In this study we investigated fecundity and survivorship of four staple Korean freshwater cladocerans for their whole life to develop a new standard method used for ecological risk assessment of pesticide. The tested Korean freshwater cladocerans were Daphnia obtusa, Daphnia sp., Moina macrocopa and Simocephalus vetulus. In addition, standard test species, Daphnia magna endorsed formally by the major international organizations was tested together in order to compare with the reproductive characteristics of Korean cladocerans. A total of 358 young was produced by D. magna, whereas, 297 young was reduced by Daphnia sp. throughout the entire life. The average life span of Daphnia magna was 50 days much longer than any other Korean species. Nevertheless all of the Korean water flea tested showed good fecundity, produced over 60 young for 21 days as the validation criterion for reproduction toxicity test in Ecological Effects Test Guidelines published by USEPA. Especially Moina macrocopa produced their first brood in 4 days. Therefore 10 day reproduction test with Moina macrocopa may be an alternative good method to save cost and time to elucidating the effects of hazardous substances on the reproduction of aquatic invertebrates.
The toxicity values heavy metals were evaluated by immobilization and chronic reproduction impairment tests, using Daphnia magna. Acute tests were evaluated by the inhibition of their mobilization after 24hrs without food addition. The tests of reproductive impairment were investigated for 21 days by food addition and exchange or water. The effect of each concentration was assessed by Probit analysis and t-test. The results obtained from this study were as follows : 1) The change of pH and DO was not significant in the acute tests, while, in the reproductive tests, pH was increased by 0.3~1.4 and DO also increased. 2) The $E_iC_{50}$ values of immobilization to Daphnia magna in artificial fresh water were $0.030mg/{\ell}(Cu),\;0.054mg/{\ell}(Cd),\;0.12mg/{\ell};(Cr),\;0.74mg/{\ell}(Pb),\;3.4mg/ {\ell}(As)$ and the $NOE_iC$ values were $0.010mg/{\ell}(Cu),\;0.018mg/{\ell}(Cd),\;0.010mg/{\ell}(Cr),\;0.10mg/{\ell}(Pb),\;and\;$1.8mg/{\ell}(As)$. 3) The $E_rC_{50}$ values of reproductive impairment to Daphnia magna were $13.8\mu\textrm{g}/{\ell}(Cu),\;2.9\mu\textrm{g}/{\ell}(Cd),\;15.5\mu\textrm{g}/{\ell}(Cr),\;61.7\mu\textrm{g}/{\ell}(Pb),\;759\mu\textrm{g}/{\ell}(As)$, and $NOE_rC$ values were $0.95\mu\textrm{g}/{\ell}(Cu),\;$0.54\mu\textrm{g}/{\ell}(Cd),\;1.2\mu\textrm{g}/{\ell}(Cd),\;$7.4\mu\textrm{g}/{\ell}(Pb),\;110mu\textrm{g}/{\ell}(As)$. The results of tests using OECD artificial culture water were more sensitive than natural water for culturing. The presented data show that an artificial culture water is suitable in the experiment of bioassay for assessing the toxicity of marterials.

A drought-tolerant transgenic rice (Agb0103) was developed using a pepper methionine sulfoxide reductase (CaMsrB2) under the control of rice Rab21 promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. Commercialization of genetically modified (GM) crops will require the evaluation of risks associated with the release of GM crops. With the potential problems associated to GM crops safety testing, the investigation of their effects on non-target organisms is necessary for environmental risk research. This study was carried out to assess acute toxicity of a GM crop using the water flea (Daphnia magna) for non-target organism risk evaluation. The effect of acute toxicity on Daphnia magna of Agb0103 rice and a non-GM rice, Ilmibyeo, were investigated at different concentrations (0, 625, 1,250, 2,500, 5,000, and 10,000 mg/L). The Agb0103 rice used for the test was confirmed to express the CaMsrB2/PAT gene by the PCR and ELISA. Daphnia magna feeding tests showed no significant differences in cumulative immobility or abnormal response with either Agb0103 rice or non-GM rice. The 48hr-EC50 values showed no difference between Agb0103 rice (2243 mg/L) and non-GM rice (2694 mg/L). These results suggest that there is no significant difference in toxicity to Daphnia magna between Agb0103 rice and its non-GM counterpart.
