.068 and 0.067, respectively. At the optimized condition, the detection limit was found to be 0.4 ngL-1 OTC. This method was applied to the hatchery fish tissue.

  • Human astrovirus (HAstV) is a major cause of acute diarrhea among children, resulting in outbreaks of diarrhea and occasionally hospitalization. Improved surveillance and application of sensitive molecular diagnostics have further defined the impact of HAstV infections in children. These studies have shown that HAstV infections are clinically milder (diarrhea, vomiting, fever) than infections with other enteric agents. Among the 8 serotypes of HAstV identified, serotype 1 is the predominant strain worldwide. In addition to serotype 1, the detection rate of HAstV types 2 to 8 has increased by using newly developed assays. HAstV is less common compared with other major gastroenteritis viruses, including norovirus and rotavirus; however, it is a potentially important viral etiological agent with a significant role in acute gastroenteritis. A better understanding of the molecular epidemiology and characteristics of HAstV strains may be valuable to develop specific prevention strategies.

Nickel(II) Determination by Spectrophotometry Coupled with Preconcentration Technique in Water and Alloy Samples

  • A micro organism Agrobacterium tumifacient as an immobilized cell on a solid support was presented as a new biosorbent in a simple and sensitive spectrophotometry determination of Ni(II) in various samples using 4-hydroxy benzaldehyde-4-bromophenyl hydrazone as a color developing agent (λmax 497 nm) at pH 4.0 ± 0.2. Beer's law was obeyed over the range of 0.01-0.1 μg L-1. The molar absorptivity and Sandell's sensitivity were 1.285 × 105 L mol-1cm-1 and 0.007245 μg cm-2 respectively. Under these conditions, the preconcentration factor obtained was 82, and the detection limit achieved was 0.05 μg L-1. The detailed study of various interfering ions made the method more sensitive and selective. The recovery of Ni(II) from various samples range from 97.75 to 99.35%. The present method was successfully applied for the determination of Ni(II) in spiked, natural water and alloy samples. The proposed method was compared with reported methods in terms of Student's ‘t'-test and Variance ratio ‘f'-test which indicates that there is no significant difference between proposed and literature method at 95% confidence level.

Involvement of Heat-stable and Proteinaceous Materials in the Culture of Pseudomonas putida JB-1 for the Inhibition of Tobacco mosaic virus Infection

  • Out of various fungi and bacteria tested for inhibition of Tobacco mosaic virus(TMV) infection using Nicotiana tabacum cv. Xanthi-nc, a bacterial isolate JB-l, identified as Pseudomonas putida had a strong direct inhibitory activity against the TMV infection. Its systemic or indirect activity was also noted at more than a half level of the direct control efficacy. Disease severity was reduced significantly in the susceptible tobacco N. tabacum cv. NC 82 by the treatment of the bacterial culture filtrate, somewhat more by the pretreatment than by simultaneous treatment, probably by inhibiting the TMV transmission and translocation in the plants, showing negative serological, which responses in the viral detection by DAS-ELISA. TMV-inhibitory substances from P. putida JB-1 were water-soluble, stable to high temperature(even boiling), and to a wide range of pH. As proteinase K nullified their antiviral activity, the TMV inhibition activity of P. putida may be derived from proteinaceous materials. In electron microscopy, TMV particles treated with the JB-1 culture were shown to be shrunken with granule-like particles attached on them. All of these aspects suggest that P. putida JB-1 may be developed as a potential agent for the control of TMV.
