Due to dermal exposure to hazardous chemicals causing potential adverse health symptoms through skin absorption, dermal monitoring has had an important role in assessing such exposure. This paper overviews comparatively a number of studies of dermal monitoring with different methodologies such as surface monitoring, skin wiping, skin washing, adhesive methods and tape stripping, fluorescence and infrared spectroscopy, skin patches, pads and clothing, video exposure monitoring and dermal exposure assessment toolkits and models. However, there is a lack of information on the relationship between exposure levels and adverse health symptoms. Therefore, more specific strategies for dermal exposure monitoring should be developed and standardized with further development of biological and ocular monitoring.

Two experiments were conducted to evaluate chloroform exposure and corresponding body burden by exposure routes while bathing. All pesticides must be assessed strictly whether safe or not when agricultural operators are exposed to the pesticides in farmland. A pesticide is commonly regarded as safe when estimated dermal absorption amount is lower than the acceptable operator's exposure level (AOEL). In this study, dermal absorption rate of chlorpyrifos, a widely used organophosphate insecticide, was investigated using rat dermal tissue model. Chlorpyrifos wettable powder solved in water (250, 500 and 2,500 ppm) was applied to freshly excised rat dermal slices ($341{\sim}413{\mu}m$ thickness) on static Franz diffusion cells at $32^{\circ}C$ for 6 hours. After exposure period of 6 hours, and then washing-at residual amount of chlorpyrifos was analyzed in dermal tissues, tape strips, washing solution, washing swabs of receptor bottles and receptor fluids at 1, 2, 4, 8 and 24 hours. Chlorpyrifos was only detected in dermal tissue but not found in receptor fluid at each concentration and time point, and the absorption rate of 250, 500 and 2,500 ppm was 2.36%, 1.96% and 1.69%, respectively. The estimated exposure level of chlorpyrifos was calculated as 0.012 mg/kg bw/day. The health risk for farmers in this condition is a level of concern because the estimated exposure level is 12 times higher than AOEL 0.001 mg/kg bw/day. However, actual health risk will be alleviated than estimated because absorbed chlorpyrifos is not permeated into internal body system and only retained in skin layer.
Objectives: This study was conducted to investigate the boron concentrations in 30 slime products and estimate children's dermal exposure to boron. Methods: Thirty slime products from online and offline stores were purchased for boron analysis. The boron content of each slime sample was analyzed using ICP-OES. A nationwide survey was performed to determine use patterns of slimes by a home-visit survey of 10,000 children divided into three age groups: 0-2, 3-6, and 7-12 years. The dermal exposure to boron was calculated using an exposure algorithm for child slime users. Results: The proportion of the child population using slime was 45.8%, and the mean use frequency was $2.5{\pm}3.8$ per week (range: 0.08-35). Mean time spent playing with slime was $19.6{\pm}11.3min$ (range: 1-100). Twenty-five of the thirty slimes contained a boron concentration exceeding the EU limit for toys made with sticky material of 300 mg/kg. Dermal exposure ranged from $0.0008-13.78{\mu}g/kg/day$ with the maximum weight fractions of boron in the 30 products. The dermal exposure estimate was the highest in 7-12 years old group. Conclusion: Slime use among Korean children may cause high exposure to boron. Regulation is needed to minimize boron exposure from slime products.
Skin is continuously exposed to external stimuli including ultraviolet radiation, which is a major cause of skin photoaging. According to recent discoveries, UVA with a lower energy but deep-penetrating properties, compared to UVB, is likely to play a major part in causing skin photoaging. The clinical and histochemical changes of photoaging are well characterized, but the biochemical mechanisms are poorly understood partly due to the lack of suitable experimental systems. In this work, three-dimensional, reconstituted skin culture models were prepared. After certain period of maturation, the equivalent models were shown to be similar in structure and biochemical characteristics to normal skin. Mature dermal and skin equivalent models were exposed to sub-lethal doses of UVA, and the effects of UVA relevant to dermal photoaging were monitored, including the production of elastin, collagen, collagenase(MMP-1), and tissue inhibitor of metalloproteinases-1 (TIMP-1). Interestingly, dermal and skin equivalents reacted differently to acute and chronic exposure to UVA. Elastin production was increased as soon as one week after commencing UVA irradiation by chronic exposure, although a single exposure failed to do so. This early response could be an important advantage of equivalent models in studying elastosis in photoaged skin. Collagenase activity was increased by acute UVA irradiation, but returned to control levels after repeated exposure. On the other hand, collagen biosynthesis, which was increased by a single exposure, decreased slightly during 5 weeks of prolonged UVA exposure. Collagenase has been thought to be responsible for collagen degeneration in dermal photoaging. However, according to the results obtained in this study, elevated collagenase activity is not likely to be responsible for the degeneration of collagen in dermal photoagig, while reduced production of collagen may be the main reason. It can be concluded that reconstituted skin culture models can serve as useful experimental tools for the study of skin photoaging. These culture models are relatively simple to construct, easy to handle, and are reproducible Moreover the changes of dermal photoaging can be observed within 1-4 weeks of exposure to ultraviolet light compared to 4 months to 2 years for human or animal studies. These models will be useful for biochemical and mechanistic studies in a large number of fields including dermatology, toxicology, and pharmacology.

