For many years, it has been demonstrated that neurotrophins regulate the adult nervous system, implicating their potential as therapeutic agents for the treatment of neurodegenerative diseases. We generated adenoviral vectors encoding brain-derived neutotrophin factor (BDNF) and neurotrophin-3 (NT3) and tested either separately or together for the ability to induce differentiation of neuronal precursor cells with two different origins. Separate transduction of adenovirus delivering BDNF (BDNF-Ad) or NT3 (NT3-Ad) induced the neuronal differentiation in hippocampal and cortical precursor cells. NT3-Ad infected cells extended short neurites, whereas BDNF-Ad infected cells had longer neurites. In the early differentiation of hippocampal precursor cells, simultaneous infection of BDNF-Ad and NT3-Ad promoted further differentiation and neurite elongation compared with the separate infection of each virus. In contrast, simultaneous infection did not show the synergistic effect in the cortical precursor cells, suggesting that the neurotrophins play distinct roles in different regions of the brain. However, the numbers of neurites and spines per differentiated cells were markedly increased in cortical as well as hippocampal precursor cells, indicating the promotion of efficient neurite elongation and formation of dendritic spine, when BDNF-Ad and NT3-Ad were co-infected. These results suggest more studies in the effect of a combinatorial use of neurotrophins on different sites of brain need to be carried out to develop gene therapy protocols for neurodegenerative diseases.

In lithium-ion batteries, separator membrane's, main role is to physically isolate a cathode and an anode while maintaining rapid transport of ionic charge carriers during the passage of electric current. As far as battery safety is concerned, the electrical isolation of electrodes is most crucial since unexpected short-circuits across the membrane induces hot spots where thermal runaway may break out. Internal short-circuits are generally believed to occur by protrusions on the electrode surface either by unavoidable deposits of metallic impurities or by dendritic lithium growth during battery operation. Another cause is shrinkage of the separator membrane when exposed to heat. If separator membrane can be engineered to prevent the internal short-circuit, it will not be difficult to improve lithium-ion batteries' safety. Commonly the separators employed in lithium-ion batteries are made of polyethylene (PE) and/or polypropylene (PP). These materials have terrible limitations in preventing the fore-mentioned internal short-circuit between electrodes due to their poor dimensional stability and mechanical strength. In this study we have developed a novel separator membrane that possesses very high thermal and mechanical stability. The cells employing this separator provided noticeable safety improvement in the various abuse tests.
Lithium-ion cells have become the go-to energy source across all applications; however, dendritic growth remains an issue to tackle. While there have been various research conducted and possible solutions offered, there is yet to be one that efficiently rules out the problem without, however, introducing another. This paper seeks to present a fast charging method and system to which lithium-ion batteries are charged while maintaining their lifetime. In the proposed method, various lithium cells are charged under multiple profiles. The parameters of charge profiles that inflict damage to the cell's electrodes are obtained and used as thresholds. Thus, during charging, voltage, current, and temperature are actively controlled under these thresholds. In this way, dendrite formation suppressed charging is achieved, and battery life is maintained. The fast-charging system designed, comprises of a 1.5kW charger, an inbuilt 600W battery pack, and an intelligent BMS with cell balancing technology. The system was also designed to respond to the aging of the battery to provide adequate threshold values. Among other tests conducted by KCTL, the cycle test result showed a capacity drop of only 0.68% after 500 cycles, thereby proving the life maintaining capability of the proposed method and system.
Aim: To study anti-tumor effects of exosomes from class II transactivator (CIITA) gene transfected CT26 cells. Methods: In this study, we established an MHC class II molecule-expressing murine colon cancer cell line (CT26-CIITA) by transduction of the CIITA gene. Immune effects in vitro and tumor protective results in vivo were tested and monitored. Results: Exosomes from CT26-CIITA cells were found to contain a high level of MHC class II protein. When loaded on dendritic cells (DCs), exosomes from CT26-CIITA cells significantly increased expression of MHC class II molecules, CD86 and CD80, as compared to exosomes from CT26 cells. In vitro assays using co-culture of immunized splenocytes and exosome-loaded DCs demonstrated that CIITA-Exo enhanced splenocyte proliferation and IFN-${\gamma}$ production of CD4+T cells, while inhibiting IL-10 secretion. In addition, compared to exosomes from CT26 cells, CT26-CIITA-derived exosomes induced higher TNF-${\alpha}$ and IL-12 mRNA levels. A mouse tumour preventive model showed that CT26-CIITA derived exosomes significantly inhibited tumour growth in a dose-dependent manner and significantly prolonged the survival time of tumour-bearing mice. Conclusion: Our findings indicate that CT26-CIITA-released exosomes are more efficient to induce anti-tumour immune responses, suggesting a potential role of MHC class II-containing tumour exosomes as cancer vaccine candidates.
Morphological evolution and growth mechanism at the solid/liquid interface during solidification were investigated in the Ni-base superalloy GTD111M by directional soldification and quenching(DSQ) technique. The experiments were conducted by changing solidification rate(V) and thermal gradient(G) which are major solidification process variables. High thermal gradient condition could be obtained by increasing the furnace temperature and closely attaching the heating and cooling zones in the Bridgeman type furnace. The dendritic/equiaxed transition was found in the G/V value lower than $0.05$\times$10{^3}^{\circ}C$s/$\textrm{mm}^2$, and the planar interface of the MC-${\gamma}$ eutectic was found under $17 $\times$ 10{^3}^{\circ}C$ s/$\textrm{mm}^2$. It was confirmed that the dendrite spacing depended on the cooling rate(GV), and the primary spacing was affected by the thermal gradient more than solidification rate. The dendrite lengths were decreased as increasing the thermal graditne, and the dendrite tip temperature was close to the liquidus temperature at $50 \mu\textrm{m}$/s.
