식물의 저온 적응 메카니즘에서 아스코브산과 관련된 효소 중 MDHA 환원효소의 활성도와 과산화수소, 아스코브산의 함량, mRNA의 발현수준과의 연관성을 연구한 결과는 다음과 같다. MDHA 환원효소의 활성도 변화는 저온에 노출되는 시간이 길어질수록 증가하였으며 6시간 이후에 엽록체분획과 세포질분획에서 급격하게 증가하는 경향을 보였으나 실온으로 회복시켰을 때 효소의 활성도가 상대적으로 감소하는 경향을 보였다. 저온에 노출된 동안 아스코브산의 함량은 비교적 일정한 경향을 보이다가 실온으로 회복시키면 그 이후에는 급격하게 증가하는 경향을 보였다. 반면 저온에 노출되는 동안 급격히 dehydroascorbate 함량이 감소하였다가 실온으로 회복되면 약간 증가하는 경향을 보였다. 아스코브산의 함량과 엽록체분획과 세포질분획의 MDHA 환원효소의 활성도와의 상관관계는 각각 정의 상관($R^2$=0.9240, 0.9108)을 나타내었으나 디하이드로아스코브산의 함량과 MDHA 환원효소의 활성도 사이에는 각각 부의 상관($R^2$=0.8638, 0.8980)을 나타내었다. MDHA 환원효소 활성도와 과산화수소의 함량과의 상관관계를 과산화수소의 생성량이 증가하면 MDHA 환원효소의 활성도가 증가하는 정의 상관($R^2$=0.9443, 0.9647)을 나타내었다. 저온스트레스 처리 시간이 증가할수록 MDHA 환원효소의 mRNA의 발현 수준과 총 MDHA 환원효소의 활성도가 증가하는 경향을 나타내었다.
The relationship between relative water contents of lettuce leaves and biochemical activities in lettuce was examined in this study to explore an adaptation response of lettuce to water stress from soils. Soil water contents and relative water contents of leaves were positively related to show $R^2$=0.8728. Hydrogen peroxide contents of leaves rapidly increased with reduction of soil water content, whereas soluble protein contents and dry matters rapidly decreased. And chlorophyll a and b contents of leaves decreased with increase in carotenoid content. Furthermore, the activities of ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), and dehydroascorbate reductase (DHAR) increased dramatically, and mRNA transcript levels of APX, MDHAR and DHAR also increased. Relationship of relative water content of lettuce leaves to hydrogen peroxide, to ascorbate peroxidase activity, to dehydroascorbate reductase activity, and to monodehydroascorbate reductase activity was shown to be positively correlated. It is highly plausible from this study that these enzyme activities could be developed as an indicator of water states in soils.

Thioltransferase, also called glutaredoxin, is a general GSH-disulfide reductase of importance for redox regulation. Previously, the protein thioltransferase, now called S-type thioltransferase, was purified and characterized from Arabidopsis thaliana seed. In the present study, a second thioltransferase, called L-type thioltransferase, was purified to homogeneity from Arabidopsis thaliana leaves. The purification procedures included DEAE-cellulose ion-exchange chromatography, Sephadex G-50 gel filtration, and glutathione-agarose affinity chromatography. The purified enzyme was confirmed to show a unique band on SDS-PAGE and its molecular weight was estimated to be 26.6 kDa, which appeared to be atypical compared with those of most other thioltransferase. It could utilize 2-hydroxyethyl disulfide, S-sulfocysteine, and insulin as substrates, and also contained dehydroascorbate reductase activity. Its optimum pH was 8.5 and its activity was greatly activated by L-cysteine. When it was kept for 30 min, it appeared to be very stable up to $70^{\circ}C$. It was activated by $MgCl_2$ and, on the contrary, inhibited by $ZnCl_2$, $MnCl_2$, and $AlCl_3$.
Dehydroascorbate (DHA) reductase (DHAR, EC 1.8.5.1) catalyzes the reduction of DHA to reduced ascorbate (AsA) using glutathione (GSH) as the electron donor in order to maintain an appropriate level of ascorbate in plant cells. To analyze the physiological role of DHAR in environmental stress adaptation, we developed transgenic tobacco (Nicotiana tabacum cv. Xanthi) plants that express a human DHAR gene isolated from the human fetal liver cDNA library in the chloroplasts. We also investigated the DHAR activity, levels of ascorbate, and GSH. Two transgenic plants were successfully developed by Agrobacterium-mediated transformation and were confirmed by PCR and Southern blot analysis. DHAR activity and AsA content in mature leaves of transgenic plants were approximately 1.41 and 1.95 times higher than in the non-transgenic (NT) plants, respectively In addition, the content of oxidized glutathione (GSSG) in transgenic plants was approximately 2.95 times higher than in the NT plants. The ratios of AsA to DHA and GSSG to GSH were changed by overexpression of DHAR, as expected, even though the total content of ascorbate and glutathione was not significantly changed. When tobacco leaf discs were subjected to methyl viologen at $5\;{\mu}M$, $T_0$ transgenic plants showed about a 50% reduction in membrane damage compared to the NT plants.