Pesticides are widely used to prevent loss of agricultural production but extensive exposure can induce health problems to pesticide operators. This study was performed to evaluate the health risk of highly produced pesticides used in fruit growing farm land by comparison of estimated exposure level with AOEL using KO-POEM program. AOEL was driven based on NOAEL of each pesticide evaluated by JMPR, EFSA or KRDA. In calculation of exposure level, types of formulation, dilution factors, spraying duration and motor type and exposure protection device were allocated according to actual condition of use. Dermal absorption rate was differently applied among EFSA default values (25% or 75%), general default value (10%) or real test result values to know the plausibility of default values and safety of pesticide to operators in outline. Twenty pesticide ingredients (fungicides and insecticides) were produced more than 30 tons per year, which were mancozeb, chlorothalonil, imidaclopirid and etc in order. Dermal absorption rates obtained from studies were various from 0.07 to 81% but mostly under 10%. The estimated exposure levels showed big differences more than 10 times higher when using EFSA default rate and up to 5 times higher when using general rate of 10% comparing using rates of test results. Mancozeb, chlorthalonil, diazinon and chlorpyrifos presented still higher exposure level than AOEL even when using test absorption rate from study, which suggests that re-evaluation of AOEL or dermal exposure absorption rate or strict management are required for health protection of operators who use those four pesticides in farm land.
직업성 천식은 시안화물의 폭로에 의해 발생되는 것으로 알려져 있다. 남호주에 있는 가구산업에서는 일반적으로 목재판 위를 피복하기 위해 이중분사도포를 할 때 이소시안화물 도포제가 사용된다. 이 연구는 이와 관련 된 분야의 자료부족으로 관련 산업에서의 환경 및 피부와 안구폭로에 대한 모니터링을 위하여 수행되었다. 이 연구결과에 의하면 저압다량(HVLP)의 분사기 사용, 페인트 용액중의 저농도 경화제 및 전면 호흡보호구와 같은 적절한 보호구의 사용시에는 대기로부터 오염물질의 유입은 거의 없었고, 분사실 주변의 일반적인 작업범위에도 HDI 가 거의 감지되지 않았다. 페인트분사시의 일회용 장갑착용으로 피부폭로는 발견되지 않았으나, 칼라 모니터링에 의하면 장갑을 착용하지 않거나 청결하게 관리할 수 있는 공정이나 안전한 장소에 보관하지 않으면 작업장으로부터 피부폭로가 있었다. 페인트분사로부터 눈의 오염은 감지되지 않았고, 호흡기, 피부 및 안구에도 폭로되지 않았으며 유입과 가장 관련성이 있는 것은 작업형태이다.