The genetic toxicity of environmental pollutants, namely, nonylphenol (NP), bisphenol A (BPA) and chloropyriphos (CP) was investigated in aquatic sentinel species, freshwater crustacean, Daphnia magna, and larva of aquatic midge, Chironomus tentans, using Comet assay. Physiological effect of such pollutants was also investigated by studying the specimens' rates of reproduction, growth and survival. Acute toxicity results showed that, as expected, Daphnia was more sensitive than Chironomus to chemical exposure. The order of acute toxicity was CP > NP > BPA in D. magna and NP > CP > BPA in C. tentans. BPA may exert a genotoxic effect on D. magna and C. tentans, given that DNA strand breaks increased in both species exposed to this compound, whereas NP- and CP-induced DNA damage occurred only in C. tentans. In vivo genotoxic data obtained in aquatic sentinel species could provide valuable information for freshwater quality monitoring. The experiments with NP-exposed D. magna showed that the pollutant has long-term effects on reproduction, whereas no short-term effect on DNA integrity was found, being an example of a false-negative result from the biomarkers perspective. This result could be interpreted that other mechanism than genetic alteration might be involved in NP-induced reproduction failure in D. magna. False-positive results from the genotoxic biomarker obtained in BPA-exposed D. magna and in NP-exposed C. tentans make it difficult to use DNA integrity as an early warning biomarker. However, as the mere presence of genotoxic compounds, which are potentially carcinogenic, is of high concern to human and ecosystem health, it could also be important to rapidly and effectively detect genotoxic compounds in the aquatic system in ways that do not necessarily accompany a higher level of alteration. Considering the potential of D. magna and C. tentans as bioindicator species, and the importance of genotoxic biomarkers in ecotoxicity monitoring, DNA damage in these species could provide useful information for environmental risk assessment.
To elucidate the mechanisms by which algal food quality affect Daphnia growths, we measured carbon incorporation rates and respiration rates of Daphnia magna with Cryptomonad Rhodomonas minuta, green algae Scenedesmus acutus and cyanobacteria Synechococcus sp. with varying physiological states as food. Carbon assimilation rates were high with R. minuta and S. acutus and low with Synechococcus sp. showing a similar pattern to the growth rate pattern. There was no clear difference among respiration rates of three algal species. Carbon assimilation rates and respiration rates of D. magna appeared to be independent on Molar C:P ratios in algal foods. Carbon growth efficiencies (incorporated carbon per assimilated carbon amount) were lower when D. magna fed with Synechococcus sp. than fed with R. minuta or S. acutus. Analysis of variance results show that carbon assimilation rates which were sum of incorporation and respiration rates and carbon growth efficiencies were only dependant on species affiliation. Overall, our results showed that algal species with varying ${\omega}3$ polyunsaturated fatty acid content led different carbon incorporation rates and overall carbon assimilation rates of D. magna.
To investigate the impacts of transgenic rice on freshwater organisms, we conducted two life table experiments using Daphnia magna for fifteen and twenty days, respectively. We examined life history traits such as population growth rates (r), reproductive rates ($R_0$), generation times, and survivorship. In the first experiment, we used non-drought-stressed transgenic and non-transgenic rice harvested in 2005. In the second study, we used non-transgenic and transgenic rice harvested in 2006 following drought stress. Each experiment involved three treatments in which D. magna neonates were fed with Selenastrum capricornutum (control treatment) and S. capricornutum with 5% aqueous extracts of non-transgenic rice (N-T) and transgenic rice (T). In the first experiment, D. magna showed reduced population growth rates and lowered fecundity in the N-T and T treatments. In the second experiment, D. magna receiving both transgenic and non-transgenic rice extracts showed very high mortality, low population growth rates and reproduction rates. We could not detect any significant negative effects of extracts from transgenic rice on D. magna life history traits at 95%.

