Pathogenetic Impact of Vacuolar Degeneration by Accelerated Transport of Helicobacter pylori VacA

  • Vacuolar degeneration of the gastric epithelial cells is a characteristic feature of the derangement of mucosa where Helicobacter pylori colonizes, and H. pylori vacuolating cytotoxin (VacA) has been suggested to playa key role in it. To elucidate the VacA-involved degenerative mechanism, VacA was purified, and its impact on degeneration of HeLa cells was determined. In the presence of ammonium chloride, cell vacuolation by VacA was dose-and time-dependent, however, no detectable degeneration of the cells was observed with the VacA concentration tested. A further increase of vacuolation was shown in cells pre-treated with diethyl pyrocarbonate (DEPC) , and this resulted in a change of the cell morphology to become spherical. Similar phenomena were also observed when HeLa cells were co-cultivated with intact H. pylori cells. It was remarkable to note that the degree of growth inhibition was proportional to the increase in vacuole formation, suggesting that the vacuolation rate would be critical for cell degeneration. Surprisingly, although VacA was itself inhibited by DEPC, its uptake was markedly increased by this agent, similar to that found in cells with Nabutyrate. These data indicate that the cell's tolerance of VacA transport may be critical for vacuolar degeneration and may be changeable during H. pylori inhabitation.

Similar Degree of Degeneration in the Articular and Bursal Layers of Delaminated Rotator Cuff Tear

  • Background: The purpose of the study was to compare the degree of degeneration of the articular and bursal layers of delaminated supraspinatus tendons based on histological examination. Methods: Fifty-four patients with a full-thickness rotator cuff tear were included in the study. Tendon specimens were harvested during arthroscopic rotator cuff repair from the lateral torn edges of the articular and bursal layers of the delaminated tear. Harvested samples were stained with H&E dye and evaluated based on a semi-quantitative grading scale. Results: There were no significant differences in the seven histological characteristics of tendon degeneration: fiber structure, fiber arrangement, round nuclei, regional variations in cellularity, vascularity, collagen stainability, and hyalinization between the articular and bursal layers of the delaminated rotator cuff tear (all p>0.05). Total degeneration scores of articular and bursal sides were $13.1{\pm}3.85$ points and $13.2{\pm}3.42$ points, respectively, and were not significantly different (p=0.958). Conclusions: The study demonstrates that tendon degeneration was similar in the articular and bursal sides of the delaminated full-thickness rotator cuff tear, suggesting that degeneration would be a main etiology for the rotator cuff tear not only in the articular side but also in the bursal side. Considering potential disadvantages of subacromial decompression, this study tentatively suggests routine use of subacromial decompression as well as the need for halting or recovery from rotator cuff degeneration for better rotator cuff repair.

An unusual presentation of non-specific cystic degeneration of craniofacial fibrous dysplasia: a case report and review of literature

  • Background: Fibrous dysplasia (FD) is a rare, sporadic, and benign congenital condition in which normal cancellous bone is replaced by fibro-osseous tissue with immature osteogenesis. FD localized in the cranial and facial bones is called craniofacial fibrous dysplasia (CFD). Cystic degeneration in CFD cases is rare; cystic degeneration appearing in both the maxilla and the mandible FD lesion is even rarer. The aim of this article was to report a case of fibrous dysplasia of the mandible and maxilla complicated by nonspecific cystic degeneration. Case presentation: A 30-year-old woman presented with a rare case of non-specific cystic degeneration in a mandible and maxilla FD lesion that occurred 11 years after surgery. She was diagnosed with polyostotic CFD and underwent maxillary and mandibular bone contouring. Cyst enucleation under general anesthesia was performed in the mandibular region due to pain and discomfort. Conclusions: In cases involving non-aggressive and non-invasive FD cystic degeneration in focal areas, conservative treatment is recommended. However, if cystic degeneration of FD develops rapidly and causes discomfort, pain, or dysfunction, surgical treatment should be considered.

