Xanthium strumarium L. is an annual plant belongs to the family Asteraceae which is is called a 'Cocklebur' that is used for medicinal purposes. Convergent phyto-activity of various extracts of Xanthium strumarium L. (Asteraceae) was examined. We estimated antioxidant activity from ground part and fruit extract of X. strumarium using 1, 1-diphenyl-2-picrylhydrazyl radical (DPPH) and ABTS assay. The extract of X. strumarium was separated each fraction that of ethanol, petroleum ether, and ethyl acetate. It showed potent radical scavenging effect against the DPPH radical and ABTS. The study revealed that X. strumarium could be used as a potential source of natural antioxidant.
The EtOH extracts of red seaweeds (Symphyocladia latiuscula, Chondrus ocellatus and Carpopeltis affinis) and solvent partitioned fractions were investigated for their 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging effects and the total phenolic contents were correlated with ABTS and DPPH radical scavenging activities. The EtOH extracts and their solvent partitioned fractions exhibited strong ABTS and DPPH radical scavenging activities. Among the solvent partitioned fractions obtained from n-Hexane (HX), methylenchloride (MC), ethylacetate (EA), and buthanol (BuOH), the HX fraction from C. affinis showed higher radical scavenging activities than other fractions. Total phenolic contents showed significant correlation ($r^2$ = 0.709) with ABTS radical scavenging activity. The results of this study suggest that the strong radical scavenging activity of HX fraction from C. affinis is a promising natural antioxidant for healthcare products.
To optimize the hydrolysis conditions in the production of antioxidant hydrolysates from tuna cooking juice concentrate (TC) to maximize the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, TC containing 48.91% protein was hydrolyzed with Alcalase 2.4 L, and response surface methodology (RSM) was applied. The optimum hydrolysis conditions included a 2.2% (w/v) Alcalase concentration and 281 min hydrolysis time, resulting in the highest DPPH radical scavenging activity of 66.49% (0.98 µmol Trolox/mg protein). The analysis of variance for RSM showed that hydrolysis time was an important factor that significantly affected the process (p < 0.05). The effects of different drying methods (freeze drying, hot air drying, and vacuum drying) on the DPPH radical scavenging activity and amino acid (AA) profiles of TC hydrolysate (TCH) were evaluated. Vacuum-dried TCH (VD) exhibited an increase in DPPH radical scavenging activity of 81.28% (1.20 µmol Trolox/mg protein). The VD samples were further fractionated by ultrafiltration. The AA profiles and antioxidant activities in terms of the DPPH radical scavenging activity, 2,2'-azino-bis(3-ethylbenzthiazoline)-6-sulfonic acid (ABTS) radical scavenging activity, ferric reducing antioxidant power, and ferrous ion chelating activity were investigated. Glutamic acid, glycine, arginine, and cysteine were the major AAs found in the TCH fractions. The highest DPPH radical scavenging activity was found in the VD-1 fraction (< 5 kDa). The VD-3 fraction (> 10 kDa) exhibited the highest ABTS radical scavenging activity and ferric reducing antioxidant power. The ferrous ion chelating activity was the highest in VD-1 and VD-2 (5 to 10 kDa). In conclusion, this study provided the optimal conditions to obtain high antioxidant activities through TCH production, and these conditions could provide a basis for the future application of TCH as a functional food ingredient.

Lactic Acid Bacteria (LAB) are among the representative probiotics that have been used for a long time in fermented food. Although there are many studies on detecting the radical scavenging activity of LAB, few studies have been conducted on the environmental factors that improve scavenging activity. This study investigated the environmental factors affecting the DPPH radical scavenging and various antioxidant activities of Kimchi-derived Lactobacillus plantarum K-21 with antihypertensive and radical scavenging activities. The optimal conditions for scavenging DPPH radicals were glucose 2%, bactopeptone 0.5%, Tween 80 0.05%, L-cysteine 0.05%, and an initial pH 6.5 at 35℃. Under optimal conditions, the DPPH radical scavenging activity was 94.8±2.2%, which was 1.5 times higher than that of the basic medium. In addition, L. plantarum K-21 had other antioxidant activities; ABTS radical scavenging (93.6±1.5%), hydroxyl radical scavenging (8.5±0.9%), metal chelating (65.9±0.5%), NO scavenging (53.1±19%), SOD-like (25.1±1.5%), and reducing power (11.7±1.4%) activities were detected. Therefore, L. plantarum K-21 may act not only as a starter for lactic acid-fermented foods with improved functionality but also as a drug for various diseases caused by oxidative stress.
A 96-well plate was applied to determine the DPPH free radical scavenging activity using 107 plant-specific enzyme inhibitors and 100 unknown plant-originated extracts. The final optimum volume was $250{\mu}L$ containing $100{\mu}M$ DPPH ethanolic solution at pH 7.8. In this condition, the radical scavenging activities were significantly increased by two known antioxidants consisting of ascorbate and a-tocopherol in a concentration-dependent manner. Among the 107 inhibitors, ampicillin and gallic acid showed 90.2% and 92.6% antioxidant activity at $100{\mu}M$, respectively, and these results were consisted with previous findings. In the tested 100 natural materials at $50{\mu}g/mL$, antioxidant activity of AT-407 resulted in the highest of 90.1%, and 10 extracts including AT-388 and AT-443 showed over 70%. Our results suggest that the use of 96-well plate for determining DPPH free radical scavenging activity would be a suitable method to select antioxidant-like substances of both synthetic compounds and natural products.

