First Report of Leaf Spot of Datura metel Caused by Alternaria tenuissima in Korea

  • In June 2013, we collected leaf spot disease samples of Datura metel from Gangneung, Gangwon Province, Korea. The symptoms observed were small circular to oval dark brown spots with irregular in shape or remained circular with concentric rings. We isolated the pathogen from infected leaves and cultured the fungus on potato dextrose agar. We examined the fungus morphologically and confirmed its pathogenicity according to Koch's postulates. The results of morphological examinations, pathogenicity tests, and the rDNA sequences of the internal transcribed spacer regions (ITS1 and ITS4), glycerol-3-phosphate dehydrogenase (G3PDH) and the RNA polymerase II second largest subunit (RPB2) gene sequence revealed that the causal agent was Alternaria tenuissima. To the best of our knowledge, this is the first report of leaf spot of D. metel caused by A. tenuissima in Korea as well as worldwide.

Control of Late Leaf Spot of Groundnut (Arachis hypogaea) by Extracts from Non-Host Plant Species

  • The effects of leaf extracts of 14 different non-host plant species on in vitro conidial germination of Phaeoisariopsis personata, the causal organism of late leaf spot(LLS) of groundnut were evaluated. Aqueous and ethanol leaf extracts of Datura metel, Lawsonia inermis and aqueous leaf extracts of Sphaeranthus indicus at 25%(w/v) concentration completely inhibited the conidial germination of P. personata both at 24h and 48h after incubation. Aqueous leaf extracts of Blumea bifoliata, Eucalyptus globules, Ocimum sanctum and Pongamia pinnata, and ethanol leaf extracts of Azadirachta indica and S. indicus inhibited the conidial germination by >90%. Aqueous and ethanol leaf extract of L. inermis and S. indicus were highly inhibitory to conidial germination up to 1% concentration. Aqueous and ethanol leaf extracts of D. metel and ethanol leaf extract of A. indica were highly inhibitory to P. personata even at 0.01% (100 ppm) concentration. Ethanol leaf extract of A. indica up to $80^{\circ}$, aqueous leaf extracts of D. metel and S. indicus up to $100^{\circ}$, and L. inermis up to $60^{\circ}$, were highly stable and retained their fungitoxic effects. Extract of D. metel was antifungal even after 180 days when it was stored both at room temperature and $4^{\circ}$. Aqueous leaf extract of D. metel at 2% concentration effectively reduced the development of LLS by >60%, under greenhouse conditions both in prophylactic and simultaneous applications. Extracts of D. metel could be a potential economical and an eco-frendly alternative for control of late leaf spot, and its efficacy under field conditions is further being evaluated.

Antifungal Activity of Withametelin, a Withanolide Isolated from Datura metel

  • Withametelin, a steroidal compound isolated from leaves of Datura metel L.(Solanaceae), showed antifungal activity against some plant pathogenic as well as saprophytic fungi tested in vitro. Except Curvularia maculans and Colletotrichum sp., spore germination of all the other 23 fungi was inhibited significantly at 125 to 1000 ppm. Out of the 25 fungi tested, Curvularia sp., Cercospora abelmoschi, Heterosporium sp., Erysiphe cichoracearum and Ustilago cynodontis were most sensitive as there was complete inhibition of germination at 1000 ppm. Similarly, spores of Alternaria brassicae, Curvularia lunata and Helminthosporium pennisetti showed less than 3% germination at the same concentration. Rest of the fungi showed more than 15% spore germination at the same concentration.

흰독말풀(Datura metel L.)종양 캘러스의 현탁배양으로부터 Scopolamine 생성 (Scopolamine Production in Suspension Cultures of Tumor Calli from Datura metel L.)

  • 흰독말풀의 tumor조직에서 유도한 tumor callus로부터 scopolamine의 생산을 극대화하기 위한 연구를 수행하였다. Tumor callus의 생장률을 증가시키기 위해서는 초기접종량을 배지의 2%로 하고 배양기간은 4주가 효과적이다. 그래서 만일 이단계배양을 추진하려면 배지교체의 시기는 3주가 가장 적당한 것으로 확인되었다. Scopolamine의 생성을 위한 광의 효과를 알아본 결과, 암상태보다 광상태가 callus생장 및 scopolamine 함량 증가에 효과적이었다. Scopolamine 함량 증가의 최적 광량은 16$\mu$mo1 m$^{-2}$ s$^{-1}$, 광질은 적색파장 그리고 연속광보다 장일조건 (16/8)의 광주기에서 tropane alkaoid인 scopolamine합성이 현저히 촉진되었다. 질소원으로는 산화형 질소인 NO$_{3}$$^{-}$ 를 4 g/L로 증가시키는 것이 가장 효과적인 것으로 확인되었다. Scopolamine의 함량을 증가시키기 위한 elicitor 공급은 10 mg/L chitosan과 15 mg/L yeast추출물이 가장 효과적이었으며, 전구물질로는 0.2 mM tropine과 0.3 mM tropic acid가 가장 우수한 것으로 확인되었다. 이러한 결과를 종합해 보면, 흰독말풀 tumor callus에서 scopolamine 대량생산을 추진할 경우 3주 동안 광상태와 무기이온 조성을 균형 있게 조절하여 생장을 최대로 증가시킨 후, 1주 동안 scopolamine생산을 위해 elicitor, precursors 등이 조합된 배지에서 biotransformation 시키는 것이 가장 효과적일 것으로 판단된다. 또한 tumor callus에 $^{60}$Co${\gamma}$-ray을 조사하여 3K, 4K및 6k tumorcallus를 확보하였으며, 이들로부터 유도한 teratoma는 형태적 변이를 나타내었다. Teratoma로부터 또 다시 새로운 teratoma-callus 세포주를 유도하여 현탁배양을 수행하였던 바, MS 기본 배지에서 1.0 mg/L 2.4-D과 0.5 mg/L kinetin을 조합하여 처리한 callus 의 액체배양에서 cell aggregation이 일어나지 않고 세포가 유리화 되면서 왕성한 생장을 보였다.