Degeneration of Ocellar Photoreceptor System on Drosophila rdgC Mutant (초파리 rdgC 돌연변이체 단안 시각계의 퇴행현상)

  • The morphological phenotype on ocellus of Drosophila rdgC mutant was observed with electron microscope. The result showed the particular phenotype that was not found in other retinal degenarative mutants. The most distinct difference was the orientation of photoreceptor cells. The photoreceptor cells did not attached to corneagenous cells but dropped under corneagenous cells and assembled around newly formed space. Enormous multivesicle bodies caused by the degeneration of photoreceptor cells were frequently found. Rhabdomeres were also severely degenerated in consequence of the mutant. Another degeneration was found in a part of photoreceptor cell, but the degeneration of subrhabdomeric cisternae (SRC) was not found. It was a ovious difference of rdgC comparing with other two retinal degenerative mutants, rdgA and rdgB. As a result, rdgC mutant was affected on the attachment between photoreceptor cells and corneageneous cells, and it suggested the defect of cell-cell attachment. In addition, rdgC mutant was accompanied by the defect not only in retina but nerve system. The results were agreed to the reference discussion that the rdgC molecule is exist in the nerve.

Senile Macular Degeneration. A Case Report (두통환자의 치료중 황반변성 변화에 관한 증례 1례)

  • This case study was performed to investigate the effects of Chongsimyonja-tang and Dong's acupuncture therapy on senile macular degeneration patient who has been treated in the Dong-Eui oriental hospital. A 57-year-old female with senile macular degeneration complained of headaches, heaviness of the ache, night sweat, dizziness. For a duration of 2 months, We prescribed Chongsimyonja-tang, and added 3-4 herbs according to the symtoms and also treated the patient with Dong s acupuncture therapy. After 2 months, the patient's eyesight improved ($0.2{\rightarrow}0.7$) and the headaches disappeared, as did heaviness of the ache, night sweat, and dizziness. In this case Chongsimyonja-tang and Dong's acupuncture therapy present valuable treatment of senile macular degeneration.

Oogenesis, Oocyte Degeneration and Sexual Maturation in Female Cyclina sinensis (Gmelin, 1971) (Bivalvia: Veneridae) in Korea

  • The ultrastructure of germ cells and follicle cells during oogenesis, oocyte degeneration, reproductive cycle, and first sexual maturity in female Cyclina sinensis were investigated for clams collected from Simpo, Jeollabuk-do, Korea, by cytological and histological observations. Vitellogenesis occured by way of endogeous autosynthesis and exogenous heterosynthesis: vitellogensis occurred through a process of autosynthesis, which involves a combined activities of the Golgi complex, mitochondria, and rough endoplasmic reticulum. The process of heterosynthesis involved endocytotic incorporation of extraovarian precursors into the basal region of the early vitellogenic oocytes prior to the formation of vitelline envelope. The follicle cells appear to play an integral role in vitellogenesis and oocyte degeneration, functioning in phagocytosis and digestion of products originating from the degenerated oocytes: these functions can permit the transfer of yolk precursors needed for vitellogenesis. Follicle cells might have a lysosomal system for breakdown and might also resorb phagosomes in the cytoplasm for nutrient storage during oocyte degeneration.

Inherited retinal degeneration in a Bengal cat

  • A 2-year-old intact female Bengal cat was presented with a 6-month history of visual impairment. The cat manifested bilateral negative menace responses and dazzle reflexes and sluggish pupillary light reflexes. Bilateral fundus changes included generalized tapetal hyperreflectivity, advanced retinal vascular attenuation, and increased pallor of the optic disc. A diagnosis of bilateral retinal degeneration was made. The clinical findings suggest that the investigated Bengal cat was most likely to have an inherited retinal degeneration. Further studies of the Bengal cat breed are needed to determine the prevalence of inherited retinal degeneration in this breed in Korea.