상추식물에서 저온 처리를 하였을 때 저온 적응성 획득 메카니즘과 관련된 APX와 DHAR의 활성도와 mRNA 발현 수준 등과의 관련성을 조사한 결과는 다음과 같다. 잎 조직내 과산화수소의 함량은 일정하게 증가하다가 $20^{\circ}C$에서 저온처리를 회복시키면 그 함량이 정상상태로 회복하는 경향을 보였으나 단백질의 함량은 반대의 경향을 나타내었다. 엽록소의 함량은 저온스트레스를 처리할 경우 엽록소 a와 b 및 총 엽록소의 함량이 점차 감소하는 경향을 보였으며 저온스트레스 회복 시 다시 증가하는 경향을 보였으나 카로티노이드 함량의 변화는 거의 일어나지 않고 일정한 수준을 유지하는 것으로 나타났다. 총 엽록소에 대한 엽록소a의 비율은 저온처리 12시간까지는 증가하다가 24시간 이후 급격하게 감소하게 되고 저온스트레스를 회복시켰을 때 정상 수준으로 회복되는 경향을 나타내었다. APX와 DHAR의 활성도는 저온 처리가 진행됨에 따라 상추의 잎 조직 내에서 급격하게 증가하는 경향을 보이고 있으나 적온으로 회복시키면 정상 수준으로 유지되었다. 저온처리 시간이 경과함에 따라 APX와 DHAR의 mRNA의 발현 수준이 크게 증가하는 경향을 보였다가 원래 수준으로 회복되었다. APX의 활성도와 과산화수소의 함량과의 상관관계는 상추의 잎 조직 내 과산화수소의 함량이 증가하면 APX의 활성도가 증가하는 경향 ($R^2$=0.8715)을 보였으며, DHAR의 활성도와 과산화수소의 함량도 동일한 경향으로 증가($R^2$=0.8643)하였다. 그러나 과산화수소의 생성량이 증가함에 따라 엽록소의 함량과 단백질의 함량은 저온 스트레스처리로 과산화수소의 생성량이 증가하면 엽록소의 함량($R^2$=0.5021)과 수용성단백질의 함량과는 감소하는 경향($R^2$=0.8915)을 보였다.
Maintaining redox balance is one of the crucial requirements for a cell to endure stress from the outside. Dehydroascorbate reductase (DHAR; EC 1.8.5.1) plays an important role in the ascorbate-glutathione cycle; one of the major ROS scavenging systems in most known biological systems. A cDNA clone of the DHAR gene from Oryza sativa (OsDHAR) was isolated and overexpressed in Escherichia coli BL21 (DE3) strain from the pET-28a(+) expression vector. The OsDHAR transformed E. coli cells showed significantly higher DHAR activity and a lower level of ROS than the E. coli cells transformed by an empty pET-28a(+) vector. Also, the DHAR-overexpressing E. coli strain was more tolerant to oxidant- and heavy metal-mediated stress conditions than the control E. coli strain. The results suggest that the overexpressed rice DHAR gene effectively functions in a prokaryotic system and provide protection to various oxidative stresses.
Tomato (Lycopersicon esculentum) seedlings exposed to various concentrations of $CdC1_2$ (0∼100 $\mu$M) in the nutrient solution for up to 9 days were analyzed with the seedling growth, $H_2O_2$ production, glutathione levels and activity changes of enzymes related to $H_2O_2$ removal. The growth of seedlings was inhibited with over 50 $\mu$M Cd, whereas the levels of $H_2O_2$ and glutathione were enhanced with Cd exposure level and time. Meanwhile, Cd exposure increased the activities of catalase (CAT) and glutathione reductase (GR) but decreased the activities of dehydroascorbate acid reductase (DHAR) and ascorbate peroxidase (APX) in both leaves and roots. These results suggest that the altered activities of antioxidant enzymes particularly involved in the $H_2O_2$ removal and the subsequent $H_2O$$_2$ accumulation could induce the Cd-induced phytotoxicity.