Effects of Plant Extracts on Conidial Germination, Mycelial Growth and Sporulation of Fungi Isolated From Poultry Feed

  • Effect of ethanolic extracts of Lawsonia inermis, Azadirachta indica, Vinca rosea, Tagetes patula, Ocimum sanctum, Colocasia antiquorum, Adhatoda vasica, Moringa oleifera, Datura metel and Curcuma longa leaf on conidial germination, mycelial growth and sporulation of Aspergillus flavus, A. niger and A. fumigatus were examined. The conidial germination of A. flavus and A. fumigatus were most inhibited by the extract of L. inermis, while that of A. niger was inhibited by A. indica. Other tested plant extracts have a good effect on conidial germination on the selected fungi. The highest mycelial growth of A. flavus(37 mm) was found in V. rosea, but in case of A. niger and A. fumigatus it(38 and 39 mm) was found in D. metel. The lowest(4, 9 and 6 mm) respectively mycelial growth of these fungi found in L. inermis. The highest sporulation($75{\times}10^4/ml$) of A. flavus was counted in V. rosea, but in case of A. niger and A. fumigatus those($45{\times}10^4$ and $55{\times}10^4/ml$) were in D. metel and the lowest($5{\times}10^4,\;12{\times}10^4\;and\;9{\times}10^4/ml$) respectively sporulation of these fungi counted in L. inermis plant extract medium.

Response of Mulberry Brown Leaf Spot Fungus Myrothecium roridum to Different Plant Extracts

  • Anti-fungal potential of 5 plant extracts viz., Eucalyptus citriodora, Allium sativum, Cassia sophera, Chromolaena odorata and Datura metel on the growth of mulberry brown leaf spot pathogen Myrothecium roridum were examined. Except fur the aqueous extract of Allium bulb, ethanolic leaf extract of all other plants more efficiently reduced the colony growth of the fungus on potato-dextrose-agar, Of which, Allium and Eucalyptus extracts were more effective. Initiation of radial growth of M. roridum on solid media was deferred maximum 6 days by ethanolic Eucalyptus extract and 4 days by aqueous Allium extract at $0.4 mg.ml^{-1}$. In the liquid media amended with Eucalyptus extract ($0.4 mg.ml^{-1}$) complete inhibition of sporulation was noticed upto 8 days, and initial inhibition of mycelial bio-mass generation was considerably diminished with time and reduction was 1.3 fold 14 days after application. While, complete inhibition of mycelial growth for 6-14 days was recorded with $\geq$0.1 mg.ml$^{-1}$ commercial eucalyptus oil. However, rejuvenation of growth appeared when fungus was re-inoculated in fresh media. Post-inoculate application of different doses Of Eucalyptus and Allium extracts significantly (p < 0.05) reduced the disease severity in pot-ted mulberry. However, persistence of the effect up to 28 days was apparent at $\geq$ 1.0 mg.ml$^{-1}$ and effectively was on par with carbendazim (1 mg.ml$^{-1}$ ). Almost equal control ability of 1.0 mg.ml$^{-1}$ Eucalyptus extracts can be achieved by ca. 10 times lowered dose of commercial eucalyptus oil. It seems, the toxic principle of E. citrodora to M. roridum is fungistatic in nature and may have essential oil based origin.

Molecular Cloning and Characterization of a New cDNA Encoding Hyoscyamine 6β-hydroxylase from Roots of Anisodus acutangulus

  • A new full-length cDNA encoding hyoscyamine $6\beta$-hydroxylase (designated as aah6h, GenBank Accession No. EF187826), which catalyzes the last committed step in the scopolamine biosynthetic pathway, was isolated from young roots of Anisodus acutangulus by rapid amplification of cDNA ends (RACE) for the first time. The full-length cDNA of aah6h was 1380 bp and contained a 1035 bp open reading frame (ORF) encoding a deduced protein of 344 amino acid residues. The deduced protein had an isoelectric point (pI) of 5.09 and a calculated molecular mass of about 38.7 kDa. Sequence analyses showed that AaH6H had high homology with other H6Hs isolated from some scopolamine-producing plants such as Hyoscyamus niger, Datura metel and Atropa belladonna etc. Bioinformatics analyses results indicated AaH6H belongs to 2-oxoglutarate-dependent dioxygenase superfamily. Phylogenetic tree analysis showed that AaH6H had closest relationship with H6H from A. tanguticus. Southern hybridization analysis of the genomic DNA revealed that aah6h belonged to a multi-copy gene family. Tissue expression pattern analysis firstly founded that aah6h expressed in all the tested tissues including roots, stems and leaves and indicated that aah6h was a constitutive-expression gene, which was the first reported tissue-independent h6h gene compared to other known h6h genes.
