동백종실(Camellia japonica L.)과 유박의 일반성분, 사포닌 및 아미노산 조성 (Proximate, Saponin and Amino Acid Compositions in Camellia (Camellia japonica L.) Seeds and Defatted Camellia Seeds)

  • To furnish basic data for the utilizatin of camellia(Camellia japonica L.) seeds as a raw material of industrial products, major chemical components of camellia and defatted camellia seeds were investigated. The moisture, crude ash, crude fat and crude protein contents of camellia seeds were 4.06%, 1.83%, 65.75% and 8.44%, respectively. Defatted camellia seeds indicates that it contains 10.31% moisture, 5.28% crude ash and 17.49% crude protein while no crude fat was detected. The curde ginseng saponin, crude saikosaponin and red ginseng saponin contents of camellia seeds were 108.0mg%, 0.4mg% and 40.3mg%, respectively. Total amino acid contents of camellia seeds and defatted camellia seeds were 7,851mg% and 13,002mg%, respectively, and the major amino acids were glutamic acid, arginine, aspartic acid and leucine. The ratio of essential/total amino acid of camellia seeds was 0.29.

동백(Camellia japonica L.)유박 추출물의 항균활성 (Antimicrobial Activity of Defatted Camellia(Camellia japonica L.) Seeds Extract)

  • To developed natural food preservatives, ethanol and water extracts were prepared from the defatted camellia(Camellia japonica L.) seeds. Antimicrobial activities were examined against 14 microorganisms which were food borne pathogens and/or food poisoning microorganisms and food-related bacteria and yeasts. Ethanol extract showed antimicrobial activities against several microorganisms tested, but not on lactic acid bactria. Especially, minimum inhibitory concentrations(MIC) for yeasts were as low as 1mg/ml. Water extract exhibited antimicrobial activities for the yeasts tested, but almost bacteria were not observed. The ethanol extract was fractionated in the order of hexane, chloroform, ethylaetate and butanol fractions to test antimicrobial activity. The strongest activity for the tested yeasts were found in the butanol fractions, but bacteria were chloroform fraction. Hansenula anomala treated with ethanol were observed by scanning electron micrograph, and the results exhibited morphological changes, including the irregularly contracted cell surface and expanded ellipsoidal shape.

품종별 유채박 단백질의 추출에 관한연구 (Studies on the Extraction of Rapesedd Protein from Some Species)

  • Studies on protein and antinutritional factors of rapeseeds are ncessary for effective utiliza-tion of defatted rapeseed meal. Proteins were extracted from seeds of several species of rape-seeds and analyzed by SDS-PAGE and the contents of glucosinolate and phytate were determi-ned. One percent solution extraction and extraction yield was relatively higherfor B. campestris and B. juncea than for other species. SDS-PAGE revealed that rapeseeds of most species were rich in low molecular weight proteins and that in particular the roteins of B. napus var. Halla and B. juncea were composed of simpler subunits as compared with other species. The content of glucosinolate was around 10mg/g of defatted meal for B. juncea however for var. Halla it was 7.26mg/g of defatted meal and 0.46mg/g of protein concentrate which were the lowest values. The level of phytate was between 2.7 and 4.6% for all species tested. Our results indicate thT B, napus var. Halla is the desired species for the utilization of rapeseed proteins.

Lipoxygenase Inhibitor from Defatted Nutmeg Seed

  • Lipoxygenase inhibitory acitivity of the methanolic extract of 60 different kinds of plant seeds was determined by a spectrophotometric method using a soybean lipoxigenase(SLO) and linolenic acid. Among the extracts examined, the methanolic extract of nutmeg(Myristical fragrans)seed showed the most potent SLO inhibitory activity. To isolate SLO inhibitor, hence, the defatted methanol extract was further partitioned with ether, ehtylacetate , and n-butanol , stepwise. The ether souble fraction was successively chromatographed on silica gel, Sephadex LH-20 and preparative TLC. Three phenolic compounds were isolated , and one of them showing a strong SLO inhibition activity was identified as a 2,6-dihydroxy-9-(3', 4', -dihydroxyphenyl)nonylphenone (IC50a=0.39$\mu\textrm{g}$/ml) by 1H-& 13C0NMR, IR, and MS spectroscopy.