In this study, the mixed toxicity of heavy metals (Cu, Pb, Hg) and surfactants (SLS, ALS) was evaluated by using Vibrio fischeri and Daphnia magna. The sensitivity of ecotoxicity to heavy metals was sensitive to daphnia but the case of surfactants were more sensitive to Vibrio fischeri. Experimental results of Vibrio fischeri show that the toxicity value P(O) was lower than the predicted value P(E), and the antagonistic effect was observed when the heavy metal and the surfactant were mixed. It seems that SLS and ALS, which are anionic surfactants, have anionic form on the hydrophilic head, so that they have an antagonistic effect that they are bonded with heavy metal ion which is a cation type and the actual toxicity is lowered. In Daphnia magna, the results showed that antagonistic, additive and synergistic effects were in order as concentrations increased. As the concentration increases, Daphnia magna, which is highly sensitive to heavy metals, seems to have a synergistic effect with a rapid increase in mortality.

Theoredoxin (TRX) transgenic soybeans were developed using the human Theoredoxin gene under the control of the -conglycinin promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. This study was done to assess the acute toxicity of a genetically modified (GM) soybean using the fresh water planktonic crustacean Daphnia magna. The acute toxicity effect of the TRX soybean and non-GM soybean (Gwangan) on D. magna was investigated at different concentrations (0, 156, 313, 625, 1,250, 2,500, and 5,000 mg·L-1). The TRX soybean used for the test was confirmed to express the TRX/PAT genes by PCR and enzyme-linked immunosorbent assay (ELISA). D. magna feeding tests showed no significant differences in the cumulative immobility or an abnormal response with either the TRX soybean or non-GM soybean. The feeding study showed a similar abnormal response and cumulative immobility of the D. magna between the TRX soybean and Gwangan treatments. Additionally, the 48 h-EC50 values for the TRX and Gwangan soybeans were 755.6 and 778 mg·L-1, respectively. The soybean NOEC (no observed effect concentration) value for D. magna was suggested to be 156 mg·L-1. These results suggest that there is no significant difference in toxicity to Daphnia magna between the TRX soybean and its non-GM counterpart.
Insect-resistant transgenic (Bt-9) rice was generated by inserting mCry1Ac1, a modified gene from the soil bacterium Bacillus thuringiensis, into the genome of a conventional variety of rice (Ilmi). With regard to potential problems such as safety, an evaluation of non-target organisms is necessary as an essential element of an environmental risk assessment of genetically modified (GM) crops. We studied the effects of the Bt-9 rice on the survival of cantor Daphnia magna, a commonly used model organism in ecotoxicological studies. D. magna fed on the Bt-transgenic rice (Bt-9) and its near non-GM counterparts (Ilmi) grown in the same environment (a 100% ground rice suspension). The Bt-9 rice was confirmed to have the inserted T-DNA and protein expression evident by the PCR and ELISA analyses. The feeding study showed a similar cumulative immobility and abnormal response of the Daphnia magna between the Bt-9 rice and Ilmi. Additionally, the 48 h-EC50 values of the Bt-9 and Ilmi rice were 4,400 mg/L (95% confidence limits: 3861.01 - 5015.01 mg/L) and 5,564 mg/L (95% confidence limits: 4780.03 - 6476.93 mg/L), respectively. The rice NOEC (No observed effect concentration) value for D. magna was suggested to be 1,620 mg/L. We conclude that the tested Bt-9 and Ilmi have a similar cumulative immobility for D. magna, a widely used model organism, and the growth of Bt-9 did not affect non-target insects.
We explored collective behaviors of indicator species to elucidate the effect of the chemical stress. After the treatments of an insecticide, diazinon, at low concentrations (1.0 and 10.0 ${\mu}g/L$), spatial dispersion patterns of Daphnia magna were checked in a test chamber. The I-index was used to characterize the movement data before (0~1 h) and after (1~2 h) the treatments in laboratory conditions. The slopes of the frequency distribution of I-index in semi-log scale decreased significantly, and the test populations appeared to be more dispersed with a lower degree of aggregation after the treatments. The index was feasible in indicating decrease in the ability of the specimens to keep desirable distances with neighbor individuals under chemical stress and showed a possibility of monitoring presence of toxic chemicals in environment through group behavior measurement.
