Effects of various Herbal Acupuncture on Adjuvant Arthritis in Rats (다양한 약침제제(藥鍼製劑)가 Rat의 Adjuvant 관절염(關節炎)에 미치는 영향(影響))

  • To investigate effects of vanous herbal-acupuncture, which were Daebangpungtang, Oyaksungisan, Dokhwalgisaengtang, and Binsosan on Adjuvant Arthritis in rats, the edema rate, the number of WBC, the quantity of TNF-${\alpa}$, COX -2, and IL-6, and histological test of the muscular tissue were measured in the arthritis part. After elicilating arthritis of Sprague Dawely(SD) rats by injection of Freund's complets adjuvant for 2 weeks, saline was injected for the control group, Daebangpungtang acua-acupuncture was injected for the Daebangpungtang group, Oyaksungisan acua-acupuncture was injected for the Oyaksungisan group, Dokhwalgisaengtang acua-acupuncture was injected for the Dokhwalgisaengtang group, and Binsosan acua-acupuncture was injected for the Binsosan group during 30days. Selected point was ST35 in all groups. 1. The number of WBC was $8.24{\pm}0.51$($10^3$/ml) in the normal group and $27.35{\pm}3.51$($10^3$/ml) in the control group. the Daebangpungtang group and the Oyaksungisan group decreased each $20.42{\pm}2.75$($10^3$/ml) and $19.78{\pm}4.99$($10^3$/ml) in the Exp.ll group. This fact showed that the group Exp.ll was more effective than the control group effectively. (P<0.05) 2. The volume of the paw were checked. The volume of paw was $0.62{\pm}0.11mm in the control group and $0.45{\pm}0.08$mm in the Daebangpungtang group and $0.47{\pm}0.07$mm in the Oyaksungisan group, the swelling of the paw was restricted significantly in the Daebangpungtang and Oyaksungisan group(P<0.05) 3. The bands of the TNF-${\alpa}$, COX -2 in the muscular tissue of the control group wcre certain and thick than other groups(except normal group), and the band of the IL-6 in the muscular tissue of all group(except normal group) were similar forms. 4. In histological finding, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in thc control and Dokhwalgisaengtang group. But the Oyaksungisan and Daebangpul1gtang group showed small amount of inflammatory cells, the refrained inflammatory state and even recovering state. From these results, it is showed Oyaksugisan and Daebangpungtang acua-acupuncturc refrain inflammatory reaction and muscular tissue necrosis in SD rats paw were induced by Freund's complete adjuvant

Effects of Daebangpungtang on galactosamine induced hepatoxity in rats (대방풍탕(大防風湯)이 galactosamine으로 유도(誘導)한 간중독(肝中毒) 흰쥐에 미치는 영향(影響))

  • In order to study the effects of Daebangpungtang on galactosamine induced hepatoxity in rats, the experimental rats divided five groups (Normal, Control, Sample A, B, C groups). Under the same condition, normal and control groups were administered normal saline for 16 days, control group was injected to abdomen with galactosamine at 8th day (800mg/kg). Sample A group was administered the Daebangpungtang aqueous solution($200m{\ell}/kg$ p.o) for 8 days and injected galactosamine(800mg/kg i.p) for the last day and was administered normal saline for 8 days. Sample B group was treated as same as group A for 8 days, and then was administered the Daebangpungtang aqueous solution($200m{\ell}/kg$ p.o) forfurther 8 days. Sample C group was administered the Daebangpungtang aqueous solution($200m{\ell}/kg$ p.o) for 16 days. The activity of GOT, GPT, ${\gamma}$-GTP, ALP, LDH and total bilirubin in serum were measured at 8th and last day. The results were obtained as follows: 1. The activity of serum GOT of the sample A group decreased effectively at the 8th day and sample B group decreased effectively at 16th day as compared with the control group. 2. The activity of serum GPT of the sample A group decreased effectively at the 8th and 16th day and sample B group decreased effectively at 16th day as compared with the control group. 3. The activity of serum ${\gamma}$-GTP of the sample B group decreased effectively at the 16th day as compared with the control group. 4. The activity of serum ALP of the sample A group increased respectively at the 8th and 16th day and sample B group decreased effectively at 16th day as compared with the control group. 5. The activity of serum LDH of the sample A, B groups decreased effectively at 16th day as compared with the control group. 6. The activity of serum total bilirubin of the sample A, B groups decreased effectively at 16th day as compared with the control group. 7. The activity of GOT, GPT, ${\gamma}$-GTP, ALP, LDH and total bilirubin in serum of the sample C group were analogous with thats of normal group.

Effect of Daebangpungtang herbal acupuncture on the carrageenan-induced arthritis in rats (대방풍탕(大防風湯) 약침(藥鍼)이 Carrageenan으로 유도한 관절염에 미치는 영향(影響))

  • Objectives : DaeBangPungTang(DBPT) is one of the prescriptions used for the treatment of knee arthritis in oriental medicine. The present study aimed to examine the analgesic effect of DBPT on a rat model of carrageenan-induced arthritis, and the relations between DBPT-induced analgesia and endogenous nitric oxide(NO) and inducible NO synthase (iNOS), cyclooxygenase-2 (COX-2), and c-Fos protein expression in the spinal cord. Methods : Carrageenan-induced arthritis rat model was used to test the effect of DBPT as a chronic pain model. After the induction of arthritis, rats subsequently showed a reduced stepping force of the affected limb for at least tile next 4 days. The reduced stepping force of the limb was presumably due to a painful knee, since oral infection of indomethacin produced temporary improvement of weight bearing. DBPT dissolved in normal saline was minted several acupoints. After the treatment, behavioral tests measuring stepping force were periodically conducted during the next 12 hours. Results : DBPT produced significant improvement of stepping force of the hindlimb affected by the arthritis lasting at least 9 hours. The magnitude of this improvement was equivalent to that obtained after an oral injection of 3mg/kg of indomethacin and this improvement of stepping force was interpreted as an analgesic effect. DBPT produced the improvement of stepping force of the affected hindlimb in a dose-dependent manner. Both NO production and iNOS, COX-2 protein expression increased by arthritis were suppressed by DBPT. DBPT on combination with electroacupuncture (EA) produced more powerful and longer lasting improvement of stepping force of the hindlimb affected by the arthritis than either DBPT or EA did. Conclusion : The present study suggest that DBPT produces a potent analgesic effect on the chronic hee arthritis pain model in the rat and that DBPT-induced analgesia modulate endogenous NO through the suppression of iNOS/COX-2 protein expression.