Scavenging effects of DPPH radical and hydroxyl radical, inhibition of linoleic acid oxidation, inhibition of NO synthesis and iNOS expression were tested with extracts and chromatographic subfractions of Canavalia lineata obtained at Jeju island. Chloroform extract and its subfractions gave moderate effects on scavenging DPPH radical and hydroxyl radical, They also inhibited linoleic acid oxidation, and NO synthesis. Inhibition of NO synthesis resulted from the repression of iNOS gene expression. Ethyl acetate extract and its subfractions showed excellent effects on scavenging DPPH radical and hydroxyl radical, while they were cytotoxic.

Vaccinium oldhamii Miq. is a Korean native tree, which is deciduous and shrub tree with broad leaf. To develop superior cultivars for edible or medicinal purposes, we evaluated total phenolic content and antioxidant activity. Total phenolic content was measured by folin-denis method and antioxidant activity was measured by DPPH free radical scavenging activity (DPPH method) and reducing power (potassium ferricyanide method). Total phenolic content, DPPH free radical scavenging activity (at 400 ppm) and reducing power (at 700 ppm) were 15.8~19.9 mg/g, 69.1~91.4% and 0.56~0.96, respectively. A linear correlation was shown between DPPH-radical scavenging activity and reducing power (r=0969).
As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.
Enzymes and non-enzymatic antioxidants are involved in defense of oxgen free radical intermediates in all aerobic cells. The non -enzymatic antioxidants and antioxidant enzyme from the extracts of Solanum nigrum L. known to be anticancer medicinal plant were examined in other to utilize the discovery in natural products as cancer chem-opereventive agents. The DPPH(1,1-diphenyl-2-picryl-hydrazyl) free radical scavening activity on plant position of Solanum nigrum L. was the highest in root, with stem, whole plant, seed, leaf and flower, at higher activities respectively. In extraction methods, the DPPH free radical scavenging activity by circulating extraction with 80 % MeOH. The DPPH activity of L6 fraction by LH-20 column chromatography showed about 6.7 times higher than that of ethyl acetate-fraction. These were identified as phenolic compounds such as 2-6-methano-3-benzazocin-11-ol, 2[1H]-phyidinethione and 2-hydroxy -5-methyl-benzaldehyde. Peroxidase(POD) and superoxide dismutase(SOD) activities of stem and root were higher than that of other plant positions and those of plant positions according to growing stage were the highest in 60 days after seeding. The numbers of isozyme pattern of POD and SOD showed 10 hands and 5 bands, respectively, especially, 8 bands of POD and 3 bands of SOC showed a difference according to plant positions.
Apple and pear are popular fruits consumed in Korea and are common fruit in daily diet. In order to compare the antioxidant activity of the apple and pear peels, total polyphenol contents, total flavonoid contents, $ABTS^+$ free radical scavenging activity, and DPPH free radical scavenging activity were measured from hot water, ethanol, and methanol extracts of the two fruit peels. The total polyphenol and flavonoid contents were highest in 95% methanol extracts of the apple peelsand 70% ethanol extract of the pear peels, respectively. Total polyphenol contents of the pear peels were higher than that of apple peels, and total flavonoid contents of the apple peels were higher than that of pear peels. The apple and pear peels had the highest $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity in 95% methanol extracts and 70% ethanol extracts, respectively. $ABTS^{+{\cdot}}$ and DPPH free radical scavenging activity of pear peels was higher than that of apple peels, and the DPPH free radical scavenging activity of apple and pear peels were detected in hot water, 95% methanol, and 70% ethanol extracts, respectively. Ascorbic acid, a synthetic antioxidant used as positive control, had significantly higher scavenging activity than the apple and pear peels. In conclusion, the apple and pear peelshave great potential as natural antioxidants. Therefore, above results should be considered to provide the possibility for the development of high functional antioxidants.