녹차씨 전처리와 추출 조건에 따른 녹차씨 추출물의 추출 수율 및 항효모 활성 (Extraction Yield and Anti-Yeast Activity of Extract from Green Tea Seeds by Pretreatment and Extraction Conditions)

  • 항효모 활성이 우수한 녹차씨로부터 천연 보존소재를 개발하기 위하여 추출 원료를 최적화하기 위한 녹차씨의 전처리 방법을 평가하였다. 과피 유무에 따른 녹차씨의 추출 결과 추출물의 항효모 활성은 같았으며, 추출 수율은 과피를 제거한 속씨에서 근소하게 높은 것으로 나타났다. 건조에 따른 녹차씨의 수분 함량은 항효모 활성에는 영향을 주지 않았으나 추출 수율은 7.3% 수분 함량에서 가장 높게 나타났다. 분쇄 공정에 따른 녹차씨 추출물의 항효모 활성은 같았으나, 원료의 입도가 작은 믹서 분쇄 원료에서 추출 수율이 더 높게 나타났다. 녹차씨 오일을 추출한 부산물인 탈지 녹차씨를 추출하여 보존소재 원료로써 활용 가능성을 평가한 결과 항효모 활성은 녹차씨 추출물과 같았으며, 탈지 방법에 따라 수율은 다르게 나타나 헥산 추출 탈지 녹차씨보다 압착 탈지 녹차씨의 추출 수율이 더 높게 나타났다. 녹차씨의 전처리 방법 평가에 따라 천연 보존소재를 추출하기 위한 원료로서 과피가 포함된 녹차씨를 수분 함량 7.3%로 건조한 후 롤밀 분쇄하여 착유기로 오일을 추출한 탈지 녹차씨를 제조하였다. 탈지 녹차씨를 추출 용매에 따라 추출한 후 수율 및 항효모 활성을 조사한 결과 경제성이 높은 추출 용매는 물이 적합하였다. 추출 온도 및 시간에 따른 탈지 녹차씨의 추출결과 $90^{\circ}C$ 추출에서는 항효모 활성이 다소 불안정한 것으로 나타났다. 추출 수율과 항효모 활성의 안정성을 고려하여 물을 용매로 하였을 때 $50^{\circ}C$, 4시간 추출 조건이 적합할 것으로 생각된다.

Comparison of Functional Constituents and Biological Activity of the Seed Extracts from Two Mulberry Fruits

  • The seeds from two mulberry fruits [Morus alba (MA) and Cudrania tricuspidata (CT)] were examined for their oil content, and fatty acid, phytosterol and tocopherol compositions and contents. Moreover, polyphenolic compounds and biological activity of the two defatted seed residue extracts were also evaluated. Oil contents of MA and CT seeds were 29.36% and 16.69%, respectively, while MeOH extracts of the defatted MA and CT seed residues were 5.10% and 6.22%, respectively. The two seed oils were composed of 81.4 and 74.37% linoleic, 5.75 and 11.39% oleic, 8.40 and 10.18% palmitic acid, and 3.52 and 3.0% stearic acids, and two other minor fatty acids, such as linolenic and arachidic acids. MA seed had higher contents of phytosterols (507.59 mg/100 g of oil), tocopherols (99.64 mg/100 g of oil), and total flavonoid (106.50 mg/100 g of seed) than CT seed, whereas CT seed had higher levels of total polyphenol than MA seed. The MeOH extract of MA seed residue showed higher antioxidant, anti-diabetic, and anti-melanogenic activity than that of CT seed residue. trans-Resveratrol (9.62 mg/100 g), quercetin (54.83 mg/100 g), and 4-prenylmoracin (48.70 mg/100 g), were found to be the main polyphenolic components in the MeOH extract of MA seed residue. These results indicate that MA seeds are good sources of essential dietary phytochemicals with antioxidant, anti-diabetic and anti-melanogenic activity.