  • Ultrastructural studies of oogenesis in oocytes, oocyte degeneration associated with the follicle cells in female Coecella chinensis were investigated for clams collected from Namhae, Geongsangnam-do, Korea. In this study, vitellogenesis during oogenesis in the oocytes occured by way of endogenous autosynthesis and exogenous heterosynthesis. Of two processes of vitellogenesis during oogenesis, the process of endogenous autosynthesis involved the combined activity of the Golgi complex, mitochondria and rough endoplasmic reticulum. whereas the process of exogenous heterosynthesis involved endocytotic incorporation of extraovarian precursors at the basal region of the oolema of the early vitellogenic oocytes prior to the formation of the vitelline coat. It is assumed that the follicle cells were involved in the development of previtellogenic and early vitellogenic oocytes and appear to play an integral role in vitellogenesis in the early and late vitellogenic oocytes by endocytosis of yolk precursors, and also they were involved in oocyte degeneration by assimilating products originating from the degenerated oocytes, thus allowed the transfer of york precursors needed for vitellogenesis (through phagocytosis by phagolysosomes after spawning). Follicle cells presumably have a lysosomal system for breakdown products of oocyte degeneration. and for reabsorption of various phagosomes (phagolysosomes) in the cytoplasm for nutrient storage during the period of oocyte degeneration.

EFFECTS OF ALCOHOL AND GLYCEROL INJECTION ON THE RAT INFRAORBITAL NERVE (백서 안와하신경에서 알콜 및 글리세롤 주입의 효과)

  • This study was performed to investigate the changes of nerve after the injection of alcohol and glycerol at the infraorbital nerve in rats. Using the eighteen Sprague-Dawley rats, weighing $200{\sim}250g$, 99% alcohol, pure glycerol, and sterile saline was injected to the epineurium of the infraorbital nerve. Glycerol injected rats were devided into 0.01ml, 0.03ml and 0.05ml groups. The alcohol and control group were injected 0.03ml at the left infraorbital nerve. The following results were obtained by histopathological examination after 1 week, 1 month, and 3 months. A few inflammatory cell infiltration and no signs of nerve degeneration were noted in control group. Total nerve degeneration was noted in the alcohol group and no regeneration was noted in 1month, and partial regeneration was noted at 3month. The nerve degeneration was noted at the periphery of nerve bundle in 0.01ml glycerol injection group. Total degeneration was noted in the 0.03ml and 0.05ml glycerol injection group and the degree was propotional to dose. These results suggest that injection of alcohol and glycerol are effective to nerve blockage by nerve degeneration, and nerve degeneration by glycerol injection is propotional to dose and nerve regeneration by glycerol injection is inversely propotional to dose.

Prevalence of Disc Degeneration in Asymptomatic Korean Subjects. Part 2 : Cervical Spine

  • Objective : Similar to back pain, neck pain has recently shown to have increasing prevalence. Magnetic resonance imaging (MRI) is useful in identifying the causes of neck pain. However, MRI shows not only pathological lesions but also physiological changes at the same time, and there are few Korean data. The authors have attempted to analyze the prevalence of disc degeneration in highly selective asymptomatic Korean subjects using MRI. Methods : We performed 3 T MRI sagittal scans from C2 to T1 on 102 asymptomatic subjects (50 men and 52 women) who visited our hospital between the ages of 14 and 82 years (mean age 46.3 years). All images were read independently by three observers (two neurosurgeons and one neuroradiologist) who were not given any information about the subjects. We classified grading for cervical disc herniation (HN), annular fissure (AF), and nucleus degeneration (ND), using disc degeneration classification. Results : The prevalence of HN, AF, and ND were 81.0%, 85.9%, and 95.4%, respectively. High prevalence of HN, AF, and ND was shown compared to previous literature. Conclusion : In asymptomatic Korean subjects, the abnormal findings of 3 T MRI showed a high prevalence in HN, AF, and ND. Several factors might play important roles in these results, such as population-specific characters, MRI field strength, and disc degeneration grading system.