Cytokines are hormone-like proteins which mediate and regulate inflammatory and immune responses. Interleukin-6 (IL-6) is involved in the final differentiation of B cells into antibody-producing cells. Interleukin-8 (IL-8) is a neutrophil chemotactic factor that plays an important role in the recruitment of neutrophil to inflammatory loci. Inflammatory mediators by cells in the gingiva have been implicated in the initiation and progression of periodontitis and oral infection. The purpose of this study was conducted to investigate the effect of lipopolysaccharide (LPS), staphylococcus enterotoxin B (SEB) on production of IL-6 and IL-8 by human gingival and facial dermal fibroblasts. Primary cultured human gingival and facial dermal fibroblasts were incubated with LPS (0.01, 0.1, $1.0{\mu}g/ml$), SEB (0.01, 0.1, $1.0{\mu}g/ml$) or LPS $(0.1{\mu}g/ml)$ plus SEB $(0.1{\mu}g/ml)$. Culture supernatants were collected at 24, 48, and 72 hrs and assessed for IL-6 and IL-8 production by enzyme-linked immunosorbent assay. IL-6 production in gingival fibroblasts stimulated with LPS was higher than that with SEB. IL-6 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-6 production in facial dermal fibroblasts was increased only by stimulation with a high concentration of LPS $(1.0{\mu}g/ml)$. Its production in facial dermal fibroblasts by exposure with SEB was decreased in comparison with control, nontreated cells. Therefore, gingival fibroblasts showed higher sensitivity than facial dermal fibroblasts in response to low concentration of LPS. Also, IL-6 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-8 production in gingival fibroblasts was enhanced greatly only by stimulation of high concentration of LPS $(1.0{\mu}g/ml)$. That by exposure with SEB was increased only in 24 hrs cultivation. IL-8 production by double exposure with LPS plus SEB was amplified in comparison with single exposure of LPS or SEB. IL-8 production in facial dermal fibroblasts was decreased by LPS and increased only in 48 hrs cultivation by SEB. IL-8 production by double exposure with LPS plus SEB was enhanced only in 48 hrs cultivation in comparison with single exposure of LPS or SEB. therefore, IL-6 and IL-8 production were released at various quantities according to bacterial toxin applied and site of fibroblast harvested. These results suggest that gingival fibroblasts may be concerned with IL-6 and IL-8 related inflammatory response more than facial dermal fibroblasts.
Chlorpyrifos is an organophosphate insecticide and one of the most commonly and widely used insecticide. However, a little known about the dermal risk of chlorpyrifos on human being. Therefore, this study was conducted for the dermal risk assessment after exposure to chlorpyrifos in Korean farmers. First, skin irritation by chlorpyrifos (10 mg/$\textrm{cm}^2$, 50 mg/$\textrm{cm}^2$, 100 mg/$\textrm{cm}^2$, 250 mg/$\textrm{cm}^2$ in acetone) was determined in rabbits for 5 days considering the usage of chlorpyrifos short term highly exposure. The index of skin irritation by chlorpyrifos was increased in each dose and length of exposure dependent manners. Next, using benchmark dose (BMD$_{5}$) approach, the dose-response relationship was assessed to calculate the reference dose (RfD). The value of RfD was 2.84 $\mu\textrm{g}$/kg/day from 142.16 $\mu\textrm{g}$/kg/day BMD5 value divided uncertainty factor 50. Finally, we assessed human dermal risk of chlorpyrifos with exposure level and RfD. Skin absorbed levels were assumed with several exposure scenarios encounting the circumstances of exposure that application method, protection equipment and cloth, exposure time and exposure frequency during chlorpyrifos spraying. By the comparison of skin absorbed dose with the reference dose, it was identified that risk values (risk index) to skin chlorpyrifos exposure were 0.958 from the point of above results and it was recommended that the occurrence of hazard effect (skin irritation toxicity) of chlorpyrifos would not be expected. Risk index was smaller than 1 in the case of spraying vehicle mounted application, 1hour exposure time and wearing protective cloth exposure. Whereas, risk index was above 1 in the case of hand-held application, 2hour exposure time and wearing common cloth. Comparing two kinds of application method, total risk index of the hand held application (1.67) was higher than vehicle mounted (0.27). Therefore, chlorpyrifos skin exposure was mainly affected by application equipment and applied form. The results of risk assessment on the human dermal toxicity of chlorpyrifos should be required to control in keeping safety rules, skin surface area available for contact, spraying time ,and spraying frequency.y.
Farmers are generally expressed to pesticides through mixing loding, application activity and harvesting of crop after application of pesticides. The present work investigated the exposure and risk of furathiocarb to workers when harvesting of cucumber was carried out in green house after application of furathiocarb EC. Glove was used for the hand exposure assessment, socks for foot and dermal patches for the other parts of body. Personal air monitor equipped with a XAD-2 resin was used for the respiratory exposure assessment. During the harvest of cucumber in green house, the initial rate of potential dermal exposure (Day 1) for methidathion was 1.3 mg/hr. The major exposure parts were hand $(78\~83\%),\;thigh\;(5\~7\%)$ and arms $(6\~9\%)$ during 3 days' harvest. No exposure was detected from the respiratory monitoring. For risk assessment, the potential dermal exposure (PDE), the absorbable quantity of exposure (AQE) and the margin of safety (MOS) and margin of exposure (MOE) were calculated. In risk assessment of harvester exposure for 7days, all MOS was > 1 and MOE was > 100 indicating that possibility of risk was little.