고콜레스테롤식에서 홍화박과 들깨박이 난소절제 흰쥐의 지질대사에 미치는 영향 (Effects of Defatted Safflower and Perilla Seed Powders on Lipid Metabolism in Ovariectomized Female Rats Fed High Cholesterol Diets)

  • To study effect of non-fat components present in plant seeds on lipid metabolism, defatted safflower and perilla seed powders were used in high cholesterol diets for ovariectomized (ovx) female Sprague-Dawley rats weighing 227$\pm$15g. Experimental groups were six as follows; normal group without ovariectomy and cholesterol-free diet, sham and ovx-control groups with high cholesterol and cellulose for dietary fiber, ovx-est group with the same diet as ovx-control but with eight subcutaneous injections of 50$\mu\textrm{g}$ 17$\beta$-estradiol. ovx-safflower and ovx-perilla with 29% and 16% (w/w) of each defatted powder in high cholesterol diets at the expense of cellulose. Weight gains were lower in normal and sham groups and food efficiencies were lower in normal,ovx-est and ovx-safflower groups compared with ovx-control. Uterus weights were dramatically reduced by ovariectomy but restored completely by 17$\beta$-estradiol and partially (~5%) by defatted safflower. Plasma levels of total cholesterol were not different among ovx-control, sham, vx-est and ovx-safflower groups (90.8~95.1 mg/dL) but that was lower in ovx-perilla (80.4$\pm$6.2 mg/dL). Plasma triglyceride (TG) levels were lower in sham (76.6$\pm$7.0 mg/dL) and ovx-perilla (79.2$\pm$5.8 mg/dL) groups. Liver cholesterol levels were lower in sham, ovx-est, ovx-safflower and ovx-perilla groups (26.6~29.8 mg/g) than ovx-control (36.5$\pm$3.2 mg/g). But liver TG levels were reduced only sham and ovx-est groups compared to control group. Fecal excretions of bile acid and cholesterol were highest in ovx-safflower group (30.8$\pm$5. and 32.1$\pm$5.7 mg/g) compared with other ovx groups (20.8~23.1 and 12.1~19.5 mg/g). These results suggest that both perilla and safflower seeds contain groups (20.8~23.1 and 12.1~19.5mg/g). These results suggest that both perilla and safflower seeds contain non-fat and non-fiber components having lipid lowering effects.

볶음온도와 시간을 달리하여 얻은 참깨박의 이화학적인 특성의 변화 (Changes in the Physico-Chemical Properties of the Meals from the Defatted Sesame Seeds at Various Roasting Temperature and Time)

  • 참깨를 여러 가지 조건에서 유은 다음 참기름을 채취하고 남은 참깨박의 이화학적인 특성의 변화를 펄사하였다. $210^{\circ}C$에서 10분간 처리한 경우 참깨약과 참기름의 최고수율은 각각 50.1%및 46.9%이었고 참깨박에 잔류되는 시름은 8.4%로 가장 낮았다. 참깨박중의 단백질 함량은 볶음온도가 높아지고 볶음시간이 길어짐에 따라 점차 감소하였다. 참깨박의 아미노산함량은 볶음조건에 따라 점차 감소하였으며 가장 크게 감소한 아미노산은 serine, cystine, Iysine이었다. 유리당을 분석 한 결과 sucrose, glucose, fructose가 각각 162.6mg%, 37.7mg%, 18.7mg% 함유되었고 유음온도가 높아지고 볶음시간이 길어짐에 따라 그 함량은 현저히 감소하였다. 수용성 갈색색소는 볶음온도가 $190^{\circ}C$인 경우는 3개의 분획(분획 I, II, III)으로 뚜렷하게 나누어졌으나 볶음온도가 높아지고 유음시간이 길어짐에 따라 분획 II는 점차 불분명해졌다.

탈지들깨박 Ethanol 추출물의 항산화 효과 (Studies on Antioxidant Activity of Ethanol Extracts from Defatted Perilla Flour)

  • The antioxidant activity of ethanol extracts from defatted perilla flour was investigated by measuring peroxide value of perilla oil during storage at $45^{\circ}C$. The antioxidant activity of ethanol extracts was also compared with BHA, BHT and tocopherol. Anti-oxidant activity of ethanol extracts was also examined in corn oil and lard. The ethanol extracts contents of defatted perilla flour and the original perilla seed were 7.69 and 4.56% respectively. The antioxidant activity of ethanol extracts was superior to that of 0.02% BHT, BHA and tocopherol in the perilla oil substrate, merely in concentration of one-twentieth as much as that contained in original perilla oil seeds. The fractions of non-polar solvent (hexane and chloroform) obtained from silicic acid column chromatography are less effective than that of polar solvent as an antioxidant. Antioxidant activity of partially purified ethanol fraction is slightly inferior to that of original crude ethanol extracts. Ethanol extracts were also effective in corn oil and lard almost same as in perilla oil. The total phenolic compound contents of crude ethanol extracts and partially purified ethanol fraction were 9.3, 6.4%, respectively.
