Lipid Peroxidation Product-Mediated DNA Damage and Mutagenicity

  • Membrane lipid peroxidation processes yield products that may react with DNA to cause mutations. Lipid hydroperoxides from linoleic acid in the presence of transition metal ions caused strand breaks in plasmid DNA. DNA damage induced by reactive aldehydes known to be produced by decomposition of lipid hydroperoxides, such as 4-hydroxynonenal or rnalondialdehyde, was repaired by endonucleases and exonuclease III which resulted in the increase of single strand breaks in DNA. Lipid hydroperoxides as well as malondialdehyde and 4-hydroxynonenal also caused mutations in the pUC18 lacZ' gene when measured as a loss of ${\alpha}-cornplementation$. In conclusion. the lipid peroxidation could be an important intermediary event in DNA damage and mutation by oxidative stress.

Protective effects of carnosine and homocarnosine on ferritin and hydrogen peroxide-mediated DNA damage

  • Previous studies have shown that one of the primary causes of increased iron content in the brain may be the release of excess iron from intracellular iron storage molecules such as ferritin. Free iron generates ROS that cause oxidative cell damage. Carnosine and related compounds such as endogenous histidine dipetides have antioxidant activities. We have investigated the protective effects of carnosine and homocarnosine against oxidative damage of DNA induced by reaction of ferritin with $H_2O_2$. The results show that carnosine and homocarnosine prevented ferritin/$H_2O_2$-mediated DNA strand breakage. These compounds effectively inhibited ferritin/$H_2O_2$-mediated hydroxyl radical generation and decreased the mutagenicity of DNA induced by the ferritin/$H_2O_2$ reaction. Our results suggest that carnosine and related compounds might have antioxidant effects on DNA under pathophysiological conditions leading to degenerative damage such as neurodegenerative disorders.

Glycation of Copper, Zinc-Superoxide Dismutase and its Effect on the Thiol-Metal Catalyzed Oxidation Mediated DNA Damage

  • The nonenzymatic glycation of copper, zinc-superoxide dismutase (Cu,Zn-SOD) led to inactivation and fragmentation of the enzyme. The glycated Cu,zn-SOD was isolated by boronate affinity chromatography. The formation of 8-hydroxy-2'-deoxyguanosine (8-OH-dG) in calf thymus DNA and the generation of strand breaks in pBhiescript plasmid DNA by a metal-catalyzed oxidation (MCO) system composed of $Fe^{3+}$, $O_2$, and glutathione (GSH) as an electron donor was enhanced more effectively by the glycated CU,Zn-SOD than by the nonglycated enzyme. The capacity of glycated Cu,Zn-SOD to enhance damage to DNA was inhibited by diethylenetriaminepentaacetic acid (DETAPAC), azide, mannitol, and catalase. These results indicated that incubation of glycated CU,Zn-SOD with GSH-MCO may result in a release of $Cu^{2+}$ from the enzyme. The released $Cu^{2+}$ then likely participated in a Fenton-type reaction to produce hydroxyl radicals, which may cause the enhancement of DNA damage.

  • Recently, to consider economical and constructive aspect, Usage of Admixture, like Blast-Furnace Slag and Fly-Ash, are increased. Blast-Furnace Slag, a by-product of steel industry, have many advantage, to reduce the heat of hydration, increase in ultimate strength and etc. But it also reduces early-age strength, it is prevented from using of Blast-Furnace Slag at cold-weather-concrete. In this study, for the purpose of increasing usage of Blast-Furnace Slag at cold-weather-concrete, it is investigated the strength properties of concrete subjected to time and period of frost damage for early age curing. According to this study, if early age curing is carried out before having frost damage, the strength of concrete, subjected to frost damage, is recovered. And that properties is not connected with the frost cause.

  • Radiation exposure may cause tissue damage by ionized or excited atoms. The molecules are transformed from water or they directly interact with cells. Antioxidants such as vitamin E, vitamin C are used to reduce the interference of radiation. Especially, vitamin E inhibits the formation and accumulation of lipid and oxidation and the activity of drug-metabolizing enzymes. After irradiation of 3 Gy to the testis, the entire shape of testis was partly depressed and an irregular vacuole was observed to the edge. The 3 Gy group after treatment with vitamin E showed the low destruction of spermatogonia and partial tissue damage as compared to the 3 Gy group. Vitamin E inhibited the production of vacuoles from the cell death and generated the block to prevent the condensation of the seminiferous tubules.

  • The purpose of this study is to present basic data in order to improve the efficiency and the objectivity of diagnosing educational structures built with the reinforced concrete. For achieving the purpose, this paper, firstly, researches damage aspects of 22 public educational of facilities in Seoul, and summarizes the results of that research. Through the analysis and the evaluation of damage aspects, this paper shows the conclusions as follows; (1) Main damage to reduce structural capacities of building is the differential settlement. (2) Though the steel corrosion is occurred by several factors, the main cause is the faulty construction. (3) To prevent the damage development, a proper repair strategy is very important.

  • In order to decide critical vibration level on structural damages induced by near blasting, deformation of existing cracks in structure was investigated and ground vibrations were measured. New criteria to evaluate blasting damage to structure are proposed considering daily deformation of cracks in structures. Results from 2 brick houses in fishing village show that new design criteria far more than current are required to cause structural damage for brick houses.

  • A debris flow is known as that flood and landslide of water cause much physical human damages worldwide to complex natural disaster that happen combining and happy event is happening mainly in urgent mountains area in domestic. Because happen about debris flow that happen from each place every year and is drift, mechanism of accumulation definitely make clear and great many damage is not running out. Must grasp actual conditions of priority debris flow to need debris flow prevention countermeasure and lay countermeasure to take away damage by debris flow. Because collecting actual conditions of debris flow that happen by objective investigation methods and accuracy, proposed about investigation calamity investigation method so that can calculate debris flow damage and prepare in subsequentness damage.

XPS STUDY ON DNA DAMAGE BY LOW-ENERGY ELECTRON IRRADIATION

  • After the first report that electrons with sub-ionization energy of DNA could cause single strand breaks or double strand breaks to DNA, there have been various studies to investigate the mechanisms of DNA damage by low-energy electrons. In this paper, we examined the possibility of using X-ray photoelectron spectroscopy (XPS) to analyze the dissociation patterns of the molecular bonds by electron irradiation on DNA thin films and tried to establish the method as a general tool for studying the radiation damage of biomolecules by low energ yelectrons. For the experiment, pBR322 plasmid DNA solution was formed into the films on tantalum plates by lyophilization and was irradiated by 5-eV electrons. Un-irradiated and irradiated DNA films were compared and analyzed using the XPS technique.

Contact Damage and Fracture of Poreclain/Glass-Infiltrated Alumina Layer Structure for Dental Application (치아 응용을 위한 /유리침윤 알루미나 이중 층상구조의 접촉손상 및 파괴)

  • Hertzian contact tests were used to investigate the evolution of fracturedamage in the coating layer as functions of contact load and coating thickness by studying crack patterns in porcelain on glass-infiltrated alumina bilayer system conceived to simulate the crown structure of a tooth. Cone cracks initiated at the coating top surface without delamination at interface and crack propagation to substrate. Preferentially the cracks made multi-cracks at the coating top surface rather than proceeding to interface. The cracks were highly stabilized with wide ranges between the loads to initiate first cracking and to cause final failure im-plying damage-tolerant capability. Finite element modelling was used to evaluate the stress distribution. Maximum tensile stress were responsible for the cracking at the coating layer and had a profound influence on the crack pattern and fracture damage in the layered structure materials.