Thioltransferase, also known as glutaredoxin, was purified from Chinese cabbage (Brassica campestris ssp. napus var. pekinensis) by a combination of ion-exchange chromatography and gel filtration. Its purity was confirmed by SDS-polyacrylamide gel electrophoresis and its molecular weight was estimated to be about 12,000 which is comparable with those of most known thioltransferases. The enzyme utilizes 2-hydroxyethyl disulfide, S-sulfocysteine, ${\alpha}-chymotrypsin$, insulin, and trypsin as substrates in the presence of reduced glutathione. The enzyme has Km values of 0.03-0.97 mM for these substrates. It appeared to contain dehydroascorbate reductase activity. The pH optimum of the enzyme was 8.5, when 2-hydroxyethyl disulfide was used as a substrate. It was greatly activated by reduced glutathione. Its activity was not significantly lost when stored at high temperature, indicating its thermostable character. It may play an important role in thiol-disulfide exchange in plant cells.
염 스트레스(0, 50, 100mM NaCl)에 의한 식물 반응을 이해하기 위하여, 본 실험은 수경법으로 2엽기까지 기른 배추를 이용하였다. 신선중과 건물중, 엽록소, 항산화 물질, 폴리아민함량, 항산화 효소 활성, 그리고 이온 레벨을 조사하였다. 배추의 생체중 및 건물중은 염의 증가에 따라 증가하였으며, 50mM NaCl 처리시 최적의 성장을 보였다. Chl a, total Chl, carotenoid 함량과 Chl a/b 비는 100mM NaCI 처리후 6일째 증가하였으나, Chl b 함량은 감소하였다. 글루타치온 함량은 염처리 후 6일째 뿌리에서 증가하였으며, dehydroascorbate는 6일째 잎과 뿌리에서 현저하게 증가하였다. 염처리후 6일째 잎에서, ascorbate peroxidase 활성은 증가하였으나, catalase와 glutathione reductase 활성은 감소하였다. 지상부에 100mM NaCI 처리후 3일째, $Na^+$ 함량은 12.5배 증가하였으나, $Ca^{2+}$, $K^+$과 $Mg^{2+}$ 함량은 43-57% 감소하였다. 또한 염 스트레스에 의한 스페르미딘 감소 현상과 스페르민 증가현상을 나타내었다. 50mM NaCl 처리에 의해 배추의 생장 촉진, 글루타치온 및 비타민 C 함량 증가 현상이 나타났으며, 또한 식물의 생장 촉진에 의한 재배 기간의 단축효과도 기대된다.
본 연구는 $H_2O_2$ 축적이 수반되는 잎의 노쇠현상과 관련하여 세포질내 ascorbate-glutathione 회로의 역할 가능성과 이때 benzyladenine(BA)의 효과를 밝히기 위하여 노쇠중인 밀 잎에서 시토졸 ascorbate peroxidase(APX) isozyme 분리 및 발달양상과 ascorbate-glutathione 회로 구성효소들의 활성도 변화를 조사하였다. 성숙한 밀 제1엽 절편을 4일간 암배양하는 동안 증가된 엽록소 분해 및 $H_2O_2$ 축적으로 규정되는 잎의 노쇠발달중 대조구 잎에서는 시토졸 APX 활성도의 유의성 있는 증가가 관찰되지 않았으며 dehydroascorbate reductase(DHAR)의 활성도는 급격히 감소되었고, glutathione reductase(GR) 활성도는 완만하게 증가하였다. 그러나 BA로 처리된 잎에서는 시토졸 APX 활성도가 현저하게 증가하였으며 DHAR 활성도의 감소가 지연되어 나타났고 GR 활성도의 증가는 대조구에 비해 증진되었으며 내재성 ascorbate 함량의 감소율과 H2O2 축적이 억제되었다. 3개의 시토졸 APX isozyme이 native-PAGE법에 의해 노쇠중인 밀 잎에서 발견되었으며 그 중 2개 isozyme은 높은 활성도를 보였다. 시토졸 APX isozyme의 발달양상의 경우 4일간의 암배양 동안 대조구 잎에서는 단지 2개의 isozyme band("a"와 "b")가 거의 같은 활성도를 지닌 채 출현하였으나 BA로 처리된 잎에서는 추가로 1개의 약한 isozyme band("c")가 더 나타났으며 "b" isozyme의 활성도가 약간 촉진되었다 그러나 "a" isozyme 활성도는 대조구 잎에 비해 암배양시간 경과에 따라 현저하게 발달하였다. 대조구 잎과 비교시 BA로 처리된 잎에서는 시토졸 APX isozyme의 발달 및 APX, DHAR, GR의 전체 활성도가 함께 증진되어 그 결과 $H_2O_2$ 제거능력이 증대된 본 실험의 결과는 ascorbate-glutathione 회로가 밀 잎의 노쇠과정에 중요하게 작용하고 있음을 제시하였다.