The effect of welding condition on microstructure and mechanical property of Plasma-MIG Hybrid Weld between Al 5083 plates(thickness : 10mm) was investigated. 1 pass weld without any defects such as puckering, undercut, and lack of fusion was obtained by 150~200A of plasma current and 5~7mm of welding speed. Gas porosities and shrinkage porosities were existed in the weld near fusion line. As welding speed and plasma current were decreasing, the area fraction of porosity was increasing. The hardness of the weld is increasing as welding speed. On the basis of microstructural analysis, Mg segregated region near dendrite boundaries tends to increase with the welding speed. In the result of hardness test, Distribution of hardness in fusion zone showed little change with the plasma current. However, when the welding speed increased, hardness in weld metal markdly increased. It could be considered that effect of heat input to growth of the dendritic solidification structures. Based on tensile test, tensile properties of weld metal was predominated by area fraction of porosities. Consequently, tensile properties can be controlled by formation site and area fraction of porosity.

N-acetyl-D-glucosamine kinase (GlcNAc kinase or NAGK) primarily catalyzes phosphoryl transfer to GlcNAc during amino sugar metabolism. Recently, it was shown NAGK interacts with dynein light chain roadblock type 1 (DYNLRB1) and upregulates axo-dendritic growth, which is an enzyme activity-independent, non-canonical structural role. The authors examined the distributions of NAGK and NAGK-dynein complexes during the cell cycle in HEK293T cells. NAGK was expressed throughout different stages of cell division and immunocytochemistry (ICC) showed NAGK was localized at nuclear envelope, spindle microtubules (MTs), and kinetochores (KTs). A proximity ligation assay (PLA) for NAGK and DYNLRB1 revealed NAGK-dynein complex on nuclear envelopes in prophase cells and on chromosomes in metaphase cells. NAGK-DYNLRB1 PLA followed by Lis1/NudE1 immunostaining showed NAGK-dynein complexes were colocalized with Lis1 and NudE1 signals, and PLA for NAGK-Lis1 showed similar signal patterns, suggesting a functional link between NAGK and dynein-Lis1 complex. Subsequently, NAGK-dynein complexes were found in KTs and on nuclear membranes where KTs were marked with CENP-B ICC and nuclear membrane with lamin ICC. Furthermore, knockdown of NAGK by small hairpin (sh) RNA was found to delay cell division. These results indicate that the NAGK-dynein interaction with the involvements of Lis1 and NudE1 plays an important role in prophase nuclear envelope breakdown (NEB) and metaphase MT-KT attachment during eukaryotic cell division.
Aluminum alloy(AC8A) matrix composites reinforced with SiC particles(10% in vol.) were fabricated by Centrifugal Spray Deposition(CSD) process. The microstructures were investigated in order to evaluate both the mixing mode between aluminum matrix and SiC particles, and the effect of SiC particles on the cooling behaviours of droplets during flight and preforms deposited. A non-continuum mathematical calculation was performed to explain and to quantify the evolution of microstructures in the droplets and preforms deposited. Conclusions obtained are as follows; 1. The powders produced by CSD process showed, in general, ligament type, and more than 60% of the powders produced were about 300 to 850 um in size. 2. AC8A droplets solidified during flight showed fine dendritic structure, but AC8A droplets mixed with SiC particles showed fine equiaxed grain structure, and eutectic silicon were formed to crystallize granularly between fine aluminum grains. 3. SiC particles seem to act as a nucleation sites for pro-eutectic silicon during solidification of AC8A alloy. 4. The microstructure of composite powders formed by CSD process showed particle embedded type, and resulted in dispersed type microstructure in preforms deposited. 5. The pro-eutectic silicon crystallized granularly between fine aluminum grains seem to prohibit grains from growth during spray deposition process. 6. The interfacial reactions between aluminum matrix and SiC particles were not observed from the deposit performs and the solidified droplets. 7. The continuum model seem to be useful in connecting the processing parameters with the resultant microstructures. From these results, it was concluded that the fabrication of aluminum matrix composites reinforced homogeneously with SiC particles was possible.
Modern technology-driven society largely relies on hybrid electric vehicles or electric vehicles for eco-friendly transportation and the use of high technology devices. Lithium rechargeable batteries are the most promising power sources because of its high energy density but still have a challenge. Graphite is the most widely used anode material in the field of lithium rechargeable batteries due to its many advantages such as good cyclic performances, and high charge/discharge efficiency in the initial cycle. However, it has an important safety issue associated with the dendritic lithium growth on the anode surface at high charging current because the conventional graphite approaches almost 0 V vs $Li/Li^+$ at the end of lithium insertion. Therefore, a fundamental solution is to use an electrochemical redox couple with higher equilibrium potentials, which suppresses lithium metal formation on the anode surface. Among the candidates, $Li_4Ti_5O_{12}$ is a very interesting intercalation compound with safe operation, high rate capability, no volume change, and excellent cycleability. But the insulating character of $Li_4Ti_5O_{12}$ has raised concerns about its electrochemical performance. The initial insulating character associated with Ti4+ in $Li_4Ti_5O_{12}$ limits the electronic transfer between particles and to the external circuit, thereby worsening its high rate performance. In order to overcome these weak points, several alternative synthetic methods are highly required. Hence, in this presentation, novel ways using a synergetic strategy based on 1D architecture and surface coating will be introduced to enhance the kinetic property of Ti-based electrode. In addition, first-principle calculation will prove its significance to design Ti-based electrode for the most optimized electrochemical performance.
